1.Protective action of grape seed proanthocyanidins combined With casein on radiation damage of rats

BACKGROUND: A great deal of free radicals can be induced,which results in peroxidation of the whole body. The antioxidant activity and the abilities of scavenging free radicals of proanthocyanidins(GSP) have been well documented,but related researches about the protective role in radiation damage were less reported.OBJECTIVE: To investigate the protective role of GSP combined with casein in 60Co-y radiation-induced peroxidative damage.DESIGN: Observing and comparative study based on animal model.SETTING: Nutrition Department of Daping Hospital Affiliated to the Third Military Medical College of Chinese PLA.MATERIALS: This study was carried out in the Third Institution of Field Operation Surgery, the Third Military Medical College of Chinese PLA.Eighty healthy Wistar rats,clearage,were adopted,ith weight from 180 to 220 g,half male and half female were adopted.METHODS: Rats were divided into four groups: normal control group,irradiation group,GSP group and GSP + casine group with 20 rats each. Rats in GSP group were fed routinely and supplemented with 1% GSP for 10days,which was replaced by 1% GSP and 5% casein. All rats except those of control group were exposed to whole-body 60Co-g amma-radiation at a dose of 7 Gy. Seven days later,6 survival rats randomly selected from each group were killed.MAIN OUTCOME MEASURES: The level of serum malondialdehyde (MDA), activities of serum superoxide dismutase(SOD),spleen index and the number of peripheral white blood cell(WBC).RESULTS: Serum MDA was obviously increased,accompanied by obviously attenuated SOD activity after irradiation; the level of MDA in both GSP group and combined group was found lower than that in irradiation group with statistical significance between them(P<0.01), the activity of SOD was significantly higher than that of irradiation group(P<0.01 ) . Markedly atrophic pathological change in spleen could be observed in irradiated rats,and meanwhile spleen index declined with the number of WBC notably decreased. But spleen index in both GSP group and combining group was markedly higher than irradiation group(P<0.05),and the same comparing result could also be found in the number of WBC(P<0.01 ).CONCLUSION: Peroxidative impairments could be obviously attenuated,and decrement of WBC could also be improved by single GSP or alternatively GSP combined with casein,with the latter showing greater effects in respect of improving immune function following irradiation,cellular chemical imbalance,and consequently the life quality.

[Result]Professor Chang considers the adolescaria functional metrorrhagia connected with stagnation of liver Qi,insufficiency of kidney and deficiency of spleen Qi,weakness,stagnation,dampness.Treatment must address both the symptoms and root causes in accordance with seasonal conditions .She regulating menstrual cycle attaches great importance to the liver , spleen and kidney,and uses the auricular points,couplet medicines.The prescription rule is convenient and specialized.The recipe case embodies the evidence-based medicines, according to the menstrual period, the late, and the inter-phase and premenstrual period of drug treatment of yin and yang change. Professor Chang's ideas and characteristics curative effect are outstanding. [Conclusion]Professor Chang thinks the adolescaria functional metrorrhagia is complicated,then perfect pragmatic value is worthy of study and understanding.
3.Alteration of biological and immunological properties of aortic homografts after de-endothelialization and decellularization

BACKGROUND:Aortic root homograft has been widely used, but it stil has a high incidence of calcification and degeneration after operation.
OBJECTIVE:To investigate the alteration of biological and immunological properties of rabbit aortic root homograft treated with de-endothelialization and decellularization methods, and to find a better decellularization method.
METHODS:Aortic root homografts obtained from rabbits were randomly divided into fresh group, de-endothelialization group, and decellularization group. The morphology of homografts was evaluated by hematoxylin-eosin, Masson and VG stainings. The biological properties such as thickness, tensile strength, broken extension rate, and heat shrinkage temperature were detected. The immunological features (inflammatory cellinfiltration, calcium content and CD152 expression) were determined with immunohistochemistry and flame atomic absorption spectrometry.
RESULTS AND CONCLUSION:As demonstrated in morphological observation, protocols of de-endothelialization and decellularization methods we used attained our desired aim of decellularization perfectly. Tensile strength and broken extension rate of decellularization group were decreased significantly compared with fresh group (P<0.01), while the difference between de-endothelialization group and fresh group was not significant (P>0.05). Hematoxylin-eosin staining showed that, inflammatory reaction was the most apparent in fresh group, showing a large amount of inflammatory cells infiltration;compared with fresh group, inflammatory reaction was significantly attenuated in de-endothelialization group, the number of inflammatory cells was significantly decreased. The inflammatory reaction was the lowest in the decellularization group, only inflammatory cells infiltrated the tissue edge. Immunohistochemistry showed that, CD152 expression was significantly increased in de-endothelialization and decellularization groups compared with fresh group (P<0.05). Calcium content in de-endothelialization and decellularization groups was significantly decreased compared with fresh group (P<0.05). It could be concluded that the immunogenicity of de-endothelialized and decellularized aortic homografts is decreased significantly, and the de-endothelialization method could better maintain the tensile property.

