1.Expression of Stathmin gene in ovarian cancer tissue and its valuation value on effect of Paclitaxel combined with cisplatin chemotherapy

Objective To study the expression of Stathmin gene in ovarian cancer tissue and its valuation value on effect of Paclitaxel combined with cisplatin chemotherapy.Methods 92 cases of ovarian cancerpatients underwent surgical and postoperative chemotherapy treatment in our hospital during July 2012 to February 2016 were selected with research subject.The expression of Stathmin gene in ovarian cancer tissues and adjacent tissues was measured by fluorescence quantitative PCR.All ovarian cancer patients were divided into high Stathmin group and low Stathmin group with 46 cases according to the expression of Stathmin gene in ovarian cancer tissues.Comparison the chemotherapy effectin ovarian cancerpatients with different Stathmin gene expression.Results The expression of Stathmin mRNA in ovarian cancer tissues was significantly higher than that in adjacent tissues(P<0.05).After chemotherapy, serum tumor markers such as HE4, CA199, CA153, β-HCG in high Stathmin group were higher than those in low Stathmin group, angiogenesis indexes such as CXCR4, SDF-1, VEGF, Ang2 were lower than those in low Stathmin group, apoptotic indexes such as Fas/Apo-1 and Bcl-2 were higher than those in low Stathmin group(P<0.05).Conclusion Stathmin gene is highly expressed in ovarian cancer tissues, and the expression of Stathmin is negatively correlated with chemotherapy.
2.Application of CD10,desmin and vimentin in differential diagnosis of special types of uterine tumors

Objective To study the application value of CD10,desmin and vimentin in the diagnosis and differential diagnosis of special types of uterine tumors.Methods The clinical data of 79 cases of special types of uterine cancer were retrospectively analyzed.CD10,desmin and vimentin were detected respectively by immunohistochemical streptavidin-perosidase(SP) method.Results The positive rates of CD10 in endometrial stromal sarcoma,epithelioid leiomyoma,atypical leiomyoma,leiomyosarcoma,carcinosarcoma were 76.2%,0.0%,13.3%,30.8%,75.0%,respectively.The positive rates of vimentin in endometrial stromal sarcoma,epithelioid leiomyoma,atypical leiomyoma,leiomyosarcoma,carcinosarcoma were 100.0%,5.6%,0.0%,46.2%,83.3%,respectively.The positive rates of desmin in endometrial stromal sarcoma,epithelioid leiomyoma,atypical leiomyoma,leiomyosarcoma,carcinosarcoma were 0.0%,61.1%,53.3%,30.8%,0.0%,respectively.The positive rates of vimentin and CD10 in endometrial stromal sarcoma and carcinosarcoma were all significantly higher than those in atypical leiomyoma,epithelioid leiomyoma(CD10:χ2=23.255,13.829,15.880,8.102,all P=0.000;vimentin:χ2=35.159,36.000,15.556,16.440,all P=0.000).The positive rates of desmin in atypical leiomyoma,epithelioid leiomyoma tissues were significantly higher than those in endometrial interstitial sarcomas,carcinosarcoma(χ2=11.480,6.717,17.875,9.097,all P=0.000).Conclusion CD10,desmin and vimentin can be used as sensitive indicators for the differential diagnosis of uterine tumors with special types.
3.Transgenic mice of Alzheimer's disease: developing and preliminary identification of transgenic mice harboring mutant amyloid precursor protein gene fused with two types of fluorescent protein genes

Objective To develop transgenic mice harboring the fusion gene of mutant amyloid precursor protein and two types of fluorescent protein for the future study on Alzheimer's disease.Methods The fusion gene CFP-54 bp-YFP-C99 was introduced into mice by mieroinjection.The presence of CFP-54 bp-YFP-C99 was confirmed by PCR in the founders.Results CFP-54 bp-YFP-C99 gene was injected into pronucleus of 2202 zygotes and 1806 injected eggs were implanted into 56 foster mothers, 13 of which were pregnant.There were 13 foster mothers who borne 52 offspring and 32 of them survived.Recipient mouse pregnancy rate was 23.2% (13/56) and the integration rate was 3.9% (2/52).Conclusion CFP-54 bp-YFP-C99 transgenic mice is obtained, but the transgenic efficiency is low.

