1.Construction of eukaryotic expression vectorinsertedby MafA and its expression in HepG-2 cells
Journal of Chongqing Medical University 2007;0(08):-
Objective:To construct eukaryotic expression vectors pEGFP-N2/MafA and pEGFP-C1/MafA,encoding the mouse musculus v-maf musculoaponeurotic fibrosarcoma oncogene family, protein A(MafA)gene, for the further research of its expression in HepG-2 cells. Methods:pRNA of the mouse MafA gene was distilled by RT-PCR from the mouse, and inserted into Hind Ⅲand SalⅠrestriction sites by PCR,then cloned into pEGFP-N2 and pEGFP-C1 vectors to obtain plasmids pEGFP-N2/MafA and pEGFP-C1/MafA.Human liver cancer cells(HepG-2) were transfected with formed plasmids by means of lipidosome. The MafA-EGFP fused protein was viewed directly with fluorensce microscope, and expression of MafA and insulin Ⅱ was detected by RT-PCR.Results:The mouse MafA gene was amplified through RT-PCR and successfully cloned into transfer vectors. The favorite gene sequence could be expressed and was completely consistent with that reported in genebank, but the insulin Ⅱ gene expression was not detected. Conclusion:The recombinant plasmids pEGFP-N2/MafA and pEGFP-C1/MafA were successfully constructed. But the insulin Ⅱgene was not expressed.

4.Effect and function analysis of sulfentanyl on tongue cancer in patients with postoperative analgesia
Chinese Journal of Biochemical Pharmaceutics 2016;36(4):92-94
Objective To investigate the effect and function analysis by sulfentanyl in patients with tongue cancer postoperative analgesia. Methods A total of 74 patients with radical resection of tongue cancer in department of anesthesiology with general anesthesia from our hospital were collected, patients or their families signed consent, according to postoperative self-controlled intravenous analgesia drugs divided into experimental group and control group with 37 cases in each group.Patients in control group were treated by fentanyl self-controlled intravenous analgesia;patients in experimental group were treated by sulfentanyl self-controlled intravenous analgesia, determination of the analgesic effect, sedative effect and the changed of vital signs postoperative 2, 4, 8, 12, 24, 48 h,at the same time recorded complications. Results After operation, at different time points analgesic effect and sedative effect were changed in the two groups, the VAS scored of the experimental group was lower than the control group postoperative 2, 4, 8, 12, 24, 48 h time points, the Ramsay Sedation scored of the experimental group was lower than the control group postoperative 2 h, 4 h, 8 h, 12 h time points ( P <0.05 ); the changed of vital signs and adverse reactions between the two groups had no difference within 48 hours ( P >0.05 ) . Conclusion Sulfentanyl in the treatment of patients with radical resection of tongue cancer the analgesic effect and sedative effect are better than fentanyl, has no effect on the vital signs, and the role is security and exact.
5.Changes of Protein Metabolism in Rat Liver after Scaldingand Effects of Insulin
Journal of Third Military Medical University 1986;0(S1):-
Full thickness scalding of 37% TBSA was inflicted io rats. Three days later, the liver of the animals was isolated and the changes of the protein metabolism in the liver and the effects of insulin on the changes were evaluated. It was found that the protein and DMA levels of the liver showed no obvious changes after scalding, but the RNA content increased by 24% and the rate of leucine incorporation by 31%. After the addition of insulin, the leucine content increased by 23.% and 25.8% in the controls and the injured animals respectively. The results indicate that the rate of protein synthesis in the liver of the scalded animals increases, which is likely to be related to the increase of RNA content, and the liver protein synthesis remains to be sensitive to insulin.
6.Changes of protein Metabolism in Rat Soleus Muscle after Burus and Effects of Leucine and Insulin
Journal of Third Military Medical University 1984;0(01):-
This paper is to report the changes of the protein metabolism in the rat soleus muscle in vitro after 37% TBSA full-thickness burns covering one hindlimb and the effects of leucine and insulin on the burned muscle. It was found that there werea significant decreases of the protein content and marked exaggeration of the rates of tyrosine incorporation, tyrosine release and tyrosine net release in the burned muscle. No changes of the above-mentioned parameters could be revealed in the muscle of the unburned limb and the controls.Elevation of leucine concentration in the medium could not improve the protein metabolism of the burned muscle.0.1u/ml of insulin could significantly stimulate the tyrosine incorporation in the muscle of the unburned limb and the controls, but no similar effect was observed on the burned muscle. The results indicate that increased protein degradation of the local burned muscle is responsible for the postburn negative nitrogen balance;administration of leucine alone cannot improve the protein metabolism in the local burned muscle;and the sensivity of the burned muscle to insulin is suppressed.
7.Permeation Enhancing Effect of Flos Caryophylli Volatile Oil Versus Azone on Diclofenac Sodium
China Pharmacy 2005;0(21):-
OBJECTIVE:To evaluate the permeation enhancing effect of Flos Caryophylli volatile oil versus azone on diclofenac sodium.METHODS:Percutaneous absorption experiment was performed on the apparatus of isolated skin, and the penetrated amounts of diclofenac sodium at different time were measured, then the steady-state flux and enhancing rate were calculated, and the deposit effect of diclofenac sodium was investigated.RESULTS:Flos Caryophylli volatile oil and azone all had promotion effect on percutaneous absorption of diclofenac sodium, especially when they used in combination.Used either in combination or alone, they all had remarkable enhancement property on deposit effect of diclofenac sodium(P
