Objective To investigate the characteristics of children′s recurrent abdominal pain(RAP) with gastroscopy and analyze the etiology.Methods Three hundred and thirteen children were investigated with gastroscopy.Gastric antrum mucosa was taken for histo-pathology and the determination of helicobacter pylori(Hp) antibody.Results Ninty-nine point six eight percent of the cases had lesions under the gastroscope.The former 4 cases had chronic superficial gastritis(CSG),CGS and bile reflux,CGS and duodenitis,CGS and bulb ulcer,and Hp infection rate was 31.36%,25%,38.64%,60.61%.Hp infection rates of active gastritis and inactive gastritis were 92% and 23.19%,which showed significant differences in 2 groups(P
2.Effect of Shuanghuanglian injection on cerebral nuclear factor-kappaB expression in mice with viral encephalitis.

OBJECTIVETo observe the effect of Shuanghuanglian injection on cerebral expression of nuclear factor-kappaB (NF-kappaB) in mice with viral encephalitis.
METHODSThe mice with experimental viral encephalitis received treatment with Shuanghuanglian injection at the dose of 0.2, 1.5, and 5 for 5, 10 or 20 consecutive days. The total RNA of the brain tissue was extracted to analyze the protein and mRNA expression of NF-kappaB using Western blotting and RT-PCR, respectively.
RESULTSCompared with the control group, the mice with experimental viral encephalitis showed significantly increased protein and mRNA expressions of NF-kappaB (P<0.01). Treatment with Shuanghuanglian injection at the doses of 0.2 and 1.5 mg/kg significantly lowered NF-kappaB protein and mRNA expressions in the brain of mice with viral encephalitis (P<0.05), and the effect was even more obvious at the dose of 5 mg/kg (P<0.01).
CONCLUSIONShuanghuanglian injection can reduce the expression of NF-kappaB in the brain of mice with viral encephalitis in a dose- and time-dependent manner.

3.Establishment of MDCK cell models expressing human MATE1 or co-expressing with human OCT1 or OCT2.

To establish single- and double-transfected transgenic cells stably expressing hMATE1, hMATE1 cDNA was cloned by RT-PCR from human cryopreserved kidney tissue, and subcloned into pcDNA3.1(+) plasmid by virtue of both HindIII and Kpn I restriction enzyme sites. Subsequently, the recombined pcDNA3.1(+)- hMATE1 plasmid was transfected into MDCK, MDCK-hOCT1 or MDCK-hOCT2 cells using Lipofectamine 2000 Reagent. After a 14-day-cultivation with hygromycin B at the concentration of 400 µg · mL(-1), all clones were screened with DAPI and MPP+ as substrates to identify the best candidate. The mRNA content of hMATE1, the cellular accumulation of metformin with or without cimetidine as inhibitor, or transportation of cimetidine was further valuated. The results showed that all of the three cell models over expressed hMATE1 mRNA. The cellular accumulation of metformin in MDCK-hMATE1 was 17.6 folds of the control cell, which was significantly inhibited by 100 µmol · L(-1) cimetidine. The transcellular transport parameter net efflux ratios of cimetidine across MDCK-hOCT1/hMATE1 and MDCK-hOCT2/hMATE1 monolayer were 17.5 and 3.65, respectively. In conclusion, cell models with good hMATE1 function have been established successfully, which can be applied to study the drug transport or drug-drug interaction involving hMATE1 alone or together with hOCT1/2 in vitro.

4.A cyclotide against influenza A H1N1 virus from Viola yedoensis.

Three cyclotides were isolated from the whole plant of Viola yedoensis in this study. The two, vary peptide E and cycloviolacin Y5, were previously reported, and a novel cycloviolacin VY1 was characterized according to the interpretation of MS/MS fragmentation of peptides which were produced from the reduced and alkylated parent peptide with the digestion of Endo Lys-C, trypsin and chymotrypsin, separately. The stability of remarkable resistance to proteolytic degradation by trypsin and chymotrypsin, and that of thermal denaturation was confirmed again. Besides, the IC50 value of cycloviolacin VY1 against influenza A H1N1 virus was (2.27 +/- 0.20) microg x mL(-1). It is the first cyclotide reported with anti-influenza A H1N1 virus activity in vitro assay.

5.Protective Effect of Shen Xi Capsule on Endotoxemia in Mice Heart

ABSTRACT:OBJECTIVE To explore the protective effects of shenxi on heart in endotoxemic mice.METHODS The experi-mental mice were equally and randomly divided into control group,LPS group,shenxi group,shenxi+LPS group.The endo-toxemia model was established by intraperitoneally injecting LPS.Mice in groups shenxi and shenxi+LPS were intragastrically administered with shenxi solution(0.75 g/kg),while the same dose of normal saline were intragastrically administrated in con-trol group and LPS group.The heart function was detected with echocardiography,and TNF-α,IL-1β,MDA,SOD in myocar-dial tissue was estimated 6 hours after LPS administration.RESULTS Compared with control group,LPS treatment could re-sult in decreased myocardial activities of SOD,and elevated levels of TNF-α,IL-1β,MDA(P <0.05);shenxi treatment could improve the above aberrant indices.Compared with control group,LPS treatment could result in reduced cardiac function, while shenxi treatment could improve it.CONCLUSION Shenxi shows a good cardio-protective effect on the cardiac injury in endotoxemic mice,which may be reached by suppressing inflammatory infiltration and oxidative stress reaction.
6.Invasion of four common periodontal pathogens into vascular endothelial cells in vitro

