Thus, the abnormity of epigenetic regulation may lead to different diseases, especially cancer.SNF5 ,which can suppress tumor significantly,is one of the core components of SWI/SNF chromosome remodeling complexes.EZH2,a histone methyltransferase,is closely and complicatedly related to cancer and has significant effect on tumorigenesis.There are many studies reporting the effects of epigenetic regulation factors-SNF5 and EZH2 on different tumors as well as the interrelation at present.In the present paper,the advances in the research on these two epigenetic regulation factors in the carcinogenesis are reviewed.
2.Apllication of high resolution electronic staining endoscopy in the diagnosis of gastrointestinal diseases

I-scan technology is the newly developed image enhanced endoscopy technology from PENTAX,Japan,which can visualize the capillary and the gap of glandular duct of gastrointestinal mucosa.i-scan images are as bright as conventional white-light images.Therefore,i-scan is able to observe much larger areas in a distant view compared with NBI.It has a broad application prospects.However,due to the lack of a unified diagnostic standard,the application of i-scan technology needs further study.This review summarizes the applications of high-resolution electron-staining endoscopy in the diagnosis of gastrointestinal diseases.
3.Significance of i -Scan in the diagnosis of Barrett′s esophagus

Objective To evaluate the value of i-Scan in the diagnosis of Barrett′s esophagus.Methods 52 patients diagnosed with Barrett′s esophagus by gastroscopies and pathological examinations were observed under the ordinary edoscopy and i-Scan respectively.Besides,the Barrett's esophageal mucosa were classified by Endo with i-scan mode, the target lesions was taken to be biopsied and pathologically examined.Results ( 1 ) In the 52 patients,mucosal lesions were found in 46 cases (88.5%) under i-Scan and 35 cases (67.3%) under ordinary edoscopy respectively,mucosal lesion detection rate in i-Scan was significantly higher than normal white mode( P<0.01).In the 33 patients which found mucosal lesions in the two modes,the definition of lesion margins,morphology of lesion cryptaeand mucosa capillaries revealed by i-Scan were superior to that by white lighted endoscopy ( P<0.05);(2) Under i-scan mode,the detection of specific intestinal metaplasia ( SIM) of typeⅣ,type V in sensitivi-ty,accuracy,specificity were respectively 90.0%,93.5%,94.4%.Conclusion i-Scan is very easy to operate and has a clear lesion of Barrett′s esophagus,which helps to target the lesion at pathological biopsy.
4.Effects of mechanical stretch on expression of pentraxin-3 mRNA and protein in A549 cells

Objective To evaluate the effects of mechanical stretch on pentraxin-3(PTX-3)mRNA and protein expression in human alveolar epithelial cells (A549 cells).Methods The human lung epithelial adenocarcinoma cells A549(A549 cells)were purchased from cell biology laboratory,Tongji Medical College,Huazhong University of Science and Technology.The cultured A549 cells were inoculated on collagen Ⅰ BioFlex plates and divided into 5 groups(n=3 wells each):group Ⅰ normal control;groupⅡsham mechanical stretch;group Ⅲ mechanical stretch;groupⅣsiRNA and groupⅤ siRNA+mechanical stretch.In group Ⅲ the cells underwent square cyclic mechanical stretch for 4 h using the Flexercell Systcm.In group Ⅳ the cells were transfected with chemosynthetic PTX-3 specific siRNA by RNAi technique.In group Ⅴ at 24 h after being transfected with PTX-3 siRNA the cells underwent mechanical stretch for 4 h.In groupⅡ mechanical stretch of the cells were prevented by Flexstep.The expression of PTX-3 mRNA in the cells was detected by real-time PCR and the expression of PTX-3 protein in the culture media was determined by Western blotting.Apoptosis of the cells was measured bv flow cytometry(Beeten-Dickinson,USA).Results PTX-3 mRNA and protein expression was signlficantly up-regulated by mechanical stretch in groupⅢand decreased by transfection with siRNA in group Ⅳand Ⅴ as compared with group Ⅰ andⅡ(P<0.05 or 0.01).The apoptosis ratio was significantly higher in group Ⅲ and Ⅴ than in groupⅡ and was significantly lower in group Ⅴ thanin group Ⅲ(P<0.01).Conclusion Mechanical stretch can up-regulate PTX-3 mRNA expression in A549 cells.
5.Effects of mechanical stretch on the expression of human beta-defensin-3 in alveolar epithelial cells elicited by interferon- gamma

