Hepatocyte transplantation is an effective method for the end stage liver diseases. This procedure includes hepatoeyte isolation, culture, cryopreservation and so on. The cryopreservation is an important aspect for hepatocyte transplantation, which determines the viability after the resuscitation. The hepatocyte cryopreservation will be summarized in this passage.

Objective To translate the English version of an abbreviated eight-item measure of Penn State Worry Questionnaire (PSWQ-A) into Chinese and to test the reliability and validity of the Chinese version of PSWQ-A in patients after total Hip Replacement.Methods Totally 218 patients after total hip replacement were recruited and were investigated by the Chinese version of PSWQ-A.Results The internal consistency coefficient of the Chinese version of PSWQ-A were 0.885.Factor analysis got two factors,which explain 68.428％ of the total variance.The content validity index was 0.924.Using SAS as an external criterion,The correlation coefficients of the PSWQ-A and SAS scores were 0.891,P＜0.05.Conclusions The Chinese version of PSWQ-A has been proved to be reliable and valid.It can be used as a valid tool for the measurement of self-efficacy in patients after total hip replacement.

Metformin has been applied for 50 year.In recent years,new studies found that metformin could reduce glucose through activating inhibition of dipeptidyl peptidase Ⅳ(DPP Ⅳ) and enhancing the biological effect of glucagon-like peptide-1(GLP-1).In addition,metformin can reduce vascular disease risk of patients with type 2 diabetes by improving insulin resistance and the effect of reducing total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),blood coagulation factor Ⅶ,plasminogen activator inhibitor-1(PAI-1) and proteinuria.Merformin can't change blood pressure of patients with type 2 diabetes,however,it can significantly reduce the salt-sensitive blood pressure of patients with obesity.Metformin has also been applied to patients with polycystic ovary syndrome(PCOS).It can induce ovulation,decrease the rate of abortion and have effect on hypertrichosis and hyperandrogenism.Besides,metformin has been the focus of the treatment of non-alcoholic fatty liver disease.

Objective To investigate the relationship between vitamin D level in plasma and concomitant symptoms in children over 4 years of age. Methods Radioimmunoassay was employed to detect the vitamin D level in plasma of 98 children over 4 years of age suffering from monoarticular joint-pain, debility, dizziness and hyperhidrosis. Results Among the 98 cases, 15 cases (rickets) were found to have a vitamin D level of 0-10ng/ml, 26 cases (subclinic rickets) with a level of 10.1-15ng/ml, 55 cases with a level of 15.1-60ng/ml, and the remaining 2 were found to have a level of 61-86ng/ml. A significant positive correlation existed between the low vitamin D level in plasma and the symptoms of joint-pain, debility, dizziness, and hyperhidrosis. Conclusion It is of a diagnostic significance to determine the 25-hydroxyvitanmin D_3 level in plasma to confirm the diagnosis of delayed vitamin D deficiency rickets in children.

Objective:To analyze clinical and pathological features of Reed nevi (pigmented spindle cell nevi) .Methods:The clinical and pathological data were collected from 26 patients with Reed nevi, who visited Department of Dermatology of Xijing Hospital from January 2015 to June 2019, and analyzed retrospectively.Results:Among the 26 patients, 15 were males, and 11 were females. Their average age of onset was 17.35 years, and median age of onset was 13.85 years. Reed nevi occurred predominantly on the lower limbs (12 cases) , followed by the face (6 cases) , upper limbs (5 cases) and trunk (3 cases) . All the skin lesions were black in color, and 7 cases presented with maculae, 19 with flat papules, 22 with quasi-circular skin lesions, and 4 with irregular skin lesions. The diameter of skin lesions ranged from 2 to 10 mm, and 23 cases had skin lesions with a diameter of ≤ 5 mm. As histopathological examination showed, pathological subtypes included junctional nevus (15 cases) and compound nevus (11 cases) ; melanocytes were spindle-shaped with obvious cytochromes in 26 cases; nevus cell nests merged with the surrounding epidermis in 4 cases, and obvious fissures were formed between nevus cell nests and the surrounding epidermis in 22 cases; no obvious cell atypia or mitotic figure was observed in any of the 26 cases, and Kamino bodies were observed in 6 cases. All the skin lesions were resected by surgery, and no recurrence was observed during the follow-up of 1 - 5 years.Conclusion:Reed nevi can manifest as irregular lesions, pagetoid distribution of spindle cells of various sizes can be observed histopathologically, and they need to be differentiated from melanoma.

