3.Detection of computer tomography perfusion in gastric cancer

Objective To evaluate the relationship between CT perfusion imaging and clinicopathological features in gastric adenocarcinoma.Methods CT perfusion was performed in 31 cases of gastric cancer diagnosed by endoscopy and biopsy one week prior to operation.After an oral intake of 1000—1200 ml of water and an injection of hypotonic agent,perfusion scan was adopted in sixteen multi- slice spiral CT (MSCT) with 60 s of cine duration.Data were analyzed by a commercial software to cal- culate tumor blood flow (BF),blood volume (BV),mean pass time (MTT) and permeability surface (PS).Microvessl density (MVD) was evaluated by using immunohistochemical staining of surgical specimens with anti-CD34.All these findings were prospectively analyzed and correlated with the clinicopathological find- ings (histological grading,presence of lymph node metastasis,serous involvement,TNM staging and MVD). Results There was significant difference of PS value not only between patients with and without lymphatic in- volvement (P<0.05),but also in different histological grade (P<0.05) and TNM staging (P>0.05). However BF,BV,MTT and MVD of gastric cancer revealed no significant correlation with the clinicopatholog- ical findings above (P>0.05).Condusion The analysis of CT perfusion imaging in gastric cancer,especially PS value,might be helpful in predicting the prognosis.
4.Investigation of correlation between systemic lupus erythematosus and human cytomegalovirus infection

Objective To study the relationship between systemic lupus erythematosus (SLE) and human cytomegalovirus (HCMV) infection.Methods HCMV was isolated from the peripheral blood leucocytes (PBL) of 63 patients with SLE.Indirect immunofluorescence (IIF) was performed to investigate HCMV pp65 antigen and polymerase chain reaction (PCH) was performed to investigate the expression of HCMV UL54 DNA in cell cultures.The clinical and Iaboratory parameters were also assessed.Results The rate of HCMV infection in SLF patients was higher than that in the healthy controls and SLE patients.It was higher in the active phase than in the inactive phase.The total amount of urine protein in 24 houm collection,ESR,SLEDA1,preyalence of arthritis and some autoantibodies were considerably higher in patients with positive HCMV infection than those with negative HCMV infection.Conclusion The HCMV infection rate in SLE patients is higher than healthy controls.HC:MV infection may contribute to the disease flare in some SLE pahents.
5.Expression of vascular endothelial cell growth factor and its receptors in vitreous of patients with diabetic retinopathy

To investigate the expression of vascular endothelial cell growth factor (VEGF) and its receptors in vitreous of patients with diabetic retinopathy ( DR), and to discuss its role in the development of DR.
●METHODS: Selected 13 patients (16 eyes) with DR and 15 healthy people (15 eyes), the expression of VEGF and its receptors (fms-like tyrosine kinase-1, Flt-1 and kinase insert domain containing receptor, KDR) were evaluated by immunohistochemistry in vitreous. The levels of VEGF, the Flt-1 and KDR in vitreous of patients with DR were examined with enzyme linked immunosorbent assay (ELlSA).
● RESULTS: lmmunohistochemical staining showed that the expression of VEGF, Flt-1 and KDR in vitreous vessel membrane of patients with DR was increased significantly. And the levels of VEGF, Flt - 1 and KDR in vitreous of patients with DR were obviously higher than that in the control group (P<0. 05 or P<0. 01).
● CONCLUSlON: VEGF, Flt - 1 and KDR were widely expressed in vitreous of patients with DR, and were positively related to micro-angiogenesis of DR patients. lt proved that VEGF and its receptors played important roles in the occurrence and development of DR.
6.Corneal strength change and keratocan expression of corneas during the healing of penetrating injury in rabbits

