1.Effect of Cold Stress on Artery Pressure and Serum Cytokines of Rabbits

Objective To know the effect of cold stress on the artery pressure and the concentration of IL-2, IL-10, IFN-?, TNF-? in the serum of the rabbits. Methods The local white rabbits of China (clean grade) were anesthetized. The mAP (mean artery pressure), mSP (mean systolic pressure), mDP (mean diastolic pressure) and the concentration of IL-2, IL-10, IFN-?, TNF-? were determined at 0 h, 0.25 h, 0.50 h, 0.75 h, 1.00 h, 1.50 h, 2.00 h, 3.00 h, 4.00 h and 6.00 h after cold stress. Results The mAP,mSP and mDP changed undulant with the delay of cold stress,but the whole tendency was descendent. The mAP,mSP and mDP went up to the peak at 1 h,however lower than control. The concentration of IL-2 increased then decreased with the delay of cold stress,the concentration of IL-10 went down then up and the concentration of TNF-?,IFN-? went down gradually. 2.Regulating Mechanism of Differentiation of Neural Stem Cells

Neural stem cells which have many characters of high self-renewal, multipotentiality, migration and good histocompatibility could differentiate neurons, astrocyte and oligodendrocyte. Therefore, the research on differentiation mechanisms of neural stem cells should be very important for the therapy of nervous system degenerative diseases and functional recovery of the central nervous system injury. The neural stem cells differentiation mechanisms regulated by the genes and environment were reviewed in the present paper.
3.EFFECT OF LINSEEDS,VITAMIN E AND ETHOXYQUINOLINE ON PRODUCTIVE PERFORMANCE AND ?-3 PUFA DEPOSITION IN THE YOLK OF LAYERS

Objective:The effect of the linseeds,VE and ethoxyquinoline in feed on the productive performance of the Hailanhong layers and ?-3 PUFA deposition in the yolk of laying eggs was studied. Method:Four hundred and eighty healthy layers of 48 weeks of age were randomly selected. The linseeds,VE and ethoxyquinoline of different levels in diet were fed on these layers. The indices of the productive performance and the fatty acids content of yolk were determined. The regression model of the fatty acids deposition was established. Results:The effect of the linseeds,VE and ethoxyquinoline in feed on the productive performance was slight. The most deposition of ?-3 PUFA was found by adding linseeds 15%,VE 0.17g/kg feed without ethoxyquinoline. Conclusion:The deposition of ?-3 PUFA in eggs could be regulated by controlling the proportion of linseeds and VE in feed with no effect on productive performance of layers.
4.Relationship between vegetable, fruit, fish and soybean dietary pattern and breast cancer: a meta-analysis

Objective To understand the relationship between dietary patterns and breast cancer and to provide a possible scientific basis for breast cancer prevention,we performed a meta-analysis in order to evaluate the current relevant literature.Method Meta-analysis was used to pool the effect of vegetable,fruit,fish and soybean dietary patterns on the risk of breast cancer.The breast cancer risk of the highest categories compared to the lowest categories of such dietary patterns was combined using fixed effect model.Searching the database (Chinese databases included CQVIP,WANFANG medicine online,CNKI and SinoMED,and English databases included PubMed,Embase,Cochrane database,Medline and Google Scholar) for articles published between January 1997 and June 2014.All statistical analyses were performed using the Stata 11.0.Result Twelve cohort studies involving 623 536 participants and 19 707 patients with brcast cancer were found to be eligible.Our results indicate that a reasonably healthy dietary pattern (which is characterized by a high consumption of vegetables,fruit,fish and soybean) is beneficial for the prevention of breast cancer.Compared to the lowest categories of such dietary patterns,relative risk (RR) of the highest categories is 0.86 (95% CI 0.80-0.92).Conclusion A reasonably healthy dietary pattern (which is characterized by a high consumption of vegetables,fruit,fish and soybean) is beneficial for the prevention of breast cancer.
5.Construction and identification of ERβ419 gene silencing recombined vectors of beagle ERβisoforms

Objective To construct and identify retroviral-mediated short hairpin RNA ( shRNA ) expression vectors of ERβ419, and explore ERβ419 unknown biological function in beagles in future.Methods To screen out the most effective gene silencing sequence of beagle ERβ419 mRNA using qRT-PCR and Western Blot assays, imitate beagle estrogen target cells.Results qRT-PCR results showed, ERβ419-shRNA1 ( P <0.01 ) and ERβ419-shRNA3 ( P <0.01)differed significantly, Western Blot result as same as qRT-PCR,ERβ419-shRNA3 is the best choice.Conclusion Beagles ERβ419-shRNA3 retrain most effectively target gene repression. It is applied to explore ERβ419 unknown biological function in beagles reproductive system, and to prevent and treat beagles reproductive function diseases.
6.Relationship between serum alpha-fetoprotein level and clinical features of hepatoblastoma in children

