1.The analysis of correlation of body mass index and atherosclerosis blood pressure in patients with maintenance hemodialysis

Objective To explore the relationship of body mass index ( BMI ) and atherosclerosis,blood pressure( BP) in patients with maintenance hemodialysis(MHD),to further elucidate the pathogenesis of cardiovascular disease in MHD patients.Methods The relationship between BP,intima-medial thickness (IMT) and BMI was analyzed in 101 patients with MHD.Results After dialysis,38% patients had systolic blood pressure over 150mm Hg,21% had diastolic blood pressure of more than 90mm Hg.Contrary to the general population,the correlation between BP and BMI were negatively (r =-0.13,P < 0.01 ).Hypertension in patients with BMI < 20 were more than that in patients with a normal body weight ( BMI 20.0 ~ 27.5 ) and overweight patients( BML > 27.5 ),and the rate was 35%,29% and 20%,respectively.Univariate analysis showed that BP and BMI were negatively correlated(r =-0.12,P <0.01),IMT was positively correlated with BMI(r=0.411,P <0.05).Conclusion In persistent hemodialysis patients,malnutrition was correlated with the occurrence and development of atherosclerosis and hypertension,and BMI decreasing was an independent risk factor,which could increase the morbidity and mortslity of cardiovascular disease.

3.Detection of P-4 and GP-46 Expression in Leishmania amazonensis Amastigotes and Promastigotes by RT-PCR

Objective To detect the expression level of stage-specific genes in Leishmania promastigotes and amastigotes. Methods Total RNAs were isolated from Leishmania amazonensis stationary promastigotes and three sources of amastigotes: freshly obtained from mouse skin lesions,infected J774.G8 macrophages,and transformed from the cultured promastigotes. mRNAs were conversely transcribed into cDNA with SuperScripII reverse transcriptase and oligo dT primers. The polymerase chain reaction(PCR)was used to amplify the specific fragments of amastigote-specific nuclease(P-4)and promastigote-specific membrane glycoprotein(GP-46). PCR products were analyzed in 1.5% agarose gel. Results A P-4-specific band(273 bp)was observed in all three types of amastigotes with similar density,but it was almost undetectable in promastigotes. In contract,a GP-46-specific band(325 bp)was expressed at a higher level in promastigotes than in all three types of amastigotes. Conclusion Promastigote-derived amastigotes express high level of P-4-specific gene and can be used as a source of amastigotes for biochemical and immunological studies.
4.UPLC-MS in the Detection of Norfloxacin in Loperamide Hydrochloride Capsules

OBJECTIVE: To detect whether there is mixing of norfloxacin in loperamide hydrochloride capsules by UPLC-MS. METHODS: The separation of norfloxacin was performed on C18 column with column temperature set at 35 ℃, and the mobile phase was composed of acetronitrile and 0.1% formic acid (gradient elution) with a flow rate of 0.3 mL?min-1. The sample size was 10 ?L. ESI source was applied with capillary tube tension at 3.0 kV and ion source temperature at 110 ℃, and scan mode was multiple reaction monitor. RESULTS: It has been proved that there was norfloxacin in 2 of the total 5 batches of samples based on the data of retention time of RP-HPLC including ultraviolet spectra, the quasi-molecular ion and the second order mass spectra split fragments. CONCLUSION: The method is highly selective and sensitive, and can be used to detect whether norfloxacin has been illegally added into loperamide hydrochloride capsules.
5.Research progress in invasive properties of malignant gliomas

Malignant gliomas are the most common intracranial tumors,with a highly invasive nature and a poor prognosis. The invasion of malignant gliomas is a complicate and consecutive multi-step process that includes adhesion, degradation and migration and involves various factors such as cell adhesion molecules, enzymes, and cytokines. Research on the invasive properties of malignant gliomas will advance our understanding of their underlying biological behavior and improve clinical treatment.
6.Clinical efficacy comparison between electroacupuncture and meloxicam in the treatment of knee os- teoarthritis at the early and middle stage: a randomized controlled trial.

OBJECTIVETo compare the clinical efficacy on knee osteoarthritis (KOA) at the early and middle stage between electroacupuncture (EA) and meloxicam.
METHODSNinety patients of KOA at the early and middle stage were randomized into an EA group and a meloxicam group, 45 cases in each one. In the EA group, EA was applied to Dubi (ST 35), Neixiyan (EX-LE 4), Liangqiu (ST 34), Heding (EX-LE 2), Xuehai (SP 10), Yan- glingquan (GB 34) and Zusanli (ST 36); the needles were retained for 20 min and EA was applied once every two days. In the meloxicam group, the meloxicam tablets were prescribed for oral administration, 7. 5 mg, once a day. The treatment lasted for 6 weeks in the two groups. The Western Ontario and McMaster University Osteoarthritis Index (WOMAC) score, rectus femoris muscle tension, the 8-foot walking test and 5-time sit-to-stand test were adopted to observe and compare the effects in the two groups.
RESULTSAfter treatment, every item score in WOMAC was reduced after treatment (all P < 0.05), but the difference was not significant between the two groups (all P > 0.05). In the EA group, the rectus femoris tension after treatment was reduced as compared with that before treatment (P < 0.05) and the reducing result was much more apparent as compared with that in the meloxicam group (P < 0.05). For the 8-foot walking test and 5-time sit-to-stand test, the time was shortened after treatment in the two groups (all P < 0.05) and the result in the EA group was much more obvious than that in the meloxicam group (both P < 0.05).
CONCLUSIONBoth EA and meloxicam are effective in the treatment of KOA at the early and middle stage. EA improves rectus femoris tension and recovers the internal mechanics balance and the efficacy is better than that of meloxicam.

