1.Expression of vascular endothelial growth factor and microvessel density in gerbil model with inoculated hepatic alveolar echinococcsis

Objective To study the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD,CD34) in inoculated hepatic alveolar echinococcsis gerbil model and explore the clinical significance.Methods Sixty healthy gerbils were randomly divided into model group,control group and sham operation group.Each gerbil in both model and control groups had liver vaccination with 0.1 ml immature alveolar echinococcus protoscolex suspension or 0.1 ml phosphate buffered saline by open abdominal surgery,respectively,and the sham operation group only had abdominal surgical procedures.Gerbils were put to death on the 120th day post operation,extracted the eyeball and drawn the blood.The alveolar hydatid tissue,surrounding and distal liver tissue were removed from the model group,and normal liver tissue was collected from the other two groups.The VEGF and MVD-CD34 expression was detected by immunohistochemistry staining,Western Blot,and enzyme-linked immunosorbent assay (Elisa).Results Liver alveolar echinococcsis tissue was only observed in model group.The VEGF expression was observed as yellow or brown yellow granules located in the cytoplasma.The VEGF score and VEGF/β-actin ratio in the different parts were 5.13 ± 1.81 and 1.12 ± 0.05,4.20 ± 1.52 and 0.91 ± 0.02,2.46 ± 1.06 and 0.28 ±0.02,2.20 ±0.77 and 0.30 ±0.02,2.13 ± 1.72 and 0.25 ±0.01,respectively.The VEGF expression in different histological sites showed a gradient change,and bubble hydatid tissues had the highest VEGF expression (P < 0.05).CD34 was expressed in alveolar hydatid tissue and surrounding liver tissue in model group and the CD34 positive cells was stained brown,while CD34 was mainly located in the cell membrane of endothelial cells.The CD34 scores and the CD34/β-actin ratio in the different parts were 5.06 ± 2.46 and 0.66 ±0.01,2.43 ± 1.26 and 0.58 ±0.02,and the other tissues did not have CD34 expression.The alveolar hydatid tissue had higher CD34 expression than any other tissues (P < 0.05).The respective serum VEGF and CD34 level in each group was (106.17 ± 14.17)pg/ml,(38.13 ± 21.36)pg/ml,(42.88 ±17.25) pg/ml,and (1.47 ± 0.28) ng/ml;(0.82 ± 0.12) ng/ml,(0.72 ± 0.16) ng/ml,both are higher in model group than those in the other groups with statistical significance (P < 0.05).Conclusion Angiogenesis is detected in gerbil alveolar hydatid tissues,which might be involved in the infiltrating growth of alveococcus.
2.Analysis on influence factors of asymptomatic cholecystolithiasis prognosis

Objective To study the prognosis of patients with asymptomatic gallstones and explore prognostic factors.Methods The clinical data of 227 patients with asymptomatic gallstones in the First Affiliated Hospital of Shihezi University from December 2013 to March 2016 were collected.All patients were followed with questionnaire,finally 192 cases were had been enrolled,of which 57 cases with clinical symptoms were symptom group and 135 cases with no clinical symptons were asymptomatic group.The prognostic factors were analyzed by un-ivariate and multivariate logistic regression.Results Univariate analysis showed that there were statistically significant difference in the num-ber of stones,stone size,diabetes,high fat diet and body mass index between the two groups(P <0.05).The Logistic regression analysis showed that multiple stones,stone diameter ≥1 cm,diabetes were the major risk factors for prognosis in asymptomatic gallstones.Conclusion Multiple gallbladder stones,stone diameter ≥1 cm,diabetes,high fat diet and obesity may be the major prognostic factors for patients with a-symptomatic gallstones.

