1.Research advances in early-onset epileptic encephalopathy

Early-onset epileptic encephalopathy (EEE) represents a group of devastating epileptic disorders that appear in neonatal or infantile period of life,characterized as pharmacoresistant generalized or focal seizures,severe electroencephalography (EEG) abnormalities,mental retardation and behavioral impairments.The interictal epileptic discharges are age-dependent and closely related to cognitive deterioration.EEE includes five epileptic syndromes,which are early myoclonic encephalopathy,ohtahara syndrome,and malignant migrating partial seizures in infancy,West syndrome and Dravet syndrome.The etiologies of EEE are highly heterogeneous,and most of them remain unknown.In many cases with EEE,seizures are resistant to treatment including anti-epileptic drugs and other methods.The prognosis for EEE is poor,and most of the children have severe mental retardation,some even are under the risk of sudden death.
3.Association of the single nucleotide polymorphism and the serum contents of interleukine-18 with ulcerative colitis

Objective To estimate the association of the single nucleotide polymorphism (SNP) and the serum concentration of interleukin (IL)-18 with ulcerative colitis (UC) . Methods Samples collected from 50 patients with UC and 128 healthy controls were detected for SNPs of IL-18 gene including rs187238, rs5744228, rs360718 and rs360717 using polymerase chain reaction (PCR) and direct sequencing. Meanwhile, the serum level of IL-18 was measured. ResultsSerum concentration of IL-18 was higher in patients with UC than in healthy controls [(3029.9±111.7) pg/ml vs ( 133.2±39.4) pg/ml,(P<0.01)], rs187238 C, rs360718 C and rs360717 A were significantly related to UC (X~2=9.26, P<0.01). There was significant difference in frequencies of rs187238 C, rs360718 C and rs360717 A between patients with UC and healthy controls (X~2=9.26, P<0. 01). Serum concentration of IL-18 was higher in UC patients with C allele than in healthy controls (P<0.05).Conclusions There is correlation between SNP and serum level of IL-18 , which may involve in susceptibility to UC.

Coronary heart disease (CHD) is a major disease greatly harmful to the health of human beings. The incidence and case fatality rate of female CHD show an increasing tendency due to lack of enough attention. Through analyzing the epidemiology, risk factors, and clinical characteristics of female CHD, we emphasized that it is necessary to pay enough attention to the particularity of female CHD, and put forward the thinking way of diagnosis and treatment of integrative medicine as "integrating Chinese medicine and Western medicine by mutual complement of advantages; combining syndrome typing and disease identification by grasping laws; treating physically and mentally by a wholism concept; spreading health education, preventing and treating comprehensively".

6.Therapeutic effects of modified Bushen Huoxue granules on ovariecto-mized osteoporosis rats

AIM: To observe the therapeutic effect of modified Bushen Huoxue granules (MBHG) on ovariec-tomized osteoporosis rats.METHODS: Female SD rats were randomly given sham operation (n =10) and ovariectomy, and then the model rats were further randomly divided into model group, MBHG treatment groups at doses of 200 mg/kg, 100 mg/kg and 50 mg/kg respectively, and positive control (estradiol valerate) group, with 10 rats in each group by intra-gastric administration for 12 weeks.The morphology, area, thickness, spacing and area percentage of trabecular bone in the rats were observed.The serum levels of calcium (Ca), phosphorus (P) and alkaline phosphatase (ALP) were mea-sured by automatic analyzer.Bone mineral density (BMD) was analyzed.Serum estradiol (E2 ), osteocalcin (BGP), os-teoprotegerin (OPG), receptor activator of nuclear factor-κB (RANK) and receptor activator of nuclear factor κB ligand (RANKL) levels were detected by ELISA.RESULTS: Compared with model group, trabecular bone significantly widened in all treatment groups with large number, and net-like structure restored partially.The thickness, area and area percenta-ges of trabecular bone in treatments groups were higher than those in model group,and trabecular spacing was less than that in model group (P <0.05).The serum Ca, P, E2 and OPG, and femoral BMD were significantly higher in treatment groups than those in model group, and the levels of ALP, BGP, RANK and RANKL were significantly lower than those in model group (P <0.01).CONCLUSION: MBHG has a significant therapeutic effect on ovariectomized osteoporosis rats. The mechanism may be related to the regulation of OPG and inhibition of RANKL secretion.
7. Role of sodium pump Na+-K+-ATPase in hypoxic injury of cortical neurons in rats

Objective: To investigate the effect of hypoxia on the activity of Na+-K+-ATPase(Na pump) and to understand the distinct functions of high- and low-affinity Na pump during hypoxia of cortical neurons. Methods: Hypoxic condition was mimicked by perfusing cortical slices or culturing cortical neurons with low oxygen solution. Sodium pump current and membrane current of neurons from cortical slices were measured by patch-clamp technique in the whole cell mode and the intracellular Ca2+ concentration ([Ca2+]i) in cultured cortical neurons was examined by video based motion edge detection system. Changes of Na pump current induced by hypoxia were also examined 4 min after hypoxia in the neurons of cortical slices. Changes of membrane current and [Ca2+]i were determined 0, 2, 4, 6, 8 and 10 min after hypoxia with or without Ouabain (Oua, inhibitor of sodium pump). Results: Total sodium pump current were significantly decreased 4 min after hypoxia ([0.265±0.068] pA/pF vs [0.160±0.046] pA/pF, P<0.01). Membrane current and [Ca2+]i were increased in a time-dependent manner 10 min after hypoxia (r=0.9803 and r=0.9734, P<0.01). The effect of hypoxia on membrane current was abolished by tetrodotoxin (TTX, 1 μmol/L, a blocker of sodium channel). Oua at 10 2mmol/L significantly promoted the hypoxia-induced increase of membrane current and [Ca2+]i through inhibiting low-affinity sodium pump (P<0.05 or 0.01), and Oua at 10 nmol/L significantly depressed the increase of membrane current and [Ca2+]i through inhibiting high-affinity sodium pump (P<0.05 or 0.01). Conclusion: Change of sodium pump activity is involved in hypoxic injury in rat cortical neurons. High-affinity sodium pump is related to the protection of hypoxic injury and low-affinity sodium pump is related to the hypoxic injury.
8.Application of point-of-care testing in blood purification and extracorporeal membrane oxygenation therapy

Point-of-care testing(POCT)is a kind of testing technology that uses portable testing instruments to test and quickly report the detection results at the bedside of testing object.Critically ill children need bedside examination, diagnosis and treatment in time according to the changes of their condition because of their critical condition and rapid progress.Therefore, the application of POCT in pediatric intensive care medicine has developed rapidly.Blood purification technology and extracorporeal membrane oxygenation(ECMO)are important treatment methods for critically ill children, while POCT is a necessary technology and an important link in the management of extracorporeal circulation.In this review, the application of POCT in blood purification and ECMO therapy is summarized.
