1.Mechanisms of curcumin protecting endothelial cells against ischemia and reperfusion injury

Aim To investigate the mechanisms in protecting HUVEC against ischemia/reperfusion(I/R) injury directed by curcumin.Methods Hypoxia/reoxgenation(H/R) model was established on HUVEC.MTT colorimetric assay was used to observe the injury degree of hypoxia and reoxygenation at the different time.With preconditioning by different concentration of Cur,the survival rate of HUVEC subjected to H/R was assessed by MTT colorimetric assay.Pretreated with Cur(5 ?mol?L-1),the expression of LC3,cathepsin B,cathepsin L,Bax and Bcl-2 were observed by fluorescent staining and Western blot in HUVEC during H/R process.Results Cur(1.25~5 ?mol?L-1) played a protective role during H/R in HUVEC in a dose-dependent manner.During H/R,the expressions of LC3,cathepsin B and the ratio of Bax/Bcl-2 increased,and the nuclear translocation of cathepsin L was induced;when cur was pretreated,LC3 was furtherstrengthened,at the same time,the up-regulation of cathepsin B,the ratio of Bax/Bcl-2 and the nuclei-location of cathepsin L were inhibited partly by Cur.Conclusions Cur can raise the survival rate of HUVEC in the process of H/R.Cur increases the autophagy activity,depresses cathepsins and Bax/Bcl-2 to protect the endothelial cells.
2.The clinical observation of reduced dose idarubicin combined with cytarabine, semustine regimen in the treatment of patients with acute myeloid leukemia

ObjectiveTo evaluate the clinical efficacy and toxicity of reduced dose idarubicin and cytarabine,semustine(IAS) regimen as induction therapy in patients with acute myeloid leukemia.MethodsA total of fifty-eight newly acute myeloid leukemia(AML) patients were randomly divided into 2 groups,including 30 cases with IAS regimen,28 cases with DA regimen The IAS regimen was treated with reduced dose idarubicin (8 ~ 10 mg/m2,days 1 to 3) and cytarabine( 100 ~ 150 mg/m2,days 1 to 7),semustine(200mg,d0).The DA regimen was treated with daunorubicin(40 ~60 mg/m2,days 1 to 3) and cytarabine ( 100 ~ 150 mg/m2,days 1 to 7).The responses ( CR and overall response rate ) were compared between the 2 groups.Results Complete remission(CR) rate in IAS and DA groups were 24 of 30( 80.0% ) and 16 of 28 (57.1% ) respectively,while the overall response rate were 26 of 30 ( 86.7% ) and 18 of 28 ( 64.3% ) respectively.There was significant difference in CR rate and overall response rate between IAS group and DA group( P < 0.05 ).Myelosuppression and infections due to neutropenia were the most frequent adverse effects,severe nonhematologic toxicity was not observed.The incidence rates of toxicities in the 2 groups were not significantly different ( P > 0.05 ).Conclusion The effect of reduced dose idarubicin and cytarabine,semustine regimen in the treatment for acute myeloid leukemia is superior to that of DA regimen,and the toxicities are tolerable.IAS regimen can be as the optional induction therapy in newly patients with acute myeloid leukemia.
3.Epidemiological features of metabolic syndrome in residents aged 20-80 years in Pudong New District of Shanghai