Objective To evaluate the effects of sevoflurane preconditioning on zonula occludens-1 (ZO-1) during myocardial ischemia-reperfusion (I/R) in rats in vitro.Methods Adult male Wistar rats,weighing 250-300 g,were anesthetized with intraperitoneal pentobarbital 30 mg/kg and heparinized.Their hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95％ O2-5％ CO2 at 37 ℃.Eighteen isolated rat hearts were randomly assigned into 3 groups (n =6 each) using a random number table:control group (group C),group I/R and sevoflurane preconditioning group (group S).At 10 min of equilibration,the hearts were perfused with K-H solution for 110 min in group C,the hearts were perfused with K-H solution for 20 min,and then subjected to 30 min of ischemia followed by 60 min of reperfusion in group I/R,and the hearts were perfused with K-H solution saturated with 3％ sevoflurane for 15 min followed by 5 min washout,and then subjected to 30 min of ischemia followed by 60 min of reperfusion in group S.At the end of equilibration,immediately before ischemia,and at 15 and 60 min of reperfusion (T1,2),HR,left ventricular end-diastolic pressure (LVEDP),left ventricular developed pressure (LVDP),+ dp/dtmax and-dp/dtmax were recorded.The development of arrhythmias was recorded during reperfusion and scored.At 60 min of reperfusion,myocardial specimens were obtained from the apex of heart for determination of the expression of ZO-1 in myocardial tissues (by Western blot) and for observation of distribution of ZO-1 and connexin43 (Cx43) (by immunofluorescence).Results Compared with group C,HR,LVDP,+ dp/dtmax and-dp/dtmax were significantly decreased and LVEDP was increased at 15 and 60 min of reperfusion,scores of arrhythmia was increased,and ZO-1 expression was down-regulated in I/R group.Compared with group I/R,HR,LVDP,+ dp/dtmax and-dp/dtmax were significantly increased and LVEDP was decreased at 15 and 60 min of reperfusion,arrhythmia was decreased,and ZO-1 expression was up-regulated in group S.ZO-1 and Cx43 were co-localized at the intercalated disk.ZO-1 was redistributed in the lateralization of the membrane and co-localized with Cx43 in group I/R.The incidence of ZO-1 lateralization was significantly decreased in group S.Conclusion The mechanism by which sevoflurane preconditioning decreases reperfusion arrhythmia is related to inhibition of down-regulation of expression and redistribution of ZO-1 and inhibition of redistribution of Cx43 in rats.

Objective To investigate the value of end-tidal carbon dioxide partial pressure (PET CO2 )in fluid resuscitation in severe acute pancreatitis(SAP)patients.Methods SAP patients under mechanical ventilation with the need of a fluid challenge test were included.Hemodynamic parameter cardiac index(CI)and PET CO2 were conducted before and after the fluid challenge test.The value of ΔPET CO2 was used to predict fluid responsiveness. Results Totally 43 patients with SAP were prospectively recruited.31 patients had volume responsiveness, 12 patients had no volume responsiveness.Compared with no volume responsiveness group,volume responsiveness group led to a greater increase in ΔCI[(0.9 ±0.3)vs.(0.2 ±0.3),t =3.24,P <0.05]and ΔPET CO2 [(4.1 ± 1.9)vs.(0.7 ±1.2),t =4.01,P <0.05].ΔPET CO2 and ΔCI were correlated(r =0.74,P <0.05).The area under ROC curve of ΔPET CO2 was 0.872(95% CI 0.754 ~0.923,P <0.05).An increase of 5% in ΔPET CO2 predicted fluid responsiveness with a sensitivity of 86.7%,and specificity of 89.5%.Conclusion The change of ΔPET CO2 induced by fluid challenge test is an effective way to predict fluid responsiveness in SAP patients.

