1.Optimization of Nisoldipine Buccal Adhesive Tablets Formulation by Orthogoral Design

OBJECTIVE:To optimize the formulation of nisoldipine buccal adhesive tablets. METHODS: The formulation was optimized by orthogonal design with the dosage ratio of carbopol to chitosan in drug-containing layer and protection layer, the content of lactose and the tabletting hardness as factors. The bioadhesiveness and in vitro drug release rate of the tablets was measured as well. RESULTS: The optimal formulation was as follows: the ratio of carbopol to chitosan in drug-containing layer and protection layer were 22.5∶22.5 and 33.3∶16.7 mg, respectively; the dosage of lactose was 10 mg, and tabletting hardness was 5 kg. The averge bioadhesiveness of 3 bacthes of optimized preparations was 79.5 g?cm-2, and the average in vitro drug release rate of the tablets in 12 h was more than 95%. CONCLUSIONS: The optimized nisoldipine buccal adhesive tablets enjoy ideal bioadhesiveness and slow-release character.

Objective To know the current status and its main influential factors of overweight and obesity among residents in Gushan county, Fuzhou city. Method The population aged above 20 years old was sampled by multistage sampling. Back- ground information was collected by face-to-face interview. Data on weight and height was obtained through physical check-ups. Results There were 2 412 objects were interviewed and physically measured. The overall prevalence was 26.0% for overweight and 7.1% for obesity. 4.Determination of the Dissolution of Compound Flavone Capsule by HPLC

OBJECTIVE:To determinate the dissolution of Compound Flavone capsules by HPLC.METHODS:Nova-Pak C18(250mm? 4.0mm,5? m) column was used with column at room temperature.The mobile phase consisted of methanol-0.4% H3PO4(50:50) at a flow rate of 1.0mL? min-1.The detective wavelength was 360nm.The dissolution of the Compound Flavone capsules was determined by basket stirring technique with 0.1mol? L-1 hydrochloric acid as dissolvent at a speed of 100r? min-1.RESULTS:The cumulative dissolution rate of Compound Flavone capsules was above 80% at 30 minutes.The linear range of Quercetin was 0.065 84~ 0.658 4?g(r=0.999 9).The average recovery was 99.35%,RSD=0.92%(n=6).CONCLUSION:The method is simple,accurate and reproducible,and suitable for the guality control of Compound Flavone capsules.

Objective:To investigate the influence of fetal liver AFT024 cells on the transfection efficiency of multidrug resistant gene 1(MDR1)and the in vitro expansion of CD34+ cells derived from umbilical cord blood.Methods:CD34+ cells were isolated from human umbilical cord blood by MACS CD34 Progenitor Cell Isolation Kit and co-cultured with AFT024 cells(AFT024 group)or cultured alone(control group)for 7 days.During the subsequent 14 days,retrovirus carrying MDR1 gene was supplemented twice a week to transfect CD34+ cells.On the 7th,14th and 21st day after culture,the number of total nucleated cells(TNC)was counted,the ratio of CD34+ cells was assayed by flow cytometry(FCM)and the number of CD34+ cells was calculated,and colony-forming cells(CFC)were counted by methylcellulose cultures.RT-PCR method was used to detect the level of MDR1 mRNA in the transfected cells.The expression and function of P-glycoprotein(P-gp)were evaluated by FCM assay and Rhodamine-123 efflux assay,respectively.The gene transfection efficiency was calculated by drug-resistant colony-forming cells assay.Results:(1)The MDR1 mRNA level in AFT024 group than that in control group.The gene transfection efficiency in AFT024 group was significantly higher than that in control group(46.0% vs 15.2%,P0.05).On the 14th day,the expansion fold of TNCs in control group was significantly higher than that in AFT024 group(P0.05).The expansion folds of CD34+ cells and CFCs in the AFT024 group were significantly higher than that of the control group(P
6.Changes of arterial ultrastructure and reaction of potassium channels in smooth muscle in aged rats