Objective To investigate the prevalence and risk factors of metabolic syndrome (MS) in residents in Pudong New District of Shanghai. MethodsA total of 5 584 residents aged 20-80 years were randomly selected from Pudong New District of Shanghai through multistage sampling and interviewed from April to July of 2008. Metabolic syndrome was defined according to three diagnostic criteria for MS, issued by the modified National Cholesterol Education Program Adult Treatment Panel Ⅲ criteria ( NCEP-ATP Ⅲ ), International Diabetes Federation (IDF), and Chinese Diabetes Society (CDS). ResultsThe crude prevalences of MS in the adult population in Pudong New District were 18.2% and 13.1% standardized ( male 19. 1%, female 17.4%, the age-standardized 15.6% and 13.2% ) with CDS criterion, 31.8% and 24.4% standardized ( male 28.4% ,female 35.1% ,the agestandardized 22. 7% and 25.0% ) with NCEP-ATP Ⅲ criterion, and 21.7% and 17.0% standardized ( male 15.9% ,female 26.7%, the age-standardized 13.8% and 19.2% ) with IDF criterion. The age-specific prevalence of MS increased according to three diagnostic criteria, and the age-adjusted prevalence was higher in males than females in junior age groups and higher in females than males in senior ones. Significant differences were present among region, education, marriage status, smoking, work intensity, recreation, and physical activity according to some diagnostic criteria. ConclusionsSubstantial proportions of adults in Pudong New District of Shanghai suffer from metabolic syndrome, and there exists a tendency for young people involved. MS has become a noteworthy public health problem. It suggests that community-integrated control strategy of MS should be made a priority.
4.Ultrasonic diagnosis of thyroid microcarcinoma and analysis on reason for its misdiagnosis

Objective To analyze the ultrasonic features of thyroid microcarcinoma (TMC) and the causes of misdiagnosis. Methods The ultrasonic features including shape, margin, echogenecity, microcalcification, vascularity and lymphadenopathy were analyzed retrospectively in 26 pathologically-proven TMC patients. Results In 26 cases, 11 cases were diagnosed correctly before operation (11/26, 42.31%), 12 cases were misdiagnosed (12/26, 46.15%) as adenoma or benign nodule, and 3 cases were missed diagnosed (3/26, 11.54%). Among the 23 cases detected on ultrasound, 21 cases were solid and hypoechoic (21/23, 91.30%);19 cases were ill-defined (19/23, 82.61%);12 cases were taller than wide in shape (12/23, 52.17%); 14 cases had microcalcification (14/23, 60.87%); 7 cases showed central or peripheral blood flow signals (7/23,30.43%) with arterial resistance index>0.70 in 3 lesions and<0.70 in 4 lesions. Conclusions Several ultrasonographic features are helpful in identiifcation of TMC, including hypo/iso-echogenecity, ill-deifned margin, taller-than-wide shape, microcalciifcation, arterial signals with high resistance index, and abnormal lymphadenopathy. Moreover, for cases with multiple lesions, to the potential co-existence of benign and malignant lesions should be considered.
5.Type-B ultrasonic characteristics of posterior vitreous detachment

Objective To observe the B-scan ultrasonic characteristics of the posterior vitreous detachment (PVD) and the relationship between the PVD and retina. Design Retrospective case series. Participants 991 cases (1107 eyes) with PVD. Methods The results of B-scan ultrasonography were retrospectively in PVD patients and according to the adhesion between the posterior vitreous cortex (PVC) and eyeball wall, complete PVD and incomplete PVD were divided. Main Outcome Measures The ratio of types and characteristics of PVD. Results 710 eyes(64.1%)were diagnosed as complete PVD, in which 69 eyes(17.3%)accompanied with peripheral ruptured detachment of retina, and 16 eyes(23.0%)PVC adhered with lid of peripheral retina hole. 397 eyes(35.9%)were diagnosed as incomplete PVD, in which 159 eyes(40.0%)as partly PVD, and 76 eyes(47.7%)concomitanted with PVC proliferation and vitreous incompletely splitting. 121 eyes(30.5%)PVC were adhered with the macula, and 117 eye(29.5%)with the optic disc. Conclusions Ultrasonography can be applied to diagnose PVD precisely,which can provide credible morphologic evidence for the clinic.
6.Effects of pravastatin, fosinopril and their combination on myocardium TNF-alpha expression and ventricular remodeling after myocardial infarction in rats.

