BACKGROUND:Potassium channel is the main ion channel to regulate vascular smooth muscle contraction and relaxation, and closely related with vascular tone. However, the reports about the mechanism of potassium channels in the body’s aging process are rare. OBJECTIVE:To investigate the effects of aging on the arterial ultrastructure and smooth muscle potassium channel reaction, and then to explore the possible mechanisms. METHODS:Sixteen healthy male Wistar rats were col ected, 19-month-old rats were assigned to the old group (n=8), 2-month-old rats were used as young group (n=8). The thoracic arteries were isolated and cut into rings to conduct contractility measurement in six rats of each group. The thoracic arteries were stimulated with specific calcium-activated potassium channel blocker tetraethylammonium, specific voltage-dependent potassium channel blocker 4-aminopyridine, specific ATP-sensitive potassium channel blocker glibenclamide, and specific inward rectifier potassium channel blocker BaCl 2 ultrastructure changes under electron microscope. , and then the arterial contractile response to the blockers were observed. The thoracic arteries of the remaining two rats in each group were taken to observe the arterial RESULTS AND CONCLUSION:Compared with the young group, the ultrastructures of the thoracic aortic endothelial cells and smooth muscle cells were changed in the old group;KCI induced the maximum thoracic aortic contractile tension, and then recovered to the baseline tension, and the recovery time in the old group was significantly longer than that in the young group;al the four kinds of blockers could increase vascular tone, and the tetraethylammonium and 4-aminopyridine induced thoracic aortic contractile response in the old group was significantly lower than that in the young group;there was no significant difference in vasoconstriction induced by glibenclamide and BaCl 2 . Aging can induce arterial ultrastructure changes and declination of vasodilatation capacity, which may partly due to the decreasing of the potassium channels function in smooth muscle cells, especial y the calcium-activated potassium channel and inward rectifier potassium channel.

AIM: To explore the feasibility of using vector-based small interfereing RNA(siRNA) to inhibit the expression of MDR1 mRNA and P-glycoprotein and to reverse the multidrug resistance of drug-resistant ovarian cancer cell line.METHODS: An adriamycin-resistant human ovarian cancer cell subline OVCAR/AR was established by stepwise inducement.Another mutidrug-resistant human ovarian cancer cell subline OVCAR/MDR was established by transfecting multidrug resistant gene 1(MDR1) into ovarian carcinoma cell line OVCAR-3.Transfection of MDR1 mRNA specific siRNA expressing plasmids(pSN/mdr1a and pSN/mdr1b) into OVCAR/AR and OVCAR/MDR cells was performed using liposome transfection reagents.MDR1 mRNA expression level was quantified using real time reverse transcription polymerase chain reaction(real time RT-PCR).Flow cytometry(FCM) was performed to assess the expression of p-glycoprotein(P-gp).Multidrug resistant to anticancer agents was evaluated by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide(MTT) assay.RESULTS: Basal MDR1 mRNA expression level in drug-resistant cell line OVCAR/MDR was higher than that in OVCAR/AR cell line, and was both higher than that in its parent cell line OVCAR-3.The expression of MDR1 mRNA and P-gp protein in both OVCAR/AR and OVCAR/MDR cells was inhibited dramatically by transfecting with pSN/mdr1a and pSN/mdr1b.The reversal rate of chemoresistance to adriamycin in OVCAR/AR and OVCAR/MDR transfected with pSN/mdr1a and pSN/mdr1b was 79.5% and 93.9%,compared with control group.CONCLUSION: MDR1 expression in the drug-resistant cell lines is partially inhibited by treatment with vector-based MDR1 specific small interference RNAs at the mRNA and protein level, which increases the chemotherapy sensitivity of these drug resistant ovarian carcinoma cell sublines.

Objective: To investigate the relationship between long-, short-term systolic blood pressure variability (SBPV) and renal damage in elder population. Methods: Our research was conducted in the 3rd physical examination of healthy population from Kailuan group by cohort study. Cluster sampling was used by 25% ratio in subjects≥60 years of age to monitor their 24-hour ambulatory blood pressure and finally, 2464/3064 participants with inclusion criteria were recruited. SBPV indexes as standard deviation of systolic blood pressure (SSD), variability independent of the mean (VIM), maximum-minimum difference (MMD) and average real variability (ARV) were examined; renal damage indexes as estimated glomerular filtration rate (eGFR) and microalbuminuria (ALBU) were detected. Relationships between different long-term, short-term SBPV indexes and eGFR, ALBU were studied by multi-liner regression analysis. Results:①The mean age of 2464 participants was (67.41 ± 6.05) years including 1667 (67.7%) male and 797 (32.3%) female.②Multi-liner regression analysis indicated that different long-term SBPV indexes were not related to eGFR and ALBU; 24h SBPV in all 4 indexes and day-time SSD, MMD, ARV were negatively related to eGFR; 24h ARV and day-time MMD, ARV were positively related to ALBU; night-time SBPV indexes were not related to eGFR and ALBU.Conclusion: Different short-term SBPV indexes were, at certain point related to eGFR and ALBU

Objective:To evaluate the efficacy and safety of narrow-band intense pulsed light in the treatment of erythromelanosis follicularis faciei et colli (EFFC).Methods:The patients with EFFC, diagnosed at the Department of Cosmetic Laser Surgery, Hospital for Skin Disease, Chinese Academy of Medical Sciences, and Peking Union Medical College, from January 2017 to December 2022, were retrospectively evaluated. There were 28 patients, including 24 males and 4 females. They ranged in age from 15 to 35 (21.0±4.2) years. All patients received three sessions of narrow-band intense pulsed light treatments, with intervals of 1-2 months. A follow-up visit was completed 1-2 months after three treatments, and efficacy was evaluated. Two dermatologists assessed the clinical improvement in erythema, pigmentation, and follicular papules on a 4-point scale (0=0-25% improvement, 1=26%-50% improvement, 2=51%-75% improvement, and 3=76%-100% improvement). Adverse events were also recorded. Results:A total of 28 patients received three sessions of narrow-band intense pulsed light treatments and were followed-up. After three treatments, the mean improvement scores for erythema, follicular papules, and pigmentation were (2.02±0.65), (1.38±0.59) and (0.80±0.61) respectively. Among these patients, 67.9% (19/28) experienced over 50% improvement in erythema, 39.3% (11/28) showed over 50% improvement in follicular papules, and 7.1% (2/28) demonstrated over 50% improvement in pigmentation. No blisters and scars were formed.Conclusions:Narrow-band intense pulsed light is a safe and effective treatment for patients with EFFC, particularly for those presenting with erythema.