BACKGROUND:Potassium channel is the main ion channel to regulate vascular smooth muscle contraction and relaxation, and closely related with vascular tone. However, the reports about the mechanism of potassium channels in the body’s aging process are rare. OBJECTIVE:To investigate the effects of aging on the arterial ultrastructure and smooth muscle potassium channel reaction, and then to explore the possible mechanisms. METHODS:Sixteen healthy male Wistar rats were col ected, 19-month-old rats were assigned to the old group (n=8), 2-month-old rats were used as young group (n=8). The thoracic arteries were isolated and cut into rings to conduct contractility measurement in six rats of each group. The thoracic arteries were stimulated with specific calcium-activated potassium channel blocker tetraethylammonium, specific voltage-dependent potassium channel blocker 4-aminopyridine, specific ATP-sensitive potassium channel blocker glibenclamide, and specific inward rectifier potassium channel blocker BaCl 2 ultrastructure changes under electron microscope. , and then the arterial contractile response to the blockers were observed. The thoracic arteries of the remaining two rats in each group were taken to observe the arterial RESULTS AND CONCLUSION:Compared with the young group, the ultrastructures of the thoracic aortic endothelial cells and smooth muscle cells were changed in the old group;KCI induced the maximum thoracic aortic contractile tension, and then recovered to the baseline tension, and the recovery time in the old group was significantly longer than that in the young group;al the four kinds of blockers could increase vascular tone, and the tetraethylammonium and 4-aminopyridine induced thoracic aortic contractile response in the old group was significantly lower than that in the young group;there was no significant difference in vasoconstriction induced by glibenclamide and BaCl 2 . Aging can induce arterial ultrastructure changes and declination of vasodilatation capacity, which may partly due to the decreasing of the potassium channels function in smooth muscle cells, especial y the calcium-activated potassium channel and inward rectifier potassium channel.
7.Expression and Its Significance of microRNA-125b and microRNA-133b in Plasma in Children with Asthma

Objective To explore the diagnostic value of the quantitative detection of plasma miRNA-125b and miRNA-133b in children with asthma.Methods Thirty asthmatic patients were enrolled in this study and collected the blood specimens during acute phase and stable phase respectively (AP group and SP group).Thirty allergic rhinitis children (AR group) and thirty healthy children were recruited to the control group (NC group).The levels of miRNA in different groups were detected by qRT-PCR.The performance of miRNA-125b and miRNA-133b were evaluated by receiver operating characteristic curves (ROC) and the area under the curve (AUC) (95%CI).Results The relative expression of miRNA-125b in AP group and SP group were significantly higher than A R group (t=3.913,3.120,P<0.01),miRNA-133b.In AP group and AR group the expression of miRNA-133b were significantly higher than control group (t=4.426,4.720,P<0.01).The detection of miRNA-125b yielded an area under the curve of ROC of 0.7989,(95 % CI:0.7111~ 0.8864) in discriminating asthmatic patients from healthy group.And the miRNA-133b was 0.7274 (95%CI:0.586 5~0.863 0) in discriminating asthmatic patients during the acute phase from healthy group.Conclusion The relative expression of miRNA-125b in children with asthma was significantly higher than that in AR group and NC group,miRNA-125b may prove to be a non-invasive biomarker for the auxiliary diagnosis of asthma especially when the relative expression up to 1.998.

The PCR product of PvpA gene was cloned into prokaryotic expression vector pET41a(+) and the recombinant expression vector was then transformed into E.coli DH5a after identified by restriction enzyme digestion and PCR.The positive recombinant plasmid was transformed into E.coli BL21 (D3) and induced to express PvpA protein.The obtained protein was analyzed by SDS-PAGE and Western blotting,purified by Ni-NTA affinity chromatography.The results showed that the purified PvpA fusion protein was obtained successfully.The expressed protein reacted to the high anti-PvpA immune serum from rabbit specially by western blotting.This study would be helpful to established a new diagnostic method for the detection of M.gallisepticum.
9.Investigation of depression disorders and its influencing factors among senile patients with type 2 diabetes mellitus in rural-urban fringe

Objective To investigate depression disorders and its influencing factors among senile patients with type 2 diabetes mellitus(T2DM) in rural-urban fringe in order to supply reference for commu-nity nursing intervention for it. Methods This study was conducted among 323 residents above 60 years old in Gushan town, Fuzhou City. CES-D (center for epidemiology survey-depression scale) and ADL (ac-tivities of daily living scale) were used to evaluate the depression condition and related factors in these pa-tients and the results underwent analysis. Results 25.7% of people with diabetes had various degree of depression, among which 13.3% might had depression and 12.4% had depression definitely. Patients with loss of spouse, living along,reduction of self- care ability and low BMI showed higher incidence of depres-sion. Variables went into the unconditional multivariate logistic regression equation included marriage status and BMI. Conclusions T2DM patients with loss of spouse, living alone,reduction in self-care ability of daily living and low BMI are high risk population of depression. It was proposed that psychological nursing for the aged should be the important content of community nursing.
10.Study on the status and influential factors of activities of daily living among 4237 elderly in Gushan town of Fuzhou countryside

Objective To describe the status of activities of daily living (ADL) and explore the influential factors among the elderly in Gushan town of Fuzhou. Methods A census was conducted by using questionnaires among people aged 60 years and above at this area. Results 17. 3% of 4237 old people investigated decreased their ADL The main influential factors were natural attribute, social economic features, living habits, depressive symptoms, psychomotility, and number of diseases. Conclusions Most of the old people in Gushan town of Fuzhou could take care of their life. Many factors showed great influence on ADL Applying to all-around measures and improving the aged's functions could help to implement successful aging.
