1.Experimental study of transfecting adenovirus carrying G250 antigen gene to dendritic cells and to activate immune effector cells in kidney cancer treatment
Chinese Journal of Urology 2010;31(3):161-164
Objective To investigate T cell-mediated effects of dendritic cells(DC)transfected with kidney associated antigen G250 gene using adenovirus(Ad)as vector in the treatment of kidney cancer. Methods Peripheral blood mononuclear cells(PBMC)were isolated by standard FicollPaque density gradient centrifugation of heparinized blood obtained from healthy donors. After removal of the nonadherent ceils, the adherent cells were divided into gene transfer group, protein pulsed group and control group. The gene transfer group was infected with Ad/G250 virus;the protein pulsed group was added with G250 protein; the control group was added with PBS.The DCs were cultured and proliferated in vitro using recombinant human granulocyte macrophage colony stimulating factor and inlerleukin-4.The autologous T cells were added into the 3 DC groups(responders:dendritics,20:1).After 5 d co-culture,3 eytotoxic T lymphocyte(CTL)groups were collected.TheG250 mRNA expression was detected in transduced DCs using RT-PCR.The G250 protein translation products and cell surface marker of DCs were analyzed by flow cytometry. The specific cytolytic activities of CTL to different target cells(renal carcinoma 786-0 cell line and lung cancer A549 cell line)were assessed by MTT method. Results Ad/G250 transfected DC successfully and the G250 expression was confirmed by RT-PCR. The expression levels of CD_(80), CD_(83). HLA-DR and CD_(86) in the Ad/G250 group were higher than those in the other 2 groups. The cytotoxicity to 786-0 of T lymphocytes activated by Ad/G250 transfecting DCs(83. 4±2. 8)% was greater than those of T lymphocytes in the protein pulsed group(79. 6±2. 4)% and control group(77. 3±2. 1)%. There were no significant differences of the cytotyxicities to A549 among the 3 cytotoxic T lymphocyte groups(F=0. 373.P=0.693). Conclusions Ad-loading DCs may be useful for immunotherapeutic protocols against self-antigens for the kidney cancer.
2.Diagnostic value of ROP with the changes of serum IGF-I, VEGF levels in premature
Chinese Journal of Primary Medicine and Pharmacy 2014;(24):3721-3724
Objective To observe the changes of serum IGF-I,VEGF levels in premature infants, and to dis-cuss the predicting values for the development of ROP;Methods Preterm infants admitted into those whose birth weight ( BW)≤1 500g and ( or) gestational age ( GA)≤31 weeks joined in ROP-screen,and those who were diag-nosed as ROP according to ICROP were included into ROP group. In the same time, those who didn't develop ROP were divided into the control group.Serum IGF-I,VEGF were measured one week from their birth or correct gestational age of 31 weeks.Serum IGF-I, VEGF were evaluated with ELISA method. Results The concentration of IGF-I, VEGF:Group ROP[(20.65 ±11.11)μg/L,(195.61 ±105.98)ng/L] were all significantly lower than the control group [(42.86 ±18.72)μg/L,(363.59 ±101.08)ng/L](all P<0.01).There were significant different of birth weight[ROP group (1 229.69 ±232.37)g,the control group (1 401.32 ±171.78)g;t=4.144)],GM[ROP group (3.20 ±1.23)mmol/L,the control group (3.70 ±1.17)mmol/L;t=2.082],the time of oxygen inhaling[ROP group (32 ±22)d,the control group (21 ±12)d;t=-2.856],oxygen inhalation mode(the percentage of mechanical ventilation)[ROP group 33/48(68.8%),the control group 16/50(32.0%);χ2 =16.363] and fertilization way(the percentage of test tube baby)[ROP group 13/48(27.1%),the control group 4/50(8.0%);χ2 =6.262] between ROP and control group (all P<0.05).Conclusion The low serum IGF-I(<28.08μg/L),VEGF(<232.89ng/L) of a premature in the first week or correct gestational age of 31 weeks and (or)BW≤1 500g would probably suggest the development of ROP.

Chinese Journal of Tissue Engineering Research 2013;(36):6521-6526
BACKGROUND:Liver cancer pathogenesis and intervention have attracted increasing attentions. Mesenchymal stem cel s become a popular tool for cel cancer research, because of their low immunogenicity and tumor tropism. At present, mesenchymal stem cel s have been applied to the study of liver cancer.
OBJECTIVE:To summarize the advances of mesenchymal stem cel s used in liver cancer in basic and clinical research.
METHODS:An online retrieval of CNKI and Pubmed database was performed by the first author for the articles about mesenchymal stem cel s and effect of modified mesenchymal stem cel s on hepatoma carcinoma cel s published from January 2004 to January 2013. The key words were“mesenchymal stem cel , liver cancer, tumor”in Chinese and English. Repetitive research was excluded, and 47 studies met the inclusion criteria.
RESULTS AND CONCLUSION:Stem cel s are seldom reported in liver cancer, and the limited present study show that mesenchymal stem cel s may have a certain influence on the hepatoma carcinoma cel proliferation, invasion and biological behavior. However, due to the differences of cel lines used by the various laboratories, experimental conditions, animal models, as wel as infusion means of stem cel s, experimental results are also inconsistent. Scholars have conducted a series of studies on the mechanism of the Wnt pathway and tumor necrosis factor-related apoptosis-inducing ligand pathway. Tropism of mesenchymal stem cel s to tumor cel s, including liver cancer is widely recognized, so scholars imported therapeutic genes and drugs into mesenchymal stem cel s to interfere with the development of liver cancer, and have achieved some progress. This evidence provides new avenues for cel therapy for liver cancer. Less safety studies in vivo and clinical trials of mesenchymal stem cel s are available, therefore security risks deserve further research.

5.Comparative analysis on composition principles of traditional Chinese medicine prescriptions for osteoporosis and osteoarthritis.
China Journal of Chinese Materia Medica 2014;39(16):3172-3175
This paper collected the traditional Chinese medicine (TCM) prescriptions of osteoporosis and osteoarthritis from CNKI to set up database and used TCM inheritance system to analyze the composing principles in prescriptions. We collected 103 and 106 prescriptions for osteoporosis and osteoarthritis respectively, then analyzed inherent principles between herbs in prescriptions by using the TCM inheritance system, and obtained the frequency of each herb, and drug combinations of two diseases' prescriptions. The result showed that the principles in TCM prescriptions of osteoporosis and osteoarthritis are similarities, buch as tonifying kidney and invigorating the circulation of blood, but differences of the specific drugs. That is to say, tonifying kidney is the main method of osteoporosis treatment, however, invigorating the circulation of blood is the main method of osteoarthritis treatment.
