1.Quantitative Study of Rutin in Different Commercial Fructus jujubae

Objective To compare the rutin contents in 7 kinds of commercial Fructus jujubae by HPLC method. Methods The analysis was carried on MN NUCLEODUR C18 column. The mobile phase consisted of acetonitrile-0.05 % H3PO4(15 ∶ 85), flow rate was 1.0 mL/min, determination wavelength was 257 nm and the column temperature was at 40 ℃ . Results The linear range was 0.050 5~ 0.404 4 ? g and r= 0.998 7.The recovery was 96.2 % and RSD 1.6 % (n=6). Conclusion This method can eliminate the interference of sugar ingredients efficiently and is with good repeatability. It is suggested that the plumper the fruits of jujubae, the higher the content of rutin.
2.Correlation between surgical outcomes and body mass index in patients with lumbar disc herniation

Objective To investigate the correlation between surgical outcomes and body mass index (BMI) in patients with lumbar disc herniation. Methods 448 patients who underwent operative treatment for lumbar disc herniation were followed-up consecutively. The height and body weight of each patient was measured and recorded preoperatively. The BMI was calculated by the following formula, BMI=weight/height2 (kg/m2). The patients were asigned into three groups (normal group, BMI 28) based on the BMI. A questionnaire evaluation form was designed according to the 60 indexes put forward by North American Spine Association and the authors' clinical experiences. The post-operative signs and symptoms, living and working ability of the patients were evaluated by the questionnaire. Results Among the total 448 patients, none was lost for follow-up and all the patients were followed-up for average 2.48 (2.08-5.16) years. The improvement rate of the normal group was significantly higher than that of the obese group(P 0.05). The post-operative working ability of the patients in the normal group recovered more quickly than that of the patients in obese group (P 0.05). Objective To explore the effects of polymethylmethacrylate(PMMA)-induced autophagy osteoclasts on bone disso-lution animal models ,and study the mechanism of PMMA particle-induced pyrophosphate osteolysis .Methods 30 8-week-old BALB / c mice were randomly divided into two groups ,sterile air with back injection on mice to form airbag and homologous skulls were implanted into experimental group mice were injected with PMMA particles ,the control group were injected with physiological saline .After 14 d ,the mice were killed ,osteoclast activation related gene (RANK / RANKL ) and autophagy morphological exami-nation of the the airbags tissue and the skull were detect .Results The tartrate-resistant enzyme staining (TRAP)-positive osteo-clasts(21 .31 ± 6 .32)s of experiment group is significantly higher than that of the control group (7 .45 ± 3 .23) ,immunohisto-chemistry showed that autophytic protein microtubule-associated protein 1 light chain 3(LC3) and Beclin 1 antibody staining score level in experimental group was significantly higher than that of control group .RT-PCR showed that the RANK mRNA level(1 .35 ± 0 .05) of experimental group was significantly increased(P< 0 .05) .Conclusion The autophagy induce by PMMA is involved in the formation of osteolysis .
4.Effect of advanced glycosylation end products on expression of connective tissue growth factor in cultured rat aortic smooth muscle cells

Objective To explore the effect of advanced glycosylation end products (AGEs) on expression of connective tissue growth factor (CTGF) in cultured rat vascular smooth muscle cells (VSMCs) and the mechanism of accelerated atherosclerosis in diabetes. Methods VSMCs were isolated and cultured from the thoracic aorta of rat. Reverse transcription polymerase chain reaction (RT-PCR) and Western immunoblot analysis were used to detect the expression level of CTGF mRNA and protein. Results The expression of CTGF mRNA and protein were increased by AGE-BSA in the time- and dose-dependent manners(P<0.05). Conclusions AGEs may increase the expression activity of CTGF in cultured vascular smooth muscle cells.
5.Gene detection and clinical study of rhinovirus isolated from children with acute low respiratory tract infection in Shanghai

