strains that could produce raw starch-digesting glucoamylase were isolated from soil and mildewed rice. The highest raw starch-digesting glucoamylase activity strain named OR-1 was identified as Rhizopus.sp.The raw starch-digesting glucoamylase activity of the strain is 90U/mL.Through UV and NTG mutagenesis, the raw starch-digesting glucoamylase activity raised to 200U/mL and 325U/mL respectively. The RDA were 70% and 65% respectively.

International Eye Science 2015;(1):134-135
AlM:To evaluate the effect of intralamellar cryolysis of cornea with amniotic membrane transplantation on painful bullous keratopathy ( PBK) .?METHODS: Randomly selected 156 cases ( 156 eyes ) with PBK who underwent surgery of intralamellar cryolysis of cornea with amniotic membrane transplantation. Followed up with 2 ~ 3mo, the symptoms of eye pain, corneal epithelial blisters, foreign body sensation and postoperative complications were observed.?RESULTS:Pain symptoms disappeared in all patients, and corneal epithelial blisters disappeared in 130 cases (83. 3%). All agonizing pain symptoms disappeared, but patients had occasional foreign - body sensation, occasional corneal epithelial blisters in 24 cases ( 15. 4%) . Two weeks after surgery, corneal stroma dissolved, 2 cases ( 1. 3%) of them were cured by conjunctival flap cover.?CONCLUSlON:The operation of intralamellar cryolysis of cornea with amniotic membrane transplantation can relieve the pain in 98. 7% of PBK patients and simple therapy for treating PBK. Hence, it's be worth to advocate for relieve the pain of patients.

Objective To observe the alteration of astrocyte and apoptosis in neonatal hypoxic-ischemic encephalopathy(HIE).Me-thods The pathological brain specimens of 25 newborns who died of HIE were observed macroscopically and microscopically, included male 14 cases and female 11 cases. The immunochemistry staining with glial fibrillary acidic protein(GFAP)and in situ end terminal dexynucleotidy 1 transferase mediated deoxyuridine triphosphate-biotion nick and labeling(TUNEL)methods were used to observe the changes of astrocyte and the neurons apoptosis in HIE.The relationship between the change of astrocyte and the neurons apoptosis and the occurrence of HIE and course of disease were further synthetically analyzed. Results The brain specimens of 25 newborn HIE had apoptosis cells at the different level. The degree of apoptosis of cerebral neurons was severe in the cases of 24 h-6 d survival(P

Objective To investigate the association of chromosome 8p copy number alteration (CNA) with postoperative survival of patients with hepatocellular carcinoma (HCC),and to screen for possible target genes in the survival-related CNA (s) in 8p.Methods 187 HCC patients were enrolled into the study, which included 66 patients whose follow-up data were available and the follow-up was 2.6 ~ 73.3 months.High-resolution Agilent 244K comparative genomic hybridization (CGH) and Affymetrix U133 Plus2.0 expression arrays were used to screen for CNAs and gene expression differences in 8p.The associations between CNAs in 8p and survival were analyzed using the log-rank test,Kaplan-Meier survival analysis and Cox proportional hazards models. The gene expression levels between the groups were compared by the Mann-Whitney U test. Results Copy number loss on 8p12 (31/66,47%) was significantly associated with reduced survival rate, and HCC patients with 8p12 loss had a 4.1-fold (95% CI =1.8 ~ 9.4,P < 0.05) increased hazard ratio (HR) for death from HCC,as compared to those without the loss. The mRNA expression levels of the 3 genes in 8p12,including TMEM66,DCTN6,and MAK16,were significantly decreased in HCCs with gene loss than in HCCs without the loss (all P < 0.05),and in non-tumorous liver tissues (all P < 0.05).Conclusion Loss of 8p12 is an independent prognostic marker of unfavorable survival for patients with HCC,and underexpression of genes TMEM66,DCTN6,and MAK16,owing to 8p12 loss, contributed to unfavorable prognosis.

OBJECTIVETo observe the effect of Chinese herbal extract HuNan A-1 (HNA-1) on the thymic output function in Simian immunodeficiency virus (SIV) chronically infected rhesus macaques.
METHODSEight Chinese rhesus macaques had been infected by SIVmac239 for 16 to 21 months, and then they were randomly divided into the treatment group and the control group, 4 in each group. Monkeys in the treatment group were administered with HNA-1 by gastrogavage, once daily for 2 successive months, while those in the control group were administered with equal volume of normal saline by gastrogavage, once daily for 2 successive months. The general condition and body weight of monkeys were observed. Plasma viral loads were detected using real-time fluorescent quantitative PCR assay. CD4 percentages and counts, as well as naive CD subsets were detected using flow cytometry. T-cell receptor excision circles (TREC) were detected using real-time fluorescent quantitative PCR assay. The thymus tissue was pathologically observed using routine HE staining. The correlation between lesions of the thymus tissue, CD4 counts, naive CD counts, and TREC were analyzed.
RESULTSThere was no statistical difference in body weight, viral loads, absolute CD ratios between the two groups after treatment (P > 0.05). The altered TREC multiple showed an obvious decreasing tendency in the control group, while it showed an increasing tendency in the treatment group (P < 0.05). In both groups, destroyed structures of the thymus tissue could be seen, filled with pink unstructured material. Increased connective tissues, lowered connective cell density, and confused arrangement could also be seen in the two groups, with no obvious difference. TREC contents were positively correlated with naive CD4 counts after removing extremum (r = 0.926, P = 0.001). Naive CD4 counts were positively correlated with CD4 counts (r = 0.961, P = 0.005).
CONCLUSIONSTREC content determination, as a marker of newly thymic emigrants, could be taken as a testing method for evaluating the thymic output function. Besides, HNA-1 treatment increased the thymic output significantly in SIV chronically infected monkeys. Correlation existed among TREC contents, naive CD4 counts, and pathologies of thymus tissues, especially in late infection stage.

Objective To integrate research training into clinical teaching effectively, the essential steps are understanding the students` demand and mastery, factors hindering learning and how to learn effectively.MethodsStudents in 7th grade in Peking Union Medical College (PUMC), graduate students and residents under clinical training, trained doctors for advanced studies from primary hospitals were surveyed with questionnaire by collecting educational background, the demand of learning medical research methodology, factors hindering learning, specific issues need in medical research methodology, learning model and assessment approaches. Results Totally 69 people were surveyed.92.75% of the respondents said they had studied medical research methodology and related courses before. However, 51 people (73.91%) answer they cannot read literatures with critical thinking.52 people (75.37%) indicated that they had difficulty in conducting research.58 (84.06%) people said it is very necessary to study the medical research methodology. No learning opportunities and enough time were the main hinders forlearning relevant knowledge. The top three high-demanding issues are sample size calculation and statistical knowledge, study design, and how to use the statistical software.44 (63.77%) people suggest e-learning with case based learning. Conclusions Integrate medical research methodology teaching into clinical teaching is in high-demand for students.E-learning is suggested with case based learning.