Background The pathogenesis of primary open angle glaucoma(POAG) and high myopia are very complex.To construct the regulatory network of virulence genes and relevant genes that involved in pathogenicity are helpful for reveal of the pathogenesis.Objective The aim of this study was to investigate myocilin(Myoc),a gene that contributes to POAG and high myopia in eyes of BXD Recombinant Inbred(BXD RI)mice and construct the regulatory network of Myoc.Methods The affymetrix microarray system was used to detect the differential expression of Myoc in the eyes of C57BL/6J(B6),DBA/2J(D2) and BXD RI mice.Expression quantitative trait loci (eQTL) mapping was performed to construct the regulatory network of Myoc gene.Results The average expression level of the Myoc gene in the BXD strains was 10.83,and the gene exhibited expression levels ranging from 8.39 in BXD55 mice tol 1.43 in B6 mice.The eQTL mapping for the Myoc gene showed a significant likelihood ratio statistic (LRS) of 21.78.The QTL was mapped in chromosome 2,and Myoc was located on chromosome 1,indicating that the Myoc gene was a trans-acting QTL.Olfml2a was identified to be a candidate upstream gene of Myoc by analysis of bioinformatics.Genetic regulatory network analysis demonstrated that a series of genes associated with Myoc probably played roles in the pathogenesis and development of POAG and high myopia.Conclusions The genetical genomics approach provides a powerful tool for constructing pathways that contribute to complex traits,such as POAG and high myopia.
4.Association of the single nucleotide polymorphisms in the calcitonin receptor-like receptor gene with primary angle closure in a Han Chinese population

?AIM: To study the association of the single nucleotide polymorphism ( SNP) rs1157699 in the calcitonin receptor-like receptor ( CRLR ) gene with primary angle closure ( PAC) in a Han Chinese population.?METHODS: All samples, involved 232 PAC cases and 306 controls, were obtained from an epidemiologic survey conducted in Funing, Jiangsu Province, China. Genotyping were carried out by TaqMan-MGB probe using the real time quantitative polymerase chain reaction system to study the relationship between SNP of rs1157699 in CRLR gene and PAC.?RESULTS: The prevalence of CRLRrs1157699 genotype was 67.4%, 30.0%, 2.6% for CC, CT, TT in cases, and 71.3%, 27.0%, 1.7% in controls respectively.There was no difference between the two groups in the distribution of genotype and allele frequencies of rs1157699 (P>0.05).?CONCLUSION:Our results do not support a significant role for rs1157699 in CRLR with PAC.
5.Primary study on executive function in children with Ping-Pong training and swimming training

Objective To investigate the executive function of children with different sports training. Methods Forty children with Ping-Pong training (Ping-Pong group) and 41 children with swimming training (swimming group), aged 6-9 years, completed GO/NOGO task. Behavioral data (reaction time and accuracy) and event related potential component N2 were collected and analyzed. Results The reaction time was significantly faster and accuracy significantly lower of GO task and NOGO task in swimming group than in Ping-Pong group (P<0.05 and P<0.01). There were significant differences in the amplitude of NOGO-N2 on site CPz between swimming group and Ping-Pong group[(-11.36±9.4) μV vs (-7.55±7.99) μV, P<0.05]. Conclusion The inhibitory function of children with Ping-Pong training is stronger than those with swimming training.
6.Clinical effect of 23 - gauge pas plana vitectomy for delay-onset endophthalmitis following cataract surgery

AIM: To discuss the clinical effect of 23-gauge pas plana vitrectomy ( PPV) for delay-onset endophthalmitis following cataract surgery.? METHODS: This retrospective chart review study included patients with delay-onset endophthalmitis that was present 6wk or more after cataract surgery, who underwent 23-gauge PPV from January 2010 to January 2016 at Renmin Hospital of Wuhan University. The diagnosis was made according to clinical symptoms and signs. The postoperative follow - up results were recorded, including anterior segment examinations, fundus examinations, and best-corrected visual acuity.?RESULTS:Totally 15 patients (15 eyes) were included. The average time between cataract surgery and their eye symptoms of endophthalmitis was 5. 13 ± 2. 41mo. The culture isolates revealed that 9 eyes ( 60%) were cultured positive, anaerobes in 4 eyes (44%), fungal infections in 3 eyes ( 33%) , aerobes in 2 eyes ( 22%) . The mean follow-up period was 12mo. The final visual outcomes were as follows:11 eyes ( 73%) achieved a better vision with PPV; 3 eyes ( 20%) achieved a better vision with PPV, total capsulectomy ( TC ) and IOL removal; 1 eye ( 7%) with PPV, silicone oil tamponade, total capsulectomy ( TC ) and IOL removal was enucleated because of uncontrolled inflammation.?CONCLUSION: The 23-gauge PPV is safe and effective for the management of delay - onset endophthalmitis following cataract surgery. Recurrence rate might be decreased with total capsulectomy and IOL removal.
7.Effects of forepaw sensorimotor deprivation in early life on spatial learning and memory in rats

