1.Research status of COX-2 and its inhibitor in breast cancer

COX-2 is the rate-limiting enzyme in prostaglandin biosynthesis,which may promote the human tumorigenesis and tumor development through a variety of pathogenic mechanisms.The over expression of COX-2 in breast cancer is significantly correlated with the clinicopathological pammeters and prognosis.COX-2 inhibitors play anti-tumor effects through different mechanisms.Recent studies indicate that COX-2 inhibitors are effective both in breast cancer chemoprevention and treatment of breast cancer combined with cytotoxic drugs and aromatase inaetivators,and therefore may provide a potential new therapy of breast cancer prevention and treatment.
2.Complications of acute Stanford type B aortic dissection and prognosis

ObjectiveTo find the death predictors of acute Stanford type B aortic dissection patients by studying relationships between complications and prognosis. MethodsEighty-four patients with acute Stanford type B aortic dissection were included, the complications of pericardial fluid, pleural effussion,conscious disturbance ,oliguria, gastrointestinal tract bleeding and the level of creatine kinase were analyzed.There were 73 men and 11 women with age 30 - 81 (52.0±12.7) years old. ResultsThe overall mortality was 9.5%(8/84). There were 33.3% (1/3) died with pericardial fluid, 35.7% (5/14) died with pleural effussion, 50.0% (2/4) died with gastrointestinal tract bleeding, all patients died with oliguria and conscious disturbance, 36.4%(4/11 ) patients died with elevated creatine kinase. The patients suffered from any of the complications had higher mortality than those free from complications. There was significant difference between patients suffering from pleural effussion, gastrointestinal tract bleeding,oliguria,conscious disturbance and high level of creatine kinase and who not suffering from the complications respectively in statistically (P < 0.05 ). Logistic regression showed that pleural effussion was independent death predictor.ConclusionsThe mortality increases when acute Stanford type B aortic dissection patients combined with pleural effussion, gastrointestinal tract bleeding,oliguria, cons cious disturbance or high level of creatine kinase. Pleural effussion is independent death predictor.

Cycloxygenase-2 plays an important role in many human tumorigenesis, and is overexpressod in breast cancer. Therefore, it is necessary to investigate the relationship between COX-2 and breast cancer, and to develop the desirable COX-2 inhibitor. These can privide a new antitumor method.
5.COX-2 and its inhibitors in lung cancer drug resistance

COX-2 is the inducible form of cyclooxygenase,which is overexpressed in non-small cell lung cancer,especially in the adenocarcinoma,and is involved in the production of lung cancer drug resistance, reducing the sensitivities of tumor cells to chemotherapy drugs. COX-2 inhibitors have the effects of antitumor and prevention of tumor formation. Therefore,in order to reverse the drug resistance of tumor cells,improve the sur-vival rate and the prognosis of patients with tumor,the application of COX-2 inhibitors as adjuvant chemotherapy and combined with chemotherapy drugs has become a new direction in the treatment of lung cancer.
6.A STUDY ON THE HUMAN INTRACEREBRAL SEGMENTS OF THE CENTRAL BRANCH OF THE MIDDLE CEREBRAL ARTERY

The origin, diameter, arrangement pattern, distribution and microvascular architecture of the intracerebral segments(IS) of the central branches of the middle cerebral artery(MCA) were examined and pictured in 27(10 infants, 5 newborns, 12 fetuses) unfixed brains under stereoscopic microscope and scanning electron microscope. 1. The extracerebral segments(ES) of the central branches run toward the anterior perforated substance in S-shaped curves. The IS arise from ES or their collateral branches. They form the fan-shaped appearance in rostromedial and caudolateral state after entering brain. 2. The IS give rise collateral branches in right or acute angles, and they are of two kinds: the longitudinal and the transverse branches. The mean diameter of the IS in original site is 0.21?0.18mm for the medial group; 0.36?1 0.21mm for the lateral group. 3. The medial group mainly distribute in the rostal part of the internal capsule and basal nucleus; the lateral group mainly distribute in the caudal part. 4. Some circular impressions of the precaillary sphincters were observed at the beginning of precapillary arteriole and capillary, converging point of tributary branches of venule. The relationship between the IS of the central branches and the hemodynamics was discussed.

