1.PPAR? activators inhibitory effects on angiotensinⅡ induced tumor necrosis factora expression in neonatal rat cardiac myocytes

Objective:To explore the effects of the nuclear receptor peroxisome proliferator-activated receptor (PPAR)? activators on tumor necrosis factor ? (TNF-?) expression in neonatal rat cardiac myocytes.Methods:Primary culture of cardiac myocytes was prepared from 1~3 days old Spraque-Dawley rats.Cardiac myocytes were pretreated with rosiglitazone at different concentrations and then stimulated with angiotensin Ⅱ(1 ?mol/L).ELISA and reverse transcripition-polymerase chain reaction(RT-PCR) were used to examine TNF-? in cultured supernatants and TNF-? mRNA in cardiac myocytes,respectively.Results:Pretreatment of cardiac myocytes with rosiglitazone inhibited the increase of TNF-? and TNF-? mRNA induced by angiotensinⅡ in a concentration-dependent manner.Conclusion:Rosiglitazone inhibits the expression of TNF-? in cardiac myocytes,which provides a new way for treating chronic heart failure.
2.Preventive Effects of Iptakalim on Brain Edema Caused by Acute Hypobaric Hypoxia in Rats.

Objective To investigate the preventive effect of iptakalim hydrochloride(Ipt) on brain edema in rats after exposure to simulated high altitude and the mechanisms involved.Method Adult Wistar rats were randomly divided into normoxic control group,simulated plateau acute hypoxia group simulated 8 000 m for 8 h and three Ipt taking groups which were pretreated with 2, 4 and 8 mg(kg?d) of Ipt respectively for 7d before exposure to hypoxia.Water content was determined by the wet-dry method;Na~+,K~+,Ca~(2+) and Mg~(2+) content in the cortex was detected with all atomic absorption spectrophotometer;inorganic phosphorus standardization was applied to decide the activities of Na~+/K~+-ATPase,Ca~(2+)-ATPase and Mg~(2+)-ATPase;Occludin and AQP4 gene expression were determined by RT-PCR.Result The water content was increased and the concentration of Na~+ and Ca~(2+),the activities of Na~+/K~+-ATPase,Ca~(2+)-ATPase and Mg~(2+)-ATPase and the gene expression of the Occludin and AQP4 were all down-regulated in acute hypoxia group compared with normoxic control group.And these changes could be significantly relieved by pretreatment with Ipt.Conclusion Ipt can prevent the occurrence of brain edema caused by acute hypobaric hypoxia possibly related to the effect on water content,electrolytes concentration,ATPase activity and gene expressions of aquaporin and tight junction proteins.
3.Investigations of synchronization of expression between apoptosis and P27~(Kip1),PCNA in esophagus squamous cell carcinoma

Purpose:To study the mutual relationships between apoptosis,cell proliferation correlative gene PCNA and tumor suppressor gene P27 Kip1 as well as to investigate if they have something to do with the occurrence,development,and prognosis in esophagus squamous cell carcinoma.Methods:47 cases of primary esophagus squamous cell carcinoma were put into investigative group whose tumor samples from operation resection were studied. 20 cases with normal esophagus cell mucous membrane tissue were put into the control group. Expression level of PCNA,P27 Kip1 was detected by immunohistochemistry method (S-ABC) in both groups. Apoptotic index was detected by TdT mediated deoxyuridylic acid(dUMP)nicking end labeling (TUNEL method) in both groups.Results:①In the group of 47 cases of esophagus squamous cell carcinoma,there were 17 cases of positive expression in P27 Kip1 ,and the positive rate was 36.17% , average proliferation index(PI) was 45.86?19.11%,average apoptotic index is 1.05%?0.59%,ratio of PI/AI is 57.37? 45.17 ,all of which were statistically significant for difference between normal tissue and carcinoma in esophagus squamous cell (P

Acute respiratory distress syndrome (ARDS) is a serious state threaten human health with a high mortality about 30%-40%. At present, there is no effective treatment for ARDS. Microvesicles derived from mesenchymal stem cells (MSC-MVs) have a heterogeneous subcellular structure secreted by MSCs. It plays an important role in the repair of tissue and organ damage.Recent studies have shown that MSC-MVs, played an important role in repairing lung injury, may replace MSC for cell therapy. Therefore MSC-MVs may bring new hope for ARDS treatment.
5.Study on Quality Standard for Huamoyan Tablet

Objective To establish the quality standard for Huamoyan Tablet. Methods Radix Salviae MiltioiThizae and Radix Cyathulaewere were identified by TLC. The content of protocatechuic aldehyde was determined by HPLC. Results The spots on TLC plates were clear without the interference of negative control. Protocatechuic aldehyde showed a good linear relationship in the range of 0. 021~0. 105 μg, r=0. 999 9. The average recovery was 98. 13 % ,and RSD=2. 41%. Conclusion The method is simple, sensitive and accurate, and can be used to control the qual-ity of Huamoyan Tablet.

