1.Correlation of enhancement radiosensitization of emodin isolated from Guangxi P. multiflorum Thunb on hypoxic nasopharyngeal cancer cells with expression of DNA repair genes

Aim To investigate the effects of emodin isolated from Guangxi P.multiflorum Thunb on the expression of KU70/KU80 in hypoxic nasopharyngeal cancer CNE-1 cells and reveal the relationship between radiosensitization of emodin monomer and DNA repair genes.Methods The expression of hypoxia inducible factor-1?(HIF-1?)and DNA double-strand break repair genes(KU70/KU80)between the experimental groups and the control group under hypoxic condition was detected by the real-time fluorescence quantitative RT-PCR.Results Expression of HIF-1? was significantly increased under hypoxia condition.HIF-1? had no change after treatment with emodin alone.The expression level of KU70/KU80 was induced by radiation.Compared with radiation alone group,radiation combined hypoxia group obviously enhanced the expression of KU70/KU80.KU70/KU80 mRNA expression significantly reduced after radiation combined with emodin under hypoxic condition.Conclusion In the hypoxic environment,emodin combined with radiotherapy can effectively inhibit the expression of HIF-1 ? and DNA double-strand break repair genes(KU70/KU80),which may be its mechanism of radiosensitization.
2.Screening analysis of positive thyroid autoantibody in waterborne high iodine ar-ea healthy people

Objective:To investigate the correlation between positive rate of thyroid autoantibodies in healthy people and thy -roid dysfunction.Methods:Fasting blood from 3218 healthy examined people in Cangzhou were collected and the serum was separa -ted.Automatic electrochemiluminescence immunoassay systems and reagents were used to determine the concentration of thyroid stimu -lating hormone(TSH),free triiodothyroninthyroid(FT3),free thyroxine(FT4),peroxidase antibody(TPOAb)and thyroglobulin antibody ( TgAb ) .TPOAb>34 U/ml and TgAb>115 U/ml was positives.Statistical analysis of test results .statistica analysis of test reults.Results:The positive rate of thyroid autoantibodies of tested population was 16.19%.The positive rates of TPOAb and TgAb were 14.57%,12.74%separately.The detection rate of TPOAb and TgAb was 11.12%in tested population.The positive rate of TPO-Ab and TgAb and TPOAb+TgAb were 6.92%,5.68%,5.03%separately in 1532 male subjects and were 21.53%,19.16 ,16.67%separately in 1686 female subjects,which were significantly higher in female group than that in male group (P<0.001).They gradually increased with age and reached a peak above 70 years old in female group and in 50-59 in male group.The positive rate and OR of thy-roid autoantibodies in hyperthyroidism and subclinical hyperthyroidism and in hypothyroidism and subclinical hypothyroidism and in hy -pothyroidism and subclinical hypothyroidism were statistically significant .Conclusion:The positive incidence of thyroid autoantibodies is higher in healthy people of Cangzhou .It is essential to follow-up these people of normal thyroid function with positive autoantibodies in order to facilitate prevention and early diagnosis and treatment of thyroid disease .
3.Induction of human ovarian cancer SKOV3 cells apoptosis by oxymatrine

AIM To observe the effects of Oxymatrine on induction of apoptosis in human ovarian cancer SKOV3 cells. METHODS Apoptosis induced by Oxymatrine in human ovarian cancer SKOV3 cells was tested by MTT assay, fluorescent microscope, and DNA gel eletrophoresis. RESULTS SKOV3 cell viability dropped down depending on the Oxymatrine concentration and treatment time. When incubated with Oxymatrine (0 189 mmol?L -1 and 0 378 mmol?L -1 ) for 48 h, SKOV3 cells showed morphological changes associated with the characters of apoptosis under fluorescent microscope. Typical DNA ladder was found during gel eletrophoresis. CONCLUSION Oxymatrine can induce apoptosis in SKOV3 cell lines.