Background Corneal walleye formation is a result of excessive corneal wound healing.Controlling the corneal trauma or excessive healing reaction after operation of penetrating corneal trauma is a focus in the study of corneal wound healing.Objective This study was to investigate the changes of corneal strength and the expression of keratocan,a marker of corneal stroma,after corneal penetrating injury operation,and to determine the optimal removal time of corneal suture.Methods Full-thickness incisions of 5 mm along the vertical diameter were done at the central cornea on 80 eyes of 80 six-month-old New Zealand rabbits in this study.Then the incisions were interruptedly sutured with 10-0 nylon thread to establish the corneal wound healing models.Each 4 rabbits were sacrificed in 1 week,2,3,4,5,6,7,8 weeks after operation,respectively,and the central corneal stripes were prepared with the size of 7 mm×5 mm.The mean maximal strength of the corneal bands was measured by uniaxial tensile test with electroforce3220-AT biomachanics machine.Then,each 6 whole corneas were obtained at the above time points,and the dynamic changes of keratocan mRNA expression in the specimens were detected by reverse trancription PCR (RT-PCR).The care and use of the animals followed the rules of ARVO.Results The mean maximal strength of corneal stripes was 0,(1.007 ± 0.041),(1.991 ± 0.034),(2.512 ± 0.030),(3.630 ± 0.049) and (4.935 ± 0.004)M Pa in 1 week,2,3,4,5,6 weeks after modeling,and the corneal strength values from 3 weeks through 6 weeks were significantly enhanced in comparison with the value at the adjacent before timepoint (q =6.35,7.54,8.21,5.86,all at P<0.01).The relative expression levels (absorbance) of keratocan mRNA in the corneas were 0.869±0.015,0.779 ± 0.065,0.621 ± 0.027,0.460 ± 0.018,0.393 ± 0.057 and 0.255 ± 0.045 in 1 week,2,3,4,5,6 weeks after operation,and each value was lower than that of the adjacent before timepoint (q =5.24,5.61,7.49,4.75,5.47,all at P<0.01).The intensity of corneal stripes and the expression levels of keratocan mRNA in the corneas were stable in 7 weeks and 8 weeks after operation.Conclusions The dynamic change of corneal strength during the repair of penetrating corneal injury is associated with the down-regulation of keratocan in cornea.Rabbit cornea reaches a maximal strength capacity in 6 weeks after penetrating injury,therefore,it is the optimal time to remove suture.
7.Effects of external counterpulsation combined with laser photocoagulation for treatment of non - proliferative diabetic retinopathy

AIM: To investigate the effects of external counterpulsation combined with laser photocoagulation for treatment of non-proliferative diabetic retinopathy.
METHODS: A prospective study method were used from Aug. 2013 to Feb. 2016. A total of 104 cases in our hospital for treatment of non - proliferative stage of diabetic retinopathy patients were selected as the research object, and all the patients were equally divided into observation group and control group, 52 cases in each group according to the order of admission. Patients in control group were treated with panretinal laser photocoagulation treatment. The observation group were given external counterpulsation combined with laser photocoagulation for treatment, observed the prognosis in the two groups.
RESULTS: The total efficiency in the observation group and the control group were 98. 1% and 84. 6%, the observation group was significantly higher than the control group (P<0. 05). The eye artery EDV and PSV values in the observation group and the control group after treatment were significantly higher than those before treatment (P<0. 05), while the observation group after treatment of eye artery EDV and PSV value were significantly higher than those in control group (P<0. 05). The CMT values in the observation group before and after treatment were 198. 13±45. 32μm and 200. 46±31. 94μm, while the control group were 203. 14±51. 94μm and 202. 90±42. 95μm that compared between the two groups were not statistically significant (P>0. 05).
CONCLUSION: External counterpulsation combined with laser photocoagulation treatment has good safety in the treatment of non-proliferative diabetic retinopathy, it can promote eye artery blood flow speed, thereby improve the therapeutic effect.
8.Effect of constant magnetic field on the osteopontin gene expression in rat aorta smooth muscle cells

