1.Biomechanics research on ATP6i gene-interference treating steroid-induced necrosis of the femoral heads in rats

Objective To explore the biomechanics mechanism of ATP6i gene-interference treating steroid-induced necrosis of the femoral heads(SINFH)in rats.Methods Forty SD rats were randomly assigned to four groups:blank control group,experimental control group,ATP6i SiRNA(+)group and ATP6i SiRNA(-)group,10 rats in each group.We established rat models of SINFH by injecting Escherichia Coli Endotoxin and prednisolone for 12 weeks,then cut along the inner side of the patella for about 1 cm and turn the patellar outside of the both knees of the experimental control group,the ATP6i SiRNA(+)group and the ATP6i SiRNA(-)group,and drill from the intercondylar until the femoral neck.Rats were treated by transfecting ATP6i small interference RNA(ATP6i SiRNA)into the osteoclasts of the femoral heads to interference the expression of ATP6i SiRNA(+),or by the ATP6i SiRNA(-)and same amount of saline.One month later,test the load,stress,modulus of elasticity,and axial stiffness of the right femoral heads of all four groups,and at the same time take the left femoral heads of all four groups for pathological study.Results The load,stress,modulus of elasticity,and axial stiffness of the ATP6i SiRNA(+)group were(307.01±27.34)N,(60.44±5.38)MPa,(125.29±11.16)MPa,(162.42±14.45)N/mm,respectively,which were much greater than the ATP6i SiRNA(-)group and the experimental control group(P<0.05).In the pathological studies,we found that the trabecula of the femoral heads of the ATP6i SiRNA(+)group was much thicker than the ATP6i SiRNA(-)group and the experimental control group,but the degeneration,necrosis and breakage of the osteoepiphysis in each group had no statistically significant differences.Conclusion ATP6i gene-interference can inhibit the acid-secreting function of osteoclasts and improve the biomechanical property of the femoral heads of rats in treating the SINFH.
2.Production of human papillomavirus type 6 virus-like particles and detection of its neutralizing antibodies

Objective To produce human papillomavirus type 6(HPV-6)virus-like particles with Escherichia coli expression system and study its immunogenicity.Methods HPV-6 L1 gene was inserted into pmkaryotic expression vector pTO-T7 and then expressed in Escherichia coli ER2566.The HPV-6 L1 protein was purified by ammonium sulfate precipitation,ion-exchange chromatography,and hydrophobic interaction chromatography.Then the purified HPV-6 L1 self-assembled into virus-like particle after removing 1,4dithiothreitol(DTr).The morphology of the virus-like particles was investigated with dynamic light scatter and transmission electron microscopy,and the immunogenicity was determined with in vitro pseudownons neutralization as8ay.Results HPV-6 L1 was expressed in soluble form in Escherichia coli.Following the removal of DTT,purified HPV-6 L1 protein could assemble into virus-like particles as 25 am in the radius.And the animal immunization test showed HPV-6 virus-like particles can elite hish titer neutralizing antibodies.Conclusion The bacterially expressed HPV-6 L1 VLP is highly immunogenieity and easy to produce.And it can be good candidate of HPV-6 vaccine.
3.Close Relationship between the 2009 H1N1 Virus and South Dakota AIV Strains

Although previous publications suggest the 2009 pandemic influenza A(H1N1)virus was reassorted from swine viruses of North America and Eurasia, the immediate ancestry still remains elusive due to the big evolutionary distance between the 2009 H1N1 virus and the previously isolated strains. Since the unveiling of the2009 H1N1 influenza, great deal of interest has been drawn to influenza, consequently a large number of influenza virus sequences have been deposited into the public sequence databases. Blast analysis demonstrated that the recently submitted 2007 South Dakota avian influenza virus strains and other North American avian strains contained genetic segments very closely related to the 2009 H1N1 virus, which suggests these avian influenza viruses are very close relatives of the 2009 H1N1 virus. Phylogenetic analyses also indicate that the2009 H1N1 viruses are associated with both avian and swine influenza viruses circulating in North America. Since the migrating wild birds are preferable to pigs as the carrier to spread the influenza viruses across vast distances, it is very likely that birds played an important role in the inter-continental evolution of the 2009 H1N1virus. It is essential to understand the evolutionary route of the emerging influenza virus in order to find a way to prevent further emerging cases. This study suggests the close relationship between 2009 pandemic virus and the North America avian viruses and underscores enhanced surveillance of influenza in birds for understanding the evolution of the 2009 pandemic influenza.
4.Therapeutic Efficacy and Safety of Compound Muni Ziqi Granules in the Adjuvant Treatment of Chloasma: A Systematic Review

