4.Advances in theoretical studies on coagulating substances secreted by vascular endothelial cells

BACKGROUND:Vascular endothelial cel s participate in the coagulation cascade reaction or contraction of blood vessels by secreting abundant coagulating substances that promote coagulation. OBJECTIVE:To overview the effects of different coagulating substances secreted by vascular endothelial cel s, and provide theoretical basis for the screening of coagulant biomaterials. METHODS:A computer-based research in CNKI and PubMed databases was performed for relevant literatures addressing vascular endothelial cel s and its secreting coagulating substances published from 1988 to 2016 using the keywords of“vascular endothelial cel s, endothelin, Ang II, TXA2, tissue factor, col agen, fibronectin, von wilbrand factor, thrombospondin, platelet activating factor, plasminogen activator inhibitor, proaccelerin, antihemophlic factor”in English and Chinese, respectively. Final y 36 articles were enrol ed for result analysis. RESULTS AND CONCLUSION:Vascular endothelial cel s can secrete numerous coagulation factors that play important roles in the process of coagulation, inflammation reaction and thrombosis fol owing vascular injury. Among them, coagulation factor V and VIII are directly involved in the coagulation cascade reaction and promote thrombosis. In the meanwhile, the vasoconstrictors narrow the lumen, thereby assisting coagulation and promoting thrombosis indirectly. 5.Expression of MT-3 mRNA in human esophageal squamous cell carcinoma

Objective To detect the expression of metallothionein-3 (MT-3)mRNA in human esophageal squamous cell carcinoma. Methods Five cell lines of human esophageal cancer,TE-1,TE-13,TTN,ECA-109 (cell lines of esophageal squamous cell carcinoma) and OE33 (cell lines of esophageal adenocarcinoma),were used in this study. RT-PCR was employed to detect the expression of MT-3 mRNA. Peripheral blood monouclear cells from normal subjects were served as controls. Results Sequencing of RT-PCR product certified the gene of MT-3 mRNA. It was revealed by gel electrophoresis that there was expression of MT-3 mRNA in each cell line. The relative expression of MT-3 mRNA was 0.230?0.023,0.516?0.020,0.140?0.009,0.176?0.015 and 0.085?0.011 in cell lines of TE-1,TE-13,TTN,ECA-109 and OE33,respectively,significantly lower than that in controls (0.762?0.026) (P
6.Effect and Interaction of Total Flavonoids and Saponins from Huang-Qi Ge-Gen Decoction on IL-12 and IL-15 of Liver among Diabetes Rats

This study was aimed to explore the effect of total flavonoids and saponins from Huang-Qi Ge-Gen (HQGG) decoction on blood glucose (BG), serum lipid, interleukin-12 (IL-12) and interleukin-15 (IL-15) of liver in diabetes mellitus (DM) rats, in order to investigate their interactions in regulating DM processes. A total of 66 SD rats were randomly divided into 6 groups, which were the normal group, model group (A1B1), control group, total flavonoids group (A2B1), total saponins group (A1B2), and total flavonoids and saponins group (A2B2), with 11 rats in each group. Except the normal group, other groups were intraperitoneal injected with streptozotocin (STZ). And the experiment was according to 2×2 factorial design experiment scheme. The BG was determined before STZ injection and 7 days after the STZ injection. After 30 days, BG, serum lipid, IL-12 and IL-15 of liver were tested. Related indexes were calculated to the weighted composite score. Main and interactive effect of total flavonoids and saponins were studied according to the factorial design experiment scheme. The results showed that compared with the normal group, all indexes of model group showed statistical differences (P<0.05). Total flavonoids and saponins from HQGG decoction can effectively reduce BG, without any interactions between them. Both the total flavonoids and total saponins can reduce serum cholesterol (CHO), triglyceride (TG), liver IL-12 and IL-15. And there were interactive effects. The single use of herb achieved better effects than the combination. It was concluded that total flavonoids and saponins from HQGG decoction can reduce BG, CHO, TG, and liver IL-12 and IL-15 levels in rats. However, the regulation of total flavonoids and saponins on indexes mentioned above showed no additive effect.
7.Studies on drug resistance genes among 49 strains of extended-spectrum beta-lactamase-producing bacteria

