Hypercholesterolemia is an important risk factor of cardiovascular disease,which is related to complications and mortality of cardiovascular disease.A lot of researches show that some genes polymorphism was related to hypercholesterolemia.This paper introduces the hypercholesterolemia,including LDLR(low-density lipoprotein),apo(apolipoprotein) and PCSK9(proprotein convertase subtilisin kexin type 9),etc,and summed up the relationship between the genes polymorphism and hypercholesterolemia.

Objective To establish a macrophage-derived foam cell model of human monocytic THP-1 cell line and to identify the foam cell.Methods THP-1 cells were differentiated into macrophages after induction with 160 nmol/L phorbol-1-myristate-13-acetate for 24 hours and then incubation with 80 mg/L oxidized low density lipoprotein(oxLDL) for 48 hours.The differentiated cells were observed after oil red O staining under light microscope.High performance liquid chromatography/atmospheric pressure chemical ionization tandem mass spectrometry(HPLC-MS) was used for quantitative analysis of cellular cholesterol and cholesteryl esters contents.Results There was a large number of red lipid droplets in phorbol-1-myristate-13-acetate and oxLDL treated THP-1 cells after oil red O staining under light microscope.Cellular contents of cholesterol and cholesteryl esters in the differentiated cells increased markedly compared with the normal THP-1 monocytes by HPLC-MS assay.Conclusion A human monocyte-derived foam cell model has been established by incubating THP-1 cells with 120nmol/L phorbol-1-myristate-13-acetate for 24 and then with 80 mg/L oxidized low density lipoprotein for 48 hours.

Objective To identify the relationship among glycogenes,N-glycans and hepatocarcinoma metastasis and drug resistance by studying the differential expressions of glycogenes and N-glycans in MHCC97-H and MHCC97-L human hepatocarcinoma cell lines,and to confirm the novel target of hepatocarcinoma metastasis and anti-tumor therapy.Methods Real-time PCR was used to quantitatively analyze glycogenes and fluorescein isothiocyanate (FITC)-lectin was used to analyze glycans characteristics.RNA interference approach was used to interfere the glycogenes,and the invasive ability in vitro and drug susceptibility of MHCC97-H cells were detected before and after interference.Modification of N-glycosylation (tunicamycin and PNGase F treatment) was done,and the invasiveness in vitro,tumorigenicity in vivo and drug susceptibility of MHCC97-H cells were detected before and after modification.Results The expressions of glycogenes and glycans were different in MHCC97-H cells and MHCC97-L cells.The silence of MGAT5 in MHCC97-H cells inhibited invasion ability and increased sensitivity to 5-fluorouracil in vitro(t =7.312,P ＜ 0.05).Modification of N-glycosylation decreased MHCC997-H cells invasion ability in vitro and tumorigenicity in vivo and increased sensitivity to 5-fluorouracil.Conclusion The differential expressions of glycogens and N-glycans in human hepatocarcinoma cell lines correlate with tumor invasion and drug resistance,and they are expected to be novel targets of tumor chemotherapy.

Objective:To enhance the management level of drug emergency inspection. Methods: The emergency inspection of plasticizers was used as the example, and the management in the whole inspection procedures was reviewed to summarize the difficulties and solutions and analyze the management features of drug emergency inspection. Results and Conclusion: There are three keys in successful drug emergency inspection, including correct top-level design, high efficient execution teams and necessary guarantee sys-tems.

Objective: To establish a method for determination of mercury (Hg) and arsenic (As) contents in biological samples. Methods: Biological samples containing Hg were pretreated with digestive agents of HNO3-H2SO4 and that of As with HNO3-HClO4 and HNO3-H2O2 under the pressure of 0.5 MPa,1.0 MPa and 1.5 MPa respectively heated for 2 minutes. And then the concentrations of Hg or As were determined by intermittent flow hydride generation-atomic spectrophotofluorimetry. Results: Recoveries of Hg and As were 97.3 %～99.1 %and 99.4 %～105.7 %respectively. Conclusion: The method is simple, rapid, accurate and sensitivite and with good reproducibility and is suitable for the determination of Hg and As contents in biological samples.