Objective To study the effect of mechanical stretch on the expression of human beta-defensin-3 (HBD-3) in alveolar epithelial cells(A549 cells) elicited by interferon-gamma(IFN-γ) and to investigate the role of HBD-3 in the pathogenesis of ventilator-associated pneumonia (VAP) . Method A549 cells cultured in vitro were treated with mechanical stretch (group S), 10 ng/ml IFN-γ (group I) ,and 10 ng/ml IFN-γ with mechanical stretch (group IS), respectively. Cells without treatment served as controls (group C). Cells were stretched by 20% amplitude of stretch at 30 cycles/mm by Flexercell-4000[TM]Unit for 2 h, 4 h, and 6 hours. The HBD-3 mRNA expression was determined by real-time RT-PCR after treatment. After 6 hours, treatment, cells were cultured for 24 hours and the expression of HBD-3 was examined by laser scanning confocal microscope. The experimental data were statistically analyzed by using one-way ANOVA analysis and q-test. Results The expression of HBD-3 mRNA in A549 cells could not significantly be changed by mechanical stretch alone. Compared with group C,the HBD-3 mRNA expression after treatment with 10 ng/ml IFN-γ for 2 hours,4 hours and 6 hours increased significantly by (2.63 C,the HBD-3 mRNA expression after treatment with 10 ng/ml IFN-γ and mechanical stretch for 2 hours,4 hours and 6 hours increased by (1.54 were significantly lower than those in group I (P < 0.01). The HBD-3 expression in group IS after mechanical stretch for 6 significantly different from than in group C. Conclusions Mechanical stretch can significantly suppress the up-regulation of HBD-3 in alveolar epithelial cells elicited by IFN-γ, and this may be one of the explaina-tions that patients under mechanical ventilatiori(MV) have a higher risk of VAP.
6.Effect of Mild Moxibustion on the Rehabilitation Efficacy in Patients with Cerebral Palsy

ObjectiveTo observe the clinical effect of warm needling based on conventional rehabilitation in treating cerebral palsy.MethodSixty patients with cerebral palsy were randomized into an observation group and a control group, 30 in each group. The control group was intervened by physiotherapy (PT), occupational therapy (OT), speech therapy (ST), conductive education (CE), etc.; the observation group was by mild moxibustion at Baihui (GV20), Zusanli (ST36), and Guanyuan (CV4) in addition tothe above rehabilitation therapies.ResultThe rehabilitation result in the observation group was superior to that inthe control group (P<0.05).ConclusionMild moxibustion can significantly enhance the rehabilitation result in patients with cerebral palsy.
7.Effects of trichosanthin on apoptosis and cytoskeleton microtubule structure reconfiguration in lung cancer A549 cells

Objective:This study aimed to explore the effects of trichosanthin on the proliferative inhibition and apoptosis induc-tion in human lung carcinoma A549 cells. Methods:A549 cells were treated with various concentrations of TCS. The inhibitory effects in proliferation were detected by the MTT method. The microfilament changes were observed by transmission electron microscopy. Apoptosis rate and cell cycle were determined by flow cytometry. Results:A549 cells treated with TCS presented apoptotic changes and decreased cell activity. When the concentration increased and time was prolonged, the inhibition rate increased correspondingly. Conclusion:Pharmacological doses of TCS inhibited the proliferation and differentiation in lung carcinoma A549 cells and affected the function in A549 cells by changes in the cytoskeleton.
8.Gemcitabine, navelbine, and therarubicin (GNT) as treatment for patients with refractory or relapsed T-cell lymphoma

Objective:This study was conducted to evaluate and discuss the curative effect and toxicity of gemcitabine, navel-bine, and therarubicin (GNT) regimen for patients with refractory or relapsed T-cell lymphoma (TCL). Methods:A total of 69 patients with refractory or relapsed TCL treated with GNT were enrolled. The treatment protocol was set as follows:800 mg/m2 gemcitabine ad-ministered at 1 and 8 d;25 mg/m2 navelbine administered at 1 d;and 20 mg/m2 therarubicin administered at 1 d. This protocol was re-peated every three weeks. The median cycle was 4 (range:2 to 6). Results:The overall response rate was 65.2%and the achieved com-plete remission was 29.0%. Hematology toxicities were the main adverse reactions observed in all of the patients. The incidence rates of grades 1 and 2 toxicity in leukopenia or neutropenia, anemia, and thrombocytopenia were 50.7%, 33.3%, and 26.1%, respectively. Grades 3 and 4 treatment-associated toxicities were detected in 23.1%of the responding patients. One-, three-, and five-year estimated overall survival (OS) of the whole cohort were 71.7%, 47.3%, and 32.4%, respectively. The median OS was 36 months. Conclusion:GNT was effective and suitable for patients with refractory or relapsed TCL.
9.Androgen receptor expression in molecular subtypes of breast cancer and its relationship with the clinicopathological features

