1.Trans-umbilical single site double-ports laparoscopy hernia sac high ligation in children (102 cases)

Objective To explore the reliability, advantages and precautions of trans-umbilical single site double-ports laparoscopy hernia sac high ligation in children. Methods Retrospective analysis of the clinical data of 102 patients of trans-umbilical single site double-ports laparoscopy hernia sac high ligation from April 2015 to June 2016, then summarize the treatment process, method and experience. Results All 102 cases of children with inguinal hernia, 84 cases of unilateral, bilateral in 18 cases. Unilateral cases, found in 24 cases of lateral occult hernia. Operation time: one side (16.17 ± 8.23) min, bilateral (23.68 ± 9.72) min. All the patients were discharged after 1 d, and the incision was beautiful. There was no swelling in the scrotum or labia, no hematoma, no adhesive intestinal obstruction, and followed up from 6~20 months without recurrence. Conclusion It is safe, reliable and effective to treat inguinal hernia in children with trans-umbilical single site double-ports laparoscopy hernia sac high ligation. It has advantages such as beautiful incision, small trauma, quick recovery, low requirements on technology and equipment, easy to grasp.
4.Significance of hypoxia induced alveolar epithelial cell apoptosis and hypoxia inducible factor-1alpha expression

Objective To investigate the effects of cobalt chloride(COCl2)induced rat alveolar epithelial cell apoptosis and the significance of related apoptosis protein hypoxia inducible factor-lalpha(HIF-1 a)expression. Methods Alveolar epithelial cells were cultured in vitro and divided into four groups with 6 samples in each.Hypoxia 4 h group:added 500μmol/L COCl2 for 4 hours inducing alveolar epithelial cell hypoxia;hypoxia 24 h group:co-cultured with COCl2 for 24 hours;hypoxia 48 h group:co-cultured with CoCl2 for 48 hours;control group:no CoCl2 added.The apoptosis rates were detected by fluorescent microscope and flow cytometry.The apoptosis morphological changes of the cell were observed by transmission electron microscopy.The protein expression of HIF-la was detected bv Western Blot. Results (1)Fluorescent microscope results showed that apoptosis rate in hypoxia 4 h、24 h、48 h group[(5.83±0.76)%,(15.00±3.28)%and(51.50±3.00)%],were higher than control group[(1.50±0.50)%](P<0.05).(2)Flow cytometry analysis reached the same results as with fluorescent microscope.(3)Typical morphological changes of apoptosis were observed in hypoxia 24 h group.(4)HIF-1α expression in hypoxia 4 h、24 h and 48 h groups[0.69±0.035,1.02±0.044 and 0.71±0.046],were higher than control group(0.21±0.026)(P<0.01).(5)Positive correlations were found between relative amount of HIF-1αprotein and apoptosis rate by fluorescent microscope and flow cytometry analysis(r=0.484,P=0.016;r=0.713,P=0.009).Conclusions Hypoxia could induce apoptosis of alveolar epithelial cells,which may participate in the pathogenesis of hypoxia-induced lung injury.
6.EFFECTS OF ADRENALINE AND DEXAMETHASONE ON PROLIFERATION OF THE LENS EPITHELIAL CELLS

To detect the effects of adrenaline and dexamethasone on proliferation of the lens epithelial cells and discuss the mechanism, the central area 5~7mm in diameter of the anterior capsule was obtained in cataract surgery (phacoemulsification, Phaco), and the specimens were divided into three parts, and were cultured in vitro and treated with 10 -6 mol/L adrenaline and dexamethasone for 48 hours. And then the positive area ratio of proliferating cell nuclear antigen (PCNA) in immunohistochemistry was estimated by medical color image analysis. The data were analyzed with the statistical software. The results indicated that the 10 -6 mol/L adrenaline and dexamethasone had obvious inhibition on LEC proliferation. The positive area ratio of PCNA was (2 614?0 922)%( t =5 594, P =0 0000) and (3 338?0 838)%( t =3 361, P =0 0013). It is suggested that adrenaline and dexamethasone could inhibit the proliferation of LEC and could be used to treat PCO after cataract surgery. The experiments provide the scientific basis for selection of drugs to prevent PCO after cataract.
7.The study of combined treatment for advanced endometrium carcinoma──A report of 45 cases

