1.Content Changes of Aristolochic Acid-A from Caulis Aristolochiae Manshuriensis and Its Processed Products

Objective To research the content changes of Aristolochic Acid-A from Caulis Aristolochiae Manshuriensis and its processed products.Method Chromatographic assay was performed on Lichrospher-C18 column (4.6 mm?200 mm,5 ?m) with methanol-0.1% glacial acetic acid solution (70∶30) as mobile phase,the flow rate was 1.0 mL/min and the detection wavelength was set at 310 nm,the column temperature was 30 ℃.Result The content of Aristolochic Acid-A was lower in three processed products than in crude drugs,and the reduction rate of the one which was boiled by NaHCO3 was the highest.Conclusion The three processed method can reduce the content of Aristolochic Acid-A,and achieve the aim of reducing the toxicity.
2. Treatment of advanced colorectal cancer with chemotherapy and recombinant human endostatin in the window period

Objective: To observe the effect of recombinant human endostatin (endostar) combined with chemotherapy on advanced colorectal cancer. Methods: A total of 120 inoperable patients with advanced colorectal cancer who were admitted to the Guizhou Cancer Hospital from June 2014 to June 2018 were selected. The patients were divided into two groups. Sixty cases were allocated to the test group and received endostar of 15 mg/d, d1-d7, which was repeated after 7 and 14 days. Chemotherapy was initiated on the 5th day of endostar (endostar window period). Sixty patients were allocated to the control group and received endostar of 15 mg/d, d1-d14, which was repeated after 7 and 21 days. Chemotherapy was initiated on the 1st day of endostar. The chemotherapy regimen used was mFOLFOX6 or FOLFIRI. Results: The objective response rate (ORR) of the test and control groups was 25.0%, and 18.3%, respectively, and the disease control rate (DCR) was 80.0% and 73.3%, respectively. The difference was not significant (P=0.375, P= 0.388). The 1-year survival rate of the test group and the control group was 69.6% and 62.5%, respectively, while the 2-year survival rate was 39.7% and 21.3%, respectively. Moreover, the 3-year survival rate was 26.8% and 13.3%, respectively, and the median survival time was 22 months (95% CI 16.817-27.183) and 16 months (95% CI 11.890-20.110), respectively. In contrast to the control group, the survival rate increased and the survival time was prolonged in the test group (P=0.033). The progression time (TTP) of median disease in the test and control groups was 9 months and 8 months, respectively. This was not statistically significant (P>0.05). Conclusions: This study found that chemotherapy with recombinant human endostatin in the window stage can enhance the 1, 2, and 3-year survival rate of patients with advanced colorectal cancer, as well as prolong the median survival time.

4.Study on the Quality Standard for Qingzao Runfei Mixture

Objective To establish quality control method for Qingzao Runfei Mixture. Methods Mulberry Leaves,Glycyrrhiza,Radix Glehniae,Ophiopogon Japonicus and Loquat Leaves were identified by TLC. Glycyrrhizic Acid was determinated by HPLC. Results The negative sample of TLC had no interference. The specifity was good. Glycyrrhizic acid showed a good linear relationship in the range of 0.203 1~1.692 1 ?g,r =0.999 8. The average recovery was 98.24%,and RSD was 2.3%. Conclusion The established methods are simple,quick and with good reproducibility. This study provides methods for the quality control of Qingzao Runfei Mixture.
5.Comparison of magnetic resonance with multislice computer tomography in T-staging of cardiac cancer

Objective To compare the diagnostic accuracy of magnetic resonance (MR) images with multislice computer tomography (MSCT) for preoperative T-staging of patients with cardiac cancer. Methods MR and MSCT were performed in 28 cases of cardiac cancer diagnosed by biopsy prior to operation within one week. After an oral intake of 1000 ml water and an injection of hypotonic agent, MR and MSCT scan were carried out. MR sequences included FSE T1W, FSE T2W, FSE T1W with fat suppression and dynamic enhanced FSPGR with fat suppression. MSCT was applied with dynamic triphasic contrast enhancement. All of the findings were prospectively analyzed by two doctors separately and compared with the surgical and pathological findings. Results According to histopathologic staging, the accuracy of MR and MSCT in T1-staging were 88.8% and 11.1%, in T2-staging were 77.8% and 22.2%, in T3-staging were 83.3% and 32.7%, in T4-staging were 100.0% and 50.0%, respectively. 6.Effect of Apigenin on Inhibiting Proliferation and Apoptosis of Human NCI-H1650 Cell Line

