1.Protective effects of total glucosides of paeony on the injury of neonatal rat cadiocytes ;induced by H2O2

Objective To investigate the protective effects and mechanism of total glucosides of paeony (TGP) on the injury of neonatal rat cadiocytes induced by H2O2. Methods Cadiocytes of 3 days neonate rat was cultivated for 72 hours and divided into the model group, the TGP low (30μg/ml), medium (60 μg/ml), high (120 μg/ml) dose groups, Shuxuening injection (100 μg/ml) group and normal control group (n=8);and except the normal control group, the cells in other groups were induced by H2O2 (100μg/ml). After 6 hours, the morphology changes were observed and the survival rates were detected;the content of AST, CPK, LDH in culture medium were detected; the activity of SOD, CAT, GSH-Px and the content of MDA in cardiomyocytes were determinted;the apoptosis rates were detected. Results Compared with the model group, the survival rates of cardiomyocytes in TGP medium and high dose groups significantly improved (64.1%± 7.3%, 81.3%± 7.8%vs. 42.3%± 6.2%;P<0.05 or P<0.01);the content of AST (26.72 ± 6.03 U/ml, 23.86 ± 5.38 U/ml vs. 34.75 ± 6.91 U/ml), CPK (1.96 ± 0.35 U/ml, 1.59 ± 0.32 U/ml vs. 2.54 ± 0.40 U/ml), LDH (811.55 ± 162.32 U/L, 683.48 ± 134.14 U/L vs. 936.07 ± 140.94 U/L) in culture medium were significantly decreased (P<0.05 or P<0.01);the activity of SOD (85.34 ± 16.87 U/mg , 94.62 ± 17.75 U/mg vs. 56.45 ± 13.76 U/mg), CAT (22.76 ± 3.38 U/mg, 26.49 ± 3.72 U/mg vs. 16.13 ± 3.18 U/mg) in cardiomyocytes were significantly increased and the content of MDA (8.74 ± 2.05 nmol/mg, 6.91 ± 1.80 nmol/mg vs. 11.56 ± 2.19 nmol/mg) were significantly decreased (P<0.05 or P<0.01), the apoptosis rate (24.2% ± 5.5%, 13.4% ± 3.9% vs. 51.2% ± 9.1%) were significantly decreased (P<0.05 or P<0.01);and the activity of GSH-Px (3.54 ± 0.83 U/mg vs. 2.50 ± 0.67 U/mg) in cardiomyocytes of TGP high dose treatment group was significantly increased (P<0.05 or P<0.01). Conclusion TGP had dose-dependent protective effects on the injury of neonatal rat cadiocytes induced by H2O2, which perhaps related to its effects of improving the activity of antioxidase, inhibiting the oxidative stress, and decreasing the apoptosis rates.
2.Diagnostic evaluation of serum P53 antibody concentration in lung cancer patients

Objective Statistically analyze the serum P53 antibody level in lung cancer patients to show its diagnostic values.Methods 53 cases of lung cancer and 32 cases of lung cancer after operation and chemotherapy are used as study group. 34 normal controls and 35 cases of lung benign disease were used as control group. All the serum P53 antibody levels of the testees are measured with a ELISA kit.Results In 53 cases of lung cancer group, the positive rate was 58.5% (31/53), while the average serum P53 antibody level was (4.551?6.074)IU/ml. In 32 cases of lung cancer after treatment including surgical and medical procedures, the positive rate was 25% (8/32), while the average serum P53 antibody level was (1.086?2.836)IU/ml. In 35 cases lung benign disease group, the average serum P53 antibody level was (0.163?0.097)IU/ml. In normal group, the average serum P53 antibody level was (0.162?0.08)IU/mL.The lung cancer group had statistical significance in comparison with the other three groups (P0.05).Conclusion The P53 antibody′s appearance is highly correlated with lung cancer. It can be used as an important marker of lung cancer and be beneficial to the differential diagnosis of lung lesions and the evaluation of the treatment.
3.Clinical value of CT-guided percutaneous fine needle aspiration for peritoneal lesions