OBJECTIVE:To establish the method for the simultaneous determination of 4 flavonoids in rat plasma sample, such as quercetin-3-O-sophoroside,isoquercitrin,hyperin,rutin. METHODS:8 SD rats were selected and given Poacynum hender-sonii leaf extract suspension intragastrically 0.1 g/kg(calculated by extract). The blood sample 1 mL was collected from orbit 1 h after medication. The contents of 4 flavonoids were determined by microemulsion liquid chromatography(MELC)after centrifuga-tion. The separation was performed on Zorbax SB-C18 column with mobile phase consisted of microemulsion [polyoxyethylene lau-ryl ether-n-butanol-ethyl acetate-triethylamine-water (volume ratio was 2.5:3.0:1.7:0.3:92.5,pH=5.0)] at the flow rate of 0.8 mL/min. The column temperature was set at 30 ℃,and detection wavelength was 360 nm. Sample size was 10 μL,and internal standard was bergenin. It was compared with HPLC method using organic solvent as mobile phase(mobile phase consisted of 0.2%phosphoric acid solution-acetonitrile,gradient elution,other condition same as MELC method). RESULTS:In rats plasma,the lin-ear range of quercetin-3-O-sophoroside,isoquercitrin,hyperin,rutin were 10-1000 ng/mL(r≥0.9980). The limits of quantitation was 10 ng/mL(S/N=10). RSDs of precision,stability and reproducibility tests were all below 7.5%(n=5). Average sample recov-eries were between 97.4%-98.5% and RSDs were less than 5.3%(n=5). Extraction recoveries were between 88.7%-100.3%(RSD≤6.14%,n=5). The methodological evaluation results of MELC and HPLC method were all in line with the regulations of pharmacopeia,and the results of blood concentration betweenthem were similar. Compared with HPLC method, MELCmethod could shorten detection time(10 min vs. 40 min)andreduce the amount of organic solvent (0.38 mL vs. 28 mL).CONCLUSIONS:Established MELC method is rapid,simpleand green,and can be used for 4 flavonoids from P. henderso-nii leaf in rat plasma sample.

Objective To evaluate the effect of sevoflurane postconditioning on the expression of Pim-1 kinase during myocardial ischemia-reperfusion (I/R) in rats.Methods Male Sprague-Dawley rats,weighing 250-300 g,were used in this study.After the animals were anesthetized,their hearts were immediately removed and retrogradely perfused with an oxygenated K-H solution at 37 ℃ in a Langendorff apparatus.Thirty-six isolated rat hearts were assigned into 3 groups (n=12 each) using a random number table:control group (group C),group I/R and sevoflurane postconditioning group (group SP).The hearts were subjected to ischemia for 30 min followed by 120 min of reperfusion to establish the model of myocardial I/R injury.In group SP,the hearts were perfused with K-H solution saturated with 3％ sevoflurane for 15 min starting from the beginning of reperfusion.Heart rate (HR),left ventricular end-diastolic pressure (LVEDP),left ventricular developed pressure (LVDP) and the maximum rate of increase and decrease of left ventricular pressure (±dp/dtmax)were recorded at the end of equilibration and 30,60,90and 120 min of reperfusion.Myocardial tissues were obtained at T2 for determination of the expression of Pim-1 kinase,Bcl-2 and cytochrome c (Cyt c) in cytoplasm and mitochondria by Western blot.The hearts were selected at T4 for determination of myocardial infarct size (by TTC staining) and for examination of mitochondrial ultrastructure (with transmission electron microscope).Results Compared with group C,HR,LVDP and ±dp/dtmax were significantly decreased,and LVEDP was increased at T1-4,the myocardial infarct size was enlarged,the expression of Pim-1 and Bcl-2 in cytoplasm and mitochondria was down-regulated,the expression of Cyt c in cytoplasm was up-regulated,and the expression of Cyt c in mitochondria was down-regulated in group I/R (P＜0.05).Compared with group I/R,HR,LVDP and ±dp/ dtmax were significantly increased,and LVEDP was decreased at T1-4,the myocardial infarct size was decreased,the expression of Pim-1 kinase and Bcl-2 in cytoplasm and mitochondria was up-regulated,the expression of Cyt c in cytoplasm was down-regulated,and the expression of Cyt c in mitochondria was upregulated in group SP (P＜0.05).The damage to mitochondria was significantly attenuated in group SP as compared with group I/R,and was aggravated as compared with group C.Conclusion The mechanism by which sevoflurane postconditioning attenuates myocardial I/R may be related to up-regulation of Pim-1 kinase expression in rats.
