1.The damaging effect of sulfur mustard on the mitochondria in rat lymphocytes in vitro

Objective To investigate the damaging effect of sulfur mustard on the mitochondria in rat lymphocytes in vitro.Methods Rat spleen lymphocytes were isolated by density gradient centrifugation and cultured with 100 ?mol/L sulfur mustard in vitro.DNA ladderring was used to detect the cell apoptosis.Cyt-c release was measured by Western blotting.The mitochondria function was detected by MTT method.Fluorescent probe labeled with Rhodamine 123 was used to study mitochondria potential.Results The mitochondria potential and cell viability decreased at 4 h after treated with 100 ?mol/L sulfur mustard.There was a liner correlation between the mitochondria potential and cell viability in lymphocytes.The content of Cyt-c increased significantly at 4 h as compared with control group in Western blotting.Conclusion Sulfur mustard could damage the rat lymphocyte mitochondria significantly,and mitochondria participates in cytotoxic effect induced by the sulfur mustard.

3.The Study of Three-dimensional Ultrasound in the Solid Spaceoccupying Lesions of Liver A Report of 63 Cases

3D ultrasound reconstruction in 63 cases of the solid space occupying lesions of liver was reported.Among them,there were 34 primary liver cancer,20 liver metastases and 9 cases hamangioma.By 30 imaging,the internal structure of the tumor could be visualized in 71%(45/63); the relation of the tumor to the surrounding vessels was apparent in 65%(41/63); the margin of the tumor could be outlined in 56%(35/63).The vesucts indicate that 3D ultrasound is superior to conventional 2D sonography to show more accurately the space localization,internal structure and morphology of the lesion thus provides meaningful imaging diagnostic evidences clinicaly.
4.Analysis of Surgical Technique and Clinical Outcome of Laparoscopic Inguinal Hernia Repair

Objective To explore surgical technique and clinical outcome of transabdominal preperitoneal hernia repair(TAPP)and totally extraperitoneal hernia repair(TEP)in inguinal hernia repair.MethodsThe clinical data of 23 patients underwent laparoscopic herniorrhaphy were retrospectively analyzed.Nine patients were underwent TAPP,and 14 patients underwent TEP.ResultsNone of patients was changed to open operation.Only one patient of TEP was changed to TAPP.Average operative time was(82.1?40.6)min.Blood loss was(5.7?3.0)ml.Disruption of peritoneum happened to 2 patients underwent TEP.The rate of postoperative complications was 21.7%(5/23),which included serum swelling(1 case),scrotal emphysema(2 cases),transient neurapraxia in the area of repair(1 case)and urinary retention(1 case).No chronic pain,foreign body sensation and infection were found.Average hospitalization after operation was(4.3?0.9)d.No recurrence was observed during a follow-up period of 1-15 months.ConclusionLaparoscope herniorrhaphy is safe due to lower recurrence and complications.It also has the advantages of slight pain and rapid recovery.
5.Diagnosis and therapy of intra-abdominal hernia

Objective To investigate the causes of intra-abdominal hernia and improve the level of early (diagnosis) and therapy of intra-abdominal hernia. Methods A retrospective analysis of the clinical data of 68 cases of intra-abdominal hernia was made. Results The postoperative confirmed diagnosis was para-occal hernia in 1 patient;herniation of efferent jejunal loop through the space between the afferent jejunal loop and the mesocolon after Billroth II gastrectomy in 6 patients,one of which had bowel necrosis,all of the 6 patients were cured after reduction of the hernia or enterectomy after and enteroanastomsis;internal henia though (ruptured) pelvic (peritoneum) after Miles operation in 4 patients;internal hernia though the space between the (descending) colon and the lateral abdominal wall after colostomy of descending colon in 6 patients;internal (hernia) caused by (adhesion) of omentum to the intestinal wall of sigmoidostomy in 1 patient;internal hernia through hiatuses caused by postoperative or post-peritonitis adhesions to the peritoneum,ovaries,urinary (bladder),uterus,(intestinal) wall or between loops of intestine in 50 patients(73.5%),including 3 cases of bowel (necrosis).All of those patients were cured after reduction of the hernias,repair of the hiatuses,release of (adhesions) or enterectomy and enteroanastomosis. Conclusions Preoperative diagnosis of internal abdominal hernia is difficult.The possibility of internal abdominal hernia should be cansidered in patients with a history of operation,and who complain of frequent abdominal pain or bowel obstruction.Early diagnosis and prompt (operation) is necessary to prevent the occurrence of bowel strangulation and bowel necrosis.
6.Roles of polymorphism in promoter region of miR-34b/c in cancer: a Meta analysis