Objective To determine the effect of apigenin on inducing apoptosis and inhibiting proliferation of non-small cell lung cancer cell line NCI-H1650. Methods NCI-H1650 cell line was cultured routinely in vitro, with blank control group and different concentrations of apigenin (10, 20, 40, 80 μmol?L-1). The blank control group was RPMI-1640 solution without apigenin. Cell proliferation was detected by MTT. Hoechst 33258 was applied to observe morphological changes of the apoptotic cells after treatment of different concentrations of apigenin. Flow cytometry AnnexinV-FITC/PI double staining method was used to determine cell apoptosis rate. The expression levels of apoptosis-related signaling molecules Bax and Bcl-2 protein were performed by Western blotting. Results MTT showed that apigenin inhibited proliferation of NCI-H1650 cell line in a concentration-and time-dependent manner (P<0.01).Hoechst 33258 nuclear staining showed typical characteristics of apoptosis in certain concentration-dependent manner, such as nuclear condensation and appearance of apoptotic bodies. In addition, the results of flow cytometry staining indicated that the apoptotic rate of NCI-H1650 cells cultured with blank control group and different concentrations of apigenin (10, 20, 40, 80 μmol?L-1) for 48 h was (5.00±0.33)%, (18.05±4.67)%, (21.48± 1.95)%, (43.24±1.11)%, (66.23±3.65)%, respectively (P<0.01).Western blotting results showed that the expression levels of Bax increased with increasing of the drug concentration, but Bcl-2 decreased with increasing of the drug concentration. Conclusion Apigenin can inhibit the proliferation and induce apoptosis of NCI-H1650 cells in non-small cell lung cancer. Mechanisms may be related to increase of the expression of apoptosis related protein Bax and decrease of the expression of anti apoptosis protein Bcl-2.
7.Feasibility study of intra-utero surgical repair of fetal lamb

Objective To evaluate the feasibility of intra-utero fetal lamb surgical repair. Methods Six twin lambs underwent surgery intra-utero at 112 days of gestation (term 140~160 days). After maternal laparotomy and Hysterotomy, fetal lamb′s toe was excised or its cleft lip was repaired in one of twin. Results At 30 to 32 days post operation,five lambs were spontamcously delivered and the other was cesarean delivery to full term gestation. Fetal wounds healed without inflammation and scar formation. Conclusions The methods of fetal lamb intro utero surgical repair is feasible.
8.Polymorphism of TNFα-308 and mutations of HBV C region in patients with chronic HBV infections

Objective To investigate the association between polymorphism of TNFα-308 and mutations of HBV C region in patients with chronic HBV infection. Methods Ninety-five patients with chronic HBV infection were recruited in the study. The single nucleotide polymorphism(SNP) of TNFα-308 Was determined by restriction fragment length polymorphism(RFLP). Mutations of nt1762/1764, nt1896, nt1899, nt1862, as60, aa87 and as97 in HBV C region were detected by direct sequencing after PCR amplification. Mutations of the above points among different genotypes were compared by Fisher's exget test. Results Three different genotypes G/G(63/95, 66.3%), G/A(28/95, 29.5%) and A/A(4/95, 4.2%) were found in TNFα-308 site. The rates of mutations of aa87 and aa97 points in patients with G/G, G/A and A/A genotype were 39.3%(24/61), 11.5%(3/26) and 50.0% (2/4), respectively(F=7.658, P<0.05);while the mutation rates of nt1762/1764, nt1896, nt1864, nt1899 and aa60 were of no statistical significance among different genotypes(F=0.669, 1.542, 1.123, 2.420 and 0.966, P>0.05). Conclusion Compared with G/G genotype, antigenicity of HBV may be more stable in patients with TNFα-308 G/A genotype, which is beneficial to HBV clearance.

10.The expression of IFN-γ and FOXP3 in the microenvironment of renal cell cancer, and its role in the tumor infiltrating lymphocytes distribution

Objective To investigate the role of IFN-γ and FOXP3 expression in subpopulation distribution and functions of tumor-infiltrating lymphocytes (TILs) in the microenvironment of renal cell cancer.Methods 30 renal cell cancer tissue samples were freshly collected from the laparoscopic radical nephrectomy in the first hospital of Jiaxing.After frozen sectioning,immunofluorescent staining was conducted to detect the infiltrating CD4 positive and CDs positive cells,and the expression of FOXP3 and IFN-γ as well.In addition,TILs were isolated from the tumor tissues by density-gradient centrifugation.TILs from tumor center or tumor invasive edge were purified independently and measured for the mRNA levels of FOXP3 and IFN-γ by qRT (quantitative reverse transcription)-PCR.Results Tumor-infiltrating CD4+ and CD8+ T cells were concentrated in the invasive edge of renal cell cancer tissues.The expression of FOXP3 was found to be inversely related to that of IFN-γ from the immunofluorescent staining.The relative FOXP3 mRNA levels for the TILs from tumor center and invasive edge were 64.6 ± 9.4 and 36.2 ± 1.8,respectively,with significant difference(P <0.05).The relative IFN-γ mRNA levels were 631.8 ± 151.4 and 1 726.0 ± 344.1 (P < 0.05).The trend of relative expression of FOXP3 was reversed in terms of IFN-γ.Conclusions The study on the renal cell cancer tissue samples suggested that the tumor-specific cytotoxic immune cells relatively concentrated in the tumor invasive edge.
