1.Correlation between abnormal methylation of IGF-2 exon 9 and IGF-2 gene expression

Object To investigate the correlation of IGF-2 exon 9 CpG island methylation and the expression of IGF-2,and further explore the mechanism of Wilms tumor. Methods The IGF-2 exon 9 methylation status and IGF2 gene expression in 42 cases of Wilms tumor and corresponding normal tissues were detected by using methylation-sensitive restriction enzyme PCR and allele-specific IGF2 gene expression analysis.Results IGF-2 exon 9 methylation rate was 15% in W ilms tumor group, and 97% in normal tissue group. The difference was significant (P < 0.01), and the difference in pathological type was also significantly different (P < 0.01). Exon 9 unmethylated tissues showing biallelic expressions of IGF-2 were significantly higher than that of methylated tissues (P < 0.05). Conclusion IGF-2 exon 9 unmethylation may be the cause of biallelic expression of IGF, which contritues to tumorigenesis in Wilms tumor.
3.Therapeutic Observation of Abdominal Acupuncture plus Moxibustion for Cervical Spondylosis of Vertebral Artery Type

Objective To observe the clinical efficacy of abdominal acupuncture plus moxibustion in treating cervical spondylosis of vertebral artery type.Method A hundred patients with cervical spondylosis of vertebral artery type were randomized into a treatment group and a control group, 50 cases in each group. The control group was intervened by abdominal acupuncture plus electroacupuncture; while the treatment group was by moxibustion in addition to the treatment given to the control group. After a treatment course, the onset and remission time of vertigo was observed, and the clinical efficacies were compared between the two groups.Result The total effective rate was 96.0% in the treatment group versus 93.8% in the control group, and the difference was statistically significant (P<0.05). There was a significant difference in comparing the onset and remission time of vertigo between the two groups (P<0.05). In comparing the relapse rates 3, 6, and 12 months after the intervention between the two groups, the difference were statistically significant (P<0.01).Conclusion Abdominal acupuncture plus moxibustion is an effective approach in treating cervical spondylosis of vertebral artery type.
4.Prescription optimization and stability investigation of urea ointment

Objective To adjust the prescription of urea ointment and investigate its stability .Methods In accordance with the original prescription , four groups of urea ointment were prepared .Group A and group B were made with yellow vaseline from different manufacturers , while groups C and D were prepared in summer and winter respectively with the same source materials.The viscosity and moldability of the four groups were evaluated through the senses .The prescription was slightly adjusted through the single-factor effect experiment .Three batches of urea ointment were prepared according to the new prescription and their stability was investigated .Results Using the original prescription , the urea ointment made with yellow vaseline provided by Manufacturer One had better viscosity and moldability than that from Manufacturer Two .The urea ointment made in summer had lower viscosity and poorer moldability than the one made in winter .The amount of beeswax increased in the adjusted prescription .Three batches of urea ointment were prepared with the new prescription and their stability was maintained .There were no changes in appearance , and the content and microbial limit remained in the standard range for 11 months.Conclusion The source of materials and change of the external environment could affect the characteristics of urea ointment .The adjusted prescription is feasible and stable .
5.Clinical studies of U8 similar bioelectricity stimulation therapy of infertility with gracile endometrium

Objective Observe the effect of of U8 similar bioelectricity stimulant therapy of infertility with gracile endometrium on endometrium reception .Methods 80 cases of patients in 252 natrual period with infertility caused by poor endometrial develop-ment for unknown reasons were randomly divided into two groups .The treatment group was intervened by U8 similar bioelectricity stimulant other day from the eighth day of menstruation until the day to inject HCG .After ovulation ,patients were sustained by progestin with a dose of 20 mg per day .The control group was given estradiol valerate with a dose of 2mg per day from the eighth day of menstruation until the day to inject HCG .After ovulation ,patients were sustained by progestin with a dose of 20mg per day as well .To compare the generral conditions on HCG injection day of the two groups anterior-posterior :both groupsal thickness and types ,parameters of intramembrane and submucosal uterine blood flow ,biochemical pregnancy rates and clinical pregnancy rates . Results The endometrial thickness and types on HCG injection day in treatment group were improved significantly (P<0 .05) . The parameters of intramembrane and submucosal uterine blood flow were better than the control ,there was a significant difference ( P<0 .05) .And the clinical pregnancy rate of treatment group was higher than that of control group there was a significant differ-ence(P<0 .05) .Conclusion U8 similar bioelectricity stimulation can promote the grow th of endometrium ,improve the endometrial receptivity and the clinical pregnancy rate .
6.Preparation, characterization and in vitro evaluation of poly(2-ethyl-2-oxazoline) modified curcumin liposomes

