Hematopoietic stem cell transplantation (HSCT) is the only effective curative therapy for patients with certain hematological diseases. Haploidentical HSCT has been practiced worldwide for approximately twenty years. Significant improvements on this therapy were made in China within the past decade, with the novel non-in-vitro, T-cell-depleted haploidentical HSCT system. This review de-scribes the current situation and provides a prospective overview of these novel HSCT systems in China.

Antibody drug conjugates (ADCs) are an emerging class of targeted therapeutics with the potential to improve therapeutic index over the traditional chemotherapy. However, it is difficult to control the site and stoichiometry of conjugation in mAb, typically resulting in heterogeneous mixtures of ADCs that are difficult to optimize. New methods for site-specific drug attachment allow development of more homogeneous conjugates and control of the site of drug attachment. In this article, the new literature on development of ADCs and site-specific ADCs is reviewed. In addition, we summarized the various strategies in production of site-specific ADCs.

4.Protective effects of bone marrow mesenchymal stem cells against the renal tubular epithelial cell damage induced by cisplatin

AIM:To investigate the protective effects and mechanism of mesenchymal stem cells on renal tubule cells treated by cisplatin.METHODS:The mesenchymal stem cells(MSCs)were separated from mouse bone marrow.The necrosis and apoptosis rate of renal tubule cells induced by cisplatin were detected by flow cytometry.After co-culture with MSCs,the above changes of renal tubule cells were detected.RESULTS:In co-culture system,the apoptotic rates of renal tubule cell were decreased obviously and the cell number increased obviously.CONCLUSION:The MSCs protect the renal tubule cells from apoptosis induced by cisplatin.This effect may be related with the paracrine effects of MSCs.
5.Relationship Between Syndrome Differentiation of Diabetic Retinopathy and Pattern of Visual Evoked Potential

Objective To explore the relationship between syndrome differentiation of diabetic retinopathy (DR) and pattern of visual evoked potential (P-VEP).Methods One hundred and eighty-six eyes from 104 cases of DR were allocated to three groups according to TCM syndrome:qi-yin deficiency (Group A),qi stagnation and blood stasis (Group B) and yin-yang deficiency (Group C).P-VEP induced by various space frequency (0.388?Hz,1.55?Hz) and pictures of various colors (black/white,red/magenta,blue/green) was observed in the three groups and compared with that in the normal control group (Group D).Results One hundred and three eyes in Group A mainly manifested as simple DR,47 eyes in Group B and 36 eyes in Group C mainly manifested as proliferative DR.The latency of P 100 was prolonged and the amplitude was decreassed in all groups of DR,but the latency of P 100 was much prolonged in Group B,the amplitude was much decreased in Group C and the changes were less in Group A.Conclusion P-VEP varies in different TCM syndrome types of DR,and determination of P-VEP under various stimulating conditions may serve as an index of syndrome differentiation of DR.

Objective To investigate the role of interferon ?on smooth muscle cells proliferation and migration after balloon injury. Methods Animal model of rabbit iliac artery balloon injury was set up, smooth muscle cells derived from injured artery were cultured. Smooth muscle cells were divided into four groups ( control, TGF ? 1, IFN ?, TGF ? 1 and IFN ?).Cells from each group were treated with medium or TGF ? 1 (10 ng/ml) and/or IFN ?(500 u/ml) for 72 h separately. Smooth muscle cell proliferation was determined by cell count and MTT, migration of smooth muscle cells was also detected. Matrix metalloproteinase 2 was also detected by zymography. 8.Oral ganciclovir for treatment of cytomegalovirus viremia in patients receiving allogeneic stem cell transplantation

Objective To investigate the efficacy and safety of oral ganciclovir on cytomegalovirus viremia in patients receiving allogeneic stem cell transplantation.Methods Thirty patients who were treated with oral ganciclovir from Feb,2006 to May,2006 were studied.Intravenous ganciclovir was used 10 mg/(kg?d)for eight days before HSCT as prophylaxis of CMV infection.Cytomegalovirus was detected after HSCT once a week using polymerase chain reaction(PCR)method.If CMV was positive and CMV-DNA load was higher than 6.0?102 copies/mL and lower than 105 copies/mL,oral ganciclovir was used for the treatment(1 g,three times a day).Results CMV viremia was positive at a median time of 42 days after HSCT;median CMV-DNA load was 4.626?103 copies/mL.Good response on CMV DNA load(reduction below 6.0?102 copies/mL)was observed in 90% of oral ganciclovir and the response rate was 70.0% within 14 days.Median duration of therapy for good response was 10 days(range 2~30 days).Severe adverse effects were not observed and CMV-related disease did not occur.Conclusion Oral ganciclovir is an attractive and safe alternative for pre-emptive CMV viremia treatment in allo-HSCT recipients.
9.Sensitization effect of insulin of chemotherapy on human adenocarcinoma cells A549

Objective To explore the sensitization effect of insulin to chemotherapy as a metabolic promoter on human lung adenocarcinoma cells A549.Methods The sensitivity of human adenocarcinoma cells A549 to DDP was observed by MTT methods,and IC30 values of the cells were calculated.The optimal concentration of insulin to promoting A549 cells' growth was determined by MTT assay.The A549 cells were pretreated with insulin of various concentrations for different time period,and then the cell sensitivity to DDP was evaluated with MTT assay.Flow cytometry was used to study the cell cycle distribution of the A549 cells treated with insulin.Results The IC30 value of DDP was about 36.87?g/ml.It was found that the cell growth was significantly promoted with pretreatment of insulin at the concentration of 4~16mU/ml for 8~16 hours.Insulin enhanced the inhibitory effect of DDP(36.87?g/ml)on cell proliferation when human lung adenocarcinoma cells A549 were treated with insulin(2.0~16.0mU/ml;8~16h)as indicated by MTT assay(P
10.Detection of HPV-DNA and SCCAg in the Follow-up of Cervical Squamous Cell Carcinoma Patients

Objective To investigate the infection of human papilloma virus(HPV) and expression of serum squamous cell antigen (SCCAg) in the follow - up of 120 cervical squamous carcinoma patients who had received operation or operation combined with radiotherapy or only radiotherapy. Methods 120 cases of therapical cervical squamous carcinoma patients were detected HPV - DNA by HC - Ⅱ and serum SCCAg using immunohistochemical method. Results The positive rate of HPV - DNA and serum SCCAg was 49.17% and 17.50% , with a significant difference between them( P 0.05 ). Conclusion HPV - DNA test with HC - Ⅱ for follow - up of cervical squamous carcinoma patients was feasible. It was more sensitive than serum SCCAg. But it suggested that high risk type HPV -DNA test combined with serum SCCAg may be the independent prognostic factors.
