1.Differentiation of human umbilical cord mesenchymal stem cells into hepatocyte-like cells with the combined induction of hepatocyte growth factor and fibroblast growth factor

BACKGROUND:Fibroblast growth factor can promote proliferation of mesenchymal stem cells (MSCs).grew along the wall.However,there are few reports on the differentiation of MSCs into hepatocytes following fibroblast growth factor induction.When mass concentration of hepatocyte growth factor was 1 μg/L,it can promote mitosis of hepatocytes,and is the strongest mitogenic agent for normal hepatocytes.OBJECTIVE:To explore the biological characteristics of human umbilical cord MSCs/n vitro and their differentiation ability to hepatocyte-like cells under the induction of chemical factors.DESIGN,TIME AND SETTING:The cytological in vitro study was conducted at the Institute of Blood,Jinan University from August 2008 to April 2009.MATERIALS:Umbilical cord was obtained from healthy full-term fetus,which was provided by the Guangzhou Huaqiao Hospital.The parturient signed informed consent.Hepatocyte growth factor and flbroblast growth factor were bought from Peprotech,USA.METHODS:The MSCs from human cord were isolated and cultured by type Ⅳ colagenase digestion+differential adherence.At the third passage,MSCs received cell surface antigen analysis and cell cycle determination to detect their ability to differentiate into adipocytes and osteoblasts.At the fifth passage,MSCs were adjusted into 5×10~9/L and assigned into 2 groups.BMSCs in the control group were incubated in DMEM/F12 containing 5% fetal bovine serum.BMSCs in the induction group were treated with above-mentioned medium supplemented with 20 μg/L hepatocyte growth factor and 10 μg/L flbroblast growth factor.MAIN OUTCOME MEASURES:The following parameters were measured:biological characteristics of human umbilical cord MSCs and differentiation of human umbilical cord MSCs into hepatocyte-like cells in vitro.RESULTS:At the third passage,MSCs derived from human cord expressed CD29,CD44,CD105,but not antigens of hematopoietic CD34,CD45,and 92.2% of them were in G_0/G_1 phase.Oil red O staining showed cytoplasm presented red granules.Alkaline phosphatase staining demonstrated that cytoplasm was black,with the differentiation ability into adipocytes and osteoblasts.Following 10 days of combined induction of hepstocyte growth factor and fibroblast growth factor,RT-PCR and Western blot results confirmed that cells expressed alpha fetoprotein and albumin.Negative expression was found in the control group.CONCLUSION:Human umbilical cord contained plenty of MSCs,with strong potential of multi-differentiation.Umbilical cord MSCs can differentiate into hepatocyte-like cells following combined induction of hepatocyte growth factor and fibroblast growth factor.
2.Changes of transcutaneous oxygen pressure and transcutaneous carbon dioxide pressure during altitude training

BACKGROUND:Generaly, higher transcutaneous oxygen pressure indicates stronger aerobic fitness. Decreased transcutaneous oxygen pressure and increased transcutaneous carbon dioxide pressure reflect severe fatigue, muscle hypoxia, and poor function state.
OBJECTIVE:To observe changes in transcutaneous oxygen pressure and transcutaneous carbon dioxide pressure in biceps brachi of excelent male rowing athletes during 6-week altitude training, and analyze relationship of the changes to training load.
METHODS:The training containsthreestages,i.e., before, during and after altitude training. Transcutaneous oxygen pressure and transcutaneous carbon dioxide pressure were determined using PeriFlux System 5000 laser Doppler flowmetryin athletesintheresting-fasting state in the morning. Lactic acid level in athletes was determined using 6-grade load test 1 week before altitude training and 2 weeks after the end of altitude training.
RESULTS AND CONCLUSION:Transcutaneous oxygen pressure and theratio of transcutaneous oxygen pressure to transcutaneous carbon dioxide pressure showed the W-shape change, and transcutaneous carbon dioxide pressure showed the V-shape change. These results suggest that the arrangement of the altitude training program is suitable, namely“reserve physical fitness at the plain stage, add amount and enhance degrees of altitude training load at the early and late stages”,andit can provide a better training load structure model for the post-altitude training. Transcutaneous oxygen pressure and transcutaneous carbon dioxide pressure, in particular the ratio of them,canbeused to evaluate the degree of exercise load that athletes receive. At the same time, capilaries oxygen can be quantified by testing transcutaneous oxygen pressure.Transcutaneous oxygen pressureis recommendedas a noninvasive index to monitor changes in aerobic capacity of athletes.
3.Study on Gastroscopic and Histologic Mucosal Changes in 2440 Children

Objective To study the gastroscopic and pathohistologic mucosal changes in children′s chronic gastritis and to improve the diagnosis of the chronic gastritis in children.Methods Two thousand four hundred and forty children with upper-abdominal symptoms who were examined by gastroscopic observation were collected,and samples of gastric mucosa were examined by the same pathohistologist.And then the pathohistologic changes were analysed.Results All the 2440 children had different mucosal inflammation confirmed by gastroscope,and the pathologic results show that there were 2132 cases of chronic superficial gastritis(90.03%) and 76 cases of chronic atrophic gastritis(3.21%).Among 2440 cases,80 cases with intestinal metaplasia(3.37%),972 cases with lymphoid follicles(41.05%),409 cases with active inflammation(17.28%),902 cases with helicobacter pylori(Hp) infection(38.09%).Moderate and severe inflammatory were more predominant in patients with Hp infection,especially with lymphoid follicles or active gastritis,and the percentage of them were 50.79%,68.73%,73.53%,respectively.Conclusions Major chornic gastritis in children are chronic superficial gastritis,and the pathohistological changes are different from the adults.It also shows that there is a close relationship between gastric mucosal inflammation and Hp.