Objective To understand human rhinovirus (HRV) etiology of acute lower respiratory tract infection (ALRTI) in children in Shanghai area and establish a nested reverse transcription- polymerase chain reaction (nested RT-PCR) assay.Methods Three hundred and forty-two naso- pharyngeal secretion (NPS) samples from ALRTI cases who were hospitalized were collected during January 2005—December 2005.Nested RT-PCR techniques were used to detect HRV-specific RNA.The PCR products were sequenced and data of nucleotides were analyzed.The proportion of HRV infection in children with ALRTI,the distribution of gender,age and season,and clinical char- acteristics were also investigated.Results Forty-six (13.5%) of 342 samples were HRV positive detected by nested RT-PCR.The sequences of 15 positive samples shared high homology of 83%- 97% with HRV sequence in GenBank.Within the 15 positive samples,nucleotide homology varied from 64.4% to 98.4%,and the ratio of genetic variation was from 1.6% to 48.3%./00.These 15 ampli- cons attribute to the two branches of HRV cladogram.The sequences of 15 amplieons were highly varied,in which single nucleotide mutation and several nearby nueleotides mutations were found. Ribonucleotide deletion and insertion in the nucleotide sequence was also found.HRV positive sam- ples were detected in 33 boys and 13 girls,respectively.The ratio of infection cases between boys and girls was 2.5:1.Of 46 HRV infected cases,27 (58.7%) were less than 12 months of age and 38 (82.6%) were less than 3 years old.HRV infected ALRTI occured all the year round and peaked from March to May.Of the patients whose NPS samples were HRV positive detected by nested RT-PCR,45 patients were diagnosed with bronchopneumonia and 1 was diagnosed with asthmatic bronchitis.Fever of most patients was moderate.The peripheral blood leukocyte counts in thirty-nine (84.8%) patients were less than 10?10~9/L.Neutrophil percentages in thirty-seven (80.4%) patients were less than 0.50.C-reactive protein of thirty-six (78.3%) patients were less than 8 mg/L. All of these features were the characteristics of viral pneumonia.The complications were not common and conditions of most patients were not severe.All the children were cured.Conclusions This nes- ted RT-PCR technique is highly specific,rapid and convenient for the detection of HRV RNA in NPS of patients with ALRTI and the genome of HRV viruses is highly variable.The incidence of HRV infection predominates in children in Shanghai area.ALRTI of HRV is short of specificity and condi- tions of most patients are not severe and their prognoses are fine.

A simple and high throughput method was developed and validated for simultaneous determination of valproic acid and its two toxicant ene-metabolites, 2-enevalproic acid and 4-enevalproic acid in epilepsy patient plasma using liquid chromatography–tandem mass spectrometry. Probenecid was used as in-ternal standard and solid-phase extraction was selected for sample preparation. A chromatographic separation was performed on an Agilent Poroshell SB-C18 column (50 mm ? 4.6 mm i.d., 2.7μm) by an optimized gradient elution at a flow rate of 0.9 mL/min. The total run time was 7 min. Electrospray ionization was used in negative ion mode by multiple reaction monitoring of the precursor-to-product ion transitions at m/z 143.0-143.0 for valproic acid, m/z 140.9-140.9 for 2-enevalproic acid and 4-enevalproic acid for their poor fragments, and m/z 283.9-239.9 for probenecid. The results showed good linearity of valproic acid, 2-enevalproic acid and 4-enevalproic acid in their respective linear ranges. The correlation coefficients were more than 0.998. The intra- and inter-day precision of the assay was less than 11.0%and the accuracy ranged from 2%to 12%. This analytical method was successfully applied to assay plasma concentrations of valproic acid and its two ene-metabolites in epilepsy patient plasma and used for therapeutic drug monitoring.
7.The effect of stress-related hyperglycemia on myocardial function of patients with cardiac disease after non-cardiac surgery

