2.Effects of constant low temperature on cold resistance of different strains Polygonatum odoratum.

In this paper, the five strains of Polygonatum odoratum were used as the experimental materials to test the supercooling point, freezing point, the degree of supercooling, the transition stage time, cooling time and water composition of the plant tissue. The cold resistance of P. odoratum was analyzed with the Gray Correlation Method. The results showed that the cold resistances of the five strains of P. odoratum were different, and the water content of plant tissue had some relevance with freezing point and supercooling point, whereas, it could not be measured when the moisture content was too low. The order of cold resistance of the five strains of P. odoratum was ZJCY, DYYZ, XYYZ, CYYZ and JZ I.

3.Rapid Detection of Medicine Based on New Dielectric Barrier Discharge Ionization Mass Spectrometry

Currently, rapid detection of effective components in synthetic drugs and herbal medicine remains an important and difficult issue of medicine research. A novel ionization technique, called dielectric barrier discharge ionization ( DBDI ) , has strong ionization ability, and is suitable for weak polar substances. Besides, this technique possesses many intrinsic advantages, such as simplicity, rapidity, no complicated sample pretreatment, etc. In this study, a new DBDI ion source, based on single electrode technique, was used to detect four weak polar synthetic drugs. The results showed that the protonated molecular ions [ M+H]+of four weak polar synthetic drugs were observed obviously. What′s more, the DBDI ion source was also used for the rapid analysis of Radix Aconiti and Radix Aconiti Preparata pieces without any sample pretreatment. The result showed that the protonated molecular ions [M+H]+ and fragment ions [M+H-60]+of aconitine, hypaconitine, and mesaconitine were detected in Radix Aconiti. And only the fragment ions [M+H-60]+were detected in Radix Aconiti Preparat. The researches indicated that diester aconitine and monoester aconitine were the main effective components of Radix Aconiti Radix and Aconiti Preparat, respectively. The new DBDI ion source provided a fast and reliable method to identify effective components of medicine, showing a broad application prospects in synthetic drugs and herbal medicine research.

The plasma visfatin,endothelium-dependent artery dilation and intima-media thickness of common carotid arteries were measured in first-degree relatives of type 2 diabetes,obese patients and control subjects.Regional body fat were detected by MRI.The result suggested that plasma visfatin levels were significantly higher in obese subjects than those in non-obese subjects,and hypervisfatinemia is independently associated with fasting blood glucose.
5.Correlation of gastroesophageal reflux and asthma-like pathophysiological changes

Objective To investigate whether gastroesophageal reflux can cause asthma-like pathophysiological changes and its mechanism.Methods Sixty BALB/c mice were randomly divided into 4 groups:group A was gastroesophageal reflux control group,group B was asthma control group,group C was gastroesophageal reflux group,and group D was asthmatic group.The asthmatic models were replicated with ovalbumin(OVA) and aluminum hydroxide,and gastroesophageal reflux models were replicated with hydrochloric acid solution pepsin.After the last inhaling ovalbumin and slow perfusion,the airway pressure was detected,and eosinophil (EOS) and neutrophil granulocyte in bronchial lavage fluid were counted.The flow cytometer was used to determinate IL-4,IFN-γ,and Th1/Th2 ratio changes of spleen cells;Lung tissue and esophagus sections were stained with HE,and pathological changes of lung tissue and esophagus were observed.Results In group C and group D,the airway hyper-responsiveness was significantly increased compared with group A and group B,and the differences were statistically significant (all P < 0.05).In group C and group D,IL-4were significantly increased,while IFN-γand Thl/Th2 ratio were significantly decreased than that in the group A and B group,the differences were statistically significant (all P < 0.05).EOS of group C and group D accounted for a high percentage of total cells and they were significantly higher than that in group A and group B,and the differences were statistically significant (all P < 0.05).Through lung tissue biopsy,in group C and group D,bronchial lumen deformation,infiltration of inflammatory cells around the wall,basement membrane thickening,inflammatory cell infiltration around the wall,peripheral vascular edema,enlarging alveolar cavity,alveolar wall thinning,fracture,part of alveolar fusion into bullace of lung,could be observed.The lung pathological section showed that the endothelial cells in group A and group B were integrated and had no denaturation or necrosis,and there was no inflammation cell infiltrate around.EOS biopsy could be observed in group A and group B of lower esophagus markedly submucosal edema,submucosal inflammatory cell infiltration,and keratin hyper function,visible bacteria group A group B and group D were basic ally normal,with no pathological changes.Conclusions Gastroesophageal reflux can induce Th1/Th2 decreasing,airway hyper-responsiveness and pathophysiological changes similar to asthma.
6.A preliminary study about the apoptostic mechanism of RNA targeting basic fibroblast growth factor in glioma U251 cells.

