Purpose:To explore the expression of metastasis suppressor gene Kai1 in ovarian epithelial tumor and its significance.Methods:The expression of Kai1 protein in 108 samples of epithelial ovarian tumors and 12 of normal ovarian tissues was examined by S-P immunohistochemical staining.Results:The positive rates of Kai1 protein expression among malignant,borderline,benign and normal tumors were 26.6%,71.4%,73.3% and 83.3% respectively. The Kai1 protein expression in malignant tumors was obviously less than that in benign ones ( P 0.05). Background The study on influences of gender,age and axial length (AL) on lens thickness (LT) and anterior chamber depth (ACD) in normal population is still lack in China.Objective This study was to measure LT and ACD in normal subjects and discuss their changes with age,gender and AL.Methods AL,ACD and LT were measured on 866 eyes of 433 normal population with Lenstar LS900 in Tianjin Eye Hospital from March through December in 2013,including 390 eyes in 195 males and 476 eyes of 238 females.ACD/AL,LT/AL and (ACD+LT)/AL were calculated after measurement.The subjects were grouped to the 10-20 years group,21-30 years group,31-40 years group,41-50 years group,51-60 years group and ≥61 years group based on age,or the 22 mm≤AL<24 mm group,24 mm≤AL<26 mm group and AL≥26 mm based on AL,and the measured parameters were compared and analyzed among the groups.Oral informed consent was obtained from each subject or child contutor prior to any medical examination.Results AL,ACD and LT values were (24.41±1.37),(2.89±0.42) and (4.05±0.52) mm in the males,and those in the females were (23.83±1.31),(2.72±0.40) and (4.20±0.44) mm,showing significant differences between them (t=6.278,6.102,-4.595,all at P=0.000).No significant differences were found in ACD,LT,ACD/AL,LT/AL and (ACD+LT)/AL values between the right eyes and the left eyes (t=-0.713,0.186,-0.754,0.296,-0.168,all at P>0.05).As the increase of the AL,ACD was remarkable deepened but LT was declined,with the considerable difference among the 22 mm ≤ AL<24 mm group,24 mm ≤ AL< 26 mm group and AL ≥ 26 mm group (F =176.640,94.569,both at P =0.000).ACD value was higher,but LT value was lower in the 24 mm≤AL<26 mm group or AL≥26.00 mm group than that in the 22 mm≤AL<24 mm group (all at P<0.05).ACD value was gradually reduced,while LT value was elevated with aging,with significant differences among the different age groups (F =104.531,373.225,all at P =0.000).Significant correlations were found between age and ACD,LT or (ACD + LT)/AL,with the regression equations of ACD (mm) =-0.015 × age + 3.434 (F =-22.979,P =0.000);LT (mm) =0.024 × age + 3.122 (F =44.856,P =0.000),LT/AL =0.001 × age + 0.122 (F=37.982,P=0.000),(ACD+LT)/AL=0.001×age+0.259 (F=22.451,P=0.000).Conclusions Compared with normal females,males have thinner LT and deeper ACD.In the normal eyes,longer AL is,thinner LT is and the deeper ACD is.LT thickens and ACD deepens with aging.
3.Long-term effect of pseudomonas aeruginosa preparation on advanced colorectal cancer during surgery

Objective: This study aimed to observe the long-term effect of Pseudomonas aeruginosa preparation used in peritoneal injection of advanced colorectal cancer patients during surgery. Methods: A total of 83 colorectal cancer patients who received surgery between September 2006 and March 2008 were enrolled in this study. The patients were divided into two groups. Palliative resection and a 10 ml P. aeruginosa peritoneal injection were performed in 30 of 83 patients in the treatment group. Simple palliative resection was conducted in the other 53 patients, which comprised the control group. Both groups were then treated by regular chemotherapy and radiotherapy. Results:The follow-up visit was completed in 79 of 83 patients, with a high follow-up rate of 95.2%. No significant difference was found in the five-year overall survival time between the two groups (P=0.403). However, the five-year median survival time in the control group was only 13.9 ± 2.14 months, whereas that in the treatment group was 17.2 ± 2.12 months. Conclusion: Within a short period, peritoneal injection of P. aeruginosa during surgery could confer certain survival advantages for advanced colorectal cancer patients. However, the long-term effect of this therapy remains unknown.
4.Ocular toxicity of intravitreal injection of triamcinolone acetonide in rabbit eyes

Objective To evaluate the toxicity of intravitreal injection of triamcinolone acetonide on the cornea,lens,ciliary body and retina in rabbit eyes. Methods Thirty-two gray rabbits were divided into four groups(n=8).The rabbits were intravitreally injected with buffered saline solution(control group),4 mg triamcinolone acetonide(group B),1.3 mg triamcinolone acetonide(group C) and vehicle(group D).Intraocular pressure,scotopic and photopic electroretinogram examinations were performed before injection and at different time points after injection.Histologic and ultrastructural changes were observed 1 week,1 month and 3 months after injection. 5.Construction of eukaryotic expression vector for rat Akt1 gene and its expression in 293 cells

