2.Construction of Essential Medicine Selection Model Based on Knapsack Theory

OBJECTIVE:To provide reference for replenishing and perfecting essential medicine selection method in China. METHODS:According to the requirements of essential medicine selection,based on the actual situation,target function and restraint conditions for essential medicine selection could be determined by the knapsack theory and the essential medicine selection model could be built. The feasibility of established model was analyzed by an sample. RESULTS:The objective function of estab-lished model was Min∑CjXj,the minimum cost of all the selected essential medicines. Other constraints included covered range of disease,ADR,contraindications,attentions,dosage forms and specifications. Nine kinds of common oral hypoglycemic drugs were selected as candidate drugs,from which glipizide,gliclazide,metformin and repaglinide were chosen as the essential hypogly-cemic medicines with the proposed model. CONCLUSIONS:Established essential medicine selection model based on knapsack the-ory is scientifical,objective and with strong practicality.
3.lncRNA-H19 induces epithelial-mesenchymal transition and enhances the invasion of non-small cell lung cancer cells

Background and purpose:In recent years, the studies have indicated that long non-coding RNAs (lncRNAs) may play important roles in the initial stage and development of tumors. H19 is one of those lncRNAs, which is already proved to overexpress in a variety of tumors such as bladder cancer, stomach cancer and hepatocellular carcinoma (HCC). And H19 also could promote tumor proliferation and increase tumor cell migration and invasion ability, but neither the expression nor the function of H19 in non-small cell lung cancer(NSCLC) are clariifed. This study aimed to observe the effects of H19 on the proliferation, epithelial-mesenchymal transition (EMT) and invasion ability of NSCLC cell line A549.Methods:H19 was overexpressed by plasmids transfection, then the effect of H19 on the proliferation of A549 was measured by cell counting kit-8(CCK-8), the invasion of A549 cells was detected by Transwell assay, and the changes of cell morphology were observed with an optical microscope, and the expression of EMT-related proteins was detected by Western blot, and the promoter activity of CDH1 was measured by luciferase assay.Results:The proliferation of A549 cells was increased under the overexpression of H19(D value in blank group was 1.64±0.02, in negative control group was 1.59±0.04, in overexpression of H19 group 1.89±0.02,P<0.05), the invasion ability of A549 cells was dramatically enhanced [negative control group (30±6)/vision, overexpression of H19 group (110±7)/vision,P<0.05], and the A549 cells developed longer pseudopodia and had wider intercellular spaces. All these morphology changes indicated that the cells were undergoing the process of EMT, and meantime, the expres-sion of CDH1 was decreased, along with the expression of VIM and SNAI2 elevated, which were also related to the progression of EMT, and H19 also could depress the promoter activity of CDH1 by 60% (P<0.05).Conclusion:The overexpression of H19 induces the EMT, and enhances the proliferation and invasion ability of A549 cells.
4.Dose Determination and Calibration of Siemens primus E LINAC

The procedures for the calculation and measurement and calibration of output dose of the Siemens primus E accelerator with IAEA regulation are introduced to probe an accurate,rapid and effective method.The parameter value of measurement is determined according to IAEA regulations and absorbed dose calculation formal is briefed to the form that the reading of instrument multiplies exposure calibration coefficient Nx and Air temperature,pressure and humidity effects Ktp and cumulative correction factor Cf the precision of accelerator conform to the standard of QA&QC by the measurement and calibration of the Hospital Primus E accelerator in output dose.The methods of measurement and calibration can be used for the determination of output dose in precise radiotherapy of digital medical linear accelerator and clinical treatment.
5.ANTIMUTAGENIC EFFECT OF P—AMINOBENZOIC ACID AND SODIUM EISULFITE ON THE MUTAGNICITY OF N-METHYL-N-NITRO-N-NITROSOGUANIDINE

Antimutagenic effect of p-aminobenzoic acid (PABA) and sodium bisulfite (NaHSO_3)on the mutagenicity of N-methyl-N-nitro-N-nitrosoguanidine (MNNG) was detected by the mehod of human lymphocytes unscheduled DNA Sythesis (LIDS). The results showed that when the concentration of PABA and NaHSO_3 was varied in presence of a constant concentration of MNNG, 5 ?10~(-5) mol/L concentration of PABA, 3 ? 10~(-4) mol/L concentration of NaHSO_3 were the most effective, while the concentration of MNNG varied in the presence of a constant concentration of PABA and NaHSO_3, antimutagenicity was the most effective at high concentration of MNNG. The results indicated that the PABA and NaHSO_3 exhibited antimutagenic activity towards MNNG-induced mutagenicity in LIDS and the extents of antimutagenicity were related with the concentration of MNNG.
6.Rapid detection of TSH and T_4 by microfluidic chip-based immunoassays

