1.Drug Consultation Calls from 2001 to 2008 in Clinical Pharmacy of Our Hospital:A Retrospective Analysis

OBJECTIVE:To investigate the status quo of the pharmaceutical care of drug consultation calls so as to facilitate the development of pharmaceutical care and promote rational drug use.METHODS:All the recordings on the pharmaceutical consultation calls from Jan.2001 to Sep.2008 were collected to analyze the compositions of consultants,problems consulted and the categories of the drugs consulted,etc.RESULTS:75.6% of the consultants were hospital employees;41.7% of the problems consulted were related to drug interactions and dosage and administration,and 22.8% of the drugs consulted were antibiotics.CONCLUSION:The drug consultation call model is convenient and feasible,and is proved to be a realistic professional pharmaceutical care.
2.Expression of FOXM1 transcription factor and its clinical significance in cervical cancer

Objective To detect the expression of FOXM 1 transcription factor in cervical cancer tissues and to evaluate its clini-cal significance .Methods Immunohistochemical staining was employed to detect the expression of both FOXM 1 and Ki-67 in a va-riety of cervical tissues respectively from 38 patients with cervical cancer ,22 with cervical intraepithelial neoplasia (ranging from CINⅠ to CIN Ⅲ) and 17 with normal cervical epithelium .Results Abnormal expression rate of FOXM 1 was respectively 5 .88% , 63 .6% and 92 .1% in normal cervix ,CIN and cervical cancer ;Deregulated FOXM1 expression in cervical cancer tissues was signifi-cantly correlated with patients’ pathological differentiation ,clinical stage ,post-operational recurrence and Ki-67 expression level . Conclusion FOXM1 expression may be correlated with the deregulated proliferation ,malignant progression ,staging and prognosis in cervical cancer .

4.Synthesis of peptide of BALF4 gene of Epstein-Barr virus and application in the detection of nasopharyngeal carcinoma

Objective To analyze the application of recombinant gp125 proteins in serological diagnosis of nasopharyngeal carcinoma (NPC). Methods B cell epitopes of EBV gp125 protein were forecasted by internet server and the peptides were synthesized and coated to microplate for the detection of EBV-IgA antibody in NPC patients and healthy donors by ELISA. Results Six peptides were synthesized, wherein two showed low specificity and others displayed significantly higher positive rate in the NPC group than in the healthy group (P <0.001). Conclusion The BALF4 gene peptides are successfully synthesized and epitopes with high specificity and strong immunogenicity are screened for the preparation of gp125 antigen with high specificity and sensitivity.
5.Efifcacy of thalidomide combined with VAD in treatment of Elderly multiple myeloma and its adverse reactions

Objective To investigate the efifcacy of thalidomide combined with VAD in treatment of elderly multiple myeloma(MM). Methods 76 cases of elderly MM were randomly divided into two groups, control group (n=38) were treated with VAD alone while the observation group (n=38) were treated with thalidomide and VAD. Clinical efifcacy and adverse reactions in two groups were compared. Results The total effective rate in observation group was 84.21%, which was signiifcantly higher than 55.26%in control group (P<0.05). After treatment, the M protein, myeloma cells andβ2-microglobulin in observation group were (20.77±4.15)×10-2μg/mL, (12.84±2.85)×10-2μg/mL and (2.48±0.53)μg/mL, which were signiifcantly lower than before treatment and control group(P<0.05). Hemoglobin in observation group was (108.83±5.81) g/L, which was signiifcantly higher than before treatment and control group(P<0.05). The adverse reactions were nausea, vomiting, drowsiness, constipation, infection and rash and so on in two groups after treatment, and there was no signiifcant difference between two groups. Conclusion Thalidomide combined with VAD regimen has a better effect in treatment of elderly multiple myeloma than single VAD, has less adverse reactions and well tolerated.
6.Preparation and identification of artificial antigen for rhein.

The Rhei Radix et Rhizoma was one of the most widely used traditional Chinese medicine for its special biological activities. The content of rhein, one of its major compounds, was an important standard for the quantity control of Rhei Radix et Rhizoma. The major method used for the detection of rhein was instrumental analysis like HPLC, but it was complex, time-consuming and cannot detect large samples at the same time. The enzyme-linked imunmosorbent assay (ELISA) was accurate, reliable, simple, low costs, and of a high-throughout. Recently, it was widely used for the determination of those small molecule compounds in some traditional Chinese medicinal plants. In this study, an artificial antigen were synthesized by the carbodiimide (CDI) method. Rhein-bovine (rhein-BSA) conju gate and rhein-ovalbumin (rhein-OVA) conjugate, were produced as the immunogen and coating antigen, respectively. The conjugate and the hapten number in the conjugate were determined by UV-Vis spectrophotometry (UV). The conjugation ratio of Rhein and BSA was about 4.0:1, rhein acid and OVA was 2.6 : 1, respectively. Rhein-BSA conjugate was used to immunize Bal b/c mice to produce antiserum. The antiserum titer of the Rhein were higher than 8000 detected by ELISA. The successfully synthesized conjugate antigen rhein-BSA implies its feasibility in the establishment of fast immunoassay for the rhein content determination.

