1.Effects of microRNA-132 on the proliferation and apoptosis in human pancreatic cancer cells SW1990

Objective To observe the effect of miR-132 transfection on proliferation and apoptosis of pancreatic cancer SW1990,and to explore the underlying mechanism.Methods The expression of miR-132 in the pancreatic cancer tissue and the adjacent tissues in 28 pancreatic cancer patients were detected by realtime quantitative polymerase chain reaction (RT-qPCR).miR-132 was transfected into SW1990 cells by using liposome method,untransfected cells and cells with missense miR-132 transfection were used as black control and negative control.The proliferation and apoptosis was detected by CCK-8 assay and DAPI staining.The transfected cells were implanted in nude mice as xenograft tumor,and TUNEL was used to detect the apoptosis; immunohistochemistry was used to detect the expression of Ki-67 and mucin-4 protein in the xenograft tumors and mucin-4,HER-2,p-FAK protein in transfected cells.Results The expression levels of miR-132 in pancreatic cancer tissue and adjacent tissues were 0.46 ± 0.11 and 1.24 ± 0.36,and the difference between the two groups was statistically significant (P < 0.05).The expression level of miR-132 in transfected SW1990 cells was 2.95 ± 0.46,which were significantly higher than those in negative control (0.84 ± 0.17) ; the survival rate of transfected cells at 48,72,96 h was 56.5%,44.7%,37.4% of negative control cells.The apoptosis rate in transfected cells was 41.6%,and the corresponding value was 5.7% in negative control,and the difference was statistically significant (P < 0.05).The expression levels of mucin-4,HER-2,p-FAK in nagative control were 2.94 ± 0.42,1.75 ± 0.37 and 2.74 ± 0.24,and the corresponding values in transfected cells were 0.76 ± 0.14,0.34 ± 0.04 and 0.27 ± 0.03,and the difference between the two groups was statistically significant (P <0.05).In vivo,the growth of xenograft tumors in transfected nude mice was significantly inhibited [(0.23 ± 0.05) vs (0.59 ± 0.13) g,P < 0.05],the apoptosis of xenograft tumor cells was significant increased [(21.7 ± 4.7) % vs (5.2 ± 0.7) %,P <0.05].The expressions of mucin-4 and Ki-67 protein in nagative control was 281.34 ± 36.62 and 148.05 ± 21.34,and the corresponding values in transfection group were 64.35 ± 7.16 and 30.75 ± 4.61,and the difference was statistically significant (P <0.05).Conclusions miR-132 transfection has an effect of inhibiting proliferation and promoting apoptosis on SW1990 cells,and the mechanism may be down-regulation of mucin-4,HER-2,p-FAK protein rxpression.
2.Multivariate statistical analysis of external medicine for hydrocephalus therapy with TCM

Objective:To research and summarize the characteristics and medication regulars of external medicine for hydrocephalus therapy with TCM. Methods:Retrieved literatures of explicitly recorded external medicine in all ages,input its drugs into the Excel sheet specifing and specified various terms used for drug,adopted spss17.0 frequency analysis,principal factor analysis,cluster analysis, and sought its regulars.Results:Frequency analysis of external medicine from higher to lower in order as Radix Ledebouriellas,Rhizoma Bletillae,Arborvitae seed,Cinnamon bark,Ackinthepulpit tuber,Rhizoma Zingiberis ect. Withdrawed 16 main factors from 50 original targets, used cluster analysis made 16 main factors into 10 types as eliminating phlegm for resuscitation,activating blood and disinhibiting fluid etc. Conclusion: External medicine for hydrocephalus therapy mainly as drugs for activating blood and dissolving stasis, expelling wind with diaphoresis, warming yang,reinforcing kidneys to strengthening bones,regulating qi.Multivariate statistical analysis can objectively and comprehensively reflect the characteristics and regulars of external medicine and provided some exploratory thoughts.
3.Relationship between the occurence of acute respiratory failure after subototal esophagectomy and three-field lymphadenectomy and the choice of anesthesia and management

Purpose:To explore the relationship between the occurence of acute respiratory failure after subototal esophagectomy and three-field lymphadenectomy and the choice of anesthesia and management.Methods:From March 2000 to Janunary 2001,thirty-seven patients scheduled for subtotal esophagectomy and three-field lymphadenectomy with e- sophageal cancer,adopting the combination of epidural,cervical nerve block,and general anesthesia were studied.Re- sults:Seven cases out of thirty-seven had acute respiratory failure,accounting for 19% in whole group.They had complete recovery after antifectious therapy and respiratory support.Conclusions:To combine the epidural,cervical nerve block and general anesthesia can reduce intravenous anesthetics use and shorten the extubation time,decrease the incidence of postop- erative respiratory failure.
4.The relation between Helicobacter pylori infection and the amount of interleukin-8 and granulocyte clolny-stimulating factor

Objective To evaluate the relationship between Helicobac ter pylori (Hp) related gastritis and gastric mucosa content of interleukin - 8 (IL-8) and granuloeyte colony-stimulating factor ( G-CSF). Methods ELISA was used to detect the amount of IL-8 and G-CSF in supernate fluid of gastromucosal tissue culture from 60 patients, and to examine the difference between Hp infected and non-Hp infected cases. These 60 cases include normal gastric mucosa (n =20) , chronic gastritis mucosa (n = 14) , gastric ulcer mucosa (n = 13) , duodenal ulcer mucosa ( n = 13 ). 6.Kinematic analysis of the upper limbs during elbow extension in C_5 and C_6 spinal cord injury patients