7.Analysis of clinical research literature about treated plague by using traditional Chinese medicine

Objective To analyze the problems of treating plague by TCM and find out the characteristics of clinical research of contemporary plague by the analysis of clinical research literature on the treatment of plague by TCM. Methods Differential treatment methods of one kind of plague in the literature as the main source of the data and the information that the pathogenesis-symptom-location of disease-nature of disease-prescriptions-drugs were taken to build the data base of the syndrome differentiation and treatment of certain kind of plague. Results Analyzed with literatures,the characteristics of plague clinical research showed: Different pathogenesis characteristics and evolution of plague were caused by diff① erent evils. ②Contemporary plague was named by western disease name. A new model which disease differentiation combined with syndrome differentiation has been formed. ③The diversification of administration promoted the treatment of emergent communicable disease with Chinese medicine. Conclusion The plague research lacked such research literature which could both reflect individualized diagnosis and embody the clinical rules. Traditional Chinese medicine theory could be researched and enriched by finding out the pathogenesis characteristics and evolution rule of the new plague and thus guide the clinical practice. It was effective to control and treat mergent communicable disease with TCM.
8.Experimental progress of signal transduction pathways in knee osteoarthritis

BACKGROUND:Studies have shown that signaling pathway plays an important role in the pathogenesis of knee osteoarthritis. OBJECTIVE: To elaborate the experimental progress of signal transduction pathway in knee osteoarthritis METHODS: A computer-based retrieval of PubMed, CNKI, VIP and Wanfang databases was performed to find the relevant literatures concerning experimental studies of signaling pathway in knee osteoarthritis. Al data were primarily screened to exclude repeated and irrelevant articles. Articles about association between knee osteoarthritis and signal pathway were included.RESULTS AND CONCLUSION:A total of 51 articles were colected, including 20 in Chinese and 31 in English. It is concluded that the signal transduction pathway related to knee osteoarthritis is mainly concentrated in the two aspects: the proliferation of chondrocytes and the apoptosis of chondrocytes. The signal transduction pathways related to the proliferation of chondrocytes mainly include Notch signaling pathway and Wnt signaling pathway. SDF-1/CXCR4 signaling pathway, TLR4 signaling pathway, and MAPKs signaling pathway are associated with apoptosis in chondrocytes. Hippo-YAP signaling pathway and ERK1/2 signaling pathway have a double role in the regulation of proliferation and apoptosis of chondrocytes.
9.Preliminary Study on Congenital Transmission of Trichinella spiralis in Mice

Objective To study congenital transmission of Trichinella spiralis in mice and observe the protection of anti-Trichinella antibodies from the infected dams to challenge infection. Methods According to the gestation (fertilization), the Kunming mice were divided into two groups: the infected group after gestation and the gestated group after infection. New-born mice were cut into small pieces to separate the larvae within 1 day after birth. One-day-old offspring born to normal dams were nursed by the infected dams, slaughtered after 21 days and examined for the larvae. Serum anti-Trichinella antibody level in offspring born to the infected dams was assayed by ELISA at different time after birth, and its immune protection against challenge infection was studied. Results Out of 6 offspring born to the dams infected at 7 days after fertilization, two were found to be infected. Among other female mice which were first infected with T. spiralis and then gestated, only the offspring born to the dams fertilized at 8 and 22 days after infection were found to be infected, the infection rate of offspring was 20% (2/10) and 25%(2/8) respectively. All larvae recovered from the young were non-encapsulated. The cross-fostering experiment showed that none of 30 offspring born to normal dams were found to be infected. The serum antibody positive rate in 27 offspring born to the infected dams at 1, 7, 24, and 40 days after birth was 100%, 100%, 77.8% and 14.8%, respectively. The worm reduction rate in the offspring 40 days after birth was 62.0% after challenge infection. 10.Changes of CBS expression in brain following ischemia and reperfusion of limbs

AIM:To detect the changes of cystathionine β-synthase (CBS) expression in the brain following ischemia-reperfusion of hind limbs in the rats and to elucidate their significance .METHODS:Hind limb ischemia was in-duced by clamping infrarenal aorta with a microvascular clip and brain injury was made by following reperfusion .The brain tissue was obtained from the animals subjected to sham operation , 4 h of ischemia without reperfusion and 3 h, 6 h, 12 h, 24 h of reperfusion following 4 h of ischemia .The expression of CBS at mRNA and protein levels was measured at different time points by reverse transcription-polymerase chain reaction ( RT-PCR) and Western blotting .The CBS activity and hy-drogen sulfide ( H2 S) concentration in the brain were determined by a universal microplate reader .The observation of path-ologic changes of the brain was made following the inhibition of CBS by hydroxylamine .RESULTS:The relative expression of CBS at mRNA and protein levels in IR group significantly increased compared with sham group .It reached a peak at 12 h (P<0.01), and returned to baseline at 24 h (P>0.05).This time course correlated with increased CBS activity and H2 S concentration .No statistically significant difference in the above indexes between sham operation group and ischemia group was observed (P>0.05).Inhibition of CBS activity induced more severe brain injury in IR group .CONCLUSION:Up-regulation of CBS/H2 S pathway is involved in the protection for neurons during the ischemia-reperfusion of hind limbs .