Objective To investigate the drug resistance genes of extended-spectrum beta- lactamase-producing bacteria in 49 strains.Methods Extended-spectrum ?-lactamase -producing strains were detected by the disc diffusion test.The techniques of polymerase chain reaction,sequence analysis, pulsed-field gel electrophoresis were used to analyze the genotype and homology of extended-spectrum ?- lactamase-producing strains.Results The incidence of ESBL-producing strains from E.coli,K pneumoniae,K oxytoca,was 20% in 2000,and 40% in 2003.Among the 49 ESBLs producers the most common genotype was CTX-M-14( n=33).The others were CTX-M-3,CTX-M-9,CTX-M-12,CTX-M-15, CTX-M-24 and SHVSa.Both two CTX-M subtypes,CTX-M-3 and CTX-M-14,were detected in one strain. However,4 ESBL-producing strains confirmed by phenotype remained untyped.The results showed that the ESBLs producers were not closely related,except for two strains of E.coli and two strains of K.pneumoniae which were homgenic respectively.Concolusions The incidence of ESBL-producing strains increases with years.The most common genotype of ESBLs is CTX-M.There is no evidence for epidemiologic spread of ESBL-producers by pulsed-field gel electrophoresis.
8.Neuroprotective effect of curcumin to Aβ of double transgenic mice with Alzheimer's disease.

OBJECTIVETo observe the changes in Aβ40, Aβ42 and ADDLs in brains of 3 month-old APPswe/PS1dE9 double transgenic mice after six-month intervention with curcumin, in order to discuss the neuroprotective effect of curcumin.
METHODAPPswe/PS1dE9dtg mice were randomly divided into the model group, the Rosiglitazone group (10 mg x kg(-1) x d(-1)) and curcumin high (400 mg x kg9-1) x d(-1)), medium (200 mg x kg(-1) x d(-1)) and low (100 mg x kg(-1) x d(-1)) dosage groups, with C57/BL6J mice of the same age and the same background in the normal control group. After 6 months, the immunohistochemical staining (IHC) and the Western blot method were used to observe the changes in positive cell of Aβ40, Aβ42 and ADDLs in hippocampal CA1 area, their distribution and protein expressions.
RESULTBoth of the immunohistochemical staining and the Western blot method showed more positive cell of Aβ40, Aβ42 and ADDLs in hippocampal CA1 area and higher protein expressions in the model group than the normal group (P < 0.01). IHC showed a lower result in the Rosiglitazone group than the model group (P < 0.05), while Western blot showed a much lower result (P < 0.01). The number of Aβ40, Aβ42 and ADDLs positive cells and the protein expressions decreased in the curcumin high group, the medium group showed a significant decrease (P < 0.01), and the low dose group also showed reductions in the protein expressions of Aβ40 and Aβ42.
CONCLUSIONThe six-month intervention with curcumin can significantly reduce the expressions of hippocampal Aβ40, Aβ42 and ADDLs in brains of APPswe/PS1dE9 double transgenic mice. Whether curcumin can impact Aβ cascade reaction by down-regulating expressions of Aβ40, Aβ42 and ADDLs and show the neuroprotective effect needs further studies.

10.Correlation between CT features and clinical severity stratification in acute pulmonary embolism

Objective To analyze the correlation factors between CT imaging features of pulmonary embolism(PE)and clinical severity stratification,to explore the value of CT pulmonary angiography (CTPA)in acute PE severity stratification.Methods According to the clinical severity,48 patients with acute PE proved by CTPA were classified into two groups,including 21 critical and 27 non-critical patients. Embolism index,ratio of central pulmonary involvement,ratio of right ventricle maximum minor axis (RVMMA)to left ventricle maximum minor axis(LVMMA),namely RV:LV,dilation of main pulmonary and/or right pulmonary trunk,and dilation of bronchial arteries in both groups were analyzed comparatively.
