2. The expressions of uPA and PAI-1 in patients with breast cancer and their clinical significance
Tumor 2013;33(4):361-367
Objective: To investigate the expressions of uPA (urokinase-type plasminogen activator) and PAI-1 (plasminogen activator inhibitor-1) in human breast cancer tissues and their clinical significance. Methods: The expressions of uPA and PAI-1 in breast cancer tissue samples from 252 patients with breast cancer were detected by immunohistochemistry. The associations between the expressions of uPA and PAI-1 with the clinicopathological factors were evaluated by using Kruskal-Wallis rank sum test. The correlation of uPA expression and PAI-1 expression was examined by using Spearman's rank correlation test. The survival analysis was conducted to assess the impacts of uPA and PAI-1 expressions on the prognosis of patients with breast cancer. Results: The expressions of uPA and PAI-1 were related to nuclear grade, tumor size, axillary lymph node status and Ki67 expression (P < 0.05), and the expression of uPA was also related to HER-2 (human epidermal growth factor receptor 2) expression (P < 0.05). In breast cancer tissues, the expressions of uPA and PAI-1 showed a positive relationship (rs = 0.773, P = 0.000). In subgroups of patients with lymph node-negative and lymph node-positive breast cancer, a positive relationship (rs = 0.760, P = 0.000; rs = 0.780, P = 0.000) was also seen between uPA and PAI-1 expressions. The tumor size (P = 0.023), nuclear grade (P = 0.008), axillary lymph node status (P = 0.000) and the expressions of HER-2 (P = 0.033), uPA (P = 0.005) and PAI-1 (P = 0.023) were associated with DFS (disease-free survival). Of these factors, axillary lymph node status was an independent prognostic factor for patients with breast cancer. The result of subgroup analysis revealed that the expressions of uPA and PAI-1 could influence the DFS of patients with lymph node-negative breast cancer (P = 0.000; P = 0.045). Conclusion: The results of this study suggest that uPA and PAI-1 may be used as prognostic markers and also be helpful to make treatment decisions, especially for patients with lymph node-negative breast cancer. 5. Synthesis of novel 3-(substituted phenyl) chromone derivatives and their anti-tumor activities
Academic Journal of Second Military Medical University 2010;31(8):864-869
Objective: To search for new isoflavones with more potent antitumor activities from the target compounds synthesized by inserting a double bond or an acetylene bond between chromone and its 3-substituted phenyl, ester and hydroxymethyl groups, or by formation of an amide of 3′ or 4′-amino group of isoflavones with norcantharidin. Methods: The key intermediate 3-iodo-7-methoxyl-4H-chromen-4-one (5) was prepared with 2-hydroxyl-4-methoxyl- acetophenone and ethyl formate. The target compounds with 3-substituted double bond or acetylene groups were synthesized by Heck coupling and Sonogashira coupling reaction; the amide compounds were synthesized by Suzuki coupling reaction and amide formation of 3′ or 4′-amino group of isoflavones with norcantharidin. All of the target compounds were confirmed by 1HNMR, MS and IR spectra. The IC50 values of target compounds were determined by the standard method using two kinds of human tumor cell lines, colon cancer cell line HCT116 and liver cancer cell line SMMC 7721 to study their anti-tumor activities. Results: All the 17 target compounds obtained had certain anti-tumor effect in vitro, with compounds 7h and 7e showing better anti-tumor effects, which were similar to that of control curcumin and more potent than that of genistein in vitro. Conclusion: The insertion of a double bond or an acetylene bond between chromone and its 3-substituted phenyl, ester and hydroxymethyl groups may promote the anti-tumor activities of isoflavone analogues. It seems that the formation of an amide of 3′- or 4′-amino group of isoflavones with norcantharidin has no noticeable promotion effect on the anti-tumor activities.

OBJECTIVE:To study the extraction rate and speciation of antivitiligo-related elements in Opercalina turperthum by water extraction and semi-bionic extraction. METHODS:Water extraction and semi-bionic extraction were respectively used. Wa-ter-soluble state and suspension state in extract of O. turperthum were separated by microporous filtering film;organic and inorgan-ic trace elements in water-soluble state were separated by macroporous resin. The contents of Cu,Zn,Fe,Ca,Mg,Mn and Sr were detected by flame atomic adsorption spectrophotometry, and speciation analysis was conducted. RESULTS:After water extrac-tion,extraction rates of 7 trace elements were 40.47%-72.49%;ratio of suspended particles was 3.69%-8.78%;ratio of organic state/inorganic state was 104.36% in water-soluble state of Sr and 3.94%-48.39% in water-soluble state of Cu,Zn,Fe,Ca,Mg, Mn. After semi-bionic extraction, except for Mn,extraction rates of trace elements were higher than water extraction, extraction rates of Cu,Zn,Fe,Ca,Mg,Sr were 77.69%-90.19%;ratio of suspended particles was 0.39%-8.57%;the ratio of organic state/inorganic state was 72.74%-180.79% in water-soluble state of elements. CONCLUSIONS:Cu,Zn,Fe,Ca,Mg,Mn and Sr in O. turperthum are dissolved easily, mainly existing in the form of inorganic state. After semi-bionic extraction, except for Mn,the dis-solution rate of other elements and the proportion of organic trace elements after dissolution increase significantly. Both dissolution and effective utilization rate of trace elements by semi-bionic extraction are higher than water extraction.