Aim To synthesize 8-bromo-ethoxy Rhein and investigate its mechanisms and inhibition effect on hepatitis B surface antigen ( HBsAg ) and e antigen ( HBeAg) in HepG2.2.15 cells.Methods 8-bromo-ethoxy Rhein was synthesized based on the chemical structure of Rhein , and its structure was identified by IR,1 H-NMR and 13 C-NMR spectra.MTT assay was used to test the inhibitory effect of 8-bromo-ethoxy Rhein on HepG2.2.15 cells.After the cells treatment by 8-bromo-ethoxy Rhein , the HBsAg and HBeAg in cell supernatant were detected by ELISA .The expres-sion of hepatitis B virus X gene ( HBx) was detected by Western blot .The cell cycles were examined with flow cytometry.The intracellular free calcium concentration was detected by laser scanning confocal microscopy . Results The structure of 8-bromo-ethoxy Rhein was confirmed by IR,1 H-NMR and 13 C-NMR.MTT results showed that synthetic product and Rhein could inhibit the cell proliferation in HepG2.2.15 cells.After trea-ted with 8-bromo-ethoxy Rhein and Rhein for 72 h,the half inhibitory concentration 50%( IC50 ) was 14.29 mg? L-1 and 11.59 mg? L-1 , respectively .Using non-cytotoxic dose of 8-bromo-ethoxy Rhein , the inhibitory effect on HBsAg and HBeAg was gradually enhanced with increasing 8-bromo-ethoxy Rhein concentration . The inhibitory effect of synthetic product on hepatitis B virus was better than that of Rhein .8-bromo-ethoxy Rhein could down-regulate the expression of HBx , in-tracellular calcium ion concentration and block the hepatitis B virus ( HBV ) replication.Flow cytometry results showed 8-bromo-ethoxy Rhein didn′t affect the cell cycle .Conclusions Compare with Rhein , the synthesis of 8-bromo-ethoxy Rhein shows stronger inhi-bition on hepatitis B virus in HepG2.2.15, and its mechanisms may involve down-regulating the expres-sion of HBx and reducing calcium ion concentration .

Objective To investigate the effect of dexmedetomidine hydrochloride ( Dex ) preconditioning on renal function in rats after renal ischemia reperfusion injury under high glucose condition. Methods SD rats were randomly divided into 6 groups:NG-Sham operated group,NG-I/R group, NG-Dex group,HG-Sham operated group,HG-I/R group,HG-Dex group. Renal ischemia reperfusion model was established except Sham groups. Dex 50 μg·kg-1 was injected intraperitoneally 30 min before ischemia in the Dex preconditioning group,25% glucose 3 g·kg-1 was given intraperitoneally before the renal ischemia reperfusion model was established in high glucose groups. Blood glucose and renal function of each group were detected . Renal pathologic changes were observed with hematoxylin-eosin staining. Apoptosis of renal tissue was detected by TUNEL method. Results BUN,Cr and apoptosis rate in NG-I/R group were higher than those in NG-Sham operated group ( P<0.05);BUN,Cr and apoptosis rate in NG-Dex group were lower than those in NG-I/R group ( P<0.05);BUN,Cr and apoptosis rate in HG-I/R group and HG-Dex group were higher than those in NG-I/R group and NG-Dex group,respectively (P<0.05); However,there was no significant difference between HG-I/R group and HG-Dex group ( P>0.05) . Conclusion Dex has a protective effect on renal function after renal ischemia reperfusion, but this effect is inhibited in high glucose condition, which may relate to the increasing of kidney cells apoptosis.
7.Analysis of abnormal thyroid function in the health check-ups of residents in Cangzhou City of Hebei Province

Objective To know about the abnormal condition of thyroid function in healthy population of urban residents in Cangzhou City,to reduce the incidence of thyroid disease to provide a reference.Methods Urban residents participated in a health checkup were selected as respondents in Cangzhou from 2013 to 2015,whose thyroid examination and laboratory test data of thyroid function were collected.Statistical analysis of thyroid dysfunction was performed.Results Totally 3 218 healthy controls with 248 cases of thyroid dysfunction were tested.The detectable rate was 7.71%.The detectable rates of hypothyroidism,subclinical hyperthyroidism,hypothyroidism,and subclinical hypothyroidism were 0.81% (26/3 218),0.68% (22/3 218),1.83% (59/3 218),and 4.38% (141/3 218),respectively.The prevalence of subclinical thyroid disease (5.07%) was higher than that of clinical thyroid disease (2.64%,x2 =32.140,P < 0.01).The detectable rates of thyroid dysfunction from 2013 to 2015 were 2.73% (24/879),6.54% (71/1 086),and 12.21% (153/1 253),respectively.The differences among the three were statistically significant(x2 =69.443,P<0.01).The detectable rate of male thyroid dysfunction was 3.20% (49/1 532)and female was 11.80% (199/1 686).The women's rate was significantly higher than that of men (x2 =12.806,P < 0.01).The majority of patients were in the 40 to under 50 and older than 70 years of age which accounted for 10.25% (65/634) and 10.40% (31/298),respectively.Conclusions The detectable rate of thyroid dysfunction in healthy population of urban residents in Cangzhou City is increasing year by year,most cases are subclinical hypothyroidism.In the future,early diagnosis and differential diagnosis of thyroid diseases should be strengthened and appropriate intervention measures should be taken to reduce the incidence of thyroid and related diseases.