OBJECTIVE: To systematically evaluate therapeutic efficacy and safety of Compound muni ziqi granules in the adjuvant treatment of chloasma, and to provide evidence-based reference for clinical treatment. METHODS; Retrieved from Chinese Journal Full-text Database (CJFD), China Scientific Journal Database (CSJD), Wanfang database, Medline and Embase, randomized controlled trials (RCTs) about therapeutic efficacy (total response rate, cure rate, the decrease level of E2, FSH and LH in serum) and safety of Compound muni ziqi granules alone or combined with routine drug (trial group) vs. routine drug (control group) in the treatment of chloasma were collected. Meta-analysis was conducted by using Rev Man 5. 3 statistical software after data extraction of clinical studies meeting inclusion criteria and quality evaluation with Cochrane systematic evaluator manual 5. 1. 0. RESULTS: Totally 12 RCTs were enrolled, involving 1 100 patients. Results of Meta-analysis showed that total response rate [OR=2. 84, 95%CI(2. 20, 3. 67), P<0. 001], cure rate [OR=2. 11, 95%CI(1. 59, 2. 79),P<0. 001],decrease level of Ez [OR=-98. 64, 95% CI (-110. 84, -86. 44), P <0. 001], decrease level of FSH [OR=-1. 85, 95%CI(-2. 58,-1. 11),P<0. 001] and decrease level of LH [OR= - 5. 85, 95% CI (-6. 83, -4. 87), P<0. 001] in trial group were significantly better than control group, with statistical significance. In trial group, a few patients suffered from the increase of menstruation, the decrease of menstruation, temporary pigmentation and transient burnout; but all symptoms did not affect the treatment. CONCLUSIONS: Compound muni ziqi granules show definite clinical efficacy and good response rate in the adjuvant treatment of chloasma with mild ADR.
5.Analysis on inheritance effect of famous doctor studio based on R software and CiteSpace

Objective:To evaluate the construction effect of famous doctor studio and the correlation among the acceptance indexes by analyzing the achievements of famous doctor studio in the First Affiliated Hospital of Guangzhou University of Traditional Chinese Medicine.Methods:R software was used to count the relevant data of national famous medical studios approved from 2010 to 2018, and the advantages were analyzed by radar chart; Spearman correlation coefficient was calculated, and R software was used to draw the correlation heat map between the evaluation indexes; papers published by studio members during the construction period were retrieved, which were transformed into refworks format, imported into CiteSpace software to draw a visual knowledge map.Results:A total number of 506 articles were included in this study, involving 25 evaluation indexes and 22 national famous doctors' studios. The statistical analysis results showed that there was a significant positive correlation between the number of academic experience papers of famous and senior TCM experts published in core journals (V3) and the number of diagnosis and treatment schemes of dominant diseases (V1), and there was a significant negative correlation between 8 groups of indexes; Tinnitus, insomnia, premature ovarian failure, acupuncture and acupuncture therapy are research hotspots.Conclusions:When evaluating the construction effect of famous doctor studio, various indicators should be considered comprehensively, appropriately control of the number of members is helpful to improve the quality of talent training, and pay attention should be paid to experience exchange and thought collision during the training process.
6.Serum antibody level and diagnostic value in convalescent patients infected with novel coronavirus

