2.Association of early prognosis and risk factors about early neurological deterioration in aged patients with cerebral hemorrhage in basal ganglia

Objective To study the association of early prognosis and risk factors about early neurological deterioration (END) in aged patients with cerebral hemorrhage in basal ganglia. Methods One hundred and thirty-nine aged patients with cerebral hemorrhage in basal ganglia were selected. The patients were divided into END group (59 cases) and non-END group (80 cases), the clinical data were retrospectively analyzed. The patients were followed up 3 months after discharge, the prognosis was evaluated by modified Rankin scale. Logistic regression analysis was used to determine the independent risk factors of END. Results The incidence of END was 42.4%(59/139). The incidence of unfavourable prognosis in END group was significantly higher than that in non-END group:78.0%(46/59) vs. 30.0%(24/80), and there was statistical difference (P<0.05). The score of Canadian stroke scale (CSS) at admission in END group was significantly lower than that in non-END group: (5.2 ± 2.1) scores vs. (6.9 ± 1.7) scores, but the white blood cell count at admission, hematoma volume and brain ventricle hemorrhage rate were significantly higher than those in non-END group: (10.7 ± 2.9) × 109/L vs. (7.9 ± 2.4) × 109/L, (17.4 ± 14.9) ml vs. (11.2 ± 10.5) ml and 42.4%(25/59) vs. 18.8%(15/80), and there were statistical differences (P < 0.05). There were no statistical differences in sex, age, systolic blood pressure, diastolic blood pressure, body temperature, random blood glucose, fibrinogen, activated partial thromboplastin time, prothrombin time and the incidences of hypertension, smoking history, stroke, diabetes mellitus, hyperlipidemia between 2 groups (P>0.05). The Logistic regression analysis results showed that brain ventricle hemorrhage and white blood cell count at admission were independent risks factor for END in aged patients with cerebral hemorrhage in basal ganglia(P < 0.05 or < 0.01). Conclusions The aged patients with cerebral hemorrhage in basal ganglia will be prone to END, the brain ventricle hemorrhage and white blood cell count at admission are independent risks factor for END, and the early prognosis is poor.
3.A clinical comparison of endoscopic surgery versus craniotomy in hypertensive basal ganglia intracerebral hemorrhage

Objective To compare the clinical effects and characteristics between hypertensive intracerebral hemorrhage (HICH) and haematoma clearance by craniotomy for basal ganglia.Methods Clinical data of 50 patients with HICH in basal ganglia were collected,including operation duration,amount of intraoperative bleeding,cleared amount of haematoma,postoperative intracranial infection,and GOS at the sixth month after operation.The patients were divided into endoscopic group and craniotomy group.Clinical effects were analyzed by using GOS at the sixth month as a prognosis index.Results Preoperatively,the two groups showed no significant difference in any type of clinical materials(all P > 0.05).Operation durations were (1.5 ± 0.8) h and (3.5 ± 1.1) h (P < 0.05),respectively; amounts of intraoperative bleeding were (40.0 ± 19.7) rnl and (40.6 ± 13.2) ml (P < 0.05),respectively; clearance rates of haematoma were (92.6 ± 9.4) % and (73.1 ± 21.1) % (P < 0.05),respectively; cases of postoperative intracranial infections were 0 and 3 (P < 0.05),respectively,for the endoscopy group and the craniotomy group.GOS prognosis at 6 months showed 7 cases of good recovery,12 cases of slight disability,2 cases of severe disability,1 case of vegetative state,and 1 death in the endoscopy group;6 cases of good recovery,9 cases of slight disability,6 cases of severe disability,3 case of vegetative state,and 1 death in the craniotomy group.Prognosis was better in the endoscopy group than in the craniotomy group(P < 0.05).Conclusion Endoscopic surgery is an efficient and minimally invasive and operating technique for the treatment of hypertensive basal ganglia intracerebral hemorrhage.
4.Site-directed mutagenesis of cis-acting element,CHR,in human NFBD1 promoter region

Objective To investigate the role of a cis-acting element,cell cycle genes homology region (CHR),in the transcriptional regulation of human NFBD1 gene (a nuclear factor with BRCT domain 1) by conducting a site-directed mutagenesis analysis on the element in human NFBD1 promoter region.Methods Wild type of NFBD1 promoter reporter,NFBD1-PS1-325,was used as template to make CHR mutant by using PCR based site-directed mutagenesis.Dual luciferase reporter assay was used to determine promoter reporter activity.Adiamycin treatment was employed to investigate the role of CHR in the transcriptional downregulation of NFBD1 after DNA damage.Results Site-directed mutagenesis of CHR caused a significant decrease in NFBD1 promoter activity,and also attenuated the transcriptional down-regulation of NFBD1 after DNA damage.Conclusion CHR element might be involved in both basic transcriptional regulation and transcriptional downregulation of NFBD1 after DNA damage.