5.Clinical significance of plasma D-dimer and fibrinogen detection in patients with AECOPD with hypoxemia

Objective:To explore the changes and their clinical significance of plasma d-dimer (D-D) and fibrinogen (Fib) in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) with hypoxemia.Methods:150 patients with AECOPD admitted to our hospital from September 2017 to October 2018 were selected and divided into simple AECOPD group (75 cases) and AECOPD combined with hypoxemia group (75 cases) according to the arterial blood gas analysis results. In addition, 75 healthy people in the same period were selected as the control group. The D-D, Fib and C-reactive protein (CRP) level were detected in venous blood, and the differences and correlation of these indexes among the groups were statistically analyzed.Results:The D-D and Fib of AECOPD combined with hypoxemia group were significantly higher than those of AECOPD group and healthy control group, and the AECOPD group was higher than the healthy control group ( P<0.001). In AECOPD group, the plasma Fib and D-D were positively correlated with CRP ( r=0.617, P<0.001; r=0.300, P=0.009), but not with platelets (PLT) and white blood cell (WBC) ( P>0.05); in AECOPD combined with hypoxemia group, the plasma Fib was positively correlated with CRP, WBC and PLT ( r=0.854, P<0.001; r=0.345, P=0.002; r=0.272, P=0.018), but there was no correlation between D-D and CRP, WBC and PLT ( P>0.05). Conclusions:The plasma level of D-D and Fib increase with the aggravation of chronic obstructive pulmonary disease (COPD). They could be considered as potential inflammatory markers for the assessment of inflammation in the progression of COPD.
6.Expressions of EOS and COX-2 in nasal polyps in patients with aspirin triad syndrome and its significance.

OBJECTIVE:
To explore the regulatory role of COX-2 and aggregation of EOS in the pathogenesis of Aspirin triad syndrome.
METHOD:
There were 34 patients with ST (group 1), 30 patients with chronic sinusitis and nasal polyps and asthma (group 2), 30 patients with sinusitis (control group). Nasal polyps of 94 patients were obtained in endoscopic sinus surgery,HE staining and immunohistochemical SP staining were performed to detect the distribution of eosinophils(EOS) and the expression of COX-2. Statistical analysis was performed using the SPSS software and then the relationship between their expression and distribution with clinical pathology and pathogenesis was analyzed.
RESULT:
EOS infiltrated extensively in nasal polyps, and EOS counts in these groups were 80.02 +/- 6.11, 76.62 +/- 5.22, 65.97 +/- 4.78,respectively. The difference between ST group, ATA group and control group are significant (P < 0.05), no significant difference between ST group and ATA group. COX-2 in the nasal polyps was mainly expressed in submucosal glands, glandular epithelial cells, epithelial cells, endothelial cells and EOS in interstitium. The positive cell count were 88.13 +/- 6.26, 89.46 +/- 5.97, 91.22 +/- 4.11, respectively. There was significant difference (P < 0.05) between ST group and control group. No significant difference (P > 0.05) was found between ATA group and control group,ST group and ATA group.
CONCLUSION
The difference of EOS infiltration in patients with ST may be associated with an inflammatory response underlying specific clinical manifestations after exposure to non-steroidal anti-inflammatory drugs, and the difference of COX-2 expression in patients with ST may be related to the conversion of metabolic pathway of arachidonic acid and the formation of nasal polyps.

7.Relationship between depression, anxiety, stress and food addiction in primary and secondary school students

Objective:
To explore the relationship between depression, anxiety, stress and food addiction among primary and secondary school students in Yixing City, so as to provide reference and suggestions for maintaining healthy eating behavior and psychological health intervention and promotion among primary and secondary school students in Yixing City.
Methods:
From December 2022 to February 2023, a stratified cluster random sampling method was used to select 4 180 primary and secondary school students from four primary and secondary schools in Yixing City, Jiangsu Province. A questionnaire survey was conducted using the Depression Anxiety Stress Scale-21 (DASS-21) and related behavior questionnaires. The data was analyzed using χ 2 test, Wilcoxon, Kruskal Wallis H rank sum test, and binary Logistic regression.
Results:
The prevalence of food addiction among primary and secondary school students in Yixing City was 0.98% (41 students), and there was no statistically significant difference in the comparison of food addiction, depression, anxiety, and stress scores among students of different genders and age groups ( Z/H = -1.34- 5.74, P >0.05). There was a positive correlation between food addiction and binge eating behavior, depression, anxiety, and stress symptoms ( r=0.14-0.23, P <0.01). The results of binary Logistic regression showed that anxiety ( OR=5.68, 95%CI =1.74- 18.55 ) and stress ( OR=5.41, 95%CI =2.20-13.32) were positively correlated with the occurrence of food addiction in primary and secondary school students ( P <0.01).
Conclusions
The risk of food addiction among primary and secondary school students with anxiety and stress symptoms is higher than that of the general population. Guidance and intervention on student mental health should cover the entire compulsory education and high school period, in order to improve the mental health level of students and reduce the occurrence of food addiction behavior.
8.Identification of a novel GPR143 mutation in a Chinese family affected with X-linked ocular albinism.

