1.The impact of endothelial progenitor cells on the osteogenesis of bone marrow mesenchymal stem cell sheet-implant complex

Objective:To study the effects of endothelial progenitor cells(EPCs)on the osteogenesis of bone marrow mesenchymal stem cell (BMSCs)sheet-implant complex.Methods:EPCs were added to the BMSC sheets,and the expression of osteogenesis-relat-ed genes was examined by real time PCR.Cell sheets were wrapped around implants to construct cell sheet-implant complexes and the complexes were subcutaneously transplanted into SCID mice.The complexes were harvested 8 weeks after operation and observed by micro-CT and histological examination.Results:The BMSC sheet with EPCs showed higher expression of Runx2,ALP,BMP2 and VEGF in the in vitro test;higher bone volume ratio,greater amount of new bone tissue and higher expression of Runx2 and BMP2 in the in vivo test.Conclusion:EPCs can improve the osteogenesis of BMSC sheet-implant complex.
2.Design of print out function for a multi-parameter patient monitor.

This paper describes the design of the printing output function for a multi-parameter patient monitor by using the graphic plot function of a Laser Printer. The ECG data are preprocessed with a notch filter and interpolation algorithm. A logical page is constructed in the extended memory for virtual page printing. This monitor is able to output a satisfied printing with ECG waveforms of high quality.

3.A study on the effect of regular aerobic exercise on artery elasticity among patients with essential hypertension

Objective To explore the effect of aerobic exercise training on artery elasticity among patients with essential hypertension.Methods A total of 155 cases of patients with essential hypertension (EH)were selected,and were divided into control group(76 cases)and aerobic exercise group(79 cases).Control group was follow routine drug therapy,and aerobic exercise group was follow aerobic exercise training besides the routine therapy for 12 weeks.CAVI,ABI values,limb blood pressure,TG,TC,LDL -C,HDL -C and U -MA were compared before and after the intervention.Results After the intervention,bilateral CAVI values of aerobic exercise group were 7.76 ±0.15,7.88 ±0.15,8.87 ±0.25 and 8.89 ± 0.22,and lower than those before the intervention(P <0.05).The decline range of bilateral CAVI value(the difference between the mean differences before and after the intervention)were 1.03 ±0.07 and 1.03 ±0.06,significantly wider than the control group of 0.33 ±1.97 and 0.15 ±0.08(P <0.05).After the intervention,limb systolic pressure,diastolic blood pressure and triglyceride,cholesterol decreased in the aerobic exercise group(P <0.05).The limb systolic blood pressure and left limb diastolic blood pressure decreased in the control group after intervention (P <0.05),but the decline range was smaller than the aerobic exercise group(P <0.05 ).Conclusion The aerobic exercise training could significantly improve arterial elasticity among patients with essential hypertension.
4.Detection of the serum level of interleukin-17 in the chronic hepatitis B patients

Objective To research serum level of interleukin-17 (IL-17)associated with the progression of hepatic injury in the chronic patients with hepatitis B. Methods The serum level of IL-17 was measured by ELISA and the serum levels of IL-6,IL-8 were measured by RIA in patient groups and healthy group, the patient groups including 42 mild patients, 37 moderate patients and 38 severe patients. Results IL-17,IL-6 and IL-8 levels in patient patients were significantly higher than those in healthy people(P< 0.01). There is no significant difference among mild patients and moderate patients. Compared with mild patients and moderate patients, the cytokines lever were significantly higher in severe patients(P<0.01). Conclusion IL-17 as a new cytokine probably play a multiple role as immune factor and inflammation element in the progression of the chronic hepatitis B disease. Maybe, it can provide a new approach to the therapy of the chronic hepatitis B.
5.Anti-depressive mechanism of Fufang Chaigui prescription based on neuroendocrine hormone and metabolomic correlation analysis.

To elucidate the anti-depressive effect of Fufang Chaigui prescription and its mechanism and investigate its effect on neuroendocrine hormone, rats were included into a chronic unpredictable mild stress (CUMS) model for 28 d, and drugs were administered at the same time. During the period, rats' behaviors were observed and the blood was collected by using ELISA to determine representative hormone concentrations of HPAA, HPTA and HPGA. The changes in endogenous metabolites were analyzed by using H NMR metabolomics to seek the potential biomarkers. Results showed Fufang Chaigui prescription could improve the behaviors of CUMS rats obviously, increase contents of ACTH, CORT, T₃and decrease contents of TSH and TESTO and regulate the levels of lactate, α-glucose, choline, N-acetylglycoprotein, trimethylamine oxide and leucine to get closer to the contents of control group. The results of correlation analysis indicated that HPTA was associated with glycometabolism, amino acid metabolism and choline metabolism. And HPAA was related to glycometabolism and amino acid metabolism. However, HPGA was only correlated with glycometabolism. In conclusion, Fufang Chaigui prescription could show an obvious anti-depressive effect and its underlying mechanism might involve regulations of neuroendocrine function and pathways of glycometabolism, amino acid metabolism and choline metabolism.

6.Design of a digital spectrometer for the MRI system.

A spectrometer is one of the most important parts in a Magnetic Resonance Imaging (MRI) system. This paper describes the design of a digital MRI spectrometer. It is constructed on a PXI platform with several data acquisition boards and a high-resolution timing board. All functions of a MRI spectrometer are realized by the specially- designed software. The software architecture and its implementing details are discussed and experimental results are introduced.

