2.Therapeutic effect of vitamin E succinate combined with tamoxifen on graft model of human breast cancer in nude mice
Academic Journal of Second Military Medical University 2000;0(11):-
Objective:To investigate the antitumor effect of vitamin E succinate combined with tamoxifen on the graft model of human breast cancer in nude mice and to analyze the possible mechanism. Methods: MCF-7 human breast cancer cells were inoculated subcutaneously in 40 nude mice. Then the mice were equally divided into 4 groups: blank control group(non-treatment),TAM treatment group(5 mg/kg),VES treatment group(150 mg/kg) and the TAM+VES treatment group(TAM 5 mg/kg +VES 150 mg/kg).The size of the tumor was measured and both the cell cycle and cell surface Fas/FasL were determined by flow cytometry 4 weeks after treatment.Fas/FasL expression in tumor tissue was detected with immunohistochemistry and cell apoptosis was detected by TUNEL method. Results: The co-administration of VES and TAM had more potent inhibitory effect on the growth of solid tumor than VES or TAM administered alone(P

5.Construction and identification of the recombinant pCD-Eg95 plasmid of Echinococcus granulosus
Journal of Chongqing Medical University 1987;0(01):-
Objective: To construct and identify the recombinant pCD-Eg95 plasmid of Echinococcus granulosus. Methods: Total RNA was extracted from hydatid cyst protoscoleces shattered by supersound. The Eg95 coding gene was amplified by RT-PCR, and then was cloned into the eukaryotic expression vector pCDNA3.1 to construct the recombinant pCD-Eg95 plasmid. The recombinant plasmid was electroporated into E. coliBL21(DE3) strain. Results: The 471 bp Eg95 coding gene was successfully amplified, and the recombinant pCD-Eg95 plasmid was successfully constructed, which was confirmed by restriction endonuclease digestion and PCR. Conclusion: The results could provide the basis for the further research of the nucleic acid vaccine of Echinococcus granulosus.
6.Construction and expression efficiency of the recombinant plasmid pBI-Eg95 of Echinococcus granulosus in the agrobacterium tumefaciens LBA4404 strain
Journal of Chongqing Medical University 2007;0(08):-
Objective:To construct the recombinant plasmid pBI-Eg95 of E.granulosus, and to study its expression efficiency in the Agrobacterium tumefaciens(At) LBA4404 strain. Methods:Total RNA was extracted from hydatid cyst protoscoleces shattered by supersound. The Eg95 coding gene was amplified by RT-PCR,and then was cloned into the plant expression vector pBI121 to construct the recombinant plasmid pBI-Eg95.The recombinant plasmid was electroporated into At(LBA4404) strain. The recombinant At(rAt) was induced by IPTG for 0,1,3,5,7,9,11or13 hour, respectively. The expression product was identified by SDS-PAGE and Western blot assay. Results:The 471bp Eg95 coding gene was successfully amplified, and the recombinant pBI-Eg95 plasmid was successfully constructed, which was confirmed by restriction endonuclease digestion and PCR.The recombinant plasmid pBI-Eg95 was expressed successfully in the rAt,and the expression product with an approximately Mr of 16.5 kD could be detected by sera from mice infected by E.granulosus, and the expression efficiency was about 20 percent of the total bacterial protein. Conclusion:The recombinant plasmid could provide the basis for further research of the transgenic plant vaccine of E.granulosus.
7.Relationships Among Narcissism,Empathy,and Prosocial Behaviors
Chinese Journal of Clinical Psychology 1993;0(02):-
Objective:To explore the relationships among narcissism, empathy and prosocial behaviors. Methods:Four hundred and eight participants finished Narcissism Personality Questionnaire,Hyper sensitivity Narcissism Scale,The Interpersonal Reactivity Index, and Questionnaire of Prosocial Behavior.Results:① Overt -narcissism was slightly correlated with covert -narcissism.② After controlling age and gender, overt narcissism could negatively predicate perspective taking, and personal distress, and positively predicate fantasy;and covert narcissism could negatively predicate perspective taking and empathic concern, and positively predicate fantasy and personal distress.③After controlling age, gender and empathy, overt narcissism could significantly predicate self-reported proscial behaviors. Conclusion:Overt narcissism and covert narcissism have differences;both overt and covert narcissists are lacking of empathy for others.
8.SAHA affects cell cycle of MCF-7 breast cancer cells byregulating acetylated levels of p21WAF1/CIP1 promoter
Chinese Pharmacological Bulletin 2017;33(10):1421-1425
Aim To study the regulation mechanisms of deacetylase inhibitor SAHA in p21WAF1/CIP1 promoter acetylation in breast cancer MCF-7 cells. Methods We used quantitative real-time PCR, Western blot and DNA-ChIP to determine the effects on the regulation of cell cycle with SAHA treatment in MCF-7 cells.By DNA-ChIP, we assessed the acetylation level of p21WAF1/CIP1 promoter. Results SAHA significantly affected the expression of cell cycle-related factors, and induced the mRNA and protein expression of p21WAF1/CIP1.SAHA could adjust the acetylation level of p21WAF1/CIP1 promoter. Conclusion SAHA regulates the cell cycle progression by adjusting the acetylation level of p21WAF1/CIP1 promoter in MCF-7 cells.
10.Biosafety and biocompatibility of a variety of biological materials
Chinese Journal of Tissue Engineering Research 2009;13(38):7559-7562
OBJECTIVE: To discuss the biosafety and biocompatibility of biomaterials. METHODS: A computer-based online search of CNKI, CBM and VIP as well as manual search were performed to collect articles about random cell control experiments and animal experiments of biological material biocompatibility published between 1990 and 2008. A total of 28 Chinese literatures were collected, and 7 were included mainly involving the cytotoxicity test methods and blood compatibility of the experimental medium, experimental grouping, experimental materials, methods of observation, experimental results, and experimental conclusion. In addition, biocompatibility of the biological material was analyzed to summarize the biocompatibility of the biological safety. RESULTS: Based on experiments of biosafety and biocompatibility of biomaterials, the cytotoxicity test and blood compatibility of various biomaterials, such as collagen, chitosan, magnetic nanoparticle, metal vascular stent, sulphurated siliastic medical grade silicon rubber, polyurethane, ceramic coatings by micro-arc oxidation, are essential for the biological safety. The experimental results have shown that the biomaterials have good biocompatibility. CONCLUSION: A variety of biological materials present good biocompatibility, including collagen, chitosan, magnetic nanoparticle, metal vascular stent, sulphurated siliastic medical grade silicon rubber, polyurethane, ceramic coatings by micro-arc oxidation, based on the evaluation criteria.