OBJECTIVETo preliminarily investigate the mechanism of small interfering RNA (siRNA) induced apoptosis in glioma U251 cells by silencing basic fibroblast growth factor (bFGF).
METHODSU251 cells were divided into the normal control group, the mock group and experiment group, the mock and experiment group were transfected with mock vector (Ad-null) and the recombinant adenovirus carrying bFGF-siRNA (Ad-bFGF-siRNA) respectively at a multiplicity of infection (MOI) of 100. After 72 hours, the expression of related proteins was revealed by the method of Western blot. Mitochondrial transmembrane potential (ΔΨm) was measured with flow cytometry and confocal microscopy, Groups were compared using single factor analysis of variance (One-way ANOVA).
RESULTSAfter U251 cells were transfected with bFGF-siRNA, the results of Western blot showed that after 72 hours of transfection the bFGF protein in the experiment group decreased obviously, meanwhile Cytochrome C, Caspase-3 and Bax showed increased expression while in the Bcl-xl and Bcl-2 proteins decreased expression. The proportion of high mitochondrial membrane potential of cells by flow cytometry, the experimental group was 74.4% ± 4.7% decreased significantly compared with the control group 92.1% ± 2.5%, the mock group 90.9% ± 1.8% (F = 28.805, P < 0.05); laser scanning confocal microscopy results showed that the red fluorescence and green fluorescence ratio of the experimental group was 0.83 ± 0.12 decreased significantly compared with 1.36 ± 0.40 of the control group and 1.32 ± 0.35 of the mock group(F = 7.920, P < 0.05).
CONCLUSIONsiRNA targeting bFGF induced U251 cell apoptosis may be achieved through the mitochondrial pathway.

7.Anti-glioma effect of combination of bFGF-siRNA and Vpr in nude mice.

OBJECTIVETo study the anti-glioma effect of recombinant adenovirus mediated combined gene therapy of bFGF-siRNA and HIV1-Vpr in vivo.
METHODSMouse glioma model was established by injecting 5 × 10(6) LN229 cells into BALB/c-nu nude mice. 30 nude mice were randomly divided into 5 groups: the negative control group, mock group, bFGF-siRNA group, Vpr group and combined therapy group, which at regular intervals were injected with PBS, rAd5-null, rAd5-bFGF-siRNA, rAd5-Vpr, rAd5-bFGF-siRNA plus rAd5-Vpr, respectively. The tumor volume was recorded every third day to draw a growth curve. After four weeks treatment, the mice were killed and specimens were taken. HE, immunohistochemical and TUNEL staining were performed to observe the cell morphology, detect the changes of relevant target proteins and cell apoptosis, respectively. Also the ultrastructural changes were observed by electron microscopy.
RESULTSThe tumor growth inhibition rates were 36.9%, 37.2% and 58.6% in the bFGF-siRNA group, Vpr group and combined therapy group, respectively, and the combined therapy group showed the most significant effect (P < 0.05). Also the results of HE, immunohistochemical and TUNEL staining revealed that the combined therapy group had the best effects on proliferation inhibition and apoptosis induced in glioma cells (P < 0.05). The most significant ultrastructural changes were observed in the combined therapy group.
CONCLUSIONThe combined gene therapy of bFGF-siRNA with Vpr shows a prominent and synergistic anti-glioma effect compared with that of mono-gene therapy in nude mice.

8.Acupuncture for premature ovarian insufficiency: a systematic review and meta-analysis