Objective To establish an immunoassay based on microfluidic chip for detection of TSH and total T4.Methods The direct-write laser micromachining technology was used to fabricate microfluidic chip,luminol-H2O2 as luminator.This method and Roche E2010 electrochemiluminescence immunoassay were used to examine TSH by double-antibody sandwich method and total T4 by competitive principle in 50 serum samples collected at our outpatient clinic.Results The detection was completed within 25 min.Ten microliter sample and reagent was required.The detection range of TSH was 0-50 ?IU/ml and its average intraassay and interassay precision was 4.7% and 4.6% respectively and its average recovery rate was 95.3%.The detection range of T4 was 9.375-300 ng/ml,and its average intraassay and interassay precision was 4.9% and 4.7% respectively and its average recovery rate was 91.27%.Conclusion Rapid detection of TSH and T4 by microfluidic chip-based immunoassays is convenient,stable and accurate and of less reagent and time consuming.
7.Construction and Empirical Study of Evaluation Model for Essential Medicine Bidding and Procurement Based on Fuzzy Group Decision Making

OBJECTIVE:To establish the essential medicine bidding evaluation model and improve the scientificity and effec-tiveness of essential medicine bidding and procurement. METHODS:Combined with the main objective of essential medicine cen-tralized bidding and procurement,a criteria bidding evaluation index system for essential medicine was established,and a bidding evaluation model for essential medicine was built based on fuzzy group decision making and the feasibility was verified by exam-ples. RESULTS:The essential medicine evaluation model based on fuzzy evaluation and group decision making was established, and empirical analysis for Atorvastatin Calcium tablet was used to show the feasibility of evaluation model. CONCLUSIONS:The essential medicine evaluation model based on fuzzy group decision making can scientifically and effectively accomplish a compre-hensive evaluation of pharmaceutical enterprises,which builds the foundation for promoting the transparency of essential medicine bidding and procurement work and reducing the economic burden for the patients.

9.Effect of ketamine on proliferation and the cell cycle of cultured rat neural stem cell

AIM To study the effect of ketamine on proliferation,cell cycle in the cultured rat neural stem cells. METHODS The growth inhibition of ketamine on neural stem cell was evaluated by an MTT assay. The effect of ketamine on cell cycle was measured by flow cytometry. RESULTS Ketamine inhibited the growth of cultured rat neural stem cells. Flow cytometry analysis showed that G 0/G 1 phase rate was increased but S phase rate was decreased. CONCLUSION Ketamine can inhibit proliferation of cultured rat neural stem cells,and this inhibitition is associated with cell cycle block.
10.Dynamic changes of serum interleukin-18 level in patients with acute cerebral infarction

BACKGROUND: It has been proved that interleukin-18 (IL-18) involves in the development of acute cerebral infarction (ACI) and is positively correlated with the time of brain stroke onset, erythrocyte sedimentation rate (ESR), as well as neurological deficit and low-density value of brain CT.OBJECTIVE: To investigate the dynamic changes of serum IL-18 in the course of ACI.DESIGN: Verification analysis with patients as subjects and healthy volunteers as controls.SETTING: Department of neurology of a municipal hospital.PARTICIPANTS: Totally 46 inpatients (29 males and 17 females) with ACI were randomly selected from the Department of Neurology of People's Hospital, Maoming City, between December 2002 and January 2004.Meanwhile, 40 healthy controls (27 males and 13 females) were recruited from those who came to the hospital for routine physical examination. All the participants signed informed consent.METHODS: Fasting peripheral vein blood of 2 mL was collected from the patients on the 1st, 7rh, 14th and 21st days of ACI onset, and from healthy controls on the day of routine examination. The blood samples were centrifuged at 3 000 r/minute for 15 minutes at 4 ℃, and then the supernatant was collected for detecting IL-18 level by ELISA.MAIN OUTCOME MEASURES: The serum IL-18 level of patients on the 1st, 7th 14th and 21st days of ACI onset and that of healthy controls on the day of routine examination.RFSULTS: Totally 46 patients and 40 healthy controls were enrolled in this study and all data were statistically analyzed. The level of serum IL-18 was significantly higher in the patients with ACI than in normal controls on the 1st and 7th days of onset [(178±41) ng/L, (104±34) ng/L, (65±14) ng/L, P < 0.01],but was similar on the 14th and 21st days [(88±36) ng/L, (72±33) ng/L,(65±14) ng/L, P> 0.05]. The level of serum IL-18 in ACI patients was significantly higher on the 1st day than on the 7th, 14th and 21 st days of the onset (P < 0.05-0.01); moreover, it was also significantly higher on the 7th day than on the 21st day of the onset (P < 0.05).CONCLUSION: The level of IL-18 increases obviously on the 1st day of ACI onset, and gradually decreases with the extended course of disease and time of treatment.