BACKGROUND: Akt gene plays an important role in cell survival, proliferation, metabolism, and apoptosis. Although mechanism of Akt gene still remains unclear, it, a major element of survival signal, has aroused much attention in the domain of molecular biological research. OBJECTIVE: To construct eukaryotic expression vector pDC316-Akt1 for Aktl gene and to observe the expression in 293 cells. DESIGN, TIME AND SETTING: An observational study was performed at Molecular Biological Laboratory of Academy of Military Medical Sciences of Chinese PLA between September and December 2006. MATERIALS: pCD316 plasmid was provided by Benyuan Zhengyang Company; DH5α and 293 cells were reserved in our laboratory. METHODS: Aktl DNA segment was cloned from total RNA of hepatic tissue using RT-PCR and inserted into eukaryotic expressing vector pDC316 between EcoRI and Hind Ⅲ sites after sequencing. And then, 293 cells were transfected with the recombinant by liposome. MAIN OUTCOME MEASURES: Aktl gene content in transfected 293 cells using Westem blotting. RESULTS: Sequencing test indicated that Aktl amino acid sequence was in accordance with wild Aktl sequence. Akt-pDC316 was transcripted in 293 cells, and Akt1 protein with relative molecular weight of 55 000 was highly expressed in 293 cells when assayed using Western blotting. CONCLUSION: Eukaryotic expression vector for Aktl gene highly expresses in 293 cells.
6.Influence of hypoxia-inducible factor-1α on the expression of neutrophil CD18 and leukocyte adhesion in diabetic rats

Objective To observe the influence of the expression of CD18 on the neutrophile and the leukocyte adhesion to retinal vascular endothelium by hypoxia-inducible factor-1 alpha (HIF-1α) in early diabetic retinopathy rats. Methods Male Sprague-Dawley rats received intraperitoneal injection of streptozotocin to induce diabetes model. 18 diabetic rats were divided into 3 groups randomly after 2 months of diabetes induction, including diabetic group (group B), HIF-1α anti-sense oligonucleotides (ASODN) injection group (group C) and HIF-1α sense oligonueleotides (SODN) injection group (group D), the age and weigh matched health rats were chosen as control group (group A), with 6 rats in each group. Then group A and B rats received 5 % glucose solution caudalis veins injection, group C and group D rats received HIF-1α ASODN and HIF-1α SODN caudalis veins injection, respectively(0.25 mg/kg). The level of CD18 on the neutrophil isolated from the peripheral blood was measured by flow cytometry. Retinal leukostasis was quantified with acridine orange leukocyte fluorography. Results The percentage of CD 18 positive neutrophil cell was (44.93±3. 60)% in group B, (18.66±1.52)% in group A, (31.66±4.72)% in group C,(51.00±5.66)% in group D. Compared with each other groups,the differences are statistically significant (F= 42. 46, P<0.001). The number of positive staining cells of retinal leukocyte was (46.16±10.68)in group A, (133.83±20.43)in group B, (99.83±9.28)in group C, (121.33±10. 23) in group C. Compared group B with group C,the number of positive staining cells raised about 2.89 times;compared group B with group C and D,the differences are statistically significant (P= 0.12, 95% confidence interval -3.69~28. 69 ). Conclusions In vivo, HIF-1α can decreased the expression of CD18 on neutrophils from diabetic rats' peripheral blood and the collection of retinal leukos- tasis in the diabetic animals. HIF-1α may serve as a therapeutic target for the treatment and/or prevention of early diabetic retinopathy.

Objective To investigate the healing mechanism of the rabbit's selerotomy sites undergoing the transconjunctival su- tureless vitrectomy(TSV),and to compare the wound healing effect of the different surgery procedures.Design Experimental study. Participants New Zealand white rabbits.Methods Thirty-two New Zealand white rabbits were divided into four groups:the core vitrec- tomy and fluid-air exchange (Group A),the core vitrectomy (Group B),the non-core vitrectomy and fluid-fir exchange (Group C) and the non-core vitrectomy(Group D).Intraocular pressure(IOP)was measured on the day preoperatively and the day 1,3,5,7,14 postopera- tively with Tonopen tonometers.Sclerotomy sites were investigated with the ultrasound biomicroscope(UBM)on the day 1,5 and 9 postoperatively,and the internal and the external diameter of the wound was estimated on the day 1 postoperatively.The pathological sections of the sclerotomy sites on the day 3,5 and 9 postoperatively were observed under the light microscope.Main Outcome Mea- sures IOP,the internal and external diameter of the wound.Results The IOP of Group A was obviously higher than the other groups on the day 1 postoperatively (P
8.Screening of CYP21 gene P459H mutation by PCR-ACRS

AIM: To investigate the frequency of P459H(CCC→CAC) in exon 10 of CYP21 gene among normal population.METHODS: The exons 3-10 of CYP21 gene were amplified with polymerase chain reaction(PCR).The PCR round products were digested by restriction enzyme Pst I to confirm that CYP21 gene was specifically amplified.PCR-based amplification-created restriction site(PCR-ACRS) was performed using the first round PCR products as template.After the second PCR products were digested by restriction enzyme Fsp I,10% polyacrylamide gel electrophoresis was used to screen the frequency of P459H in exon 10.RESULTS: The codon 459 in exon 10 of CYP21 gene was all CCC among 100 normal cases tested.CONCLUSION: P459H(CCC →CAC) of CYP21 gene might be a novel point mutation causing CAH.Furthermore,PCR-ACRS was a fast and safe method for gene mutation screening.
