Objective To explore the establishment methods of animal models of intestinal multidrug-resistant bacterial (Pseudomonas aeruginosa, PA) infection, and to provide a good model for experimental research and treatment for infection caused by drug-resistant bacteria .Methods Male BALB/c mice were divided into four groups:Normal con-trol group, MDR-PA group, MDR-PA +antibiotics group, and MDR-PA +fasting group.They were given saline, PA suspension, PA suspension after free drinking with ceftriaxone water and PA suspension after fasting , respectively , by ga-vage.Colon tissues were collected for pathological examination and inflammation scoring .The expressions of TNF-αand INF-γin the colon tissue was assessed at the end of the experiment .Results Pathology showed different degrees of inflam-matory changes in the colon tissues .Compared with the normal control group , the expressions of TNF-αand INF-γhad sig-nificant differences with that of experimental groups .Conclusions All the three methods used in this experiment can be successfully used to establish mouse models with intestinal infection caused by multidrug-resistant Pseudomonas aeruginosa, and may be chosen for experimental studies according to different research purposes .

Objective To investigate the relationship between the calcium ion and the effects of ECA on the contraction of the smooth muscles of Oncomelania foot, and explore the mechanism of ECA in decreasing the snail climbing adhesion and increasing snail-killing. Method The in vitro experiment was used to observe the effects of ECA on contraction activities of foot smooth muscles of Oncomelania. Results ECA 5. 0, 10. 0, 20. 0 mg/L. enhanced the resting tension, decreased the contraction range and increased the contraction frequency of the foot smooth muscles of Oncomelania in Ringer's solution with or without Ca2+ , with a characteristic of the dose-dependent, but stronger in solution with Ca2+ than that in solution without Ca2+. Conclusions ECA can enhance the resting tension, decrease the contraction range and increase the contraction frequency of the foot smooth muscles of Oncomelania. This suggested that ECA could be related to increasing the intracellular Ca2+ release and influx of the extracellular Ca2+. This could provide an explanation why ECA decreases the rate of Oncomelania climbing adhesion and enhances snail-killing.

OBJECTIVE: To observe the effects and mechanisms of sodium tanshinone II A sulfonate (STS) on angiotensin II (Ang II)-induced cardiomyocyte oxidative stress. METHODS: In the primary culture of neonatal rat cardiomyocytes, the content of reactive oxygen species (ROS) was measured by 2, 7-dichlorofluorescein diacetate (DCFH-DA). 8-hydroxydeoxyguanosine level in the supernatant was measured by ELISA. As indexes of cardiomyocyte oxidative stress, the cellular contents of MDA and SOD, cell vialibity and LDH release were measured. NADPH oxidase (NOX) activity was measured by chromatometry. The expressions of p47phox was assessed using Western blot. RESULTS: STS can decrease Ang II-induced elevations of ROS level and oxidative stress, and inhibit the expression of p47phox and NOX activity. CONCLUSION: The inhibitory effects of STS on Ang II-induced cardiomyocyte oxidative stress may be associated with depressing NOX signaling pathway via down regulation of p47phox expression. Using 54 traditional Chinese medicine (TCM) listed companies in China as a research sample, panel data of 54 companies from 2012 to 2016 was used, the 11 financial indicators was selected to build a Chinese medicine listed companies growth evaluation index system by the factor analysis method. On this basis, Topsis method was used to calculate the comprehensive factor of TCM listed companies and the optimal solution of the closeness degree, in order to analyze the growth of TCM listed companies. The results showed that about 79.63% of TCM listed companies’ comprehensive factor score and the optimal solution of the close degree near the distribution from 0.4 to 0.7, indicating that the growth of TCM listed companies have a greater room for improvement; Only 3.7% of TCM listed companies comprehensive factor score and the optimal solution near the distribution from 0.8 to 0.9. China's TCM listed companies did not play its leading role. Finally, this paper puts forward relevant policy suggestions for promoting the healthy development of TCM companies.

