Journal of Third Military Medical University 1984;0(01):-
Objective To explore the effect of hypoxia on cultured rat Sertoli cells in vitro. Methods In a Sertoli-germ cells co-culture system, the effect of hypoxia on the morphological characteristics of Sertoli cells were observed, including the number of Sertoli cells, the cytoskeleton changes after Coomassie brilliant blue staining and the detachment of germ cells. Results The loosing and retracting of cytoskeleton in Sertoli cells was found. Both survival rate of Sertoli cells and detachment of germ cells showed time-response relation. Conclusion Hypoxia damages Sertoli cells.

Objective To investigate the effects of the polluted organic substances in urban water environment and genetoxicity of the organic pollutants in drinking water on DNA damage in primary cultured hepatocytes in rats. Methods Qualitative analysis of the organic extracts from source water and drinking water in five waterworks in city C with GC/MS was conducted and the effects of organic extracts on DNA damage in primary cultured hepatocytes in rats were tested with single cell gel electrophoresis assay. Comet length means and percentage of tail cell were observed. Results A total of 98 organic pollutants were detected in all samples including 60 in source water and 58 in drinking water. The pollutants were mainly esters, ketons, phenols, benzenes and the derivatives. Comet length means was found to be significantly higher ( P

Objective To study the related influencing factors of MCF 7 cells proliferation test in vitro to provide some criteria for the standard detection of environmental estrogens (EE) by cell proliferation test of MCF 7 cells in vitro . Methods The sensitivity to E 2 stimulation of different sources of MCF 7 cell lines was detected. Cell proliferation test was performed on the screened sensitive cells in different culture conditions. Results In the estrogen free medium, C cell line of MCF 7 was more sensitive to E 2 stimulation than A and B cell lines ( P

Objective To determine the effects of organic extracts of drinking water on female mice reproductive system. Methods The Kunming female mice were randomly divided into4groups,one as reagent control group, the other three as treated groups,15female mice per group. The treated groups were treated with the organics extracted from the chlorinated drinking water by the solid phase extraction at the doses of12.5,25and50L/kg respectively, once a day, continuously for5d.The pathological changes of ovary were observed by optical microscopy. Results At the10th day after treatment, the number of intermediate follicles andⅡatretic follicles in ovary in the treated group significantly increased, especially the number ofⅡa-tretic follicles in25L/kg group, which was10times as high as that in the control group. At the30th day after treatment, the number of normal follicles revealed no significant changes, the number ofⅡatretic follicles in the25and50L/kg group was still significantly higher than that in the control group, the number of corpora lutea in ovary significantly decreased with the in-creases of doses. The estrous cycle delayed at the level of50L/kg.Conclusion The organic extracts from drinking water had adverse effects on the female reproductive system by affecting the development and maturity of the follicles.

Objective To explore the potential hazards of organic pollutants in chlorinated drinking water on human reproduction. Methods The reproductive toxicity of the organic pollutants extracted by solid phase extraction technique from chlorinated drinking water in a city, was determined by the co-culture of sertoli cells and spermatogenic cells. Results No significant variation of the rate of detached spermatogenic cells was noticed compared with the control rats. The organic extracts stimulated the secretion of lactate from sertoli cells of testis in rats. Conclusion The organic extracts of chlorinated drinking water revealed potential reproductive toxicity.

Journal of Third Military Medical University 2003;0(08):-
Objective To observe the estrogenic activity of octylphenol(OP) in vitro and to conduct a preliminary study of its mechanism. Methods The estrogenic activity of OP was detected by cell proliferation test of MCF 7 cells in vitro and the mechanism was preliminarily studied by growth curve analysis, cell cycle analysis, tamoxifen(Tam) antagonistic test and apoptosis detection. Results OP was found to have estrogenic activity to stimulate the proliferation of MCF 7 cells. Cell cycle analysis revealed that the cell proliferation indexes of OP and 17? estradiol(E 2) were higher than those of alcohol. The estrogenic activities of OP and E 2 to stimulate the proliferation of MCR 7 could be antagonized by Tam. Both OP and E 2 could inhibit the cell apoptosis of MCF 7 cells. Conclusion OP possesses estrogenic activity to stimulate the proliferation of MCF 7 cells. The mechanism may be due to binding to the estrogen receptor, which may have effect on cell proliferation and apoptosis.

Objective To assess the value of a fibrin-targeted contrast agent (EP-2104R) for MR detection of thrombus, and to compare this modality with non-contrast-enhanced (NCE) MRI and Gd-DTPA injection at acute period after thrombus generation. Methods Thrombus was induced with external injury and stasis in 5 rabbits. MRI was performed before and after contrast agent injection at 6.0 h after injury, and the MRI findings were compared with that of histopathologically examinations. Results EP-2104R enhanced MRI accurately detected thrombus, which was superior to both NCE and Gd-DTPA injection (P<0.001). Gd-DTPA injection was not associated with improvement of thrombus detection. Conclusion Being a fibrin-targeted MR contrast agent for in vivo detection of acute thrombus, EP-2104R is superior to NCE MRI and Gd-DTPA injection.

Abstact:Objective To study the relationship between acute mountain sickness and geographic factors in young males, and provide no-vel materials for screening susceptible population and exploring novel research ideas and methods. Methods Total 294 young men came from low altitude were randomly selected, questionnaire was conducted and military AMS standard was applied for diagnosis. Binary logistic re-gression was used to analyze the relationship between 10 different geographical factors and AMS incidence and figure out the risk factors for medical geographical differences of AMS. Results AMS incidence was 52. 04% (153 people),with most commonly seen dizziness,head-ache,shortness of breath, chest tightness and lip empurples, the occurrence was 89. 54%,84. 31%,66. 01%,65. 36% and 60. 78%,respec-tively. Binary logistic regression analysis revealed that altitude was the only geographical risk factor (OR=0. 998,P<0. 05). Conclusion Among the population who entered plateau, lower native altitude they used to live brings higher risk of AMS,therefore corresponding preven-tions should be adapted for those who come to high altitude from low altitude in order to ensure their health and safety.

Objective To explore the teaching effects of high altitude military hygiene protection, and to provide the basis for the reform of teaching methods. Methods A questionnaire survey was con-ducted among 55 students who had attended the training program of high altitude military hygiene protec-tion from 2015 to 2017. The survey included teaching effects, curriculum design, teaching methods and suggestions. The results of the survey were processed with SPSS 18.0, and the results were expressed in number and rate. Results 55 questionnaires were distributed and all of them were valid. 69.1%of the stu-dents considered that it was very rewarding to take part in the training class. 67.3%of the students believed that it was very helpful to improve post competence. 71.8% of the students believed that the knowledge module was necessary. The teaching methods that students wanted to experience are:case teaching, practice teaching, discussion and communication, teacher lecturing, visiting teaching, equipment teaching and litera-ture studying. Conclusions The curriculum of the training program of high altitude military hygiene pro-tection is reasonable and helpful in improving post competence. Students recommended flexible teaching methods, which are suitable to try the reform of teaching methods.