Objective To investigate the clinical procedural performance of CT-guided needle biopsy for peritoneal lesions.Methods CT-guided needle biopsy was performed in 84 consecutive patients (M : F=26 : 58) with peritoneal lesions.Results Among 84 cases,60 lesions were malignant (55 metastatic tumor,4 mesothelioma,1 lymphoma) and 24 were benign (11 tuberculosis, 13 inflammation).3 cases failed to get clear pathologic diagnosis because of lost data.Diagnostic accuracy was 91.7% (77/84).The major complications were noted in 3 patients(1 with bleeding,2 ascites exosmosis).Conclusion CT-guided needle biopsy for retroperitoneal lesions is highly practical and useful,should be considered complementary in the investigation of abdominal lesions.
4.Low-frequency magnetic fields promote neovascularization in a rabbit model of atherosclerosis combined with limb ischemia

BACKGROUND:Studies on basic research of magnetic treatment of limb ischemic disease are not much, because poor compliance of animals and the stability of the magnetic field strength are difficult to control, resulting in big experimental error and decreased credibility of the results. For this kind of problem, experimental study on low-frequency electromagnetic magnetic cages for treatment of ischemic limbs was conducted, thus overcoming the two major issues of poor compliance of animals and difficult control of the stability of magnetic field strength.
OBJECTIVE:To investigate the effects of self-made low-frequency magnetic fields of rabbit cages on neovascular growth-promoting factor of rabbits with limb ischemia.
METHODS:A total of 96 rabbit models of atherosclerosis were constructed, numbered and randomly divided into ischemia group and non-ischemia group (12 treatment combination in each group). Experiments in each group were performed four times according to the requirement of factorial design. Electromagnetic field intensity factor A (0, 3, 6, 12 mT) and the time factor B (3, 5, 7 days) were set.
RESULTS AND CONCLUSION:Low-frequency magnetic field could apparently promote hypoxia inducible factor-1α, vascular endothelial growth factor and CD34 expression in ischemic limb of rabbits. Electromagnetic field intensity factor A was a key factor for contributing to the expression of hypoxia inducible factor-1α, vascular endothelial growth factor and CD34, and the time factor B was secondary factor. Low-frequency magnetic field also promoted hypoxia inducible factor-1αexpression in non-ischemia limb, but did not promote vascular endothelial growth factor and CD34 expression. Thus, the expression of vascular endothelial growth factor and CD34 was regulated by hypoxia inducible factor-1α, as wel as other factors, in the ischemic state.
5.Establishment of biotin-streptavidin time-resolved fluoroimmunoassay method for the measurement of heparanase

Objective To establish a novel TRFIA for the measurement of heparanase (HPA) in serum samples,and investigate its clinical application.Methods The micro-pore plate wells were first coated with partially recombinant murine anti-human HPA monoclonal antibody.Biotin-labeled recombinant HPA protein was then used to compete with HPA in serum samples,and the prepared europium (III)-labeled streptavidin (Eu3+-SA) was used as signal readout for establishing the BSA-TRFIA assay.Using this assay,the serum HPA levels in healthy subjects (n=32) and tumor patients (n=54) were measured.The results of BSA-TRFIA were compared with those of ELISA.Two-sample t test (or t' test),and linear correlation analysis were used to analyze the data.Results The sensitivity of BSA-TRFIA for measuring HPA was 0.33 ug/L.The CV values for intra-batch and inter-batch were 5.29% and 7.54%,respectively.The average recovery rate was 105.5%.The standard curve range was 0-1 000 ug/L.The serum HPA level measured by the BSA-TRFIA method in healthy subjects was (2.03_+ 1.47) Iug/L.In tumor patients,the HPA level was significantly higher:(22.13_+7.38) ug/L (t'=19.388,P
6.Comparative study on mammography between triple negative and triple positive breast cancer

