1.Molecular diagnostics---the fourth revolutionary breakthrough in in vitro diagnostic technology

Since the Plan of Human Genome, the project as famous asthe Manhattan Project and the Apollo Project, has finished, it brings about rapid development in molecular diagnosis and leads to revolutionary breakthrough in in-vitro diagnostics directly. Molecular diagnostics have already applicatedextensively at present in tumor , genetic diseases, congenital anomalies, hematologic diseases, infectious disease, neuropsychiatric disorders and organ transplantation .The new molecular diagnostic technologies make much contributions to clinical decision in clinic practice ,whichmeanwhile brings with new challenges to medical laboratory (clinical laboratory) as well.The medical laboratory nowadays will face the demands of high quality and expectation from clinic as well as high risks .The risk consciousness, risk management and quality control should be enhanced in medical laboratory . Strict management and standardize operations should go through pre -examination processes, examination processes and post -examination processes.Every stuff should realizes the essence of “every number is a life”.Only in this way, the medical laboratory can appropriate the development of technology and medicine and the increasing demand of clinical diagnosis , treatment, prognosis, prediction and health management.
2.Research progress of polycystic ovary syndrome with insulin, leptin and androgen

Polycystic ovary syndrome (PCOS) is an endocrine disorder with a variety of etiopathogensis and clinic manifestations. Hitherto its pathogenesy is still uncleared. However, there have been increasing supports for the contribution of the hyperinsulinism and hyperandrogenaemia in the pathological process of the PCOS. Recent years, some literatures reported that hyperleptinaemia is also an important factor. This review seeks to bring together the pothogenesy of the PCOS with these factors.
3.Research progress in international standard materials used for molecular diagnosis

Benefit from the completion of human genome project and the rapid development of bioinformatics,the clinical molecular diagnostics continues to expand its scope of application which involves more widely use in infectious diseases,genetic diseases and tumors and other aspects.In the clinical molecular diagnostics,the application of international standard materials could effectively guarantee the quality of laboratory tests.This paper reviews the research progress in well-established international standard materials used for molecular diagnosis.
4.Current mocular diagnostic technologies in drug related gene detection and its quality control system

Personalized medicine has revolutionized patients ′care, and results in a personalized drug choice and dosage, through integration of pharmacology , drug information and individual ′s genetics information.The genetic testing for genes involved in drug metabolisms and targets become more and more important.However, the variety of the tests as well as the complexity of the technology brought great challenges in laboratory managements and quality assurance for individual clinical laboratory .This article briefly described the development status of pharmacogenomics and focused mainly on the routine quality assurance principle for four different techniques adopted by related genetic testing , and then presented recent research progression of related reference materials in genetic testing .Finally, the author prospects briefly the future development aspect of the quality control system .
5.A clinical study of autologous tumor vaccine in patients with malignant tumors

Objective To evaluate the clinical significance and the mechanism of immunotherapy with autologous tumor vaccine. Methods 400 patients with pathological diagnosis of malignant tumor were enrolled in this study. 5 days after operation, the patients received vaccination of autologous tumor vaccine every 2-3 days for 5 times. 3 days before and 7 days after administrations, the peripheral blood mononuclear cells (PBMC) were isolated to assay the alteration in the proportions of CD3,CD4,CD8,CD4/CD8, and NK cells. Meanwhile, serum IL-2 and IFN-? were measured. Results After active immunotherapy with autologous tumor vaccine, the proportions of CD4 and CD4/CD8 were significantly elevated, while the proportion of CD8 and NK showed no difference. All the patients tolerated the vaccine well without serious side effects, such as skin ulceration. Conclusion The autologous tumor vaccine can elicit specific cellular immune response, and may be a potential approach to treat the cancer.
6.Improvement of natamycin production in an industrial strain by heterologous expression of the afsRS(cla) global regulatory genes.

The afsRS(cla) global regulatory genes from Streptomyces clavuligerus activate the production of two antibiotics in Streptomyces lividans. In this study, we gained an increase of 38% in the production of natamycin (3.56 g/L) in an industrial strain Streptomyces gilvosporeus TZ1401 through the integration of pHL851 that bears the afsRS(cla) global regulatory genes into its genome. We discovered by quantitive real-time reverse transcription PCR (qRT-PCR) that the expression of 6 genes of the natamycin biosynthetic gene cluster were improved from 1.9 to 2.7 times. This suggests that afsRS(cla) improve the production of natamycin through increased transcription. This study provides a good example for applying afsRS(cla) in high yield breeding of industrial antibiotic producers.

