1.Differentiation of porcine amniotic fluid stem cells into the beating cardiomyocytes.

The aim of this research is to find an effective cardiomyocyte-induced method derived from porcine amniotic fluid stem cells (pAFS). For cardiac differentiation, the cells were formed embryoid bodies (EBs) firstly, then cultured in induced-medium including 5-azacytidine (5-aza) and vitamin C (Vc). We detected the specific markers of cardiomyocyte by immunocytochemistry, RT-PCR and transmission electron microscope. The results showed that some embryoid bodies beat rhythmically after 10 days of induction. Furthermore, analysis of t test revealed that the percentage of beating cardiomyocyte-like cell clusters was highest (33%) when induction using 0.1 mmol/L Vc and 5 micromol/L 5-aza. Immunocytochemistry analysis demonstrated that cardiomyocyte-like cell clusters expressed alpha-actin, Tnni3. RT-PCR analysis also illustrated that TbX5, Gata4, alpha-MHC and Tnni3 were expressed positive in cardiomyocyte-like cell clusters. Especially, we observed basic structures of myocardium, such as myofilament, glycogen granule and so on by transmission electron microscope. In conclusion, 5-azacytidine and vitamin C could promote differentiation of pAFS into myocardium.

2.Nursing countermeasure and prevention and control of Zika virus disease

Since the first local infection case of Zika virus appeared in Brazil from May 2015, the disease quickly spread in Brazil. The writer would introduce Zika virus from 4 aspects and they are history, biological property, physicochemical property and physicochemical property. Combined the writer′s years of clinical nursing experience in tertiary hospitals, experience during Ebola period and study on Ebola, this paper would analyze how to nursing patients with this disease from 5 aspects; it also put forward some suggestions on prevention and control of Zika virus disease from aspect which includes society, government, hospitals and the public, combined with the prevention and control of infectious disease, so that to make everyone realize the importance of learning, prevention and control of epidemic, nursing, standard nursing operation and popularized knowledge of infectious diseases.
3.Causes and nursing strategies of hepatocellular carcinoma patients with fever after cryo-ablation therapy

Objective To explore the susceptible factors of fever and infection among hepatocellular carcinoma (HCC) patients who received cryo-ablation (CA) therapy,and to propose intervention countermeasures accordingly.Methods The fever and infection status of totally 428 HCC patients who received CA therapy was examined systematically;then the influencing factors were analyzed by single factor analysis and logistic regression analysis;and finally,nursing strategies were proposed accordingly.Results Totally 229 patients developed fever (53.5%),and 6 suffered from complicated infection (1.4%) after CA therapy;tumor size,CA area,Child-Pugh classification and preoperative leukocyte level were factors that contributed to the fever of HCC patients who received CA therapy.Conclusions We should attach importance to the susceptible factors of fever and infection among HCC patients who received CA therapy,and scientific intervention and nursing are the key to reducing the possibilities of fever and infection among patients who received CA therapy.