7.Origin of acoustically evoked short latency negative response in guinea pigs

Objective To establish a model of acoustically evoked short latency negative response (ASNR) in guinea pigs, a model of profound hearing loss with normal saccular functions, and verify the correlation between ASNR and vestibular evoked myogenic potential (VEMP). Methods Thirty-two healthy guinea pigs were employed in the experiment, which were randomly divided into control group ( 16subjects) and deafened group (16 subjects). Each animal experienced auditory and vestibular tests including auditory brainstem response ( ABR), VEMP and caloric test. A quick treatment was employed for deafened group consisting of a subcutaneous injection of kanamycin at a dose of 400 mg/kg followed by a jugular vein injection of ethacrynic acid at a dose of 40 mg/kg one hour later. The animals were received ABR, VEMP and caloric test 7 - 10 days following the drug administration. The deafened group was further divided into ASNR group and non-ASNR group, based on the presence of ASNR. Results In deafened group, five subjects died postoperatively, 11 subjects (22 ears) provided full data, ASNR was elicited in eight ears (36.4%), the threshold was 120- 130 dB SPL with mean of (124.4 ±4.96) dB SPL. Its latency range was 1.75 - 2. 60 ms with mean of ( 2. 15 ± 0. 27 ) ms. The mean latency of threshold was (2. 34 ±0. 18) ms. All eight ASNR ears presented with VEMP. The VEMP threshold, positive and negative potential latencies proved no statistical difference (P > 0. 05 ) between ASNR group and control group.Significant difference was detected between the VEMP presence of ASNR group and non-ASNR group ( P =0. 002). There was no statistically significant correlation between VEMP and caloric test neither between ASNR and caloric test in deafened group. Conclusions This study evoked ASNR in an ototoxicity guinea pig model which has profound hearing loss with normal saccular functions. The presence of ASNR correlated with VEMP, however, not correlated with caloric test, suggesting that ASNR and VEMP are both originated from the saccule.
8.Expression of Survivin protein in extrahepatic cholangiocarcinoma and its relationship with the prognosis.

OBJECTIVESTo investigate the expression of Survivin in patients with extrahepatic cholangiocarcinoma (EHCC) and its relationship with clinicopathological features of EHCC, and the correlation between the expression of Survivin and lymph node micrometastasis, tumor markers, and the prognosis of EHCC.
METHODSThe expression of Survivin protein in paraffin-embedded specimens of 59 patients with EHCC and their 20 para-carcinoma tissues were evaluated by S-P method of immunohistochemical staining. The correlation between the expression of Survivin and the lymph node micrometastasis, clinicopathological features of EHCC and the prognosis of EHCC were analyzed.
RESULTSThe positive expression rate of Survivin protein was 67.8% (40/59) in paraffin-embedded specimens of 59 patients with EHCC and was 20.0% (4/20) in para-carcinoma tissues, and difference between carcinoma tissues and para-carcinoma tissues was significant (P<0.01). Histological differentiation in EHCC had a negative correlation with the expression of Survivin protein, while the expression of Survivin protein in EHCC had a positive correlation with TNM of EHCC, lymphatic vessel infiltration, lymph node metastasis and perineural invasion (P<0.05). The serum CA19-9 levels in the positive group with expression of Survivin protein was (290,300+/-55 500) U/L and was obviously higher than that in the negative group [(113,300+/-31,400) U/L, P<0.05]. The mean survival time of the patients with negative expression of Survivin protein was higher than that of the patients with positive expression (43.5 vs. 21.1 months, P<0.01). Screened to significance univariate, the multivariate analysis through Cox proportional hazard model analysis showed that lymph node metastasis, residual tumor margins, and expression of Survivin protein were independent prognosis factors of the patients with EHCC (P<0.05, P<0.01, P<0.01).
CONCLUSIONSThe expression of Survivin protein in EHCC has a negative correlation with histological differentiation, while has a positive correlation with lymphatic vessel infiltration and serum CA19-9 concentrations. The expression of Survivin protein maybe an independent prognosis factor of the patients with EHCC.

9.Controlling of the high-voltage generator in a direct radiography system.

A proposal is presented to improve the original complicated control of the exposure parameters in the direct radiography system (DR). The interface circuitry of the high-voltage generator is redesigned using RS-232 serial COM port. The data and signals are transmitted between the personal computer and the high-voltage generator in the serial mode. A program package is designed to realize the control of the exposure parameters of the high-voltage generator in DR on the PC machine. Experiment results have shown that the proposed console operates steadily and it is capable of providing a more convenient operation console for the direct radiography system.

10.Establishment of a hydrogel chip for high-throughput detection of Y chromosome microdeletions.

OBJECTIVETo establish a high-sensitivity, high-specificity and low-cost hydrogel chip platform for the clinical screening of Y chromosome microdeletions.
METHODSSite-specific extended primers with a common sequence at the 5' end were used for hybridizing with the target. The Cy5-dUTP was incorporated into the products by primer extension, and the products were labeled with fluorescence. Then the extended products were added to the chip for hybridizing with acrylamide-modified common probes immobilized on the chip. After removal of the free Cy5-dUTP by electrophoresis, the signals were obtained by fluorescence scanning. And the detecting conditions of this method were optimized.
RESULTSSY254 of 9 samples was successfully detected with the hydrogel chip. The results showed that 3 were normal and the other 6 with microdeletions (1 female sample as a negative control), which coincided with the results of conventional multiplex PCR-electrophoresis.
CONCLUSIONThe hydrogel chip platform we established has provided a new technique for the detection of Y chromosome microdeletions, and is beneficial to the diagnosis and treatment of male infertility.