8.Expression of hypoxia-inducible factor 1 alpha and core binding factor alpha 1 in rat models of femoral fracture combined with cerebral trauma

BACKGROUND:The low oxygen environment after femoral fracture and cerebral trauma wil induce series of related cytokines expression, including hypoxia-inducible factor 1αand core binding factorα1, which play key roles in regulating bone healing. However, whether the accelerated bone healing is correlated with the expression of hypoxia-inducible factor 1αand core binding factorα1 is stil unknown.
OBJECTIVE:To construct rat models of brain injury, to compare the expression level of hypoxia-inducible factor 1αand core binding factorα1 in femoral fracture combined with cerebral trauma rats and simple femoral fracture rats, and to assess the influence of cerebral trauma on bone healing.
METHODS:Rats were randomly divided into blank group, simple femoral fracture group and femoral fracture combined with cerebral trauma group. At 1, 2, 3 and 5 weeks after modeling, rats were executed. Bone healing was evaluated using femoral fracture end X-ray score and hematoxylin and eosin staining at cal us tissues. Besides, the expression levels of hypoxia-inducible factor 1αand core binding factorα1 of three groups were determined with immunohistochemistry.
RESULTS AND CONCLUSION:Bone healing in the femoral fracture combined with cerebral trauma group was better than that of simple femoral fracture group. There was significant difference in the expression level of hypoxia-inducible factor 1αand core binding factorα1 between the simple femoral fracture group and femoral fracture combined with cerebral trauma group (P<0.05). At the same time, the level of simple femoral fracture group and femoral fracture combined with cerebral trauma group was significantly higher than that of blank group, and that in femoral fracture combined with cerebral trauma group was significantly higher than that of simple femoral fracture group (P<0.05). Results verified that the expression levels of hypoxia-inducible factor 1αand core binding factorα1 of rats with femoral fracture combined with cerebral trauma were significantly high, which may be the major reason why the bone healing was accelerated after fracture combined with brain injury.
9.Analysis of the function of vascular endothelial cells in coronary heart disease patients of blood-stasis syndrome.

OBJECTIVETo explore the function of vascular endothelial cell (VEC) in patients with coronary heart disease (CHD) of Xin-blood-stasis syndrome.
METHODSSome vasoactive substances produced by VEC were detected and analyzed in patients with CHD of or without Xin blood stasis syndrome in group A (n=112) and group B (n=108) respectively, also in patients with non-CHD but of Xin-blood-stasis syndrome in group C (n=110), and healthy persons in group D (n=100), including nitric oxide (NO), endothelin (ET), angiotensin H (Ag II), soluble intercellular adhesion molecule-1 (sICAM-1) and soluble vascular cell adhesion molecule -1 (sVCAM-1).
RESULTSThe abnormality degree of ET, Ag II , sICAM-1 and sVCAM-1 in various groups showed such a tendency as group A> group B> group D (P < 0.01 or P < 0.05), while no significant difference in these criteria between group A and group C was shown (P > 0.05).
CONCLUSIONThe vasoactive substances secreted by VEC are closely related to the formation and progression of CHD, and are likely to be important pathological markers of blood-stasis syndrome in CHD.

10.Effect of fibrous root extract of Coptis chinensis on soil microbes and enzyme activities.

Coptis chinensis is widely used as Chinese medicine herbs and serious soil problems occur after continual cultivation of this medicinal plant. In the preset experiment, fibrous root extract of C. chinensis (REC) was added into soil to study the effect of REC on microbes and enzyme activity in soil. The results showed that both bacteria and actinomycetes decreased by about 2 times in contrast to fungi, which increased by about 3 folds. Phosphorus bacteria, potassium bacteria, azotobacter, ammonia bacteria, and nitrifying bacteria were also reduced significantly by REC, suggesting the inhibition of nitrogen biofixation and supply, mobilization of phosphorus and potassium, ad plant growth promotion as REC added into soil. There were multiple influences of REC on soil enzyme activities. Invertase activity was stimulated, while urease was inhibited and dehydrogenase unchanged by REC, indicating the interference of biochemical reactions in soil. In addition, type and total content of phosphorus lipid fatty acids (PLFAs) , the signature of microbes, decreased while the ratio of bacterium to fungus PLFAs increased as REC increased in soil, which suggested that fungi increased relatively with bacteria decreased thereby leading to easy occurrence of crop fungus diseases following cultivation of C. chinensis. The decrease in diversity and evenness indexes of microbial community in soil by REC indicated soil ecosystem deterioration and reduction of microbial groups and densities in soil. Therefore, allelopathic chemicals released from the roots of C. chinensis could change microbial community structure and resulted in serious soil problems by continual cropping of this medicinal plant.