Objective:To investigate the levels of serum IgM, IgG, IgA and neutralizing antibodies (NAbs) in convalescing patients infected with the novel coronavirus and their diagnostic significance.Methods:Five hundreds and eighty one infected patients and 219 non-infected patients were detected by chemiluminescence method . The positive rate of antibody and antibody level were statistically analyzed for statistical significance. The correlation between the antibodies was compared, and the ability of each antibody to identify breakthrough infection in the vaccinated population was evaluated by combining ROC curve.Results:The antibody titers of IgM, IgG, NAbs and IgA in infected patients were higher than those in uninfected individuals. The antibody positive rate and antibody level of infected persons who were vaccinated and not vaccinated, with different ages, different doses of vaccines, and different intervals of vaccination were statistically significant ( P<0.05); IgA antibody levels were positively correlated with IgG and NAbs, and NAbs is positively correlated with IgG antibody; In breakthrough infection analysis, IgG, IgA and NAbs antibodies were good at identifying infected persons in the vaccinated population. Conclusions:The positive rate of IgM antibody is low in the convalescence period, which is of little significance for monitoring the course of COVID-19 and evaluating the efficacy of vaccines. In addition, the antibody level and positive rate can be significantly improved by the booster injection of COVID-19 vaccine. Differences in IgG, IgA, and NAbs antibody levels can be used to identify breakthrough infection cases from vaccinated populations.
7.Bibliometric analysis of literatures of researches on palliative care for neoplasms from 2009 to 2018 based on CNKI

Objective:To conduct the bibliometric analysis of researches on palliative care for patients with neoplasms from 2009 to 2018 based on China National Knowledge Internet (CNKI) so as to understand the research status and hot spots in this field and provide references for related researches.Methods:Chinese journal literatures related to palliative care for neoplasms published by CNKI were searched. Bibliographic Items Co-occurrence Matrix Builder (BICOMB) and Ucinet 6.0 software were used to analyze the number, year, journal, author and key words of the included literatures.Results:A total of 1 380 literatures were included, and the number of literatures was generally increasing year by year. The distribution of journals was scattered. There were 9 kinds of journals with published article volume of 20 or above, including 5 nursing journals. There were 1 265 first authors and no core author. 16 authors published 3 or more papers, and no cooperative relationship was formed. A total of 1 687 keywords were extracted from 1 380 literatures. In researches on palliative care for neoplasms in China, the hot groups were patients with advanced tumors, end-of-life patients and the elderly, and the single disease types involved were lung cancer, liver cancer, pancreatic cancer and stomach cancer. Research hotspots included quality of life, influencing factors, death education and symptoms of pain, depression, anxiety and cancer-related fatigue.Conclusions:The researches on palliative care for neoplasms in China is in the early stage of rapid development. It is necessary to strengthen cooperation, learn from foreign ideas and methods and promote the further development of domestic palliative care for neoplasms.

Nursing homes treat a large number of elderly people such as frail, dementia, disability, and advanced age. During the occurrence of major public health emergencies, it is a key institution for management, prevention and control. This paper proposes the concept of "resilient nursing homes" and analyzes the connotation and structure of resilient nursing homes based on the complex adaptive system theory. This paper also proposes strategies to improve the resilience of nursing homes in order to help nursing homes optimize their existing management systems and models, and improve their ability to respond to public health emergencies.
9.Rapid genetic characterization of a novel Enterobacteria phage and determination of its host recognizing genes.

We isolated a novel Enterobacteria phage IME08 from hospital sewage, then confirmed it was a double-stranded DNA phage by digesting its genetic material with DNase I, RNase A and several restriction endonucleases respectively. BLAST results of random fragments generated by a random PCR cloning method revealed that it belonged to T4-like virus. We subsequently determined the host recognizing genes (g37 and g38) sequence with a PCR-based "genome jumping" protocol based on highly conserved region at 5' terminus of g37 from four other T4-like Bacteriophages (T4, JS98, T2 and K3). These molecular biological methods enabled us to readily characterize the bacteriophage and efficiently determine the sequence of the genes of interest based on very limited conserved sequence information.

10.Expression of anti-avian influenza virus H5N1 secretory IgA in Chinese hamster ovary cells.

Secretory IgA (SIgA) antibodies in external secretions play an important role in mucosal immune response. Polymeric SIgA was advantageous over monomeric IgA (mIgA) and IgG in several aspects. To express secretory IgA antibody against H5N1 virus, we constructed the secretory component and immunoglobulin J expressing plasmids and co-transfected the plasmids into the Chinese hamster ovary cells (CHO) stably expressing immunoglobulin A. Then we used Zeocin to select the positive clone cells, monoclonal cells stably secreting SIgA was screened through fold dilution method at last. The SIgA antibody secreted from the CHO cells was confirmed by Western blotting, which demonstrated that we had got the complete SIgA molecular. The successful expression of this polymeric anti-H5N1 SIgA in CHO cells will contribute to the production of recombinant SIgA as a preventive agent for infectious disease control.