Objective To investigate the adhesive and invasive ability of four common periodontal pathogens,Pg33277,Pi25611,Aa29522 and Fn10953 in human umbilical vein endothelial cells(HUVEC).Methods The model of infection of HUVEC by periodontal pathogens was established in vitro.The invasive ability of four periodontal pathogens in HUVEC was tested by scanning electron microscope(SEM)and antibiotic protection assays-colony-forming units(CFU).Results All of the four periodontal pathogens were found to adhere to HUVEC by SEM and invaded HUVEC at invasion numbers of(0.8±0.1)×10~8,(4.1±0.5)×10~6,(1.6±0.3)×10~6 and(5.0±0.4)×10~6 CFU/L respectively by antibiotic protection assays-CFU.The invasion efficiencies were(0.400±0.050)%,(0.021±0.003)%,(0.008±0.002)% and(0.025±0.002)%,respectively.The invasive ability of Pg33277 was significantly greater than those of the other three periodontal pathogens(P<0.001).There was no difference in invasive abilities among Pi25611,Aa29522 and Fnl0953(P>0.05).Conclusions All of the four common periodontal pathogens,Pg33277,Pi25611,Aa29522 and Fnl0953 could adhere to and invaded HUVEC,with Pg33277 being the strongest.
7.A study on burn-out of operating room nurse and their job stresses

Objective To characterize the current status of operating room nurse' s burn-out, explore the relationship between the nurse burnout and job stress sources. Methods Applying cross section survey to do the descriptive study, choosing 445 operating room nurses were investigated by questionnaires, including MBI-HSS, and work pressure-source questionnaire (SSRS). Results Among the three burnout inventories,operating room nurses' high fatigue including emotional exhaustion which occupied 43.6%, depersonalization tendency which occupied 31.7%, low level of efficacy which occupied 54.6%. Compared with North American norm (33% for high fatigue), there was just depersonalization tendency which similar to North American norm,while the emotional exhaustion and low level of efficacy were highly above the norm; The top three factors which leading to the job stress were worrying about mistakes and accidents( 3.32 ± 0. 69), large work amount( 3.18 ±0.76), and social position ( 3. 12 ± 0.87). Conclusions The operating room nurse working pressure was generated from job burn-out, further influence nursing work quality and work efficiency.
8.Effect of scene defect teaching model on the training of clinical nurses

Objective To explore the effect of the scene defect teaching on the training of clinical nurses.Methods Totals of 480 nurses were randomly divided into intervention group ( n =235 ) and control group (n=245 ).Traditional training model was adopted in the control group including lessons,demonstration of nursing skills and practice.The scene defect teaching model was adopted in the intervention group.The teaching effect was compared between the two groups before and after intervention.Results There was significantly difference in operating skill and critical thinking between the two groups after 3 months of intervention ( t =7.76,9.14,7.92,10.21,14.33,respectively ; P < 0.05 ).Patients' satisfaction of the intervention group was significantly higher than that of the control group (t =7.04,9.33,5.86,9.06,4.43,4.05,3.85,7.08,respectively;P <0.05).Conclusions The scene defect teaching model can enhance the learning interest,promote the nurses to think and solve the problems in a critical way,and it also improve the degree of satisfaction of patients.
9.Effect of UCP2-siRNA on inflammatory response of cardiomyocytes induced by septic serum.

OBJECTIVETo study the effect of uncoupling protein 2 (UCP2)-siRNA on the inflammatory response of rat cardiomyocytes (H9C2) induced by septic serum and to investigate the possible role of UCP2 in the development of septic cardiomyopathy.
METHODSSerum samples were separately collected from normal rats and septic rats. Cultured rat cardiac cells (H9C2) were randomly divided into blank control, normal serum, 10% septic serum, UCP2-siRNA+10% septic serum and negative siRNA+10% septic serum groups. Stimulation with 10% septic serum was performed for 12 hours in relevant groups. The mRNA expression of tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β) was measured by RT-PCR. The expression of phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK) and nuclear factor-kappa B (NF-κB) was measured by Western blot.
RESULTSThe expression levels of p-p38 and NF-κB in the UCP2-siRNA+10% septic serum group were significantly higher than in the 10% septic serum group (P<0.05). The UCP2-siRNA+10% septic serum group had a significantly higher TNF-α mRNA expression than the 10% septic serum group (P<0.01), but IL-1β mRNA expression showed no significant difference between the two groups.
CONCLUSIONSUCP2 plays a regulatory role in the activation of p38 MAPK and NF-κB and the expression of downstream inflammatory mediators in H9C2 cells stimulated with septic serum.

10.Chemical constituents from the ethanol precipitated sediment of Radix Isatidis and their anti-inflammatory activities

AIM To study the chemical constituents from the ethanol precipitated sediment of Radix Isatidis and their anti-inflammatory activities.METHODS The effects of ethanol precipitated sediment on IL-6 and TNF-α levels were detected by LPS-induced RAW264.7 cell inflammation model and enzyme-linked immunosorbent assay.The chemical constituents were analyzed by UPLC-QTOF-MS/MS and high performance molecular exclusion method.RESULTS When the concentration of Radix Isatidis ethanol sediment were 200 and 400 μg/mL,it could significantly inhibit the release of IL-6 and TNF-α.The ethanol sediment of Radix Isatidis was mainly composed of polysaccharides and proteins,including trace amounts of lipopeptides,indoles and amino acids,among which polysaccharides were mainly glucans.CONCLUSION The constituents from the ethanol sediment of Radix Isatidis are sugars,alkaloids(indoles),amino acids,and organic acids(fatty acids),and they may exert anti-inflammatory effects by synergistic manner.
