Hematopoietic stem cell transplantation (HSCT) is the only effective curative therapy for patients with certain hematological diseases. Haploidentical HSCT has been practiced worldwide for approximately twenty years. Significant improvements on this therapy were made in China within the past decade, with the novel non-in-vitro, T-cell-depleted haploidentical HSCT system. This review de-scribes the current situation and provides a prospective overview of these novel HSCT systems in China.

3.Leiomyosarcoma with coronary fistulae and ventricular septal perforation:A case study

Coronary fistulae and ventricular septal perforation are very rare clinically, and even less caused by cardiac leiomyosarcoma. A case is reported that a 67-year-old female had cardiac leiomyosarcoma with progressive heart failure and coronary fistulae and ventricular septal perforation. This case was special since all ante-mortem examinations and cardiac surgery failed to detect the presence of any abnormal car-diac mass. Therefore, the malignant cardiac tumors could appear in an invasive form without mass and be one of the causes of the coronary fistulae and ventricular septal perforation.
4.Effect of excessive iodine intake on bone metabolism in male rats

Objective To explore possible poisonous and harmful effect of excessive iodine intake on skeleton in male rats. Methods Male rat models receiving various amounts of excessive iodine were established for 3,6, 12 months, and quantitative bone histomorphometric parameters and bone mineral density (BMD) of femur and lumbar spine( L1-1 ) were measured by means of dual-energy X-ray absorptimetry in vitro, and rats receiving normal diet were considered as control group. Results There was no significant difference in bone mineral quantity and structural parameters after feeding excessive iodine for 3 months in comparison with control group. 5.Oral ganciclovir for treatment of cytomegalovirus viremia in patients receiving allogeneic stem cell transplantation

Objective To investigate the efficacy and safety of oral ganciclovir on cytomegalovirus viremia in patients receiving allogeneic stem cell transplantation.Methods Thirty patients who were treated with oral ganciclovir from Feb,2006 to May,2006 were studied.Intravenous ganciclovir was used 10 mg/(kg?d)for eight days before HSCT as prophylaxis of CMV infection.Cytomegalovirus was detected after HSCT once a week using polymerase chain reaction(PCR)method.If CMV was positive and CMV-DNA load was higher than 6.0?102 copies/mL and lower than 105 copies/mL,oral ganciclovir was used for the treatment(1 g,three times a day).Results CMV viremia was positive at a median time of 42 days after HSCT;median CMV-DNA load was 4.626?103 copies/mL.Good response on CMV DNA load(reduction below 6.0?102 copies/mL)was observed in 90% of oral ganciclovir and the response rate was 70.0% within 14 days.Median duration of therapy for good response was 10 days(range 2~30 days).Severe adverse effects were not observed and CMV-related disease did not occur.Conclusion Oral ganciclovir is an attractive and safe alternative for pre-emptive CMV viremia treatment in allo-HSCT recipients.
6.Effect of recombinant human interleukin-11 on neutron irradiation injury of small intestine in mice

Objective To evaluate the effect of rhIL-11 on radiation injury to the intestinal epithelium and cells cycle of intestinal epithelial cell in mice irradiated with 3.5 Gy neutron. Methods The morphology of the small intestinal epithelium, crypt cells necrosis, and cell proliferation were observed of the epithelial cells of the irradiated mice. Cell cycle of the epithelial cells of the small intestine of the mice was examined by flow cytometry. Results rhIL-11 pretreatment before and treatment after irradiation could accelerate the repair of small intestinal mucosa in irradiated mice. G 2/M block which occurred in the irradiated small intestinal epithelial cells and the rhIL-11 treatment might significantly increased the proportion of cells at S phase. Conclusion rhIL-11 could significantly exert a preventive effect on the small intestine against radiation injury in neutron irradiated mice, with an impact on cell cycle of the intestinal epithelial cells.
7.Long-term effects of articular cartilage derived scaffold/autologous chondrocytes composites for repairing full-thickness defects of knee articular cartilage in rabbits

[Objective]To observe the histological characteristics of autologous chondrocytes/human articular cartilage derived scaffold (HACDS) composites for repairing chondral defects in rabbit models. [Methods]A full-thickness articular-cartilage defect (diameter 4 mm,thickness 2 mm) was created in the femoral condyle of rabbits. The rabbits were divided into two groups: control group,HACDS scaffold only,and experimental group,chondrocytes/HACDS scaffold. Fifteen months after implantation,the specimens were observed by inverted phase contrast microscopy,and assessed by staining with haematoxylin-eosin,alcian blue,toluidine blue,safranin O,as well as by the immunohistochemistry of collagen type Ⅱ.[Results]Histologically,the generated neo-cartilage integrated well with its surrounding normal cartilage and subchondral bone in the defects of experimental group at 15 months post-implantation,whereas only fibrous tissue was filled in the defects of control group. Histochemical and immunohistochemical analysis revealed that alcian blue,toluidine blue,safranin,and were obviously positive in the experimental groups. However,alcian blue,toluidine blue,and safranino O were negative in the control group.[Conclusion]The current histological examination demonstrated that an engineered cartilage composed of autologous chondrocytes on HACDS scaffold could be successfully obtained and further applied to repair an articular cartilage defect in a rabbit model.

9.Protective effects of bone marrow mesenchymal stem cells against the renal tubular epithelial cell damage induced by cisplatin

AIM:To investigate the protective effects and mechanism of mesenchymal stem cells on renal tubule cells treated by cisplatin.METHODS:The mesenchymal stem cells(MSCs)were separated from mouse bone marrow.The necrosis and apoptosis rate of renal tubule cells induced by cisplatin were detected by flow cytometry.After co-culture with MSCs,the above changes of renal tubule cells were detected.RESULTS:In co-culture system,the apoptotic rates of renal tubule cell were decreased obviously and the cell number increased obviously.CONCLUSION:The MSCs protect the renal tubule cells from apoptosis induced by cisplatin.This effect may be related with the paracrine effects of MSCs.
10.Advances in the study of site-specific antibody-drug conjugates.

Antibody drug conjugates (ADCs) are an emerging class of targeted therapeutics with the potential to improve therapeutic index over the traditional chemotherapy. However, it is difficult to control the site and stoichiometry of conjugation in mAb, typically resulting in heterogeneous mixtures of ADCs that are difficult to optimize. New methods for site-specific drug attachment allow development of more homogeneous conjugates and control of the site of drug attachment. In this article, the new literature on development of ADCs and site-specific ADCs is reviewed. In addition, we summarized the various strategies in production of site-specific ADCs.