Objective: To analyze the effect of acupuncture versus hormone replacement therapy (HRT) for premature ovarian insufficiency (POI). Methods: China National Knowledge Infrastructure (CNKI), Wanfang Academic Journal Full-text Database (Wanfang), Chongqing VIP Database (CQVIP), China Biology Medicine Disc (CBM), Web of Science, Cochrane Library, PubMed, and Excerpta Medica Database (EMBASE) were searched up to January 31st, 2019 to identify randomized controlled trials (RCTs) evaluating the effect of acupuncture for POI. The primary outcome was the level of basal serum follicle- stimulating hormone (FSH). Secondary outcomes included serum levels of luteinizing hormone (LH), estradiol (E2) and anti-Müllerian hormone (AMH). Two authors extracted data independently and assessed the risk of bias and the methodological quality using the Cochrane's tool. Meta-analysis was conducted by RevMan version 5.3. Results: Eight eligible RCTs with a total of 496 POI patients were included in the meta-analysis. The pooled results showed that there was a significant reduction in the basal serum FSH level (MD=-5.82, 95%CI:-9.76 to -1.87, I2=82%, P=0.004) and a remarkable elevation in the basal E2 level (SMD=0.93, 95%CI: 0.34 to 1.52, I2=88%, P=0.002) in the acupuncture group when compared with the control. Subgroup analysis showed that compared with HRT, a significant decrease in the FSH level was observed in both acupuncture alone (MD=-4.53, 95%CI:-8.96 to -0.10, I2=73%, P=0.04) and acupuncture plus HRT (MD=-9.60, 95%CI:-17.60 to -1.61, I2=50%, P=0.02), while a remarkable elevation of E2 was only found in acupuncture plus HRT (SMD=1.43, 95%CI: 1.03 to 1.82, I2=0%, P<0.00001). There was no significant difference in the LH level between acupuncture and HRT (MD=-3.16, 95%CI:-9.41 to 3.10, I2=0%, P=0.32), only one trial reported AMH, and no significant difference was found between acupuncture and HRT. Conclusion: The present study indicated that acupuncture had an advantage over HRT in reducing serum FSH level and increasing serum E2 level in women with POI. However, evidence supporting the finding is limited due to the small sample size, potential methodological flaws and significant heterogeneity. Hence, this conclusion still needs to be verified by high-quality RCTs.
9.Relationship of fatty liver with HBV infection, hyperlipidemia and abnormal alanine aminotransferase

Objective To investigate the prevalence of fatty liver in Guangzhou and its relation to hepatitis B virus( HBV)infeetion, hyperlipidemia and abnormal alanine aminotransferase (ALT). Methods A retrospective analysis was made on clinical data of 4365 participants who received health check-up in our hospital. Results The prevalence of fatty liver was 18.2%, 19.2% in males and 17.1% in females, respectively. There was no significant difference between males and females( P = 0.07). Among 793 subjects with fatty liver, 440 were males and 353 were females. The prevalence of fatty liver was 16.7% in HBV infection group and 18.3% in the group without HBV infection, no significant difference was seen between these two groups ( P = 0.45). The prevalence of fatty liver was 42.1% in hyperlipidemia group, and 11.6% in the group with normal serum triglyceride (TG)and total cholesterol (TC), respectively, there was a significant difference between these two groups( P < 0.05). The abnormal ALT was seen in 32.5% of the fattty liver group, which was significantly higher than that (8.6%) in the group without fatty liver ( P < 0.05 ). Conclusions The prevalence of fatty liver was not significantly different between males and females in Guangzhou. Fatty liver was not related with HBV infection but closely related with age and hyperlipidemia. Fatty liver was a common cause of abnormal ALT.
10.The relationship between the levels of HBV DNA loads and both the clinical characteristics and 48-week prognosis in patients with decompensated cirrhosis due to hepatitis B

Objective To evaluate the relationship between the levels of HBV DNA loads and beth the clinical characteristics and 48-week prognosis in patients with decompensated cirrhosis due to hepatitis B. Methods One hundred and forty-three patients with deeompensated cirrhosis of hepatitis B virus infection were divided into lowlevel HBV DNA group [HBV DNA < 105 copies/ml = (46 cases) and high-level HBV-DNA group(HBV DNA 105 copies/ml)(97 cases)]. 21 cases in low-level group and 52 cases in high-level group treated with nucleoside analog. Results There was no significant difference between the two groups on the demography and the baseline in ALT, ALB, TBil, CHE before treatment, while in AST and HBeAg were statistically different. At 48-week, there was no significant difference between the two groups on the liver function. The mortality rate in low-level group was similar to that in high level group. In the low-level HBV DNA patients, hepatocellular carcinoma, spontaneous peritonitis and gnstrointestiual hemorrhage were higer than that in the high-level HBV DNA patients. Conclusion In patients with decompensated cirrhosis due to hepatitis B, those who were in low-level HBV DNA had not got better than that in high-level HBV DNA, which indicated that earlier treatment was also needed in low-level HBV DNA patients.