Objective To investigate the effect of stress-related hyperglycemia on myocardial function of patients with cardiac disorder after non-cardiac surgery.Methods The clinical data of 211 patients having undergone thoracic or abdominal operations in Department of Critical Care Medicine of Sun Yat-sen University Cancer Center were retrospectively analyzed. According to the postoperative average blood glucose level in the following 3 days after surgery and the patients' history of cardiac disorder, they were divided into four groups: without hyperglycemia (blood glucose ≤ 10 mmol/L) and cardiac disorder group (HG0CV0 group), without hyperglycemia but with cardiac disorder group (HG0CV1 group), with hyperglycemia (blood glucose > 10 mmol/L) but without cardiac disorder group (HG1CV0 group) and with hyperglycemia and cardiac disorder group (HG1CV1 group). The correlations between the blood glucose and each level of the following items: high-sensitivity cardiac troponin I (hs-cTnI), brain natriuretic peptide (BNP), myocardial zymogram aspartate aminotransferase (AST), creatine kinase (CK), MB isoenzyme of creatine kinase (CK-MB), lactic dehydrogenase (LDH), lactic dehydrogenase isoenzyme (LDH-1), myoglobin (MYO), α- hydroxybutyrate dehydrogenase (HBDH) in each group were analyzed.Results The postoperative blood glucose levels of all 211 patients were significantly higher than those before operation (mmol/L: 8.7±0.2 vs. 5.7±0.2,P < 0.05), and the blood glucose level more than 6.1 mmol/L in postoperative 179 patients accounted for 84.8% of the total. In HG0CV0 group, the postoperative blood glucose level was positively correlated with MYO and AST [r values were 0.193, 0.307; 95% confidence intervals (95%CI) were 0.010 - 0.362, 0.096 - 0.479;P values were 0.038, 0.003]. There were no correlations between postoperative blood glucose level and hs-cTnI, BNP, LDH, LDH-1, HBDH, CK and CK-MB (allP > 0.05). In HG0CV1 group, the postoperative blood glucose level was negatively correlated with hs-cTnI level (r = -0.609, 95%CI = -0.810 to -0.264,P = 0.001). There were no correlations between postoperative blood glucose level and the levels of BNP, MYO, AST, LDH, LDH-1, HBDH, CK and CK-MB (allP > 0.05). In HG1CV0 group, there were no correlations between postoperative blood glucoselevel and hs-cTnI, BNP, MYO, AST, LDH, LDH-1, HBDH, CK and CK-MB (allP > 0.05). In HG1CV1 group, the postoperative blood glucose level was positively correlated with hs-TnI level (r = 0.837, 95%CI = 0.476 - 0.984,P = 0.001). There were no correlations between postoperative blood glucose level and the levels of BNP, MYO, AST, LDH, LDH-1, HBDH, CK and CK-MB (allP > 0.05).Conclusion The early stress-related hyperglycemia after non-cardiac surgery may have a protective effect on myocardial function of patients with cardiac disorder.

Objective Evaluate the efficiency of scientific research output of the 54 departments in a hospital,to put forward improvement suggestions based on the evaluation results.Methods Select appropriate indicators of scientific input and output,use the Data Envelopment Analysis method to evaluate and analyze the efficiency.Results According to the analysis of DEA,calculate the values of overall efficiency,technical efficiency,scale efficiency and scale income.Then compare and analyze the relative efficiency of different units scientific output,to identify the relatively superior department a mong the various categories.Conclusions According to the evaluation results,to find out the input surplus and insufficient output of each decision units.Then we will put forward suggestions on hospital resource allocation to optimize the scientific input and output,to improve the competitiveness of the hospital,and to activate the potential of each department's scientific research.
9.USP14 regulates H2O2 induced oxidative stress in H9c2 cells

AIM: To evaluate the effect of inhibiting ubiquitin-specific protease 14 (USPl4) activity on oxidative stress induced by H2O2 of H9c2 cells.METHODS: The H9c2 cells were incubated with H2O2 at 25 μmol/L for 2 h to establish the oxidative stress injury model.The cells were divided into control group, H2O2 group, IU1 group (25 μmol/L or 50 μmol/L) and IU1+ H2O2 group.The H9c2 cells activity was measured by MTS assay.The level of intracellular reactive oxygen species (ROS) and cell survival rate were analyzed by flow cytometry assay.The changes of the mitogen-activated protein kinase (MAPK) family related proteins were detected by Western blot.RESULTS: Compared with control group, the cell activity and the viability rate in H2O2 group were decreased (P<0.05), while the intracellular ROS, the protein levels of Bax/Bcl-2, P53, p-ERK1/2, p-JNK and p-P38 were increased (P<0.05).Compared with H2O2 group, the cell activity and the viability rate of the H9c2 cells in IU1+H2O2 group were increased (P<0.05), while the intracellular ROS, the protein levels of Bax/Bcl-2, P53, p-ERK1/2, p-JNK and p-P38 were decreased (P<0.05).CONCLUSION: Inhibition of USPl4 activity reduces the oxidative stress injury of the H9c2 cells.The mechanism may be related to inhibition of the MAPK signaling and down-regulation of apoptosis related proteins.

Peer review anti assessment includes the evaluation of expert and index system in two parts.The current status of anti-assessment study mainly focused on expert,much of the index system anti-assessment study reports.As can be seen from the status,the anti-assessment system not yet formed and the application is rare.In urgent need of further research to improve our peer-reviewed scientific research.