Objective To explore the effects of forepaw sensorimotor deprivation in early life on hippocampus-dependent spatial reference learning and memory in rats. Methods Newborn SD rats were randomly assigned to experiment group (deprivation of forepaw sensorimotor function, n=53) and control group(n=55). Rats of postnatal day 13 (PN13) in experiment group were seleeted, and models of forepaw sensorimotor deprivation were established by microsurgical technique. Open field tests and Morris water maze tests were performed during the time periods of PN25(PN21-31), PN35 (PN31-39), PN45(PN41-50) and PN60(PN56-64) to evaluate the locomotor activity and spatial reference learning and memory, respectively. Results In open field tests, there was no significant difference in parameters of locomotor activity and exploratory behavior between the two groups (P>0.05). In Morris water maze tests, eontrol group performed significantly better than experiment group during training sessions and probe tests on PN25 and PN35 (P<0.05). While on PN45, although there was no significant difference between the two groups during training sessions, control group performed significantly better than experiment group during probe tests (P<0.05). Conclusion The deprivation of forepaw sensorimotor in early life has no signifieant effect on the locomotor activity and exploratory behavior of rats, but can impair the spatial reference learning and memory.
8.Quantitative assessment of DNA damage directly in age-related cataract patients

Background Age-related cataract is one of the common causes of blindness.Although the pathophysiology of age-related cataract is far from clearly understood,it is well accepted that DNA damage plays an important role in the disease pathogenesis.Objective The purpose of this study was to quantitatively evaluate the DNA damage in peripheral lymphocytes of age-related cataract.Methods A cross-sectional study was carried out.This study complied Declaration of Helsinki and approved by Ethic Committee of Affiliated Hospital of Nantong University.Written informed consent was obtained from each subject.Two hundred and eleven patients with agerelated cataract and 147 normal subjects were enrolled from a “ Jiangsu Eye Study:Funing 2011 Eye Disease Epidemic Survey”.All the subjects aged from 50 through 80 years with matched age and gender between the two groups.The percentage of tail DNA and Olive tail moment (OTM) were detected by comet assay to assess the extent of DNA damage in peripheral lymphocytes.Statistical analyses were performed with SPSS 17.0 software,and the differences of the percentage of tail DNA and OTM were compared between the age-related cataract group and normal control group by independent sample t test as well as among the 50-59 years group,60-69 years group and ≥70 years group by one-way analysis of variance.Results Comet assay showed a round lymph cell with the clear border in the normal group;while in the age-related cataract group,the cell was bigger with a comet-like tail.The percentage of tail DNA and OTM in peripheral lymphocytes were (21.75 ± 3.51) % and 6.54 ± 1.65 in the age-related cataract group,and those in the normal control group were (9.31 ±3.60)% and 2.18 ± 1.10,respectively,with significant differences between them (t =32.67,P =0.00 ; t =28.02,P =O.00).In the 50-59 years subgroup of the age-related cataract group,the percentage of tail DNA and OTM in peripheral lymphocytes were (20.04±2.86) % and 5.92± 1.14,and in the 60-69 years subgroup of the age-related cataract group,the percentage of tail DNA and OTM in peripheral lymphocytes were (20.77 ±2.93) % and 6.13 ± 1.14,which were significantly reduced in comparison with (22.79 ± 3.67)% and 6.95±1.91 of the ≥70years subgroup(TailDNA%:q=2.75,P=0.00; q=2.02,P=0.00;OTM:q=1.03,P =0.02 ; q =0.82,P =0.00).Conclusions The pathogenesis and development of age-related cataract probably is associated with DNA damage.
9.Ten-year changes in pathogen, antimicrobial susceptibility and clinical feature of children with bacterial meningitis.

