BACKGROUND: There are several ways to measure content of pseudoephedrine, such as titration, spectrophotography,gas chromatography, capillary electrophoresis, high performance liquid chromatograpy (HPLC), combination of liquid chromatography, etc. Therefore, an exact method to measure concentration of medicine in blood is of significance for reasonable and safe administration in clinic.OBJECTIVE: To establish a reversed-phase-HPLC (RP-HPLC) method for determining the content of pseudoephedrine in blank plasma.DESIGN: Simple exponent study.SETTING: Department of Pharmacology, Renmin Hospital of Wuhan University.MATERIALS: The experiment was carried out in the Department of Pharmacology, Renmin Hospital of Wuhan University from May to July 2005. Pseudoephedrine was provided by Hubei Institute for Drug Testing. Blank plasma was provided by Central Blood Station of Xiaogan.METHODS: Standard plasma exponent was dispensed containing 50, 100, 200, 500, 750 and 1 000 μg/L pseudoephedrine, respectively. Pseudoephedrine was extracted from standard plasma exponent with organic solvent and its content was measured with HPLC. ZorbaxSB-C18 column (4.6 mm × 250 mm, 5 μm) was also used in this study [mobile phase: 0.2% phosphoric acid solution-acetonitrile (95:5); flow rate: 1.0 mL/min], and the detective wavelength was 194 nm.MAIN OUTCOME MEASURES: Veracity and feasibility through evaluating standard curve, precision and retrieved rate.RESULTS: Linearity ranged from 50 to 1 000 μg/L (r=0.999 3) and average retrieved rate was 79.20%. The relative standard deviation of within-day and between-day was less than 5%. The minimal detectable concentration of pseudoephedrine was 25 μg/L in plasma.CONCLUSION: RP-HPLC which is characterized by convenience and precision is suitable for researching pharmacokinetics and bioavailability of pseudoephedrine.

Aim To investigate the effects of Liensinine on platelet aggregation and coagulation function in rats,as well as the effect on experimental thrombosis.Methods Inhibition rates of platelet aggregation for Liensinine in vivo were determined by the model of platelet aggregation induced by adenosine diphosphate.Coagulation time of mice was measured by capillary vessel method,and bleeding time of mice was measured by tail-cutting method.The effects of Liensinine were also evaluated on prothrombin time(PT),activated partial thromboplastin time(APTT)and thrombin time(TT).The model of artery-vein bypass thrombosis and Chandler's model were established to observe the effect of Liensinine.Results The result showed that Liensinine 5 and 10 mg?kg-1 had significant effect on inhibition of platelet aggregation and markedly prolonged bleeding time,coagulation time,PT,APTT and TT.Liensinine 5 and 10 mg?kg-1 inhibited the artery-vein bypass and Chandler's thrombus in different degree,reduced the thrombus weight significantly (either wet ordry).Conclusion Liensinine exerts remarkable effect against thrombosis and possesses strong effect against platelet aggregation and coagulation.

Aim To study the influence of Liensinine(LIE) on the oxidative damage of erythrocyte ghost. Method The erythrocyte ghost oxidative injury caused by super oxide anion (O?_2) from the auto-oxidation of pyrogallic acid was used as model. The effect of LIE on oxidated erythrocyte ghost was observed by the measurement of the malonidial aldehyde (MDA)-the degradation product of lipid peroxidation, blocking degree, speed of auto-oxidation and membrane fluidity. Result O?_2 caused lipid peroxidation of erythrocyte ghost, increased the content of MDA and the speed of auto-oxidation, decreased membrane fluidity, and alleviated blocking degree. But LIE(80, 100, 120 mg?L -1 )could decrease the content of MDA and speed of auto-oxidation markedly, improve the membrane fluidity and blocking degree significantly, and inhibit the lipid peroxidation. Conclusion LIE has protective action on the oxidative damage of erythrocyte ghost initiated by O?_2

Objective To investigate the effect of radiofrequency ablation on serum tumor necrosis factor alpha-α(TNF-α),interferon gamma-γ(IFN-γ)and interleukin 17 (IL-17)levels in patients with lumbar disc herniation.Methods We studied 276 patients with lumbar disc herniation admitted to the First Affiliated Hospital of Xi'an Jiaotong University from January 2010 to December 2015.The experimental group received radiofrequency thermocoagulation and target ablation decompression treatment.In the same period,133 healthy subjects were selected as control group.We collected the general information of the admitted patients and their serum before and after operation as well as serum of the control group.The changes of TNF-α,IFN-γand IL-17 levels were detected by ELISA method.Results The levels of serum TNF-α,IFN-γand IL-17 in the patients with lumbar disc herniation were significantly higher than those in the control group (P<0.05 ).Compared with those before operation,the serum levels of TNF-α,IFN-γand IL-17 were decreased significantly in the recovery phase after radiofrequency ablation (P<0.05).Conclusion Frequency thermal coagulation target ablation decompression treatment of lumbar disc herniation can not only improve pain symptoms but also reduce the systemic inflammatory reactions.Therefore,it deserves clinical application.