6.Short incision and tencutaneous relaxing suture in treating closed rupture of the Achilles tendon

Objective To explore a new method in treating acute closed rupture of the Achilles tendon. Methods 22 cases of closed rupture of the Achilles tendon, which had been wounded within one week, were treated with short incision and tenocutaneous relaxing suture from 1994 to August 2001. Results The duration of follow up ranged from eight months to five years. According to Arner Lindholm criteria of therapeutic evaluation, excellent results were obtained in 19 (86.4% ), good in 3 (13.6% ), with the excellent and good rate being 100% . Conclusion Short incision and tenocutaneous relaxing suture is an effective treatment for acute closed rupture of the Achilles tendon because of its less suture tension and more blood supply protection. 7.Clinical study of autologous bone marrow transplantation in the treatment of 35 patients with acute leukemia

Objective To evaluate the clinical effects of autologous bone marrow transplantation(ABMT) in the treatment of patients with acute leukemia.Methods A retrospective study was accomplished on the ABMT in the treatment of 35 patients with acute leukemia from Oct 1999 to Oct 2004.The median age of the patients was 32.5(9~55) years.Of the 35 patients,26 cases were acute non-lymphocytic leukemia(ANLL) and 9 cases were acute lymphoblastic leukemia(ALL).The patients were pretreated with melphalan(140~180mg/m~2),cyclophosphamide(120mg/kg) and arabinosylcytosin(3g/m~2).Results All patients engrafted successfully.The median follow-up duration was 756(186~1950) days.The 3-year probabilities of disease-free-survival(DFS) for ANLL and ALL were(65.4%?8.9)% and(33.3?13.6)% respectively,and the probabilities of relapse were(30.6?9.2)% and(60.7?25.5)%,respectively.Conclusion To decrease relapse and increase DFS,patients with acute leukemia who have no chance for allogene haemopoietic stem cell transplantation are recommended for ABMT.
8.Screening Analysis of Neonatal Congenital Hypothyroidism and Phenylketonuria in Lianyungang Area

Objective To explore the incidence and distribution features of neonatal phenylketonuria(PKU) and congenital hypothyroidism(CH) in Lianyungang area.Methods The heel blood samples dried on filter paper was collected from newborns after 72 hours.As the screening indicators,PKU was detected through serum phenylalanine concentration(Phe) detection by Guthrie′s bacterial inhibition assay and CH was detected through serum thyroid stimulating hormone(TSH) levels detection by time-resolved fluorometry(TRFIA) or enzyme linked immunosorbent assay(ELISA).Results Sixty-four children with CH were diagnosed and the rate of CH was 1∶2423,41 patients were examined by thyroid gland nuclein kinescope picture,among which there were 23 patients whose thyroid glands were unusal.These patients lived in 45 towns(streets),which had no difference between men and women,city and country side.A couple of twins were screened for CH;15 cases of PKU was detected with an incidence of 1∶10 339.These patients′ parents were neither consanguineous marriage nor dominance hereditary family history.Their physical and mental development were similar to the same age children after interventive treatment.Conclusions The distribution of CH and PKU is diffuse in Lianyungang.Neonatal screening is the only valid means to find patients with CH and PKU.
9. Surface-enhanced raman scattering technology-based graphene oxide/gold nanorods in detection of circulating tumor cells

Objective To develop a method combining surface-enhanced Raman scattering (SERS) spectrum and electrostatic adsorption for detecting circulating tumor cells. Methods Graphene oxide was non-covalently functionalized by poly(sodium-p-styrenesulfonate) (PSS), and graphene oxide/gold nanorod (GO/GNR) hybrids were in situ synthesized via gold seeding growth approach. Then, GO/GNR hybrids were non-covalently functionalized by poly dimethyl diallyl ammonium chloride (PDDAC) to make the surface of GO/GNR positively charged. GO/GNR hybrids would target the tumor cells by electrostatic interaction. SERS technology was used to detect the composites of GO/GNR-tumor cells. The blood samples of healthy volunteers were collected, and the tumor cells of different densities were added to the blood samples to make simulated blood samples. The tumor cells in simulated blood samples were detected using the above methods. Results Positively charged GO/GNR hybrids could efficiently target the tumor cells. SERS spectroscopy could detect tumor cells within 50 to 10 000 cells. However, white blood cells might interfere the detection of tumor cells. Conclusion GO/GNR hybrids may serve as SERS probes for detection of circulating tumor cells via Raman spectroscopy.
10.Site-directed mutagenesis of cis-acting element,CHR,in human NFBD1 promoter region

Objective To investigate the role of a cis-acting element,cell cycle genes homology region (CHR),in the transcriptional regulation of human NFBD1 gene (a nuclear factor with BRCT domain 1) by conducting a site-directed mutagenesis analysis on the element in human NFBD1 promoter region.Methods Wild type of NFBD1 promoter reporter,NFBD1-PS1-325,was used as template to make CHR mutant by using PCR based site-directed mutagenesis.Dual luciferase reporter assay was used to determine promoter reporter activity.Adiamycin treatment was employed to investigate the role of CHR in the transcriptional downregulation of NFBD1 after DNA damage.Results Site-directed mutagenesis of CHR caused a significant decrease in NFBD1 promoter activity,and also attenuated the transcriptional down-regulation of NFBD1 after DNA damage.Conclusion CHR element might be involved in both basic transcriptional regulation and transcriptional downregulation of NFBD1 after DNA damage.