OBJECTIVETo detect mutation of GPR143 gene in a Chinese patient affected with ocular albinism.
METHODSPeripheral blood samples were collected from the proband and his parents. The coding regions of the GPR143 gene were subjected to PCR amplification and Sanger sequencing.
RESULTSA previously unreported mutation (c.758T>A) was found in exon 6 of the GPR143 gene in the proband and his mother. The same mutation was not found in his father. As predicted, the mutation has resulted in a stop codon, causing premature termination of protein translation.
CONCLUSIONA novel mutation of the GPR143 gene related to X-linked ocular albinism has been identified.

9.The mechanism,safety and application of berberine in promoting bone regeneration

BACKGROUND:Berberine has the potential to induce osteogenic differentiation of various mesenchymal stem cells under normal conditions and special conditions such as high glucose,infection and inflammation.It is a natural small molecule drug that can induce bone formation in seed cells instead of growth factors,and has great application prospect in bone tissue engineering. OBJECTIVE:To review and summarize the research progress in the osteogenic mechanism and efficacy of berberine,especially its osteogenic potential under high glucose,infection and inflammation conditions,and its biological safety,so as to provide theoretical basis for its development and application in bone tissue engineering. METHODS:PubMed,WanFang,and CNKI were searched for relevant literature using the keywords of"berberine,bone defects,bone repair,bone regeneration,osteoinductive,osteoporosis,osteoblast,osteoclast,bone tissue engineering,bone,high glucose,diabetes,inflam*,infect*"in English and Chinese,respectively.A total of 105 literatures were selected for review. RESULTS AND CONCLUSION:Berberine can be used to treat multiple diseases including bone diseases,and it has the ability to promote bone regeneration.This article systematically reviews the mechanism of berberine on bone regeneration and in vivo and in vitro studies.Studies have shown that it can play a role in bone repair by promoting osteogenesis,inhibiting osteoclast formation and activity,and preventing osteoporosis.It shows excellent osteogenic differentiation potential mainly via Wnt/β-catenin,PI3K/AKT,EGFR/MEK/p38MAPK,cAMP/PKA/CREB,ERK and other signaling pathways.Berberine can also relieve the inhibition of osteogenic differentiation caused by high glucose,infection and inflammation,which provides more possibilities for the treatment of bone defects in patients with diabetes or infection and inflammation in the bone defect site.Berberine also has the advantages of low toxicity,low price,easy access(currently it can be synthesized),which is a relatively ideal bone induction potential drug.In recent years,the application of berberine in the treatment of bone defect tends to be localized,mainly through the combination with bone tissue engineering technology to improve bioavailability,and has shown good bone repair effect and excellent biological safety in animal experiments.In addition,preclinical experiments have shown splendid bone regeneration potential in the conditions of diabetes,local infection and inflammation.In the future,more studies are needed to fully reveal the osteogenic mechanism and biological safety of berberine,and seek the most suitable controlled release loading system to make artificial bone replacement materials with good mechanical strength,efficacy and biological safety.
10.The effect and mechanism of epigallocatechol gallate combined with trastuzumab on the proliferation of HER2 overexpressing breast cancer cells

Objective To study the effect and mechanism of epigallocatechol gallate (EGCG) combined with trastuzu-mab on the proliferation of human epidermal growth factor receptor 2 (HER2) overexpressing breast cancer cells. Methods Trastuzumab was expressed and purified. The cell proliferation of HER2 overexpressing breast cancer cells BT474 and SK-BR-3 treated with trastuzumab, EGCG, or trastuzumab plus EGCG was evaluated by CCK8 assay. The effects of EGCG and trastuzumab on the expression of HER2, epidermal growth factor receptor (EGFR), mitogen-activated protein kinase (MAPK), protein kinase B (Akt), and their phosphorylated proteins in BT474 breast cancer cells were detected by Western blot. Results The results of cell proliferation assay indicated that EGCG and trastuzumab, alone or in combination, effectively inhibited the proliferation of BT-474 and SK-BR-3 cells. And within a certain concentration range, EGCG and trastuzumab showed a synergistic proliferation inhibitory effect on HER2 overexpressing breast cancer cells. Consistent with these results, Western blot results showed that trastuzumab and EGCG, alone or in combination significantly reduced the phosphorylation levels of Akt, MAPK, EGFR, and HER2 in BT474 cells. Moreover, the inhibition effect of EGCG plus trastuzumab was significantly more potent than either EGCG or trastuzumab. Conclusion EGCG and trastuzumab could synergistically inhibit the proliferation of HER2 overexpressing breast cancer cells, which may be related to the regulation of Akt and MAPK signaling pathways.