OBJECTIVE:To investigate the effect of extract of Ginkgo biloba(EGb) on levels of Endothelin-1(ET-1) and transforming growth factor ?1(TGF-?1) in renal tissue of diabetic rats.METHODS:Twenty-four male Sprague-Dawley rats were randomly divided into three groups:normal control group,diabetic model group and EGb-treated group.The levels of blood glucose,insulin,total cholesterol(TC),total triglycerides(TG),creatinine clearance(Ccr),urinary albumin excretion(UAE),and urinary ?2-MG were measured after 8-week corresponding treatment.Expression of SET-1,UET-1,and RET-1 was examined by radioimmunoassay technique.Expression of STGF-?1 and UTGF-?1 in serum and urine was examined by enzyme linked immunosorbent assay(ELISA).RESULTS:The concentrations of blood glucose,blood insulin,TC and TG increased significantly in diabetic group,which were down-regulated by EGb.Levels of ET-1 and TGF-?1.in both blood and urine and the ET-1 level in renal tissues were significantly higher in diabetic model group than in normal control group(P

9.Impact of mice apoptosis antigen of lung cancer induced by paclitaxel on function of dendritic cells

Objective To investigate the in vitro effect of low dosage of paclitaxel on normal murine bone marrow-derived dendritic cells(mDCs)and its role in reactivating tumor-pulsed DCs. Methods The concentration of paclitaxel which could induce 30% apoptosis of 3LL cell lines was figured out.mDCs were generated from murine bone marrow precursors.Cell culture insert system was used and four groups were divided as following: mDC,mDC+3LL,mDC+ low dose of paclitaxel,and mDC+3LL with 30% apoptosis induced by low dose of paclitaxel.The phenotypes,chemoattractive function to MIP1? and MIP-3?,and viability in activating allogeneic T cell proliferation of DCs in the four groups were analysed. Results Paclitaxel of 50 nmol/L could induce 30% apoptosis of 3LL,and had protective effects on DCs.It could stimulate the maturation of mDCs by up-regulating the phenotypes of CD11cCD80,CD11cCD86,CD11cCD40 and CD11cCDIab,and could enhance the chemoattractive function to chemokine MIP-3?.Compared with those cocultured with 3LL,DCs pulsed with apoptosis antigen of 3LL cell which was induced by 50 nmol/L paclitaxl up-regulated the phenotype of CD11cCD40,enhanced the the chemoattractive function to MIP1? and MIP-3?,and activated the proliferation of T cells. Conclusion Paclitaxel of 50 nmol/L can stimulate the maturation of DC,and can partially recover the phenotype and function of tumor-pulsed DC.
10.Analysis of treatment and prognosis of 352 lung cancer patients with brain metastases

Objective To analyze the treatment and prognosis of lung cancer patients with brain metastases.Methods The clinical data of 352 lung cancer patients with brain metastases were retrospectively reviewed.According to the treatment modalities,patients were divided into palliative therapy group(n=28),simple whole brain radiotherapy(WBRT)or chemotherapy group(n=49)and comprehensive treatment group(n=275).Comprehensive treatment group was subdivided into WBRT plus chemotherapy group(n=192),stereotactic radiosurgery(?knife)plus chemotherapy/WBRT group(n=72,n=16 for?knife plus chemotherapy and n=56 for?knife plus WBRT and chemotherapy)and neurosurgical resection plus chemotherapy/WBRT group(n=11).In comprehensive treatment group,111 patients received chemotherapy≤3 cycles,and the other 164≥4 cycles.Survival curves of each group were drawn respectively,and both survival time and survival rates were compared among groups.Results The median survival time of palliative therapy group,simple WBRT or chemotherapy group,WBRT plus chemotherapy group,?knife plus chemotherapy/WBRT group and neurosurgical resection plus chemotherapy/WBRT group was 1.7,3.2,9.0,11.6 and 17.1 months,respectively.It was revealed by survival analysis that WBRT plus chemotherapy group was better than simple WBRT or chemotherapy group (P=0.0000),?knife plus chemotherapy/WBRT group was better than simple WBRT or chemotherapy group(P=0.0000),and neurosurgical resection plus chemotherapy/WBRT group was better than simple WBRT or chemotherapy group and WBRT plus chemotherapy group(P=0.0001,P=0.0229).There was no significant difference in survival rates between neurosurgical resection plus chemotherapy/WBRT group and?knife plus chemotherapy/WBRT group(P=0.2543),and there was no significant difference in survival rates between those with?knife plus chemotherapy and those with?knife plus WBRT and chemotherapy(P=0.3804).In comprehensive treatment group,the survival rates of those with chemotherapy≥4 cycles was significantly higher than that of those with chemotherapy≤3 cycles(P=0.0000). Conclusion Both WBRT plus chemotherapy and?knife plus chemotherapy and WBRT are effective modalities for the treatment of lung cancer patients with brain metastases,and the latter has the tendency to gain more survival benefit.There is no significant difference in the survival time between patients receiving?knife with WBRT and those without.It is proper for the patients to have no less than 4 cycles of chemotherapy.