7.Determination of 15+1 European Priority Polycyclic Aromatic Hydrocarbons in Edible Oil by Online Solid Phase Extraction and High Performance Liquid Chromatography

A novel method was developed for the determination of the 15+1 European priority polycyclic aromatic hydrocarbons in edible oil by online solid-phase extraction coupled with high performance liquid chromatography-ultraviolet/ fluorescence detection ( online-SPE-HPLC-UV/FL-D ) . The edible oil samples were diluted with isopropyl alcohol, and then filtered. The online extraction was performed on a solid phase extraction ChromSpher Pi column (80 mmí3 mm) and the separation was carried out on a C18 reversed-phase PAH column (250 mmí4. 6 mm i. d, 5μm) using ultraviolet detection at 220 nm and fluorescence detection. Isopropyl alcohol, acetonitrile and water were served as mobile phase in gradient elution. The results showed good linearity for the 15+1 polycyclic aromatic hydrocarbons with all the correlation coefficients (R2)>0. 99. The limits of detection ( LODs ) were between 0. 03 and 12. 23 μg/kg. The recoveries of the sixteen components in the three levels of spiked samples were in the range of 65 . 3%-110 . 5% with the relative standard deviation (RSD, n=6) from 0. 1% to 9. 8%.
8.An clinicopathological analysis and literature review of pleuropulmonary blastoma in an adult

Objective To explore the clinicopathological features ,diagnosis and differential diagnosis of pleuropulmonary blasto-ma(PPB) .Methods A case of PPB was reported by light microscopic observation ,immunohistochemistry and molecular pathology study with review of related literature .Results A 45-year-old female was admitted to the hospital because of cough and dyspnea . Chest radiogram revealed a solid mass in the left lung .Grossly ,the tumor was described as a firm lesion with lumina or multicystic components and well-circumscribed margins .Microscopically ,the tumor was composed of sheets of malignant primitive small cells and fascicles of embryonal rhabdomyosarcoma-like cells with foci hyalinized stroma .Beneath the benign epithelium ,the primitive mesenchymal cells showed as mixed blastematous and sarcomatous characteristics that plump spindle shaped cells presented poor differention with abundant eosinophilic cytoplasms and brisk mitotic activities .Immunohistochemically ,vimentin and MyoD-1 were positive in malignant small cells but some epithelial markers are negative .Meanwhile ,K-RAS extron 3 mutation was detected by high resolution melting analysis(HRMA) .Conclusion Pleuropulmonary blastoma(PPB) is a rare malignant tumor with unique clinicopathological features .It should be distinguished from some mimics such as pulmonary blastoma and embryonal rhabdomyo-sarcoma .
9.Observation of Histomorphology and Biomechanics on Tendons Preserved by Vitrification

Objective To investigate the effect of vitrification on the biomechanics of tendon tissue in rabbit.Methods Two frozen methods were adopted.The first group was treated with Cryoprotective Agent,which was composed of 18.64%DMSO(V/V),13.37% Acetamide(V/V),9.17% 1,2 Propylene glyco(V/V),0.10mmol/LTrehalose and 10% Calf serum.The tendon tissue with three steps of preliminary treatment,preserved in Cryoprotective Agen was conserved in liquid nitrogen(-196℃)for 14 days;The second group treated with 15% DMSO and 10% Calf serum served as control group.Transmission electron microscope was used to observe the Histiooytic shape of tendon.Each group was performed with tendon tensile test,which could detect maximum load,the maximum shifting quantity and Young's Elastic Modulus.Results There was no significant damage in the tissue's micromechanism of vitrified tendon.But in cryopreservation group,the tissue's micromechanism was apparently damaged.There was no significant difference between test group and control group in maximum load(P=0.256).The same was the maximum shifting quantity(P=0.065).There was significant difference between test group and control group in Young's Elastic Modulus(P=0.006).Conclusion The damage of vitrification to tendon is less than that of profound hypothermia preservion,especially to tendon's corpuscular shape,but there is no significant difference between test group and control group in tendon's biomechanics.
10.Application of phosphates and phosphonates prodrugs in drug research and development.

Based on the character of the molecular structure, the prodrugs of phosphates and phosphonates were divided into two categories. The first is the drug which contained the phosphate group, introducing protected groups to increase lipophilicity and improve bioavailability. The other one is the drug which had no phosphate group, introducing the phosphate group into molecules to enhance the solubility, regulate the distribution coefficient and enhance the drug-like property. This review focuses on the application of phosphates and phosphonates in drug research and development based on improvement of physico-chemical property, drug safety and the pharmacokinetics.