Objective To investigate the association between the genetic variant in the promoter region of miR-34b/c and cancer risk.Methods Databases such as PubMed、EMBase、Wanfang、VIP、CNKI and so on were searched comprehensively.Based on the including and excluding criteria,literatures that were eligible were screened and data were retrieved.Meta analysis was performed by RevMan 5.1 software.Results Through searching and manually searching relevant references,a total of 9 articles with 10 independent studies were included.No significant associations were detected in C vs T,CC vs TT,CC + CT vs TT and CC vs CT + TT comparison models.However,in the CT vs TT comparison model,the result showed a significant association (Z =2.33,P =0.02).Meanwhile,subgroup analysis of hepatocellular carcinoma and colorectal cancer both showed significant associations with the polymorphism,with C vs T (OR =1.11,Z =2.10,P =0.04),CT vs TT (Z=2.40,P =0.02),CC +CT vs TT (Z=2.45,P=0.01) and CC vs TT (OR =0.66,Z=2.43,P=0.02),CC vs CT + TT (OR =0.67,Z =2.40,P =0.02) respectively.The C allele increased significantly the susceptibility of hepatocellular carcinoma,and the CC genotype reduced significantly the susceptibility of colorectal cancer.Conclusion The genetic variant in the promoter region of miR-34b/c rs4938723 T > C is significantly associated with the susceptibility of hepatocellular carcinoma and colorectal cancer.
7.Meta analysis for the association between a genetic variant in gene Dicer and cancer susceptibility

Objective To investigate the association between a genetic variant in Dicer and cancer risk .Methods Both English and Chi-nese databases were carefully searched comprehensively .Based on the including and excluding criteria ,literatures that were eligible were carefully screened and data were retrieved .Meta analysis was performed by RevMan 5 .1 software .Pooled odds ratios(OR) and 95% confi-dential interval(CI)were used for analysis .Results Ten eligible literatures were included in the meta analysis .Meta analysis showed that the mutant allele C in 3′untranslated region polymorphism loci rs1057035 of Dicer gene might reduce the cancer risk(OR=0 .92 ,95% CI=0 .87-0 .97 ,P=0 .003) .Conclusion The variant rs1057035 T>C in Dicer gene could be related to cancer susceptibility .

9.Synergistic suppressive effects of matrine and 5-fluorouracil on growth of implanted gastric cancer in nude mice

Objective To study the effects of matrine in combination with 5-fluorouracil (5-FU) on inhibiting transplanted gastric cancer (cell line SGC-7901) in the nude mice and their myelotoxicity effects. Methods The two dosages of matrine (50 mg/kg and 100 mg/kg) combined respectively with 50 mg/kg of 5-fluorouracil (5-FU) were injected intra-abdominally, and 50 mg/kg or 100 mg/kg matrine or 5-FU injection alone groups served as controls. The relative tumor volume (RTV) and tumor inhibition rate (IR) were calculated. The nude mice bone marrow was taken, the number of the nucleated cells were calculated, and bone marrow colony was cultured. 10.Mechanisms of quinolones resistance in Acinetobacter baumannii and homology analysis among the strains

Objective To explore the mechanisms of quinolones resistance in Acinetobacter baumannii and homology analysis a-mong the strains .Methods 25 strains of quinolones-resistant Acinetobacter baumannii isolated clinically were collected .Kirby-Bauer(K-B) detection was utilized to detect the sensitivity of conventional drugs ,and polymerase chain reaction (PCR) was em-ployed to detect quinolone resistance-related genes gyrA and parC which were verified by restriction enzyme digestion and sequen-cing ,repetitive extragenic palindrome(REP)-PCR was adopted to analyze the strain homology .Results Multiple resistances to 12 kinds of antibacterial agents were found among the 25 strains of Acinetobacter baumannii which were sensitive only to minocycline and amikacin ,with sensitive rates were 48 .0% and 32 .0% ,respectively ,and were all sensitive to polymyxin B [minimal inhibitory concentration(MIC)≤2 μg/mL] .gyrA and parC genes were found in the all strain .Mutation TCA→TTA(Ser→Leu) at coden 83 in gyrA gene existed in 25 strains ,mutation TCG→TTG(Ser→Leu) at coden 80 in parC gene existed in 23 strains ,mutation GAA→GGA(Glu→Gly) at coden 84 in parC gene existed in 2 strains .REP-PCR showed that the strains had high degree of homology . Conclusion Quinolone-resistant Acinetobacter baumannii has high degree of homology ,existing gyrA and parC gene mutations .