Objective To prepare Curcumin liposome (Cur-L) and poly(2-ethyl-oxazoline)-cholesteryl methyl carbonate (PEOz-CHMC) was used to modified Cur-L and to evaluate their associated properties in vitro.MethodsEncapsulation efficiency and particle size were taken as evaluation indicators to optimize the formulation and preparation conditions of Cur-L by orthogonal test.The EE, particle size and shape of the liposomes were determined by sephadex G-50 mini-column centrifugation method, ZLS dynamic light scattering instrument and transmission electron microscopy (TEM), respectively.The release of the liposome in vitro was detected by The dialysis method.MTT assay was used to determine the cell inhibition of two Cur-L.ResultsThe optimized preparing method of Cur-L is as following: 1.56(w/w) as drug-lipid ratio, 5.1(w/w) as the ratio of mass of phosphatide and cholesterol, 7.4 as the pH of PBS buffer.The EE of Cur-L was (75.05±0.64)%, while the modification of PEOZ hasno influences on EE.Through TEM, PEOZ-Cur-L has aobviouslipid bilayer structure.The average particle diameter of PEOZ-Cur-L was 84.89 nm.In vitro release experiments showed that in 24h, the accumulative release rate of Cur-L is more than 70% with pH 7.4, while that of PEOZ-Cur-L was less than 25%.The cytotoxicity experiment showed that PEOZ-Cur-L can inhibit HCT116 Human colon cancer cells more effectively.ConclusionThe optimized preparing method of Cur-L is reasonable.PEOZ can provide stability to liposomes well and does not hamper its inhibitive effects.
7.Characteristics of CD8+ stem memory T cells in patients with chronic HIV-1 infection during antiretroviral therapy

Objective To investigate the characteristics of CD8+ stem memory T cells (CD8+Tscm) in patients with chronic HIV-1 infection before and after antiretroviral therapy (ART) and to analyze their associations with progression of HIV-1 infection.Methods Thirty-six patients with chronic HIV-1 infection and 20 healthy subjects were enrolled in this study.Flow cytometry was performed to detect the percentages and absolute numbers of CD8+Tscm in patients with chronic HIV-1 infection before and after antiretroviral therapy (ART) as well as in healthy subjects.Correlation analysis was used to demonstrate the relationships between CD8+Tscm and markers for progression of HIV-1 infection (CD4+T cell count, HIV-1 viral load and level of activated T cells).Results The percentages and the absolute numbers of CD8+Tscm in patients with chronic HIV-1 infection had no significant change before and after ART.They were respectively positively correlated with the percentages and the absolute numbers of CD4+Tscm.The percentage of CD8+Tscm was proportional to the percentage of CD8+ central memory T cells (CD8+Tcm), but was inversely proportional to the percentage of CD8+ effector memory T cells (CD8+Tem).In addition, the percentages of CD8+Tscm in patients with HIV-1 infection were negatively correlated with the viral loads before ART.Conclusion CD8+Tscm are responsible for maintaining the homeostasis of other CD8+T cell subsets.CD8+Tscm play an important role in inhibiting viral replication.
8.PHARMACOKINETIC STUDIES OF 20(R)-GINSENOSIDE RG3 IN HUMAN VOLUNTEERS

AIM To study the pharmacokinetics of 20(R)-ginsenoside Rg3 (GRg3) in human. METHODS High-performance liquid chromatography-ultraviolet detection method was used in this study. RESULTS The pharmacokinetics of GRg3 in 14 healthy volunteers were investigated. After a single oral dose of 3.2 mg.kg-1 in 8 male volunteers, the plasma concentration-time course fitted well to a two-compartment open model, with the following pharmacokinetic parameters: Tmax (0.66±0.10) h, Cmax (16±6) ng.mL-1, T1/2α (0.46±0.12) h, T1/2β (4.9±1.1) h, T1/2(Ka) (0.28±0.04) h, AUC0-∞ (77±26) ng.mL-1.h. No kinetic analysis was made after an oral dose of 0.8 mg.kg-1 GRg3 in 6 other volunteers because of the low concentration, but a good correlation between Cmax and dosage of the two groups was found. CONCLUSION The absorption of GRg3 was rapid in man, and its elimination was rapid after oral administration of ginsenoside Rg3. The pharmacokinetic results shows that it exhibited first order kinetic characteristics.
9.The clinical study of stimulation of milk secretion by body massage combined with massage of breast point

Objective We discussed the effect of body massage combined with massage of breast point (ruzhong point and rugen point) to facilitate the milk secretion, alleviate the swelling pain of breast, recover physical strength of mother, establishment of the confidence of breast feeding and increase the success rate of breast feeding. Methods We divided 212 parturients into the test group (104 cases) and the control group (108 cases). The test group received body massage combined with massage of breast acupuncture point based upon routine postpartum nursing. The control group received routine postpartum nursing We observed the time and volume of milk secretion in the two groups. Results The time of milk secretion of the test group was ahead of that of the control group and volume of milk secretion was increased in the test group than that of the control group (P<0.01). Conclusions Body massage combined with massage of breast point (ruzhong and ruzhong point) could place the time of milk secretion ahead, increase the volume of milk secretion. Besides, it could alleviate the swelling pain of breast and ensure the success d breast feeding.
10.Advanced research on characteristics of the classical subtype of human glioblastoma

Glioblastoma(GBM)is one of the most common primary intracranial tumor that has high de-gree of malignancy ,invasive ability and a fatal prognosis .In recent years ,with the development of modern technol-ogy in biomedical sciences ,the understanding on GBM has developed gradually from pathological diagnosis to mo -lecular classifications ,which is based on the molecular characteristics of genetic signatures .Based on gene expres-sion and DNA methylation patterns , primary glioblastoma is divided into four subtypes , including the classical , neural,proneural and mesenchymal .These molecular classifications are closely relevant to the biological charac-teristics of glioblastoma .This review briefly introduces the molecular classifications of primary glioblastoma , but mainly focuses on the changes of the major genetic EGFR ,PTEN and PI3K,CDKN2A in the classical subtype of GBM,and discusses the treatment strategies for primary glioblastoma .