OBJECTIVETo investigate the effects of pravastatin, fosinopril and their combination on ventricular remodeling, cardiac function, tumor necrosis factor-alpha (TNF-alpha) mRNA expression, and matrix metalloproteinases (MMPs) activities after myocardial infarction (MI) in rats.
METHODSAcute myocardial infarction (AMI) was established by ligation of the anterior descending coronary artery in male Sprague-Dawly (SD) rats. Twenty-four hours after the procedure, the 48 surviving rats were grouped randomly as AMI control, fosinopril (10 mg.kg(-1).d(-1)), pravastatin (20 mg.kg(-1).d(-1)) and a combined use of the 2 drugs. Sham-operated group (n = 8) was taken randomly as non-infarction control. Six weeks after treatment with the drugs by gastric gavage, heart function and left ventricular remodeling were assessed. Left ventricular weight (LVW)/body weight (BW) ratio was determined. The relative expression of myocardium TNF-alpha mRNA was assessed by reverse transcription-polymerase chain reaction. Left ventricular myocardium MMPs activities were assessed by Zymography.
RESULTSThere were no significant differences among the four AMI groups in infarction size (P > 0.05). In comparison with the AMI group, left ventricular end-diastolic pressure, left ventricular end-diastolic diameter, LVW/BW all decreased significantly (P < 0.05 - 0.01); while dp/dtmax, dp/dtmin, fractional shortening (FS) and ejection fraction (EF) increased significantly in all three drug-treated groups (P < 0.05 - 0.01); increments of FS, LVEF and dp/dtmax were more evident in the combination group than either the fosinopril or pravastatin group (P < 0.05). The levels of TNF-alpha mRNA in AMI rats treated with fosinopril, pravastatin and their combination reduced 29%, 26% and 33%, respectively (P < 0.01); MMP-2 activity reduced 25%, 30% and 35%, respectively (P < 0.01); MMP-9 activity reduced 20%, 18% and 24%, respectively (P < 0.01). There were no significant differences in other variables among the 3 treatment groups (P > 0.05).
CONCLUSIONPravastatin, fosinopril and their combination showed favorable effects on left ventricular remodeling after AMI in rats and demonstrated improved cardiac function. The combined treatment group yielded better results in the context of improving left ventricular systolic function. These effects could be relevant to the attenuation of increased MMP-2 and MMP-9 activities and left ventricular expression of TNF-alpha.

7.Effect of different irrigants on radicular dentin cleansing and resin tag formation after post space preparation.

OBJECTIVETo evaluate the effect of different root canal irrigants on smear layer removal and dentinal tubule opening of root canal surfaces after post space preparation.
METHODSThirty maxillary anteriors were endodontically treated. After post space preparation, the teeth were assigned to three groups with ten teeth each: group A, B, C. 3% H2O2, 15% ethylenediamine tetraacetic acid(EDTA), 15% EDTA and 3% NaClO were used as irrigant of group A, B, C. After post space irrigation, eight teeth of each group were examined under scanning electron microscope (SEM) for smear layer removal and dentinal tubule opening. The left two specimens were restored with fiber posts and prepared for evaluation of resin tag formation.
RESULTSThe cleansing effectiveness of group A, B and C was respectively 3.89 +/- 0.36, 1.77 +/- 0.30, 1.25 +/- 0.21. The dentinal tubule opening of group A, B and C was respectively (2.53 +/- 0.19), (3 11 +/- 023) (3.83 +/- 0.52) microm. The cleansing effectiveness and dentinal tubule opening of root canal surfaces were significantly affected by different root canal irrigants (P < 0.05). Excessive erosion of the dentin was observed in group C. Resin tag formation differed between the three irrigant groups, and group B showed excellent resin tag formation.
CONCLUSIONIrrigation with 15% EDTA after post space preparation can excellently remove the smear layer and improve dentinal tubule opening, permitting well penetration of resin adhesive into the dentinal tubules and collagen fibrillar structure.

With human chromosomes,as antigen,anti—human—chromosome antibodies (AhChrA)were detected specifically from SLE patients sera by the methods of immunogold—silver staining(IGSS)and immnuofluorescenee tese (IFT)。To SLE,the sensitivity and specificity of serumAhChrA was 58.1%and98.5%respeetively in IGSS,34.9%and99.5%respectively in IFT。Boththe incidence and titer of AhChrA were found to be colsely related to the pathoactivity and thedamages of some important organs or tissues,such as kidney damage,abnormal immunity andhematocytopenia.A preliminary analysis of the antigen components reacting to AhChrA was alsoperformed。