Purpose To investigate the expression of androgen receptor ( AR) in molecular subtypes of breast cancer and its relation-ship with clinicopathological features. Methods The expression of estrogen receptor (ER), progesterone receptor (PR), HER-2, CK5/6, CK14 in breast cancer was detected by EnVision immunohistochemical staining, and they further be divided into 84 Luminal A subtypes, 27 Luminal B subtypes, 14 HER-2 overexpression subtypes, 11 Basal-like subtypes, and 17 Normal-like subtypes, respec-tively. The distribution of AR in 5 molecular subtypes and its relationship with clinicopathological factors, such as age, histopathologi-cal grading, tumor size, lymph node status and clinical staging, were analyzed. Results In 153 cases of breast cancer, the positive rates of AR expression were 73. 20%, Luminal A was the majority ( 62. 50%) in 5 molecular subtypes. In the HER-2 negative and positive breast cancer, the positive rates of AR expression were 75. 00% and 68. 29% and the difference between the two groups was no statistical significant (P=0. 196). Of the 153 breast cancers, 23 (15. 03%) were triple-negative breast cancers (ER-, PR- and HER-2-), 15 (9. 80%) breast cancers were negative for ER, PR and AR, 8(5. 23%) breast cancers were quadruple negative (ER-, PR-, AR- and HER-2 -) . There was significant difference of the expression of AR in ER/PR positive and negative breast cancer (P=0. 001),it was positively correlated with ER/PR protein expression (rs =0. 874, P=0. 024). The difference of AR ex-pression between histological grading was statistically significance ( P=0. 022 ) , the expression of AR was negatively correlated with histopathologic grading (rs = -0. 201, P=0. 013). The positive expression of AR was related to tumor size, but not correlated with lymph node metastasis ( P=0. 211 ) and clinical staging ( P=0. 065 ) . Conclusion The expression of AR in breast cancer tissues and its relationship with clinicopathological features may provide a basis for the further improvement molecular typing of breast cancer, and also provide the theoretical basis for clinical treatment and judging prognosis.
10.Quality evaluation of tandem mass spectrometry screening for neonatalinherited metabolic diseases in Zhejiang Province from 2009 to 2021

Objective:
To evaluate the quality of tandem mass spectrometry (MSMS) screening for neonatal inherited metabolic diseases in Zhejiang Province from 2009 to 2021.
Methods:
The data pertaining to MSMS screening for neonatal inherited metabolic diseases in Zhejiang Province from 2009 to 2021 were collected from the database created by Zhejiang Provincial Center for Neonatal Disease Screening. The percentage of MSMS screening, percentage of recall of suspected screening-positive infants and incidence of neonatal inherited metabolic diseases were analyzed retrospectively to evaluate the quality of MSMS screening for inherited metabolic diseases.
Results:
A total of 4 706 916 newborns were screened among 8 297 039 live births by MSMS in Zhejiang Province from 2009 to 2021, and the percentage of MSMS screening increased from 5.48% to 97.54%, with a mean percentage of 56.73%. Of 46 838 suspected screening-positive infants, 45 527 infants were recalled, and the percentage of recall increased from 94.57% to 98.62%, with a mean percentage of 97.20%. A total of 1 038 infants were definitively diagnosed with inherited metabolic diseases in Zhejiang Province from 2009 to 2021, with an overall incidence rate of 1/4 535, and the incidence rates of amino acid metabolic disorder, fatty acid oxidation metabolic disorder and organic acid metabolic disorder were 1/11 767, 1/13 763 and 1\15 902, respectively. Of the 11 cities in Zhejiang Province, the highest percentage of tandem mass spectrometry screening for inherited metabolic diseases was found in Hangzhou City (83.01%), and the highest percentage of recall of suspected screening-positive infants was se en in Zhoushan City (99.08%).
Conclusions
A high percentage of MSMS screening for neonatal inherited metabolic diseases was observed in Zhejiang Province from 2009 to 2021; however, there was a region-specific percentage of screening, and the recall of suspected screening-positive infants remains to be improved.
