2.Studies on Plantlet Regeneration of Monogolian Snakegourd (Trichosanthes kirilowii)in Tissue Culture

When petiole segment,blade and cotyledon slice of Trichosanihes kililhai were cultured on MS medium with different growth regulators,callus and adventitious root were induced easily but with difficulty for adventitious bud. Yet adventitious bud can be induced when whole small blade,large blade segment and adventitious root tip were cultivated on MS medium with 0. 5mg/L of 6~13 A and propagated extensively whentransplanted on new media for successive transfer culture. When adventitious root bud was cultivated on MS medium wth 0. 5mg/L of NAA and 0. 5mg/L of 6~ 13A,shoot appeared,which can be grown into plantlet though the survival rate was not very satisfactory when further transplanted. Effect of blade segment size and its slicing technique,on the differentiation of adventitious bud was discussed.

4.Changes of neuronal glutamate transporter in the rat hippocampus during acute cerebral ischemia

AIM: To investigate the changes of neuronal glutamate transporter (EAAC1) at different ischemic times in the rat hippocampus in early stage. METHODS: Brain microinjection of EAAC1 antisense oligodeoxyneucleotide (EAAC1 antisense) was adopted and focal transient ischemia was produced by the filament method of middle cerebral artery occlusion (MCAO). Western blotting and TTC staining analysis were adopted for observing EAAC1 expression and infarction volume in ischemic region .The expression of EAAC1 mRNA and protein at different ischemic times in the rat ischemic hippocampus was assessed by RT-PCR and Western blotting analysis. RESULTS: Compared with EAAC1 sense group, the volume of brain ischemic infarction [(105.67?8.70) mm~3] was reduced after brain microinjection of EAAC1 antisense. Compared with the sham-operated control, EAAC1 mRNA was significantly higher at 6 h and at (24 h) in the hippocampal regions during ischemia, while protein expression was higher at 24 h only. EAAC1 mRNA and protein expression were unchanged at other ischemic times. CONCLUSION: EAAC1 is associated with ischemic injury and its expression is increased in the hippocampal regions after focal cerebral ischemia.
5.Progress in the study of Her2-targeted cancer therapeutic antibodies.

Tumor surface antigen human epidermal growth factor receptor 2 (Her2) is a type I receptor tyrosine kinase, which belongs to human epidermal growth factor receptor family. Her2-overexpression is associated with tumorigenesis and metastasis. Due to significant clinical effects, Her2-targeted cancer therapy especially therapeutic antibody has become the hot spot in the field of cancer treatment. Anti-Her2 antibody drugs include monoclonal antibodies, antibody-drug conjugates, bispecific antibodies and emerging "two in one" antibody. Based on structure and function of Her2, this review focuses on recent advances in active mechanisms and clinical researches of these antibodies.

6.Value of 4-item examination of gastric function in evaluation of disease severity and course of peptic ulcer

Objective To study the value of 4‐item examination of gastric function in the evaluation of disease severity and course of peptic ulcer .Methods The 4‐item examination of gastric function ,including PGⅠ ,PGⅡ ,PGⅠ /PGⅡ ,gastrin 17 and He‐licobacter pylori(Hp) positive rate ,were detected by adopting enzyme linked immunosorbent assay (ELISA) in 175 cases of peptic ulcer(study group) and 60 persons undergoing physical examination(control group) .The Spearman correlation coefficient was used to analyze the relationship between the 4‐item gastric function examination with the severity and course of peptic ulcer .Results The levels of PGⅠ ,PGⅡ ,gastrin 17 and Hp positive rate in the study group were significantly higher than those in the control group(P<0 .05) ,while PGⅠ /PGⅡwas significantly lower than that in the control group(P<0 .05) .The values of PG Ⅰ /PGⅡfrom high to low were in turn the scarring stage group ,healing stage group and active stage group ,other indexes from high to low were in turn active stage group ,healing stage group and scarring stage group .The levels of PGⅠ ,PGⅡand gastrin 17 and Hp posi‐tive rate in the patients with disease course ≥3 years were significantly higher than those in the patients with disease course<3 years(P<0 .05) .The severity and disease course in the patients with peptic ulcer were positively correlated with the levels of PGⅠ ,PGⅡ ,gastrin 17 and Hp positive rate(P<0 .05) and negatively correlated with PG Ⅰ /PGⅡ(P<0 .05) .Conclusion In the pa‐tients with peptic ulcer ,4‐item gastric function examination is closely correlated with the severity and course of illness ,which can serve as the index for evaluate the severity and course of disease ,thus increases the clinical treatment pertinence ,reduces the occur‐rence of antibacterial drugs abuse ,and is worthy of being promoted and applied in clinic .
8.Methylation of SYK Gene Promoter Region in Nasopharyngeal Carcinoma

Objective To detect methylation of spleen tyrosine kinase (SYK)gene promoter region in nasopharyngeal carcino-ma,and to explore relationship between carcinogenesis of nasopharyngeal carcinoma and methylation of SYK gene promoter region.Methods A total of 52 patients with nasopharyngeal carcinoma and 26 patients with chronic rhinitis from Baoan People’s Hospital of Shenzhen Hospital of Peking University were enrolled in this study between February 2012 and August 2014.All cases were diagnosed by pathological examination.Methylation specific-polymerase chain reaction assay was per-formed to detect methylation status of SYK gene promoter region,the rate of methylation was compared for patients with nasopharyngeal carcinoma and patients with chronic rhinitis.Results The methylation of promoter region of SYK gene was detected for 11 biopsy samples among 52 nasopharyngeal carcinoma patients,the methylation frequency was 21.2% in naso-pharyngeal carcinoma biopsy samples,while methylation was not found in 26 chronic rhinitis biopsy samples.Conclusion Low methylation of promoter region of SYK gene was found in nasopharyngeal carcinoma.It suggests that methylation of SYK gene promoter region may contribute to carcinogenesis of nasopharyngeal carcinoma.
9.Theory analysis and clinical application of spirit-regulating and pain-relieving acupuncture method.

The theoretical foundation and scientific connotation of spirit-regulating and pain-relieving acupuncture method as well as its clinical application for pain are discussed. During spirit regulation, attention should be paid on regulating heart and brain, while acupoints should be selected mainly from the Heart Meridian, Pericardium Meridian and Governor Vessel. It has significant efficacy for refractory pain in clinical treatment. Spirit-regulating and pain-relieving acupuncture method is development of acupuncture treating spirit, and it is an important method for pain in clinic. Improvement on sensitization of pain center and brain function is considered as one of the mechanisms in spirit-regulating and pain-relieving acupuncture method.
