1.The diagnostic value of serum angiopoietin-2 level for gastric cancer

Objective To evaluate the diagnostic value of serum angiopoietin-2 (Ang-2) level for gastric cancer.Methods Serum Ang-2 level waa measured by enzyme-linked immunosorbent assays (ELISA) in samples from 108 patients with gastric cancer,50 patients with chronic gastritis and 50 normal controls.Results The serum Ang-2 level in patients with gastric cancer [(1539.0 ± 449.3) ng/L] was significantly higher than that in patients with gastritis [(1044.6 ± 246.1) ng/L,P < 0.01] and normal control [(1075.6 ± 228.2) ng/L,P < 0.01)].There was no significant difference of serum Ang-2 level between patients with gastritis and normal control.Gastric cancer patients with lymph node metastasis had significantly higher serum Ang-2 level than those without metastasis (P <0.01).The area under ROC curve of serum Ang-2 level for diagnosis of gastric cancer was 0.819.Conclusion The serum Ang-2 level will be a useful indicator for diagnosis of gastric cancer.
2.Construction and Expression of the eukaryotic expression recombinant plasmid of CadF gene of Campylobacter jejuni Pen: 19

Objective: To construct the eukaryotic expression recombinant plasmid of cadF gene of Campylobacter jejuni Pen: 19 and to identify its express in COS-7 cells.Methods: Total DNA of Campylobacter jejuni Pen:2?Pen:8?Pen:19 and Pen: 21 were used as templates respectively and cadF gene DNA with KpnI and EcoRI sites were amplified by PCR reactions using same primer.Because of its closest association with Guillain-Barr? syndrome,the PCR product of Pen:19 was selected as target gene and inserted into the eukaryotic expression vector pcDNA3.1(+) and the recombinant plasmid pcDNA3.1(+)-cadF was constructed.The recombinant plasmid was identified by restriction enzyme digestion and PCR reaction and was confirmed by sequencing.The recombinant plasmid was then transfected into mammalian cell COS-7 for mRNA expression and the expression were investigated by RT-PCR reaction.Results: It was confirmed with restriction enzyme digestion and PCR reaction and sequencing that cadF gene was cloned into pcDNA3.1(+) successfully.The objective gene was amplified from COS-7 cell transfected with the recombinant plasmid pcDNA3.1(+)-cadF by RT-PCR.Conclusion: The eukaryotic expression recombinant plasmid pcDNA3.1(+)-cadF is constructed and expressed in COS-7 cell successfully.The results obtained lay the foundation for research on development of Campylobacter jejuni DNA vaccine.

4.Effect of chronic stress on long-term potentiation of hippocampus and neurotransmitter of amino acid in rats

BACKGROUND: Severe or prolonged stress is harmful to health and even induces many mental and physical disorders and then impairs cognitire functions of the organism.OBJECTIVE: To investigate the effects of stress on the changes in amino acid and chronic stress on long-term potentiation (LTP) of hippocampus of rats.DESIGN: Completely randomized controlled experiment.SETTING: Mental Health Center of Shantou University.MATERIALS: The experiment was completed in the Medical College of Shantou University in December 2000. Totally 16 adult male SD rats were divided randomly into control group and stress group with 8 in each group.METHODS: Mice in stress group were swum compulsively for 4 weeds.Chronic stress model was established, and isolated hippocampal slice(500 μm) combined with electrophysiological technique was used to observe the changes of LTP in hippocampal CA1 field. High-frequency stimulation (HFS) was applied to Schaffer collaterals in hippocampus CA3, then the changes of PS amplitude and fEPSPs slope in CA1 pyramidal neurons were observed. High performance liquid chromatography (HPLC) coupled with UV detection was used for the quantificative analysis of hippocampal amino-acid neurotransmitters.MAIN OUTCOME MEASURES: ① Population spike (PS) amplitude and field excitatory postsynaptic potentials (fEPSPs) slope were used to indicate the changes of LTP. ② Changes of amino acid of hippocampus.RESULTS: One rat in stress group died because of drowning in the second week of experiment was supplied, and others entered the final analysis. ① The increases of PS amplitude and fEPSPs slope after HFS in control group were significantly higher than those in stress group (P< 0.05). ② The levels of aspartate and glutamate in control group were significantly lower than those in stress group [(2.425±0.211) μmol/g,(6.016±0.677) μmol/g. P< 0.01; (4.746±0.609) μmol/g. (8.094±1.035) μmol/g.P < 0.01]; but there was no significant difference of γ-aminobutyric acid between control and stress groups [(4.229±0.449) μmol/g, (4.249± 0.463) μmol/g, P > 0.05].CONCLUSION: It is suggested that chronic stress can inhibit the development of LTP in hippocampal CA1 field and increase the levels of hippocampal aspartate and glutamate, but does not affect the level of γ-aminobutyric acid. The accumulation of hippocampal excitatory amino acids induced by chronic stress may be the neurobiochemical basis of impairments in learning and memory abilities.
5.Preoperative assessment strategies of hilar cholangiocarcinoma

