1.Determination of Baicalin in Shangsuyu Spray by HPLC

Objective:To establish a HPLC method for the determination of baicalin in Shangsuyu Spray.Methods:A Shim pack CLC ODS column was used. The mobile phase consisted of NH 4H 2PO 4 buffer (3.0) methanol (50∶50). The detective wavelength was 276nm.Results:The linear range was from 1.08 to 20.60?g/ml, r =0.9999. The recovery of the added sample was 100.5%. RSD was 2.5%. This method was easy to operate with accurate result, high sensitivity and good reproducibility.Conclusion:The method can be used for the determination of the contents of baicalin in Shangsuyu Spray.
2.Effectiveness of Danning Tablet in patients with non-alcoholic fatty liver of damp-heat syndrome type: a multicenter randomized controlled trial.

OBJECTIVE: To evaluate the efficacy and safety of Danning Tablet (DNT) in patients with non-alcoholic fatty liver disease (NAFLD) of damp-heat syndrome type. METHODS: A multicenter, randomized, double-blinded and positive drug parallel controlled trial was performed. One hundred and thirty-five patients were enrolled into the study and divided into two groups: DNT-treated group (n=102) and ursodeoxycholic acid (UDCA)-treated group (n=33). Body mass index (BMI), principal symptoms, liver function, blood lipids, iconographic, and compositional parameters were measured before and after treatment, respectively. RESULTS: In the two groups, BMI, distress in hepatic region, fatigue, anorexia, liver function, blood lipids and iconographic parameters were significantly improved, and the improvements of BMI, distress in hepatic region were better in DNT-treated group than in UDCA-treated group. The histological study also showed that DNT had positive effect in treatment of NAFLD. CONCLUSION: DNT is an effective drug to treat patients with NAFLD of damp-heat syndrome type and is more effective than UDCA.
3.Study on the Fingerprint and Attribution of Original Herbs of the Volatile Oil in Xiangjuganmao Granules

Objective:To establish the fingerprint of the volatile oil in Xiangjuganmao granules and study the attribution of original herbs. Methods:The fingerprint of the volatile oil was established by GC-MS,and the characteristic peaks were used to study the attribution of original herbs by comparing with those of single herb and negative herb. Results:Totally 22 characteristic peaks were shown in the fingerprint of Xiangjuganmao granules. The similarities of 18 batches of Xiangjuganmao granules were within the range of 0.892-0.998. Among the 22 peaks,19 ones were defined the attribution of original herbs, and 22 chemical components were identified. Conclusion:The method is with good precision,reproducibility and stability,which can be used to control the quality of Xiangjuganmao granules.
4.Application value of MRI susceptibility weighted imaging (SWI) and diffusion weighted imaging (DWI) in the diagnosis of liver lesions

Objective To investigate the application value of MR SWI and DWI in the diagnosis of liver lesions.Methods 1 300 patients underwent 1.5T MRI scan of liver,no pathology or follow-up examination results of patients with focal liver lesions were excluded,147 patients were included and underwent conventional magnetic resonance(MRI),DWI and SWI.By 2 senior radiologists with double blind method of conventional MRI and MRI combined with DWI and SWI image reading,the feature and enhancement pattern of signal intensity were evaluated.Kappa test was used to evaluate the consistency of the diagnosis of 2 physicians.The pathological examination and comprehensive follow-up results were used as the gold standard.The diagnostic accuracy of the two groups was compared.Results 2 doctors had good consistency in the image diagnosis of the two groups.The Kappa values of conventional MRI and conventional MRI combined with DWI and SWI images were 0.912 and 0.936,respectively.The conventional MRI diagnosed primary hepatocellular carcinoma(HCC) in 40 cases,bile duct cancer cells (ICC) in 13 cases,liver metastasis (HMs) in 14 cases,cavernous hemangioma of the liver (CHL) in 17 cases.The conventional MRI combined with DWI and SWI diagnosed HCC in 50 cases,ICC in 20 cases,HMs in 20 cases,CHL in 24 cases,the differences were statistically significant (x2 =5.660,5.250,5.560,6.640,all P < 0.05).Conclusion Conventional MRI combined with DWI and SWI can provide additional valuable information and improve diagnostic performance for liver lesions.
5.Protective effect of marein against alcoholic fatty liver and its mechanism

OBJECTIVE To explore the protective effect of marein against alcoholic fatty liver (AFL) and its potential mechanisms. METHODS AFL mice model was established with strong wine by gavage. The mice were randomly divided into normal control group (n=9, 0.5% sodium carboxymethyl cellulose solution), model group (n=10, 0.5% sodium carboxymethyl cellulose solution) and marein 75 and 150 mg/kg groups (n=9). Mice were given relevant medicine intragastrically, once a day, for consecutive 30 days. After the last medication, the levels of triglyceride (TG), malondialdehyde (MDA), and superoxide dismutase (SOD) in liver tissue were determined, and hepatic histopathological changes of liver tissue were observed; the protein expression levels of peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase-1 (CPT-1), and diacylglycerol acyltransferase (DGAT) were determined in liver tissue. BRL hepatocytes injury model was induced by ethanol combined with ferrous sulfate and oleic acid; after treatment with 3, 6 and 12 μmol/L of marein for 24 h, the distribution of lipid droplets, the levels of TG, MDA and SOD and protein expressions of PPARα, CPT-1 and DGAT in hepatocytes were examined. After pretreatment with MK886 (PPARα inhibitor, 10 μmol/L),modeled hepatocytes were treated with 12 μmol/L of marein for 24 h, and the protein expressions of PPARα, CPT-1 and DGAT were determined. RESULTS As the results showed in vivo, compared with the model group, after treatment with 75 and 150 mg/kg of marein, the degree of steatosis was significantly reduced, and the levels of TG and MDA and protein expression of DGAT were significantly decreased(P<0.05 or P<0.01); the levels of SOD, protein expressions of PPARα and CPT-1 were significantly increased(P<0.05 or P<0.01). As the results showed in vitro, after treatment with 3, 6 and 12 μmol/L of marein, the lipid accumulation of hepatocytes was significantly inhibited, and the levels of TG and MDA, protein expression of DGAT were significantly decreased(P<0.05 or P<0.01), while the levels of SOD, protein expressions of PPARα and CPT-1 were significantly increased(P<0.05 or P<0.01). After MK886 pretreatment, the effects of marein on the above protein expressions were abolished. CONCLUSIONS Marein might exert a protective effect against AFL. The mechanisms might be related to inhibiting oxidative stress-mediated injury and improving PPARα-mediated lipid metabolism signaling pathway.
