3.Thrombolytic properties of a conjugate of liposomal urokinase with a monoclonal antibody specific for cross-linked fibrin in the model of rabbit artery thrombosis

AIM: To observe the targeting thrombolytic effect of a monoclonal antibody specific for cross-linked fibrin connected with liposomal urokinase(UK) in the model of rabbit artery thrombosis.METHODS: Preparation of thrombolytic solutions: with the method of controllable dialysis eradicator in liposomat, empty liposomes, liposomally entrapped urokinase and liposomally entrapped urokinase linked with D-dimer antibody were made. Experiment in vivo: a rabbit thrombosis model of abdominal aorta was induced by ferric chloride. When the blood pressure fall to the lowest, 5 different solutions were separately imported (PBS, maximal-level UK, Ab/Lip/UK, Ab/UK-Lip, Ab-UK-Lip) and observed for 40 min continuously. 4.Nursing of Double Interventional Therapy of Portal Hypertension with Liver Cirrhosis

Objective:To evaluate the nursing of portal hypertension patients with hepatic cirrhosis,who were treated by percutaneous transhepatic variceal cmbolization (PTVE) and partial splenic embolization (PSE). Methods: 12 portal hypertension patients with post- hepatitis cirrhosis underwent PTVE and PSE. Their averge age is 51.7 years old.Result :The operation of all patients were successful. The free pressure of portal vein decreased 3.75 mmHg averagely after operation. The patients were followed up for 2~6 monthes. There wasno recurrence of esophageal variceal bleeding. The symptoms of hypersplenism were relived. Conclusion:We considered some segments could help the operation successful:the psychological nursing before operation, the relieving pain, stopping hemorrhage, relieving vomit, sedation and carefully observing patients in the operation,systemic antibiotic dropping, noticing complication appearing and so on after operation
5.Significance of detection of Hcy,hs-CRP and cTnT in patients with coronary heart diseases

Objective To study the significance of plasma homocysteine(Hcy),high sensitivity C-reactive protein(hs-CRP)and cardiac treponin T(cTnT)changes in the patients with coronary heart diseases(CHD).Methods From August 2012 to November 2013,240 patients with CHD in our hospital were collected as the CHD group and 250 individuals of healthy physical examination served as the control group.Serum Hcy,hs-CRP and cTnT were detected by using the enzymatic cycle assay,immune scatter turbi-dimetry and electrochemiluminescence assay respectively.The data were statistically analyzed by the SPSS software.Results The serum levels of Hcy,hs-CRP and cTnT in the CHD group were (24.89±17.78)μmol/L,(17.43±35.89)mg/L and (0.76±2.36) ng/mL respectively,which were significantly higher than those in the control group(P <0.001).Moreover,the positive rate of Hcy in the CHD group was the highest (81 .25%),followed by hs-CRP(64.58%)and cTnT (58.75%).Conclusion Hcy,hs-CRP and cTnT played important roles in the development and progression of CHD,so reducing Hcy level may be an important pathway for preventing and treating CHD.

7.GnRH analog resensitizes cisplatin-resistant human ovarian cancer cells

Objective:To study the effect of GnRH analog triptorelin in resensitizing cisplatin-resistant human ovarian canc- er cells and to discuss the related mechanism.Methods:Cisplatin-resistant human ovarian cancer cell line OVCAR-3/CDDP was established in vitro.MTT assay was used to assess the inhibitory effects of triptorelin,cisplatin alone or a combination of both on OVCAR-3/CDDP cells.Flow cytometry was employed to observe the expression changes in epidermal growth factor receptor (EGFR)in different groups.Results:The drug restant index of OVCAR-3/CDDP cells was 13.42.The resensitizing fold of cisplatin combined with triptorelin was 3.80.The expression of EGFR had the most prominent decrease in OVCAR-3/CDDP cells in the combination group.Conclusion:Triptorelin can partially resensitize cisplatin-resistant OVCAR-3/CDDP cells,which might be related to the down-regulation of EGFR.
9.Effect of Clonorchis sinensis infection on the coagulation indexes of patients with chronic hepatitis B

Abstract: Objective To analyze the changes of coagulation indicators in patients with hepatitis B complicated with Clonorchis sinensis (C. Sinensis), and provide reference value for diagnosis, drug using and prognosis monitoring. Methods The patient samples were collected from the Third Affiliated Hospital of Sun Yat-sen University from June 2018 to February 2022 and divided into six groups. They were 40 healthy patients, 47 patients with simple chronic hepatitis B, 47 patients with post-hepatitis B liver cirrhosis, 40 patients with C. Sinensis mono-infection, 30 patients with chronic hepatitis B patients co-infected with C. Sinensis and 27 patients with post-hepatitis B liver cirrhosis patients coinfected with C. Sinensis. Four coagulation indexes, prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT) and fibrinogen (FIB), were detected and compared among the groups. Results Compared with the healthy group, C. Sinensis mono-infection group had higher level of FIB and delayed PT, APTT; chronic hepatitis B group and chronic hepatitis B patients co-infected with C. Sinensis group had delayed PT, APTT, TT and significant lower FIB, these differences were statistically significant (all P<0.05). Compared with simple chronic hepatitis B group, post-hepatitis B liver cirrhosis and chronic hepatitis B patients co-infected with C. Sinensis had significant delayed PT, APTT, TT and lower FIB (all P<0.05). Compared with the post-hepatitis B liver cirrhosis and chronic hepatitis B patients co-infected with C. Sinensis, post-hepatitis B liver cirrhosis patients coinfected with C. Sinensis group had significant delayed PT, APTT, TT and lower FIB (all P<0.05). Conclusions The coinfection of C. Sinensis will further aggravate the coagulation dysfunction of HBV patients, leading to poor treatment and prognosis. HBV patients will have worse coagulation function in the process to post-hepatitis B liver cirrhosis; Therefore, it is important to pay attention to C. Sinensis co-infection when treating HBV patients, so that further guidance on clinical use and monitoring of prognosis can be provided.
10.Effect of Yijing Recipe on apoptosis of testis spermatogenic cells and the expression of Bcl-2 and Bax protein in infertility rats.

OBJECTIVETo observe the effect of Yijing Recipe (YR) on the apoptosis of testis spermatogenic cells and the protein expression of Bcl-2/Bax in rats with adenine induced infertility.
METHODSTotally 75 Wistar rats were randomly divided into 5 groups, i.e., the blank control group, the model group, the high dose YR group, the middle dose YR group, and the low dose YR group, 15 in each group. Except those in the blank control group, rats in the rest groups were intragastrically administered with adenine for 10 successive days. From the 11th day, rats in the blank control group and the model group were fed with equal volume of normal saline. Rats in the YR groups were intragastrically administered with YR at different doses (3.38 g/100 g; 1.69 g/100 g; 0.85 g/100 g), once daily for 20 consecutive days. All rats were killed by the end of the experiment and their testes extracted. The apoptosis of spermatogenic cells and the expression of Bcl-2/Bax proteins were detected by terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) and SABC method.
RESULTSCompared with the blank control group, the Bcl-2 protein expression decreased, the Bax protein expression increased, and the apoptosis index increased in the model group, showing statistical difference (P <0.01). Compared with the model group, the Bcl-2 protein expression increased in the three YR treated groups (P <0.01, P <0.05). The Bax protein expression level decreased in the high and middle dose YR groups (P <0. 01, P <0. 05). The apoptosis index decreased in the middle dose YR group (P <0.01).
CONCLUSIONYR could inhibit the apoptosis of spermatogenic cells through regulating the expression of Bcl-2 and Bax protein in the testis.