Objective To analyze the mammographic findings of triple-negative breast cancer [TNBC,which is estrogen receptor (ER) negative,progesterone receptor (PR) negative,and human epidermal growth factor receptor 2 ( HER2 ) negative ] and triple-positive breast cancer ( TPBC,which is ER positive,PR positive,and HER2 positive ),and to evaluate the relationship of immunohistochemologic receptor status and mammographic findings.MethodsThe immunohistochemistry results of 631 cases with breast cancers were reviewed,including 117 cases of TNBC and 44 cases of TPBC.All of the patients took mammography at initial diagnosis.We retrospectively evaluated the visibility,morphology,distribution and size of the lesion (masses and calcifications) and breast density on mammography of TNBC,and compared them with those of TPBC.The age onset and tumor sizes of TNBC and TPBC were compared by using Chi-square test and t test.ResultsThe visibility rate of TNBC and TPBC on mammography were 88.0%(103/117) and 90.9% (40/44),and the difference between them was insignificant ( x2 =0.055,P >0.05).TNBC was more frequently associated with merely a mass (56/103) than TPBC (12/40) (x2 =6.860,P<0.01 ),and the mean diameter of the mass of TNBC [ ( 2.6 ± 1.4 ) cm ] was larger than that of TPBC [(2.0 ± 0.6) cm](t =2.087,P < 0.05). TNBC were less frequently associated with microcalcifications (37/103) than TPBC ( 24/40 ) ( x2 =7.423,P < 0.01 ).Mammographic density and lesion visibility were similar between the two different immunophenotypes of breast cancers.The mean age of TNBC (52±9) was more than that of TPBC (48 ±8) (t =2.759,P <0.01).Infiltrating ductal carcinoma was the main pathologic type of both groups.Basal-like breast cancer accounted for 49% (57/117 ) of TNBC while none happened in TPBC.ConclusionsTNBC shows merely a mass with indistinct margins,lager size and is less associated with microcalcifications.These mammographic features might be useful in diagnosing triple negative breast cancer.
7. Progress of radiogenomics in lung cancer

Radiogenomics aims at investigating the relationship between radiomics features and genomic features, which has certain practical value in the individualized molecular targeted therapy. Meanwhile, it is noninvasive, repeatable and inexpensive. In recent years, a large number of studies have shown that radiomics features have certain predictive values for the mutation status of driver genes of lung cancer. The application of radiogenomics is insufficient at present, but with its continuous improvement and development, it will play an increasingly important role in the precise therapy of lung cancer in the future.
8.Effect of subchronic inhalation of ethylbenzene on expression of heme oxygenase-1 in rat renal tissues.

OBJECTIVETo investigate the effect of ethylbenzene on the expression of heme oxygenase-1 (HO-1) intrarenal tissues.
METHODSForty male Sprague-Dawley rats were randomly and equally allocated to control group, low-dose exposure group, moderate-dose exposure group, and high-dose exposure group to inhale different doses of ethylbenzene (0, 433.5 mg/m(3) (100 ppm), 4335.0 mg/m(3) (1000 ppm), and 6500.0 mg/m(3) (1500 ppm)) for 6 h per day, 5 days per week, for 13 weeks. After the rat model of subchronic ethylbenzene exposure was established, the activities of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT) in renal tissues were measured, and the mRNA and protein expression levels of HO-1 in renal tissues were measured by real-time PCR and Western blot.
RESULTSCompared with the control group, all exposure groups showed significantly decreased activities of GSH-Px and CAT in renal tissues and the moderate- and high-dose exposure groups showed significantly decreased activity of SOD in renal tissues (P < 0.05). All exposure groups showed significantly higher expression of HO-1 than the control group (P < 0.05). The high-dose exposure group showed significantly higher expression of HO-1 than the low- and moderate-dose exposure group (P < 0.05), and the moderate- and high-dose exposure group had significantly higher expression of HO-1 than the control group and low-dose exposure group (P < 0.05).
CONCLUSIONA certain dose of ethylbenzene can induce elevated expression of HO-1 and decreased antioxidant levels in rat renal tissues, thus leading to oxidative stress damage.