Objective To analyze the relationship between the clinical features of hepatoblastoma and serum alpha-fetoprotein level.Methods We did a retrospective study in 74 cases into the relationship of clinical stages,treatment,prognosis,and AFP value in hepatoblastoma.Results In Stage Ⅰ and Ⅱ patients,the average AFP was (36 333 ± 13 782) ng/ml and in Stage Ⅲ and Ⅳ,the average AFP was (78 346 ± 27 956) ng/ml,P < 0.05.68 patients received preoperative chemotherapy with alpha-fetoprotein determinations before and after chemotherapy respectively.In 44 cases AFP decreased > 50% after chemotherapy,and in 20 cases AFP declined >90%,while in 13 cases AFP increased after chemotherapy.15 cases had serum AFP measured on the first postoperative day,among them 10 cases had a > 50% AFP decline,3 cases had a < 50% AFP decline,while in 2 cases AFP value increased.52 cases were postoperatively followed-up,with overall 3 year-survival rate of 86.5% ; 7 cases recurred,4 died.The average preop AFP level in 7 recurred cases was (27 060 ± 3 569) ng/ml,while the average preop AFP level in those of 3-year recurrence-free was (29 865 ± 5 867) ng/ml,P > 0.05.Only 57% tumor recurred cases had back to normal AFP level within one month postop,while 89% long term survivals reported normal range of AFP during that period,P < 0.05.Conclusions Serum AFP level in patients with hepatoblastoma is related to the course of disease.It can be used to estimate the effect of clinical chemotherapy.The speed of alpha-fetoprotein decline postop can be used as an indicator of prognosis.The postoperative normal AFP level within 4 weeks predicts a favorable prognosis.
7.Research advances in molecular biological mechanisms of neuroblastoma

Neuroblastoma(NB)is the most common solid extracranial tumor of children,which is obviously heterogenous in the positions,histopathological features,and biological characteristics. One of the most intractable issues in the current clinical treatment is the poor prognosis in high - risk and advanced stage of NB. In recent years,numerous studies have been conducted on molecular biological mechanisms of NB. Researches demonstrate that tumorigenesis is related to various factors,including chromosome,oncogene,and antioncogene abnormalities;carcinogenesis related genes (involving cell proliferation,apoptosis,angiogenesis,chemoresistance and intracellular signal transduction system)ab-normalities. Some specific information(such as chromosome 1p status,MYCN expression,and DNA ploidy)has become the important basis for judging prognosis and grouping the patients by risks. Meanwhile,there are still many discovered NB - related molecular biological mechanisms which are not yet included in the risk stratification system. Further development of targeted drugs for those molecules may be of great significance in increasing the survival rate of patients and improving the prognosis.
8.The regulatory mechanism and clinical significance of RASSF5 in skin melanoma

Objective:To investigate the regulatory mechanism and clinical significance of RASSF5 in cutaneous melanoma.Methods:We used GEPIA and HPA online tools and TCGA and GSE53118 data sets for differential expression analysis and prognosis analysis; cBioPortal was applied to analyze the correlation between RASSF5 expression and gene mutation and methylation; "limma" R package was used to screen differential genes for GO analysis, KEGG analysis and protein-protein interaction analysis, and we also performed GSEA analysis on all genes; finally we used ESTIMATE and CIBERSORTx online tools to evaluate immune cell infiltration. The kruskal test was used to compare the differences between the groups for measurement data, the Kaplan-Meier method was used to draw the survival curve, and the Cox proportional hazard regression model was used for univariate and multivariate analysis.Results:This study found that in cutaneous melanoma, the expression level of RASSF5 was significantly lower than that of normal tissues, and the methylation level was negatively correlated. KEGG analysis and GSEA analysis showed that RASSF5 can be related to multiple signaling pathways such as immune regulation, KRAS, and P53. Further analysis showed that the expression level of RASSF5 was positively correlated with the degree of infiltration of a variety of tumor immune cells. Survival analysis found that the expression level of RASSF5 was correlated with the overall survival rate of patients with cutaneous melanoma, and multivariate regression analysis found that RASSF5 was an independent predictor of cutaneous melanoma. Finally, using the GSE53118 dataset to verify again, RASSF5 is related to the overall survival rate of patients with cutaneous melanoma and can be used as an independent predictor.Conclusions:RASSF5 may regulate the occurrence and development of cutaneous melanoma cells through a variety of ways. It is a potential biological prognostic marker and therapeutic target.

10.Risk factors for clinically relevant postoperative pancreatic fistula of pancreatic tumor in children

Objective:To analyze the influencing factors of clinically relevant postoperative pancreatic fistula (CR-POPF) in children with pancreatic tumors after surgery.Methods:The clinical data of 123 children undergoing surgery for pancreatic tumor in Beijing Children's Hospital, Capital Medical University from January 2007 to March 2020 were retrospectively analyzed, including 39 males and 84 females, with a median age of 9.8 years (6.7 to 11.8). Patients without pancreatic fistula and with biochemical leakage were included in control group ( n=95), while patients with grade B and C pancreatic fistula were divided into CR-POPF group ( n=28). The independent influencing factors of CR-POPF were analyzed by univariate and multivariate logistic regression. Results:Among 123 children, 28 cases (22.8%) developed CR-POPF, including 24 cases (85.7%, 24/28) of grade B pancreatic fistula and 4 cases (14.3%, 4/28) of grade C pancreatic fistula. There were significant differences between CR-POPF and control groups in the age > 8 years and 4 months, tumor location, operation time >390 min and procedures (all P<0.05). Multivariate logistic regression analysis showed an increased risk of CR-POPF in children aged > 8 years and 4 months ( OR=8.226, 95% CI: 1.813-37.333, P=0.006) and undergoing duodenum-preserving pancreatic head resection (DPPHR) ( OR=3.353, 95% CI: 1.282-8.767, P=0.014). Conclusion:Age>8 years and 4 months and DPPHR are independent risk factors for CR-POPF in children with pancreatic tumors after surgery.
