Objective To study the improvement of hemorheology and prognosis of vertebral artery type cervical spondylosis by combination of strong Dingxuan tablet.Methods106 cases of vertebral artery type cervical spondylopathy were randomly divided into observation group and control group.Fifty-three patients in each group were treated with manipulation in the control group, and the patients in the observation group were enrolled. The blood viscosity, whole blood low shear viscosity, plasma viscosity, whole blood reduction viscosity, hematocrit, erythrocyte aggregation index, fibrinogen and so on were compared between the two groups before and after treatment.Rheological parameters of the situation, the two groups of patients for 1 year follow-up observation, compared with 1 year recurrence rate.ResultsThe total effective rate was 92.45% in the observation group and 77.36% in the control group (P<0.05).The whole blood low shear viscosity (7.01±1.52) mPa·s, the whole blood viscosity (1.18±0.28) g/L was significantly higher than that in the control group (7.88±1.65) mPa·s, (1.47±0.34) mPa·s, (4.51±0.98) g/L, respectively (P<0.05).The recurrence rate was 13.21% in the observation group and 30.19% in the control group (P<0.05).ConclusionThe combination of strong Dingxuan tablet combined with manipulation of vertebral artery type cervical spondylopathy is significant, the blood viscosity of the patients is reduced, the blood circulation function is improved, the recurrence rate after treatment is low, and it is worthy of clinical application.
4.In-vitro Antibacterial Activity of the Combination of Herba Violae and Herba Taraxaci in Different Proportions
Journal of Guangzhou University of Traditional Chinese Medicine 2004;0(06):-
【Objective】In-vitro antibacterial activity of the combination of Herba Violae(HV)and Herba Taraxaci(HT)in different proportions was compared and analyzed for the optimization of the combination proportion of Herba Violae and Herba Taraxaci and the influencing factors.【Methods】The minimum inhibitory concentration(MIC)of the combination of HV and HT was determined by agar double dilution method and the minimum bactericidal concentration(MBC)by broth dilution method, and bactericidal curve was traced.Meanwhile, the resistance of the combination of HV and HT induced by sub-inhibitory concentration was analyzed, and the influencing factors of in-vitro antibacterial activity were investigated.【Results】The combination of HV and HT in the proportion of 1∶4 had a better antibacterial activity on four kinds of gram-positive bacteria and on four kinds of gramnegative bacteria, and exerted an bactericidal effect on Escherichia coli and Klebsiella Pneumoniae(both MBC_(50)/MIC_(50) and MBC_(90)/MIC_(90) being 2).The bactericidal curve showed that 2?Log cfu/mL bacteria strain could be killed by the combination of the proportion being 1∶4 at 8~(th) hour, the effect being superior to that of other two combinations.Inoculation dosage, serum amount and pH value had no effect on in-vitro antibacterial activity.【Conclusion】The combination of HV and HT in the proportion of 1∶4 has a better antibacterial activity.
6. Identification of cytochrome b Bene of Chinese herb Testudinis Carapax et Plastrum
Chinese Pharmaceutical Journal 2012;47(3):182-185
OBJECTIVE: To establish a practical, convenient and accurate method of DNA molecular marker for the identification of Testudinis Carapax et Plastrum. METHODS: Phenol extraction method and salting-out method were used to extract mtDNA from Testudinis Carapax et Plastrum and its adulterants. Cyt b mtDNA sequences of Chinemys reevesii and other turtle species were downloaded from GenBank. Species specific PCR primers were designed according to the differential DNA fragments of Cyt b genes, and Cyt b gene segment was amplified by PCR technique and sequenced. RESULTS: The complete 16.6 kb mtDNA was extracted from all samples by the two extraction methods. The specific primers could only amplify the Cyt b gene sequence of Chinemys reevesii, and its fragment was about 319 bp. The result of sequencing indicated that the homologous similarity was 100% with Chinemys reevesii. CONCLUSION: Salting-out method is suitable for the DNA extraction from Testudinis Carapax et Plastrum. The primers are specific to Chinemys reevesii and can be used to distinguish Testudinis Carapax et Plastrum from its adulterants. Copyright 2012 by the Chinese Pharmaceutical Association.

Objective To explore the effects of fibroblast transdifferentiation for myofibroblast (MFB) in the pathogenesis of systemic sclerosis (SSc) and to explore the antifibrotic mechanism of interferon γ (IFN-γ) in SSc.Methods The fibroblasts derived from the skin lesions of SSc patients and healthy adult controls were cultured in vitro and the MFB proportion in fibroblasts was examined by qualitative and quantitative α-smooth muscle actin (α-SMA) detection.By adding IFN-γ to the culture system with several doses, the influence on fibroblast proliferation and transdifferentiation for MFB in SSc was observed with MTT and enzyme linked immunosorbent assay (ELISA) respectively.Differences in the means of two independent samples were tested by Student' t-test.The means among multiple independent samples were com-pared by ANOVA.Results The means of positive α-SMA in SSc fibroblasts were higher than those in the controls (P<0.01).With extended culture time,α-SMA levels of the two groups all increased gradually (P< 0.01 all),but there were higher α-SMA levels in SSc fibroblasts (24 h:130±19,48 h:183±21,72 h:249± 22) than those in controls (24 h:98±21,48 h:143±16,72 h:174±19) (P<0.05 all).Although fibroblast proliferation and α-SMA levels were not influenced after adding of IFN-γ 10 U/ml (P>0.05 all),but IFN-γ at concentration of 100 U/ml and 1000 U/ml could obviously repress fibroblast proliferation and α-SMA levels (P<0.05 all),and 1000 U/ml had the strongest inhibiting effect at 24,48,72 h.Conclusion The fibroblasts in the skin of SSc patients have a strong potency to transdifferentiate to MFB.Early appropviate dose of IFN-γ could repress fibroblast proliferation and transdifferentiation in SSc.

Objective To investigate relationship between different degrees of asymptomatic carotid artery stenosis and cognitive impairment. Methods The cognitive function were evaluated with the montreal cognitive assessment scale (MoCA) in 80 patients with asymptomatic carotid artery stenosis and 56 controls, the total MoCA scores and subprojects scores were compared between the patients with different degrees of ACS and controls. Results The total MoCA score, visual space and executive function, attention and delayed recall had significant difference among the mild, moderate, severe ACS patients and control group (all P < 0.05). There was no statistical significance of the total MoCA score and the scores of all subprojects between the patients of mild ACS patients and the control group (all P > 0.05). The total MoCA score and the scores of visual space and executive function , attention and delayed recall in moderate and severe ACS patients were significantly lower than the control group and mild ACS patients (P < 0.05). Furthermore, the severe ACS patients, compared with the moderate ACS patients , performed worse on the cognitive tasks which involved visual space and executive function, attention and delayed recall (P < 0.05). Conclusions Cognitive function is impaired in the patients with moderate and severe ACS,the neuropsychological characteristics are mainly impaired in visual space and executive function, attention and delayed recall.