9.Establishment and application of light initiated chemiluminescence assay of high mobility group box 1

Objective To establish a fast and quantitative light initiated chemiluminescent assay (LICA) method for high mobility group box1 (HMGB1) determination.Methods Two strains of paired HMGB 1 monoclonal antibodies were used.One was used to coat receptor microspheres.The other was labeled with biotin first and then composed with chain mildew element of affinity donor microsphere to form LICA method for HMGB1.After optimizing the reaction system,the technical specifications of the method was evaluated.Serum HMGB1 levels of common pneumonia patients (CPP) and severe pneumonia patients (SPP) were measured and compared with that of health controls.Two-sample t test was used.Results The sensitivity of LICA was 0.1 μg/L,with linear measurement ranging from 0.1 to 1 000 μg/L.The precisions of intra-and inter-analysis were 1.74%-2.92% and 1.93%-3.73% respectively,both were lower than 5%.The recovery rate was 99.74% (range:94.53%-106.37%).The correlation coefficient of LICA and enzyme-linked immunosorbent assay (ELISA) was 0.888 2.The LICA method had good specificity and no obvious cross reaction with HMGB2 and HMGB3.The serum HMGB1 level in CPP (n=35) and SPP (n=25) was significantly higher than that in health controls (n=35):(6.76±3.13),(19.69±+9.04) vs (1.49±+0.74) μg/L;t values:-5.447 and-5.186,both P<0.01.The HMGB1 levels between CPP and SPP were also significantly different (t=-3.500,P<0.01).Conclusions The established LICA method of HMGB1 has high sensitivity and specificity with reliable results.This method is also homogeneous,fast and cleaning-free,thus has a good prospect in clinical application.
10.Finite element analysis of the correlation between tilt angle of titanium cage and postoperative subsidence of titanium cage after anterior subtotal cervical corpectomy,decompression and fusion

BACKGROUND:Anterior subtotal corpectomy,decompression and fusion is a conventional method to treat cervical degenerative diseases.A titanium cage is an important implant to maintain the stability of the cervical spine after subtotal corpectomy.In recent years,many patients have complications such as titanium cage sinking,which are highly controversial. OBJECTIVE:To investigate the internal biomechanical relationship between the tilt angle of the titanium cage and postoperative titanium cage subsidence after anterior subtotal cervical corpectomy,decompression and fusion. METHODS:A three-dimensional finite element model of the C4-C6 segment was established by CT images of a normal human cervical spine,in which the anterior subtotal resection,decompression and fusion of the C5 vertebral body were simulated,and titanium cages with different tilt angles(-6° to-1° negative angle,that is,the front edge of titanium cage is shorter than the rear edge of titanium cage;1° to 6° positive angle,that is,the front edge of titanium cage is longer than the rear edge of titanium cage)were placed.After setting the boundary conditions,preloads of 50,100 and 150 N were applied respectively on the C4 vertebral body.The stress value of each contact point between the titanium cage and C4 lower-end plate and C6 lower-end plate(seven stress contact points on the contact surface of titanium mesh)was recorded and statistical analysis was conducted. RESULTS AND CONCLUSION:(1)The tilt angles of the titanium cage of the positive angle group and negative angle group under 50,100 and 150 N stress respectively were found by Mann Whitn test,with P<0.05,which was statistically significant.The dispersion coefficients of the positive angle group were smaller than those of the negative angle group under 50,100 and 150 N stress conditions.(2)Under 50,100 and 150 N stress conditions,the Wilcoxon sign rank test in the positive angle group of titanium cage tilt angle found that when the angle was set to 1° to 5°,the difference was not statistically significant(P>0.05).However,when the tilt angle of the titanium cage was set to 6°,the difference was statistically significant(P<0.05).(3)Under 50,100 and 150 N stress conditions,the Wilcoxon sign rank test in the negative angle group of titanium cage tilt angle found that when the tilt angle was set to-1° to-6°,the difference was not statistically significant(P>0.05).(4)It is concluded that in the sagittal position,the titanium cage with a positive tilt angle is more stable than with a negative tilt angle,which is more suitable for clinical use.The tilt angle of the titanium cage is relatively stable in the range of 1° to 5°.When the tilt angle is 6°,the stability starts to decline,which is easy to cause complications of titanium cage sinking after surgery.It is more suitable to select the titanium cage with a tilt angle of 1° to 5° according to the clinical situation during surgery to improve the efficacy.