OBJECTIVEDespite progress in antibiotic therapy and intensive care, childhood bacterial meningitis (BM) remains a devastating disease. We conducted this study to investigate the changes in clinical characteristics, the etiologic agents and antimicrobial susceptibility of BM during the past 10 years in children under 14 years of age.
METHODSThese 126 patients were divided into two groups according to their date of admission. Group 1 included 64 patients admitted from January 1998 to December 2002, and group 2 included 62 cases admitted from January 2003 to December 2007. All pediatric medical charts of them were reviewed.
RESULTSThe predominant isolated bacteria from CSF were coagulase-negative staphylococcus (17/62, 27.4%) and Escherichia coli (9/62, 14.5%) in group 2. The resistance rate of staphylococcus against oxacillin (MRS) was 68.4% (13/19) in group 2, significantly higher than that of group 1 (16.7%, 2/12). Among 126 cases, 42 had seizure attack and 16 had consciousness disturbance, the proportions of them in group 2 (11/62, 17.7%; 4/62, 6.4%) were lower than those in group 1 (31/64, 48.4%; 12/64, 18.8%, P < 0.05). Cases in group 2 survived with complications [13/62 (21.0%)] and sequelae [11/62 (17.7%)] were lower than those in group 1 (24/64, 37.5%, 23/64, 35.9%, P < 0.05), but the rate of empirical therapy modification in group 2 (21/62, 33.9%) was higher than that in group 1 (7/64, 10.9%, P < 0.01).
CONCLUSIONThe predominant bacteria in children with BM are staphylococcus and Escherichia coli in recent years. The antibiotic resistance rate of bacteria has been higher year after year. The clinical patterns of pediatric BM have changed with a decrease in clinically serious cases, complications and sequelae, but an increase in modification of empirical therapy.

10.Qualitative and quantitative determination of the main components of huanglianjiedu decoction by HPLC-UV/MS.

AIMTo establish a comprehensive HPLC analytical method of Huanglianjiedu decoction.
METHODSThis study was performed by HPLC-UV/MS to identify the chemical constituents of the whole and individual herbs of the "Huanglianjiedu decoction". Zorbax Extend C18 (150 mm x 4. 6 mm ID, 5 microm) column was used; the mobile phase was composed of acetonitrile (A) and water (B, with 0.5% acetic acid) with gradient elution; the flow rate was 1.0 mL x min(-1) and the column temperature was setup at 25 degrees C. The detection wavelength was 254 nm.
RESULTSThe chromatogram of Huanglianjiedu decoction showed 21 main peaks. Peaks 1, 2, 5 and 18 were from Gardenia jasminoides Ellis, Peaks 8, 13, 14, 15, 16, 17, 19 and 21 from Scutellaria baicalensis Georgi. While 10 from Coptis chinensis Franch and 20 from Phellodendron amurense Rupr., Peaks 3, 4, 6, 9, 11 and 12 came from them together. Peak 7 presented in the chromatograms of the herbs except Gardenia jasminoides Ellis. By comparison of the retention time, the on-line UV spectra and MS spectra, 11 peaks were identified as 5 (geniposide), 9 (jatrorrhizine), 10 (coptisine), 11 (palmatine), 12 (berberine), 13 (baicalin), 15 (oroxin A), 17 (wogonoside), 19 (baicalein), 20 (obaculactone), 21 (wogonin), then eight of them were quantified by HPLC-UV.
CONCLUSIONThe method could represent the characteristics of Huanglianjiedu decoction, and it could be used to evaluate the quality and quantity of Huanglianjiedu decoction. It distinguished between Coptis chinensis Franch and Phellodendron amurense Rupr. by HPLC for the first time.
