Objective To study whether hyperthermia and ionizing radiation could produce combined effects on the neurobehaviors of neonatal rats.Methods These rats who were pregnant for 8 or 9 days received respectively total body irradiation by 60Co-rays .When the rats were pregnant for 10 days,the temperature of rat anus was kept at (37?0.5) ℃,(41?0.5) ℃,(42?0.5) ℃,respectively in warming boxes and lasted for 2,3,4,5 minutes.The neurobehaviors of neonatal rats were observed.Results In contrast to control group and 37 ℃group ,when pregnant rats were exposed to high temperature.the neonate rats were delayed for the appearance of four physiologic markers,the age of acquisition for four reflexes and the time of learning and memory(P

7.Effects of Saussurea involucrata extract pretreatment on the expression of the Toll-like receptor 4/nuclear factor-κB in focal cerebral ischemia/reperfusion in mice

Objective To investigate the effects of Saussurea involucrata extract pretreatment on the expression of the Toll-like receptor 4 (TLR4)/nuclear factor-κB (NF-κB) in focal cerebral ischemia/reperfusion in mice and its possible neuroprotective mechanism.Methods Seventy-two Kunming mice were randomly divided into four groups:sham operation,saline,Saussurea involucrata extract,and edaravone groups (n =18 in each group).Saussurea involucrata extract 0.8 g/kg was given intraperitoneally in the Saussurea involucrata extract group; edaravone 3 mg/kg was given in the edaravone group; and the same volume of saline was given in the saline group.A model of middle cerebral artery occlusion (MCAO) was induced after 7 days of continuous injection.Cerebral infarct volume was determined by 2,3,5-triphenyltetrazolium staining.Immunohistochemical staining was used to detect TLR4-positive cells in ischemic brain tissue.Reverse transcriptase polymerase chain reaction was used to detect the expression of TLR4/NF-κB mRNA.Results The cerebral infarct volume in mice in the saline,Saussurea involucrata extract and edaravone groups was 131.55± 28.25 mm3,84.10 ±13.92 mm3 and 65.10 ± 6.78 mm3,respectively.There were significant difference (F =10.158,P =0.012).The infarct volume in the Saussurea involucrata extract group (P =0.020) and edaravone group (P0.005) was significantly less than that in the saline group,and there was no significantly difference between the 2 groups.The numbers of cortex and TLR4 positive cells in hippocampus area at the ischemic sides in the saline group were significantly more than those in the sham operation group (all P <0.001).The numbers of positive cells of cortex and TLR4 in the Saussurea involucrata extract group and the edaravone group were significantly decreased compared to the saline group (all P < 0.05),and there was no significant differences between the Saussurea involucrata extract group and the edaravone group.The expressions of TLR4,p50,and p65 mRNA in the saline group were significantly up-regulated compared to the sham operation group (all P =0.000).Saussurea involucrata extract could significantly down-regulate the expressions of TLR4,p50,and p65 mRNA at 24 hours after ischemia/reperfusion (all P =0.000).Edaravone could significantly down-regulate the expressions of TLR4 and p65 mRNA (all P =0.000) and it had a down-regulated trend for the expression of p50 mRNA (P =0.053); while there was no significant difference in the expressions of TLR4 and p65 mRNA between the Saussurea involucrata extract group and the edaravone group.Conclusions Saussurea involucrata extract pretreatment may significantly reduce the cerebral infarct volume,down-regulate the expressions of TLR4 and NF-κB subunit,and play a neuroprotective effect by inhibiting inflammatory response after ischemia.
8.Effect of iterative reconstruction techniques on the volume measurement accuracy of pulmonary ground-glass density nodule on low-dose CT

Objective To evaluate the potential of three commercial CT iterative reconstruction methods (ASIR,MBIR and iDose4) for the image quality and accuracy of volumetric measurement of ground-glass opacity (GGO) nodules on low-dose chest computed tomography (CT) using a phantom study.Methods A phantom containing GGO nodules was examined in volumetric mode on 2 multi-detector CT scanners at low dose.CT images were reconstructed with adaptive statistical iterative reconstruction (30% ASIR),model-based iterative reconstruction (MBIR),iDose4 L3,L6 reconstruction algorithm.Then two radiologists measured CT value of the region of interests (ROI) on reconstruction images,and calculated average noise and contrast to noise ratio (CNR) of reconstruction images.And they applied the lung nodules evaluation software to measure the volume of GGO nodules on the reconstruction images,and calculated the absolute percentage error (APE) of nodule volume measurement.The difference of noise and CNR at different reconstructions was compared with ANOVA test,and Kruskal-Wallis H ranking test was used to the difference of GGO measurements at different reconstructions.Results The average noises of CT image reconstructed with the standard FBP algorithm,30% ASIR algorithm,MBIR algorithm were 40.38 ± 4.49,30.06 ± 20.17,10.55 ± 1.56,and the CNRs were 5.00 ± 2.36,8.43 ± 3.89,20.27 ± 7.35,respectively.Compared to CT image reconstructed with the standard FBP algorithm and 30% ASIR algorithm,CT image noise reconstructed with MBIR algorithm decreased by 73.9% and 64.9%,while the CNR increased by 75.3% and 58.4% with effective dose of 0.36 mSv in HDCT,respectively(F =13.053,9.459,P <0.01).When effective dose was equal to 0.64 mSv in iCT,the image noise reconstructed with iDose L6 algorithms decreased by 36.5% compared with the standard FBP algorithm,and CNR increased by 78.4%.The reconstruction algorithm,NI and tube current were no significant difference in lung nodule volume measurement of absolute percentage error(P > 0.05).Conclusions Three kinds of iterative reconstruction algorithms are able to reduce the image noise in low-dose CT and improve the image quality.

Mian-Zao-Er was collected from the bulbs of Scilla scilloides (Lindl. ) Druce, belonging to the Hyacinthaceae family. 17 compounds were obtained using various column chromatographies on macroporus resin (HPD100), silica gel, Sephadex LH-20 and ODS, as well as semi-preparative HPLC. Their structures were elucidated on the basis of physicochemical properties and spectral data as 2-hydroxy-7-methoxyscillascillin (1), scillascillin (2), 5,7-dihydroxy-3',4'-dimethoxyspiro 2H-1-benzopyran-7'-bicyclo[4.2.0 ] octa [1,3,5 ] -trien } -4-one (3), socialinone (4), 4-methylresveratrol (5), (E)-resveratrol (6), scillavoneA (7), 3,9-di- hydroeucomnalin (8), 3-(3-hydroxy-4-methoxybenzyl) -5,7-dihydroxychroman-4-one (9), (3R)-5,7,3'-trihydroxy-4'-methoxyspiro (2H-1-benzopyran-7'-bicyclo[4, 2, 0] octa [1, 3, 5]-trien} -4-one (10), scillabene A (11), 2-hydroxyscillascillin (12), 3-(4-hydroxybenzyl) -5,7-dihydroxychroman-4-one (13), 3-( 4-hydroxybenzylidene) -5, 7-dihydroxychroman-4-one (14), 3-( 4-hydroxybenzyl) -5-hydroxy-7,8-dimethoxychroman-4-one (15), 3-(4-hydroxybenzyl) -5-hydroxy-6, 7-dimethoxychroman-4-one (16), and 3-(4-hydroxybenzyl)-5,8-hydroxy-7-methoxychroman-4-one (17). Among them, compounds 3, 4, 6, 9, 13 and 15-17 were isolated from this plant for the first time.