Objective:To investigate the different effects of Angong Niuhuang Wan on normal and pathological body. Methods:Cerebral infarct rat models were established by photochemically initiated thrombosis. The rats were randomly allocated to four groups. The control group and the model group were treated with 0.9%NaCl for seven days, and the control+drug group and the model+drug group with Angong Niuhuang Wan 0.13g/kg for seven days. Then amounts of idoenzymes of serum lactate dehydrogenase(LDH1～5) were determinated by the method of gel electrophoresis. Objective:To investigate the correlation between positive rate of thyroid autoantibodies in healthy people and thy -roid dysfunction.Methods:Fasting blood from 3218 healthy examined people in Cangzhou were collected and the serum was separa -ted.Automatic electrochemiluminescence immunoassay systems and reagents were used to determine the concentration of thyroid stimu -lating hormone(TSH),free triiodothyroninthyroid(FT3),free thyroxine(FT4),peroxidase antibody(TPOAb)and thyroglobulin antibody ( TgAb ) .TPOAb>34 U/ml and TgAb>115 U/ml was positives.Statistical analysis of test results .statistica analysis of test reults.Results:The positive rate of thyroid autoantibodies of tested population was 16.19%.The positive rates of TPOAb and TgAb were 14.57%,12.74%separately.The detection rate of TPOAb and TgAb was 11.12%in tested population.The positive rate of TPO-Ab and TgAb and TPOAb+TgAb were 6.92%,5.68%,5.03%separately in 1532 male subjects and were 21.53%,19.16 ,16.67%separately in 1686 female subjects,which were significantly higher in female group than that in male group (P<0.001).They gradually increased with age and reached a peak above 70 years old in female group and in 50-59 in male group.The positive rate and OR of thy-roid autoantibodies in hyperthyroidism and subclinical hyperthyroidism and in hypothyroidism and subclinical hypothyroidism and in hy -pothyroidism and subclinical hypothyroidism were statistically significant .Conclusion:The positive incidence of thyroid autoantibodies is higher in healthy people of Cangzhou .It is essential to follow-up these people of normal thyroid function with positive autoantibodies in order to facilitate prevention and early diagnosis and treatment of thyroid disease .

Objective: To research and establish an analysis model for chemical drug standards to provide the reference for the standard exploration jobs. Methods:The standard variation was studied by comparing and analyzing every version standard for the same drug to look for the change rule. Results:The standard items of the drug had some change rules in indicator numbers, testing methods and limits according to the order of issue time. The standard development trends could be described by making specific indicators and drawing evolution figures of the items. Conclusion:By the standard analysis model, the same item of the same drug can be compared to provide the reference for data collecting of experts. Meanwhile, formulate standard can be calculated to provide the reference for the standard improvement without drug standards.

Objective:To investigate the effect of different ventilation time in the prone position on patients with endogenous/exogenous ARDS.Methods:30 endogenous/30 exogenous ARDS patients were randomly devided into 4 groups,ventilation in the prone position for 2 h and 4 h.Recording the score of APCHEII,oxygenation index,the absorption situation in X-ray,HR,MAP,extubation time,the time out of ICU.Results:The APCHEII scores HR and MAP in four groups have no significant statistics (P＞0.05);4h ventilation for endogenous ARDS patients has a better indicators than 2 h in oxygenation index,the absorption situation in X-ray,extubation time and the time out of ICU (P＜0.05);2 h and 4 h ventilation for exogenous ARDS patients can improve indicators above,two groups have no significant statistics (P＞0.05),the results of exogenous groups are precede than endogenous group (P＜0.05).Conclusion:Ventilation in the prone position can improve the situation of ARDS patients,both endogenous patients and exogenous patients.Exogenous ARDS patients have a better treatment effect after the ventilation of 2h,however,endogenous patients need longer time and have a non-ideal prognosis.