6.Utilize the simplified POP-Q system in the clinical practice of staging for pelvic organ prolapse:comparative analysis with standard POP-Q system

Objective To determine the association between simplified pelvic organ prolapse quantification system (S-POP-Q) and the standard pelvic organ prolapse quantification system (POP-Q) in describing pelvic organ prolapse. Methods This was an observational study. From Jan. 2010 to Jan. 2014, 256 subjects with pelvic floor disorder symptoms underwent two exams: a POP-Q exam and a S-POP-Q exam. For the S-POP-Q system, vaginal segments of the exam were defined using points Ba, Bp, C, and D. For the POP-Q system vaginal segments of the exam were defined using points Aa, Ba, Ap, Bp, C, and D. The inter-system consistency between the overall ordinal stages, the anterior vaginal wall stages, the posterior vaginal wall stages, the cervix stages, the posterior fornix or vaginal cuff stages from each two kind of exam were compared. Results The Kendall tau-b correlation coefficient for overall stage was 0.81, the Kendall tau-b correlation coefficients were 0.81, 0.81, 0.85, 0.88 for the anterior vaginal wall, for the posterior vaginal wall, for the cervix, for the posterior fornix or vaginal cuff, respectively. Conclusion There is almost perfect association between S-POP-Q and POP-Q in describing pelvic organ prolapse.
7.The Outcomes Measures of the Cochlear Implantation in Patients with Waardenburg Syndrome

Objective To measure the outcomes of the cochlear implantation for patients with Warrensburg syndrome in comparison with non-syndromic patients with sensorineural hearing loss. Methods 16 of 1 300 Waardenburg syndrome patients who received cochlear implants at PUMC hospital were reviewed in this papers. The authors will discuss the effects between Warrensburg syndrome and non- syndromic patients with sensorineural hearing loss by means of the Parents Evaluation of Aural-Oral Performance of Children (PEACH). Results There were no statistical difference in quiet, in noise, in overall results and in conversation on the phone between Warrensburg syndrome and non-syndromic patients. There was statistical difference between in quiet and in noise in every group. Conclusion The auditory perception and speech recognition between Warrensburg syndrome and non-syndromic patients with sensorineural hearing loss is same, and the results in quiet is better than that in noise.
8.Progress in Elimination of Elemental Sulfur by the Acidophilic Sulfur-oxidizing Bacteria

Metal sulfides are chemically attacked by Fe~ 3+ and H~+, resulting in the formation of elemental sulfur via polysulfides or thiosulfate pathway. Elemental sulfur may aggregate and even form a layer on the metal sulfide surface, which will inhibit leaching metals from the sulfides minerals. Elimination of inert elemental sulfur in a typical acidic environment can exclusively be by way of oxidation of acidophilic sulfur-oxidizing bacteria, such a way includes the attachment, transport and oxidation process of elemental sulfur by acidophilic sulfur-oxidizing bacteria. On the basis of analysis on the pertinent researches, the molecular mechanism of sulfur elimination by the acidophilic bacteria is far away from elucidated, and to attain that target, there are still much work to be done for elucidating the molecular mechanism on the attachment, transport and oxidation process of elemental sulfur by the acidophilic sulfur-oxidizing bacteria.
9.Cystic fibrosis in 2 children with Pseudomonas aeruginosa infection

Clinical data of 2 children with cystic fibrosis (CF) and Pseudomonas aeruginosa infection in the Department of Pediatrics of Sichuan Provincial People′s Hospital from April 2018 to June 2019 were retrospectively analyzed.Patient 1 was an 11-year-old girl with no history of recurrent respiratory infections. Pseudomonas aeruginosa was found in the first sputum culture.A large number of yellow and white secretions were visible under repeated fiberoptic bronchoscopy.Chest CT showed multiple spots and tree-in-bud signs around the bronchi of both lungs. CFTR gene test results revealed 3 heterozygous mutations: c.2909G>A (chr7: 117246728), c.*133T>A (chr7: 117307295) and c. *125delT (chr7: 117307285). The other patient was a 7-year-old boy with a history of recurrent respiratory infections.His parents were close relatives.Multiple cultures of sputum and bronchoalveolar lavage fluid of the boy were Pseudomonas aeruginosa, and chest CT suggested dilation and inflammation in bronchi of both lungs.Gene detection showed that the c. 380T>G homozygous mutation at chromosome chr7-117171059 resulted in an amino acid change p. leu127stop (nonsense mutation). This article suggests that CF should be considered for Pseudomonas aeruginosa infected children having signs of bronchiectasis on chest CT and a large number of secretions under bronchoscopy.Besides, it is necessary for such kind of children to perform genetic testing in time to confirm the diagnosis as soon as possible.
10.Synthesis and antinociceptive activity of seselin derivatives.

Natural product seselin and related derivatives with an angular pyranocoumarin skeleton were synthesized from 8-acetyl-7-hydroxycoumarins by condensation with acetone, reduction, and dehydration successively under mild conditions with total yield of > 50%. Twelve seselin derivatives were tested by the writhing response assay induced by acetic acid at a dose of 40 mg x kg(-1). Seselin (4a) and 4,8,8-trimethyl-9,9-dihydro-pyran[2,3-f] chromene-2,10-dione (2b) showed obviously antinociceptive activity with inhibitory effect of 85% and 50%, respectively, more or quite potent than aspirin in the same assay, suggesting that seselin derivatives could be a novel kind of potential antinociceptive agents.