5.Effects of Grape Seed Proanthocyanidin and Casein Peptide on the liver function and protein nutritional status in rats with radiation injury

Objective: To investigate the effects of grape seed proanthocyanidin(GSP) and casein peptide on the liver function and protein nutritional status in rays irradiated rats. Methods: Adult wistar rats were randomized into four groups with 20 rats in each group including normal control, radiation control, GSP and GSP+casein peptide combined groups. The GSP group was fed with basic diet containing 1% GSP, the combined group was fed with basic diet containing 1% GSP plus 5% casein peptide. Animals except normal control were irradiated by 60 Co? rays(7Gy) 10 days later. 6 survivors were randomly selected from each group and killed on the 7 th d after radiation. The levels of the total protein (TP) and the albumin (ALB), the activities of the glutamate pyruvate transaminase(GPT) and the glutamate oxaloacetale transaminase(GOT) in serum were measured. The change of the body weight of the animals was observed during the experiment time. Results: After radiation the body weight of the rats dropped sharply. Objective To investigate the intracellular mechanisms by which 3,6-dihydroxyflavone inducing apoptosis of HL-60 cells.Methods After the HL-60 cells were treated with 3,6-dihydroxyflavone at the concentration of 5.10,20 or 40 mol/L,the cells then were inoculated under the selected optimal concentration.The survival rate of HL-60 cells was analyzed using a haemocytometer with standard trypan blue dye exclusion and a CASY cell counter and analyzer.The contents of malondialdehyde(MDA)and the activities of superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)in the cells were measured by chemochromatometry.The expression of phosphorylation of ERK,JNK and p38MAPK were examined by Western blotting.Results The survival rate of HL-60 cells was significantly reduced in a dose-and time-dependent manner after 3,6-dihydroxyflavone treatment.When the dose was over 10 mol/L or the time was longer than 12 h under the concentration of 20 mol/L,significantly decreased survival rate was observed(P
8.Expression of COX-2,Ki-67 and VEGF in bladder transitional epithelial cancers and their significance

Objective To study COX-2 expression in bladder transitional cell carcinoma in order to explore the relationship between the immunohistochemical staining and biological behavior of bladder cancer. Methods The expression of COX-2,Ki-67,and VEGF was examined in 50 cases of bladder transitional cell carcinoma and 10 normal bladder mucosa by means of immunohistochemical technique.Results The expression of COX-2 in BTCCs was higher than that in normal bladder mucosa.The positive rate of COX-2 was 80% in BTCCs while it was not detected in normal bladder mucosa.COX-2 was highly positively related to the grade,stage and prognosis of tumors.The expression of COX-2 in poorly differentiated,invasive or re- curring BTCCs was higher than that in well differentiated superficial or nonrecurring ones.But there was not significant difference between the solitary tumour and multiple ones.There were positive correlation both be- tween COX-2 and Ki-67 and between COX-2 and VEGF.Conclusion The high expression of COX-2 may benefit increase in bladder cancer cell proliferative activity.VEGF expression in COX-2 positive tumor was significantly higher than that in COX-2 negative tumor.
9.Cone-socket hematoma drainage in ward before operation

Objective To explore the treatment to craniocerebral injury.Methods 6 patients suffered from serious, developing head injury with epidural and subdural hematoma were processed by cone socket hematoma drainage in ward before hematoma was eliminated.Results Hematoma drainage can be performed within 20 min after hospitalization. No case died and no infection occured. The score of Glasgow Outcome Scale were Ⅴ(2 cases), Ⅳ(2 cases),Ⅲ(1 case) and Ⅱ(1 case) 3 months after operation.Conclusion Cone socket hematoma drainage in ward is a good method to treat craniocerebral injury.