BACKGROUND:Constant magnetic field of proper intensity can inhibit the proliferation of vascular smooth muscle cells,and it can be used to inhibit the restenosis following percutaneous coronary intervention.OBJECTIVE: To observe the effect of constant magnetic field of different intensities on the expression of osteopontin gene in rat aorta smooth muscle cells, so as to investigate whether magnetic field can be used to prevent and treat restenosis following percutaneous coronary intervention.DESIGN: A randomly grouping and controlled observation.SETTING: Department of Cardiology, Xijing Hospital of the Fourth Military Medical University of Chinese PLA.MATERIALS: The experiments were carried out in the laboratory of Department of Cardiology (Military Institute of Cardiovascular Disease), Xijing Hospital of the Fourth Military Medical University of Chinese PLA from February to December in 2006. Male pure SD rats of 200-250 g were used.METHODS : Rat aorta smooth muscle cells were cultured in vitro in DMEM medium containing fetal bovine serum (0.1 in volume serum), and then the cells were randomly divided into control group, constant magnetic field of 1, 5, 10 and 50 Gs groups, those in the control group were not treated with magnetic field, and those in the other groups were treated with magnetic field and cultured for another 48 hours. Reverse transcription-polymerase chain reaction and Western blotting were combined with absorbance (A) scanning analysis to observe the effect of constant magnetic field on the expressions of osteopontin and its mRNA in smooth muscle cells.MAIN OUTCOME MEASURES: Expressions of osteopontin and its mRNA in smooth muscle cells.RESULTS: The expressions of osteopontin and osteopontin genes in the constant magnetic field groups were significantly decreased as compared with that in the control group (P < 0.05), there were also significant differences among the constant magnetic field groups of different intensities (P < 0.05). It was indicated that the stimulation of constant magnetic field was in an intensity-dependent manner, and the expressions of osteopontin and osteopontin mRNA were enhanced as the intensity of magnetic field was increased.CONCLUSION: Constant magnetic field of proper intensity can inhibit the osteopontin expression in vascular smooth muscle cells on the gene level, and magnetic field may play a role in preventing and treating the restenosis following percutaneous coronary intervention.
9.Evaluation of Myocardial Ischemia of Kawasaki Disease at Convalescence Period by Radioactive Nuclide Myocardial Perfusion Imaging

Objective To discuss the clinical value of 99Tcm-sestamibi(99Tcm-MIBI) myocardial perfusion imaging on detecting myocardial ischemia in children with Kawasaki disease(KD) at convalescence period.Methods Twenty-one children wih KD at convalescence period were divided into 2 groups according to results of echocardiography.Four cases with coronary artery dilation,17 cases without coronary artery dilation.All cases accepted dipyridamole stress myocardial perfusion imaging.These patients who had positive results were given rest myocardial perfusion imaging again next day.Results Among 21 cases,9 cases(42.8%) were positive in perfusion imaging.Four cases with coronary artery dilation showed myocardial ischemia in different degree detected by myocardial perfusion imaging.Among 17 cases without coronary artery dilation,5 cases(29.4%) were positive.Conclusions Compared to echocardiography,99Tcm-MIBI myocardial perfusion imaging can objectively evaluate the location,extent and degree of myocardial ischemia of children with KD.It will be a routine test in observing its phase development.
10.Effect of stimulation of STN on the firing activities of the SNr neurons in rats.

AIMTo research the spontaneous firing activities during different-frequency stimulation of subthalamic nucleus and microelectrophoresis GABA, Glu and their antagons respectively, approaching the mechanism of DBS in the treatment of Parkinson's disease further.
METHODSUsing extracellular recording to investigate the effect of different-frequency stimulation of STN and microelectrophoresis several drugs on the spontaneous firing activities of the SNr neurons.
RESULTSFor STN stimulation at low frequency, there was no difference on the spontaneous firing activities of SNr neurons between pro-stimulation and meta-stimulation (P > 0.05). With the increasing of stimulation frequency, most of the SNr neurons were inhibited. While during the STN stimulation frequency at high-frequency, the firing rates of inhibited SNr neurons were changed (P < 0.05). Glu had catatonic excitement effect on the SNr neurons, whereas GABA had tonic inhibition effect. 80% of SNr neurons which were inhibited by STN-HFS were not inhibited by STN-HFS on the basis of excitatory effect of BIC.
CONCLUSIONTo treat the motor symptoms of PD, when SIN is selected as the target nucleus, the electrical stimulation with high-frequency should be chosen. It is possible that SIN-HFS modulate the activity of SNr by inhibitory effect of GABA predominantly.