Objective To discuss the mechanism of injury,treatment decisions,complications and surgical results of the floating hip injuries (FHI).Methods Thirty-three patients with FHI were retrospectively analyzed in our hospital from June 2001to June 2011.There were 25 males and 8 females,with an average age of 34.6 years (range,4-73 years).The mechanism of injury was motor vehicle collision for 20 patients,fall down from a height for 11,crushed injury for 1,and sports-related injury for 1.According to Liebergall classification for the floating hip injuries,there were 7 cases for type Ⅰ (21.2％),9 for type Ⅱ (27.2％)and 17 for type Ⅲ (51.6％).All patients had associated injuries,the average injury severity score (ISS) was 28.9.Thirty-two of the femoral fractures underwent surgical stabilization of their femoral fractures.Surgical stabilization methods included ORIF or intramedullary nailing (29 cases),external fixation (1 case),total hip arthroplasty (2 cases).Acetabular fractures were treated with open reduction and internal fixation (ORIF) in 16 cases.Thirteen patients suffered pelvic fractures underwent surgical stabilization for at least one component of ring disruption.Results Twenty-eight cases (84.8％) were followed up for 2.3 to 10.1 years (average,3.5 years).Thirteen patients were followed up in the 16 patients who suffered type Ⅰ or Ⅱ fractures.According to Majeed functional evaluation,the results were excellent in 8 cases,good in 3 and fair in 2,with the excellent and good rate being 84.6％.Fifteen patients with type Ⅲ were followed up,according to D'Aubigné and Postel evaluation criterion.The joint functions were excellent in 7 cases,good in 4,fair in 3,and poor in 1,with the excellent and good rate being 73.3％.According to Ma Yuanzhang functional evaluation,the knee functions of the all patients were excellent in 23 cases,good in 3,and fair in 2,with the excellent and good rate being 92.9％.Complications seen in these patients included post-traumatic osteoarthritis of the hip developed in one hips,2 patients developed heterotopic ossification (HO),one case of avascular necrosis of the femoral head,and two femoral nonunions.The most severe complication was traumatic sciatic nerve palsy,which was found in 5 patients.Conclusion Ipsilateral injuries to the femur and the pelvis or acetabulum (floating hip) are severe injuries usually caused by high-energy trauma.The femur fracture will most commonly be addressed first,and surgeons should be aware of the high incidence of sciatic nerve palsy as well as treatment options and potential complications associated with this devastating combination of injuries.

Objective To study the chemical constituents of Bombycis Feculae. Methods Chemical constituents were isolated BPLC-ELSD. The structures of the isolated compounds were determined by spectral means. Results Two compounds were isolated and identified as 1-deoxynojirimycin (1) and (2R,3R,5R)-2-(hydroxymethyl)piperidine-3,5-diol, named as 1,3-dideoxygalatonojirimycin (2). Conclusion Compound 2 is a new alkaloid. The extract of Bombycis Feculae, compound 1 and compound 2 show inhibitory activities against a-glucosidase.

Objective To evaluate the efficacy and safety of endoscopy-assisted percutaneous interventional procedure with a 980-nm laser for the treatment of venous malformations.Methods A total of 167 patients with venous malformation were enrolled,including 102 females and 65 males,whose age ranged from 0.5 to 60 years.Skin lesions occurred on the limbs in 66 patients,on the head and face in 48,on the chest and abdomen in 24,on the back in 13,on the neck in 10,as well as on the scrotum in 6.A 980-nm laser operated at pulse width of 0 ms-2.5 s and pulse energy of 0-20 W with a 400 μm-diameter optical fiber.Puncture was performed after circular infiltration anesthesia around skin lesions,and then the optical fiber was inserted at the same puncture point to directly or indirectly destruct the vascular wall by photothermal effects and thermal coagulation.For deep lesions around important anatomical structures,the interventional therapy was performed with the assistance of endoscopy or ultrasound.According to the size and shape of tumors,these tumors were irradiated back and forth with speeds of 2 cm/s-4 cm/s.The optical fiber was also inserted in different directions in a fan-shaped pattern for 3-5 sessions until tumors were shrunken or even disappeared.The time interval between two treatments was 3 months,and all the patients were followed up for 4 months on average.Results Of the 167 patients,148 (88.6％)were cured,11 (6.6％) markedly improved,4(2.4％) improved,and 4(2.4％) unimproved.The response rate was 97.6％.Totally,159 patients were satisfied with the therapeutic effects after 1 session of treatment,and 8 were improved after 2 sessions of treatment with a time interval of 3 months.In addition,mild postoperative pigmentation occurred in 6 (3.6％) cases,and skin burns in 3 (1.8％) cases.After 3 months,the above symptoms were improved significantly.Two (1.2％) patients had vascular recanalization and residual lesions,but they were improved after reoperation.No adverse reactions were observed,such as bleeding,scar,tendon and nerve injuries,and dysfunction.Conclusion A 980-nm laser percutaneously delivered through an optical fiber is safe and effective for the treatment of venous malformation.

Objective To study the effects of sinomenine on scavenging oxygen free radical and anti-lipid peroxidation in vitro. Methods Superoxide anion free radical(O_2~?) induced by the auto-oxidation of pyrogallol and hydroxyl free radical(?OH) induced by Fenton system were determined by spectrophotometry.The inhibition on homogenate and mitochondrial lipid peroxidation,malondialdehyde(MDA),of rat liver induced by VC-Fe~(2+) system was measured by thiobarabituric acid(TBA) colorimetric method.Results Sinomenine could significantly scavenge O_2~? and ?OH,it's EC_(50) were 1.35 and 0.136 mg/mL respectively.Sinomenine could also remarkably inhibit the lipid peroxidation of rat liver homogenate and mitochondria induced by auto-oxidation and VC-Fe~(2+) system.The scavenging and inhibitory effect exhibited in a concentration-dependent manner.Conclusion The results indicate that sinomenine possesses the effect of scavenging oxygen free radicals and anti-lipid peroxidation,which shows the protection of liver mitochondria.