Purpose:To study the effect of combined treatment on advanced endometrium carcinoma. Methods:45 cases of stage Ⅲ endometrium carcinoma with combined treatment were retrospectively reviewed. Operation was the most used mode in the combined treatment. Depending on the pathology, metastatic site etc. 31 cases were treated in combination with chemotherapy, 11 cases with operation and radiotherapy, 3 cases with radiotherapy. All cases had hormonotherapy. The multi-drug chemotherapy was dominated by platinum agents. CFP 17 cases (54.83%),EFP 5 cases (16.12%), CAP 4 cases (12 90%), CP 3 cases(9.68%), others 2 cases. Survival analysis were done by Kaplan-Meier model of SPSS 8.0. Results:15 cases of Ⅲ stage were treated by operation and chemotherapy, 16 cases by operation chemotherapy and radiotherapy, 11 cases by operation and radiotherapy. The 2-year survival rate was 69.23%,78.97%, 70.71%. There was no difference in statistics. The 5-year survival rate was 30.29%,47.38%,14.14%. 8.Clinical Analysis on Neoadjuvant Chemotherapy of Advanced Breast Cancer

Objective To study the long-term effect of neoadjuvant chemotherapy on advanced breast cancer. Methods The CAF neoadjuvant chemotherapy 〔CTX 500 mg/m2(1st day, 8th day), 5-FU 500 mg/m2(1st day, 8th day), and ADM 30 mg/m2 (1st day) every 3 weeks〕 was carried out in 31 breast cancer patients (stageⅢ,Ⅳ) for 2 cycles before operation, compared with 30 patients (stage Ⅲa) whose therapies were never done and operations could be feasible. Results The overall response rate was 87.1%(27/31). The stages of 19 patients among 31 (61.3%) declined (6 patients to stage Ⅲa, 8 to stageⅡb, 4 to stageⅡa, 1 to stage 0, 1 to complete response and none to pathological complete response). 9.The role of type 3 muscarinic acetylcholine receptor polypeptide in the pathogenesis of Sj(o)gren's syndrome

Objective To immunize BALB/c mice with type 3 musearinic acetylcholine receptor polypeptide (M3RP) and to evaluate the role of M3RP in SS. Methods Four-week-old BALB/e mice were immunized with M3R polypeptide 213-228 (M3RP) on days 0, 14, 35, 56, and were re-immuniged on days 65, 84, 105, and one mouse was killed every 2 to 3 weeks. The mice of the control group were immunized with submaxillary gland homogenate, GST, and PBS. The animals were analyzed for the presence of anti-SSA,anti-SSB, RF, ANA, anti-a-fodrin and anti-M3RP in sera by immunofluorescenee or ELISA. The cytokines of IFN-γ IL-2 and IL-10 were measured with ELISA. Salivary glands were examined by H&E staining and immunohistochemical analysis. Volume of water drinking by each groups were calculated. Results BALB/e mice immunized with M3RP and submaxillary gland homogenate developed an immune response directed against M3RP, α-fodrin and ANA, but no antibodies against SSA, SSB and RF were found. Furthermore, lym-phocytic infiltrates in the salivary glands of immunized animals were observed 50 days after first immunization of M3RP and submaxillary gland homogenate. The serum IFN-α in mice of M3RP, submaxillary gland ho-mogenate, GST and PBS was (62±6), (89±5), (30±5) and (19±6) pg/ml respectively, and IL-2 was (12.6±1.6), (19.8±0.4), (3.9±0.9), and (4.9±1.1) pg/ml respectively (P<0.05). No difference was found in the level of serum IL-10 among the four group. Expression of α-fodrin was found only in submaxillary gland in M3RP and submaxillary gland homogenate groups of mice, but not in PBS and GST controls when studied by immunohistochemical analysis. Conclusion These results suggest that BALB/c mice immunized with M3RP are reminiscent of human SS, and M3RP as an autoantigen participates the development of SS.