At present,in the opinions of the domestic and foreign scholars,the only hope of long-term survival even cure of hilar cholangiocarcinoma was obtained by radical resection(ie R0 resection).The application of preoperative assessment with appropriate and comprehensive strategies can judge the respectability and Intraoperative safety accurately to the tumor,thereby it can improve the radical resection rate and safety of patients with hilar cholangiocarcinoma.In this paper,preoperative assessment strategies are reviewed.
6.Pregnancy outcome in women with lupus nephritis

Objective To study the outcome of pregnancy with lupus nephritis. Methods The clinical data of 9 pregnancies with lupus nephritis were retrospectively malyzed. Results Hypertension,proteinuria,injured renal function and abnormal serological marker were complicated with nine patients with lupus nephritis in pregnancy. The outcome of pregnancy was as following:abortion 2 cases,stillbirth 1 case,premature delivery 4 cases,term delivery 2 cases. All neonate survived without malformation. There was no postpartum hemorrhage or death. Patients with active SLE had very high fetal mortality( 100% ). And SLE got worsen in 50% of them. However,fetal mortality rate in pa-tients with controlled or quiescent SLE was 11.1% ,which was much lower than it in other patients(P<0.05). Con-clusion Lupus nephritis should be controlled before pregnancy. Pregnancy outcome could be improved by careful multidisciplinary monitoring and proper usage of adrenal cortical hormone.

8.Mechanism of Xiao Chaihu Decoction in the Treatment of Endometriosis

Objective To explore the mechanism of Xiao Chaihu decoction in t he treatment of endometriosis in rats.Methods The rat model of endometriosis w as established. Fas and Caspase- 3 protein expression in the eutopic endometriu m and eutopic endometrium were observed by immunohistochemical method.Results The protein expression of Fas and Caspase- 3 in ectopic endometrium of Xiao Cha ihu decoction group was higher than that in the eutopic endometrium, while the r esult in the control group was just the opposite. No significant difference was shown in Danazol group. Conclusion The therapeutic mechanism of Xiao Chaihu de coction for endometriosis may be related to the promotion the apoptosis of ectop ic endometrium tissue by increasing Fas protein expression.
9.Effect of Xiao Chaihu Decoction on the Expression of Cyclooxygenase-2 in Endometriosis Rats

Objective To explore the therapeutic mechanism of Xiao Chaihu Decoction for endometriosis in rats. Methods The rat models of endometriosis were treated with Xiao Chaihu Decoction, Danazol, and Xiao Chaihu Decoction plus Danazol for 4 weeks,respectively. And then cyclooxygenase-2(COX-2) expression in endometrium and endometriotic tissue was observed by immunohistochemistry method. Results The expression of COX-2 in the endometriotic tissue was lower than that in endometrium in Xiao Chaihu Decoction group and Xiao Chaihu Decoction plus Danazol group, while the results in Danazol group and control group were opposed to those in the above two groups. Conclusion The mechanism of Xiao Chaihu Decoction for rats endometriosis may be related to the decrease of COX-2 expression in ectopic Endometrium, thus reducing the formation of prostaglandins.
10.Evaluation of left ventricular longitudinal axis systolic asynchrony using RT-3PE QTVI and STI in patients with dilated cardiomyopathy

Objective To explore left ventricular longitudinal axis systolic asynchrony by real-time triplane quanti-tativetissue velocity imaging(RT-3PE QTVI) and two-dimensional speckle tracking imaging(STI) in patients with dilated cardiomyopathy. Methods Thirty normal adults were included as the control group and thirty adults with DCM. The images from enough frame rates RT-3PE QTVI and STI were obtained from series long-axis views. Simp-son method was used to measure LVEF. Q-lab software was used to measure peak systolic tissue velocity(Vs) and peak systolic longitudinal strain (Ls),its time to peak point from each segment(Ts,Tls) and the maximal temporal difference of Ts,Tls(Ts-diff,Tls-diff),and the standard deviation of. Ts,Tls(Ts-SD,Tls-SD),which were served as systolic asynchrony indexes in assessing LV longitudinal axis systolic asynchrony. Results ① Compared the con-trol group and DCM group,there were significant differences in LVEDd,LVESd,LVEDL,LVESL,LVEF,FS (P<0.01). ② In comparison with the control group,the Ls and Vs of each segment were decreased in the DCM group (P<0.01). Compared with the control group,the parameters of Tls and Ts in DCM group had obvious ahead or de-layed(P<0.05). ③ Compared with the control group,the parameters of Ts-SD, Tls-SD, Ts-diff, Tls-diff,were significantly different in DCM group (P<0.01). Ts-SD,Tls-SD,Ts-diff,Tls-diff corrrlated negatively to LVEF(r=-0.67,-0.72,-0.75, -0.77,P<0.01). Conclusion The prevalence of left ventricular longitndinal axis systolic asynchrony exits in patients with DCM. There is a better technique to evaluate LV longitudinal axis systolic asynchrony by RT-3PE QTVI and STI than only one.