Objective To investigate motor patterns in the upper limbs of C_5 to C_6 spinal cord injury (SCI) patients during elbow extension. Methods Fifteen C_5 and C_6 SCI patients and fifteen healthy subjects formed an SCI group and a control group respectively. A three-dimensional kinematic analysis was performed on videos of the subjects performing four elbow extension activities: grasping a cup, reaching for a light switch, propelling a wheelchair, and upper limb weight bearing. A Peak Motus motion analysis system was employed. Results Compared with controls, the movement time of the SCI group was significantly longer during grasping, reaching for a light switch and wheelchair ambulation. Their angular velocity was significantly slower during reaching for a light switch and during wheelchair ambulation. The patients with spinal cord injury performed the motor tasks primarily by relying on increasing or decreasing the angular displacement of the shoulder. But they compensated for their functional deficiency by changing the movement direction of the shoulder and elbow in sagittal projection during upper limb weight bearing activities. Conclusion C_ 5-6 SCI patients use motor patterns and strategies different from those of control subjects in completing elbow extension activities.
7.The angular displacement and angular velocity of the upper limb during elbow extension activities in spinal cord injury patients and normal individuals

Objective To study upper limb motor strategies during elbow extension activities in C5-6 spinal cord injury (SCI) patients and compare them with the strategies of normal individuals. Methods A Peak Motus motion analysis system was used to measure the angular displacement and the angular velocity of the shoulder, elbow and wrist of fifteen C5-6 SCI patients and fifteen healthy subjects during four elbow extension activities including grasping a cup, reaching for a light switch, wheelchair ambulation and upper limb weight-bearing. 8.Expression of stromlysin in breast cancer tissue and its relationship with microvessel density

Objective To investigate the relationship between the biological characteristics and the expres-sion of stromlysin (MMP-7) and microvessel density (MVD) in breast cancer tissue and the effect of MMP-7 on MVD of breast cancer. Methods 60 fresh samples were obtained from patients with breast cancer,and then the ex-pression of MMP-7 and vascular endothelial cell CD34 were studied by means of immunohistochemical assay (SP method). The relationship between MMP-7, MVD and age, tumor size, histological type, lymph node metastasis and expression of ER, PR, PCNA, p53 and CerbB2 was studied. The relationship between the biological characteristics and the expression of MMP-7 and MVD in breast cancer were evaluated. Results The positive expression rote of MMP-7 in the d>2 cm group,the group with lymph node metastasis,the group with positive expression of PCNA, p53 and C-erbB-2 (78.1%,74.2%,71.8%,67.6% and 72.2%)were all higher than that in the d≤2 cm group, the group without lymph node metastasis,the group without positive expression of PCNA,p53 and C-erbB-2(32.1%, 41.3%,38.1%,38.5% and 33.3%) (P<0.05). MVD was higher in the positive breast cancer tissue of the group of d>2 cm(34.61±6.97), the group with lymph node metastasis (34.37±7.50), PCNA ( 33.24±8.39), p53 (33.28±8.94), C-erbB-2 (33.55±8.57) than in the negative breast cancer tissue of the d≤2 cm group ( 28.60±9.82), the group without lymph node metastasis (27.48±8.66), PCNA (26.88±7.89), p53 (21.71± 7.59),C-erbB-2(27.42±27.69) (P<0.05). MVD(33.62±7.36)/ high power lens in the MMP-7 positive group was higher than that (27.86±9.45)/high power lens in the MMP-7 of negative group(P<0.05). The posi-tive expression of MMP-7 was correlated with MVD (r=0.380, P<0.05). Conclusion The high expression of MMP-7 and MVD is associated with the development and metastasis of breast cancer. MMP-7 could promote the mi-crovessel development in mammary cancer.
9.Adverse Drug Reactions of 6 Kinds of Quinolones

OBJECTIVE To analyze the adverse drug reactions of 6 kinds of quinolones in order to provide information for clinically rational use of drugs.METHODS The adverse drug reactions of quinolones such as norfloxacin,ofloxacin,levofloxacin,ciprofloxacin,fleroxacin and rufloxacin used in outpatients during Feb to May 2007 were collected and retrospectively analyzed.RESULTS The main adverse drug reactions were seen in gastrointestinal system,nervous system,immune system,respiratory system,urinary system etc.From them the symptoms of gastrointestinal system and nervous system were prominent.CONCLUSIONS In use of quinolones it should think highly to their adverse drug reactions.
10.Antimicrobial Use in Cardiac Surgical During Perioperative Period:An Investigation and Analysis

OBJECTIVE To find out the situation of antimicrobial usage in cardiac surgical perioperative period.METHODS The use of antimicrobials in cardiac surgical perioperative patients during Jan-Jun 2007,and the data of reasonable use of antibacterial were surveyed and evaluated.RESULTS Of the 276 patients,100.00% of them had received antimicrobials involving 12 kinds,59.78% of them used one kind only and 34.78% used two kinds antibiotics and 5.43% used more three kinds.CONCLUSIONS The use of antimicrobials in cardiac surgical perioperative period is reasonable and scientific.