An ECG waves separation technique based on math ematical morphology is suggested and discussed in this paper. Without detection of QRS complexes, the QRS complexes can be eliminated by series morphological o perations. Then the start points and end points of P-wave and T-wave can be de cided and the waves of ECG are separated qualitatively and quantitatively. The r esults of qualitative separations are rather satisfied and the SDs of quantitative separation results are not very large. Also, ECG signals are filtered and bas eline is normalized by morphological method.

Objective To investigate the role of suppressor gene p16 in the process of differential regulation of retinoic acid (RA) on the A549 lung cancer cells.Methods Tumor suppressor gene p16 was transferred into A549 cells and the cells were treated with all-trans retinoic acid (ATR) at the dosage of 5×10-6 mol/L for 4 d. After that, the proliferation and differentiation of A549 cells were examined by growth curve and cytometry analysis, the change of lung lineage-specific marker MUC1 was tested by immunohistochemical staining. Meanwhile, Western blot was used to observe the change of p16 protein expression in A549 cells treated with ATRA.Results ATRA could obviously inhibit the growth and induce the differentiation of A549 Cells that were transferred with p16 gene. There were more cells arrested in G1/G0 phase and the expression of MUG1 was markedly down-regulated than in control cells. The expression of p16 protein was up-regulated in A549 cells treated with ATRA.Conclusion Suppressor gene p16 could enhance the effects of RA and proliferated suppression and differential induction of A549 cells.

Rabbit osteoblasts were transfected with pCD-hVEGF165 plasmid by lipofectin mediated gene transfer. The transient expressive results were detected by immunohistochemical method. It was observed that the expression of human VEGF gene was detected 72 h after transfecting distinctly.

In the development of chemo-immunotherapy, many efforts have been focusing on designing suitable carriers to realize the co-delivery of chemotherapeutic and immunotherapeutic with different physicochemical properties and mechanisms of action. Besides, rapid drug release at the tumor site with minimal drug degradation is also essential to facilitate the antitumor effect in a short time. Here, we reported a cancer cell membrane-coated pH-responsive nanogel (NG@M) to co-deliver chemotherapeutic paclitaxel (PTX) and immunotherapeutic agent interleukin-2 (IL-2) under mild conditions for combinational treatment of triple-negative breast cancer. In the designed nanogels, the synthetic copolymer PDEA-co-HP-β-cyclodextrin-co-Pluronic F127 and charge reversible polymer dimethylmaleic anhydride-modified polyethyleneimine endowed nanogels with excellent drug-loading capacity and rapid responsive drug-releasing behavior under acidic tumor microenvironment. Benefited from tumor homologous targeting capacity, NG@M exhibited 4.59-fold higher accumulation at the homologous tumor site than heterologous cancer cell membrane-coated NG. Rapidly released PTX and IL-2 enhanced the maturation of dendritic cells and quickly activated the antitumor immune response in situ, followed by prompted infiltration of immune effector cells. By the combined chemo-immunotherapy, enhanced antitumor effect and efficient pulmonary metastasis inhibition were achieved with a prolonged median survival rate (39 days).

To evaluate the value of detection of DNA aneuploidy in exfoliated airway epithelia cells of sputum specimens by the automated image cytometry for the identification of lung cancer, 100 patients were divided into patient group (50 patients with lung cancer) and control group (30 patients with tuberculosis and 20 healthy people). Sputum was obtained for the quantitative analysis of DNA content of exfoliated airway epithelial cells with the automated image cytometry, together with the examinations of brush cytology and conventional sputum cytology. Our results showed that DNA aneuploidy (DI>2.5 or 5c) was found in 20 out of 50 sputum samples of lung cancer, 1 out of 30 sputum samples from tuberculosis patients, and none of 20 sputum samples from healthy people. The positive rates of conventional sputum cytology and brush cytology were 16% and 32%, which was lower than that of DNA aneuploidy detection by the automated image cytometry (P<0.01, P>0.05). Our study showed-that automated image cytometry, which uses DNA aneuploidy as a marker for tumor, can detect the malignant cells in sputum samples of lung cancer and it is a sensitive and specific method serving as a complement for the diagnosis of lung cancer.