Objective The purpose of the present study is to illustrate the effect of olfactory ensheathing cells(OECs) on the survival and neurite outgrowth of GABAergic neurons in vitro. Methods OECs were dissociated from olfactory bulb and neurons from spinal cord of E12 mouse. On the sixth day in vitro,the Millipore cultue blank with OECs was transferred to the neuron culture mediam and continue the co-culture for another 6 days.The cultured neurons were stained with anti-GABA antibody.The neurite of neurons was observed with an image system.The number of GABAergic positive neurons was counted under the microscope. 7.Mycoplasma infection situation of genital tract and medication countermeasures among gynecological patients in hospital of traditional Chinese medicine

Objective To study the infection rate and sensitivity of genitourinary tract mycoplasma among the patients in the traditional Chinese medicine(TCM) hospital ,and its similarities and differences with the western medicine hospital ,thus to increase the therapeutic effect by combining with the related therapeutic measures of TCM .Methods The mycoplasma identification and drug susceptibility test results in 4 086 female genital tract specimens were performed the retrospective analysis .Then the medica‐tion strategy was investigated by combining with clinic .Results Of the 4 086 specimens ,1 891 cases were Mycoplasma positive with the total positive rate of 46 .3% ;in which the positive rate of ureaplasma urealyticum(UU) was higher(40 .7% ,1663 cases) , the most sensitive drugs were pyostacin and josamycin with the sensitive rates of 99 .11% and 99 .01% respectively ;mycoplasma hominis(M H) was less with the positive rate of 2 .7% (110 cases) ,josamycin and doxycycline were sensitive;the positive rate of UU and M H was 2 .9% (118 cases) ,doxycycline and pyostacin were sensitive with the sensitive rates of 95 .77% and 95 .26% re‐spectively .Conclusion Mycoplasma has higher infection rate in the genitourinary tract among gynecological patients ,the results are similar between the hospital of TCM and Western medicine hospital ;UU is mainly Mycoplasma type;the drug susceptibility test re‐sults reveal that the empirical medication for anti‐mycoplasma infection can select doxycycline and josamycin;it is recommended that the combined therapy with syndrome differentiation of traditional Chinese medicine by combining with the drug sensitivity test re‐sults has better clinical efficacy .
8.Angiotensin Ⅱ stimulates TNF-? and NO production in peripheral blood mononuclear cells in heart failure patients

AIM: To examine the change of serum tumor necrosis factor-? (TNF-?), nitric oxide (NO) in patient with congestive heart failure (CHF) and the effect of angiotensin Ⅱ (AngⅡ), valsartan on TNF-? and NO production in culture peripheral blood mononuclear cells (PBMC), to assess the relationship between the renin-angiotensin system and cytokines. METHODS: Venous blood of both healthy volunteers (n=12) and patients with CHF (n=16) were collected. Serum TNF-? and NO were examined. Peripheral blood mononuclear cells (PBMC) were obtained from both the control and the patients groups and cultured with AngⅡ at concentrations of 0, 0.01, 0.1, 1 ?mol/L, respectively. AngⅡ at concentration of 0.1 ?mol/L combined with 0.1 ?mol/L of valsartan was also used. After 24 h incubation, the contents of TNF-? and NO in the culture supernatants were measured. RESULTS: Serum TNF-? and NO production in CHF group were significantly higher than that in control group (P0.05) were observed. AngⅡ stimulated TNF-? and NO release from PBMC of patients with CHF and normal person, which was inhibited by valsartan. CONCLUSIONS: AngⅡ obviously increases TNF-? and NO production from PBMC, which indicates there is relationship between the renin-angiotensin system and TNF-?, NO. The fact that valsartan inhibits TNF-? production may be one of the mechanisms in treating CHF.
9.Health status and family functioning in elderly outpatients with chronic diseases in Gushan town Fuzhou city

Objective To investigate the relation of physical,mental,and social health status with family functioning in elderlv outpatients with chronic diseases to provide theoretic guidance for their community-based health care.Methods Cluster sampling methods were used to assess 568 elderly patients with at least one chronic disease at Gushan Town of Fuzhou City by Activity of Daily Living Scale(ADL),Center for Epidemiologic Studies Depression Scale(CES-D),self made daily social activity questionnaire,and family APGAR index questionnaire.Results The daily activity dependence rate was 23.8%.The prevalence of depression symptoms was 22.9%.Those with normal functions.and modest or severe dysfunction accounted for 56.9%,36.4%,and 6.7%,respectively.The daily activity dependence rate,the prevalence of depression symptoms,and daily social activity were found to be significantly associated with family functioning (P<0.05).Conclusion The health situations of elderly outpatients with chronic diseases may be correlated with family functioning.The community health care providers should concern both individuals and households to improve overall physical and memal health.
10.Cost-effectiveness analysis of morphine-midazolam, propofol and midazolam used for sedation in ICU patients with mechanical ventilation

Objective To evaluate the cost-effectiveness of Morphine-midazolam, propofol and midazolam used for sedation in patients with mechanical ventilation. Methods Ninety-three patients with mechanically ventila-Morphine-midazolam group:priming dose 0.05 mg/kg and 0.05 mg/kg of morphine and midazolam,then continuous The index of ideal level of sedation was on the Ramsay scale. The sedation time, the time from discontinuation to ex-tubation, sedation costs, blood pressure were measured. Results The time in midazolam group (6.0±2.4) h was longer than that of propofol (4.6±1.7) h (P<0.01), but there was no significant relationship between morphine-midazolam group (5.6±2.7) h and midazolam group (4.6±1.7) h (P>0.05). The sedation costs in morphine-mi-dazolam group (101.7±20.4) yuan were lower than those of midazolam group (127.7±21.3) yuan (P<0.05) and propofol group(199.7±65.9) yuan (P<0.01). The ratio of hypotension in propofol group (35.4%, 11/31) hap-pened more frequent than that of midazolam group (3.2%, 1/31) (P<0.01) and morphine-midazolam group (9.7%, 3/31) (P<0.05). Conclusions Morphine-midazolam is a safe, effective and economic drug compared with midazolam and propofol used for sedation in patients with mechanical ventilation.
